Preparative isoelectric focusing of reduced wheat gluten proteins
Proteins extracted from gluten of the bread wheat cultivar Fiorello 2 in the presence of 2-mercaptoethanol or dithiothreitol were separated by isoelectric focusing in a free solution in a pH 3–10 gradient containing 50% v/v 1-propanol or urea. The collected fractions were analyzed by sodium dodecyl sulfate-polyacrylamide gel electrophoresis in 10% gels (high and medium molecular weight glutenin subunits) and 16% gels (low molecular weight gliadins). The isoelectric focusing pattern of gluten polypeptides in 50% v/v 1-propanol was comparable to that obtained on two-dimensional gel electrophoresis, based on isoelectric focusing and polyacrylamide gel electrophoresis or nonequilibrium pH gradient electrophoresis and polyacrylamide gel electrophoresis. A similar isoelectric focusing pattern was also observed when 3M urea was used as solvent. New gluten polypeptides, similar in mobility to the high molecular weight subunits of glutenin were detected at acidic pH

La reintroduzione del grano monococco (Triticum monococcum L)in Sicilia: risultati di un'analisi preliminare delle caratteristiche agronomiche e qualitative di tre genotipi.
Il grano monococco (Triticum monococcum L.), conosciuto come “farro piccolo”, è una specie diploide con genoma A (2n=2x=14), le cui cariossidi, anche dopo trebbiatura, si presentano rivestite dalle glume. Insieme a farro ed orzo, dal Neolitico fino al termine dell’età del Bronzo (900 a.C.), è stato alla base della dieta delle popolazioni mediterranee, per essere poi sostituito dal più produttivo farro (T. dicoccum) e successivamente completamente dimenticato. Il crescente consumo di alimenti con caratteristiche dietetico-nutrizionali elevate, unito alla possibilità di introdurre nel panorama cerealicolo siciliano una coltura mai sperimentata prima, ci ha indirizzato nella realizzazione di un progetto specifico sulla caratterizzazione agronomica, tecnologica e nutrizionale di questa specie.
Per la caratterizzazione agronomica, condotta con metodo tradizionale, sono stati allestiti una prova sperimentale con tre genotipi di grano monococco (Monlis, ID331 e Stendhal) e due campi dimostrativi destinati alla coltivazione del genotipo Monlis. La caratterizzazione tecnologica, condotta per valutare l’attitudine alla trasformazione degli sfarinati ottenuti dalle suddette varietà, si è incentrata sull’analisi alveografica e farinografica, l’analisi dell’indice di glutine e la determinazione dell’indice di caduta.
La caratterizzazione dietetico-nutrizionale si è basata sulla determinazione di diversi componenti. Nello specifico, il contenuto proteico delle varietà di monococco e il contenuto in ceneri si è rivelato più elevato rispetto a quello delle varietà di grano tenero e grano duro messe a confronto, mentre è stato riscontrato un contenuto di fibre totale pressocchè uguale rispetto alle varietà di grano tenero

Going Beyond Counting First Authors in Author Co-citation Analysis
The present study examines one of the fundamental aspects of author co-citation analysis (ACA) - the way co-citation
counts are defined. Co-citation counting provides the data on which all subsequent statistical analyses and mappings
are based, and we compare ACA results based on two different types of co-citation counting - the traditional type that
only counts the first one among a cited work's authors on the one hand and a non-traditional type that takes into
account the first 5 authors of a cited work on the other hand. Results indicate that the picture produced through this non-traditional author co-citation counting contains more coherent author groups and is therefore considerably clearer. However, this picture represents fewer specialties in the research field being studied than that produced through the traditional first-author co-citation counting when the same number of top-ranked authors is selected and analyzed. Reasons for these effects are discussed
IN VITRO ASSESSMENT OF ACETIC ACID-SOLUBLE PROTEINS (GLUTENIN) TOXICITY IN CELIAC DISEASE
Acetic-acid-soluble storage proteins from gluten of the bread wheat cv. Sprint 3 were fractionated by adsorption chromatography on 2000 Å controlled-pore glass (CPG) beads, and glutenin polymers with molecular mass higher than 107 Da and free from monomeric gliadins were recovered. The glutenin polymers were found to consist of high-molecular-weight (HMW) and low-molecular-weight (LMW) glutenin subunits. Peptic-tryptic (PT) digests of glutenins were examined for their agglutination activity on human myelogenous leukemia K 562(S) cells, agglutination being strongly correlated with toxicity for the celiac intestine. The peptide fraction at a concentration of 1 g/L of culture medium was able to agglutinate 30% of K 562(S) cells, suggesting a moderate toxic effect. This toxicity may be accounted for by homologies in amino acid sequences between glutenin subunits and /β-and -gliadin
Variations on the Author
“Variations on the Author” discusses two of Eduardo Coutinho’s recent films (Um Dia na Vida, from 2010, and Últimas Conversas, posthumously released in 2015) and their contribution to the general question of documentary authorship. The director’s filmography is characterized by a consistent yet self-effacing form of authorial self-inscription: Coutinho often features as an interviewer that rather than express opinions propels discourses; an interviewer that is good at listening. This mode of self-inscription characterizes him as an author who is not expressive but who is nonetheless markedly present on the screen. In Um Dia na Vida, however, Coutinho is completely absent form the image, while Últimas Conversas, on the contrary, includes a confessional prologue that moves the director from the margins to the center of his films. This article examines the ways in which these works stand out in the filmography of a director who offers new insights into the notion of cinematic authorship
