Aims: We reported that pancreatic ductal adenocarcinomas (PDACs) without high-grade pancreatic intraepithelial neoplasia (PanIN) in the vicinity had worse prognoses than PDACs with high-grade PanIN. However, the molecular characteristics of PDACs with and without high-grade PanIN have not been compared. The aim of this study is to clarify the molecular characteristics of PDACs with and without high-grade PanIN. Method and Results: We reviewed all of a consecutive series of 100 patients with PDACs and divided them into 2 groups: the PDACs with PanIN-2 or PanIN-3 in the background (the PanIN-high group, n = 60) and the PDACs without PanIN-2 or PanIN-3 in the background (the PanIN-low group, n = 40). We evaluated the p53, p16, and SMAD4 expressions in the invasive ductal carcinoma (IDC) components by immunohistochemical staining. KRAS mutation was also analyzed in 80 tumors. The PanIN-low group showed significantly more frequent “high p53 expression” and “loss of SMAD4 expression” than the PanIN-high group (p = 0.048 and p = 0.019, respectively). Loss of p16 expression was not significantly different between the groups. The rate of KRAS wild type was significantly higher in the PanIN-low group than the PanIN-high group (p = 0.024). Conclusions: Our results demonstrated that the molecular characteristics in the PDACs with high-grade PanIN were different from those in the PDACs without high-grade PanIN. PDACs without high-grade PanIN may develop via a pathway other than the PanIN-carcinoma sequence.</p

Expression of MUC16 in PanIN lesions and normal ducts.

<p>(A) MUC16 expression was evaluated in tissues containing both the normal pancreas, and adjacent dysplastic lesions. While MUC16 expression was weak in the low-grade, early stage PanIN lesions (PanIN I), it progressively increases with increasing dysplasia with the highest expression observed in high-grade dysplasia (PanIN III) and PC. Note the predominant membrane staining of MUC16 in all grades of PanINs (Original magnification ×200). (B) Box plot representing the composite score of MUC16 across the different grades of PanIN lesions and normal ducts. From the box plot we observe that MUC16 is strongly expressed in PanIN II and III when compared to PanIN I.</p

Role of Galectin in the Regulation of Neural Excitability by Drosophila sialyltransferase

    No full text
Galectins are major sugar binding proteins that can be blocked from interactions with its sugar epitopes, galactose, by sialylation. Previous research from Panin lab has showed that sialylation affects the neural excitability. Since galectins and sialylation are so intimately linked, this study examines whether galectin has a role in neural excitability. To test the hypothesis, possible mutant fly lines with galectin gene deletion were created with hobo deletion method. Next, we will screen for the fly lines that contain galectin gene deletion. Then, we will conduct functional and behavioral analysis of the galectin mutants to investigate if neurological functions of galectin mutant flies are compromised. Finally, the galectin mutant flies will be crossed with sialyltransferase mutants to examine whether the defect is rescued or amplified. The results will shed light on the molecular and genetic mechanism of galectin in the nervous system and the mode of interaction of galectin with sialylation

The claustrum of the bottlenose dolphin Tursiops truncatus (Montagu 1821)

The mammalian claustrum is involved in processing sensory information from the environment. The claustrum is reciprocally connected to the visual cortex and these projections, at least in carnivores, display a clear retinotopic distribution. The visual cortex of dolphins occupies a position strikingly different from that of land mammals. Whether the reshaping of the functional areas of the cortex of cetaceans involves also modifications of the claustral projections remains hitherto unanswered. The present topographic and immunohistochemical study is based on the brains of eight bottlenose dolphins and a wide array of antisera against: calcium-binding proteins (CBPs) parvalbumin (PV), calretinin (CR), and calbindin (CB); somatostatin (SOM); neuropeptide Y (NPY); and the potential claustral marker Gng2. Our observations confirmed the general topography of the mammalian claustrum also in the bottlenose dolphin, although (a) the reduction of the piriform lobe modifies the ventral relationships of the claustrum with the cortex, and (b) the rotation of the telencephalon along the transverse axis, accompanied by the reduction of the antero-posterior length of the brain, apparently moves the claustrum more rostrally. We observed a strong presence of CR-immunoreactive (-ir) neurons and fibers, a diffuse but weak expression of CB-ir elements and virtually no PV immunostaining. This latter finding agrees with studies that report that PV-ir elements are rare in the visual cortex of the same species. NPY- and somatostatin-containing neurons were evident, while the potential claustral markers Gng2 was not identified in the sections, but no explanation for its absence is currently available. Although no data are available on the projections to and from the claustrum in cetaceans, our results suggest that its neurochemical organization is compatible with the presence of noteworthy cortical inputs and outputs and a persistent role in the general processing of the relative information. © 2014 Cozzi, Roncon, Granato, Giurisato, Castagna, Peruffo, Panin, Ballarin, Montelli and Pirone

PanIN neuroendocrine cells promote tumorigenesis via neuronal crosstalk

    No full text
Nerves are a notable feature of the tumor microenvironment in some epithelial tumors, but their role in the malignant progression of pancreatic ductal adenocarcinoma (PDAC) is uncertain. Here we identify dense innervation in the microenvironment of precancerous pancreatic lesions, known as pancreatic intraepithelial neoplasms (PanIN), and describe a unique subpopulation of neuroendocrine PanIN cells that express the neuropeptide substance P (SP) receptor Neurokinin 1-R (NK1-R). Using organoid culture, we demonstrated that sensory neurons promoted the proliferation of PanIN organoids via SP-NK1-R signaling and Stat3 activation. Nerve-responsive neuroendocrine cells exerted trophic influences and potentiated global PanIN organoid growth. Sensory denervation of a genetically engineered mouse model of PDAC led to loss of Stat3 activation, a decrease in the neoplastic neuroendocrine cell population, and impaired PanIN progression to tumor. Overall, our data provide evidence that nerves of the PanIN microenvironment promote oncogenesis, likely via direct signaling to neoplastic neuroendocrine cells capable of trophic influences. These findings identify neuroepithelial crosstalk as a potential novel target in PDAC treatment
