Validation data for the quantification of the Annonaceous acetogenin annonacin in Rat brain by UPLC-MS/MS

    No full text
AbstractAnnonaceous acetogenins (AAGs) are environmental neurotoxins from the fruit pulp of several Annonaceae species, whose consumption was linked to the occurrence of sporadic atypical Parkinsonism with dementia. The quantification of the prototypical AAG annonacin in Rat brain homogenates was performed by UPLC-MS/MS in selected reaction monitoring (SRM) mode, using a triple quadrupole mass analyzer. A natural analog of annonacin was used as an internal standard. Analyzed data set of the analytical validation of this method is presented, including stability of the samples, extraction recovery and matrix effect, supporting the results described in the article “Quantification of the environmental neurotoxin annonacin in Rat brain by UPLC-MS/MS” N. L’antimysticisme de Port-Royal et la mystique de Fénelon

    No full text

Effects of different anti-aging dietary restrictions on endothelin-1 receptors in rat lung.

The effects of aging on ET-1 receptors and the possible modifications induced by restriction dietary regimens were evaluated in lung of 3, 12 and 23 months-old male Sprague-Dawley rats, fed nd libitum (AL) or subjected to quantitative restriction to 60% of daily food intake (DR), and to every-other-day feeding (TF). Competition experiments with ET-1 (1pM-100nM) and with BQ-123 (1pM-50mM) were performed. Affinity was in the range of nM. Density resulted 4038+/-1185 and 4656+/-1100 fmol/mg protein for 3 and 24 months-old AL rats, and 1970+/-556 and 1533+/-270 for DR and IF rats while ET-A/ET-B was 49:51*, 65:35#, 48:52, 37:63 respectively (*p< 0.05 vs 24 months AL, #p < 0.05 vs DR, p< 0.01 vs LF). Aging seems to be associated with a tendency to increase of the density of ET-1 receptors and with a significant alteration of receptor subtype distribution

Reasons why it is time to change imaging guidelines on endometriosis

Abstract: In light of the rising number of patients referred for magnetic resonance imaging (MRI) due to suspected endometriosis and the high expectations of these patients, there is a need for new imaging guidelines to optimally protocol and indicate MRI and transvaginal ultrasonography (TVUS) examinations. This is crucial for accurately addressing the inquiries of gynecologists, encompassing complete mapping and preoperative staging, and facilitating effective communication with patients. In this context, the development of a standardized lexicon, as well as dedicated imaging classifications, is recommended to aid in the comprehensive management of patients. Clinical relevance statement: The radiologist should use a standardized lexicon and provide a score along with details about the specific compartments affected by endometriosis disease. This helps in offering clearer guidance to the surgeon. Key Points: • An optimal staging is based on the combination of clinical examination, transvaginal US, and MRI. • MRI is able to detect location that is hidden at the beginning of a laparoscopic surgery and thus the need for dedicated MR classifications to correctly stage the disease. • Deep pelvic endometriosis index (dPEI) classification is externally validated and highly correlated with operating time, hospital stay, and postoperative complications

Carotid atherosclerosis and risk of subsequent coronary event in outpatients with atherothrombosis.

The presence of carotid plaque reflects overall atherosclerotic burden and may predict coronary artery disease events. We examined the association among carotid atherosclerosis, history of atherothrombotic events, and risk of coronary events. Among 45 227 patients in the Reduction of Atherothrombosis for Continued Health (REACH) Registry with 4-year follow-up, 23 364 patients with information on carotid atherosclerosis at baseline were analyzed. The primary outcome was the composite of first occurrence of cardiovascular death, myocardial infarction, or coronary hospitalization. The carotid atherosclerosis was present in 46% of patients (n=10 725) and was associated with increasing conventional cardiovascular risk factors and extent of symptomatic vascular disease. During 4-year follow-up, 4304 patients experienced ≥1 coronary event. After adjustment for cardiovascular risk factors and geographic region, the risk of coronary events increased by 22% (95% confidence interval [CI], 14%-30%) in patients with versus without carotid atherosclerosis. The relative increase was 18% (95% CI, -7%-51%) in patients enrolled with multiple risk factors only, 25% (95% CI,16%-35%) in patients with coronary artery disease, 46% (95% CI,28%-65%) in patients with cerebrovascular disease, and 37% (95% CI,17%-60%) in patients with peripheral artery disease. Carotid atherosclerosis was associated with increased risk, even among patients with previous myocardial infarction but no known stroke (P=0.001) or among patients with previous stroke but no known myocardial infarction (P<0.001). Carotid atherosclerosis was an independent predictor of coronary events across all types of symptomatic vascular disease and had an incremental effect on risk regardless of risk factors or location of vessel disease
