    An improved method for intracellular DNA (iDNA) recovery from terrestrial environments

    The simultaneous extraction of intracellular DNA (iDNA) and extracellular DNA (eDNA) can help to separate the living in situ community (represented by iDNA) from background DNA that originated both from past communities and from allochthonous sources. As iDNA and eDNA extraction protocols require separating cells from the sample matrix, their DNA yields are generally lower than direct methods that lyse the cells within the sample matrix. We, therefore, tested different buffers with and without adding a detergent mix (DM) in the extraction protocol to improve the recovery of iDNA from surface and subsurface samples that covered a variety of terrestrial environments. The combination of a highly concentrated sodium phosphate buffer plus DM significantly improved iDNA recovery for almost all tested samples. Additionally, the combination of sodium phosphate and EDTA improved iDNA recovery in most of the samples and even allowed the successful extraction of iDNA from extremely low-biomass iron-bearing rock samples taken from the deep biosphere. Based on our results, we recommend using a protocol with sodium phosphate in combination with either a DM (NaP 300 mM + DM) or EDTA (NaP + EDTA 300 mM). Furthermore, for studies that rely on the eDNA pool, we recommend using buffers solely based on sodium phosphate because the addition of EDTA or a DM resulted in a decrease in eDNA for most of the tested samples.     Microbial ecology of subsurface granitic bedrock: a humid–arid site comparison in Chile

    Abstract Subsurface microorganisms face extreme challenges such as anoxic, xeric, and oligotrophic conditions. In igneous systems, nutrient limitation is critical, as biomass input relies on surface-derived fluids via tectonic fractures. Despite growing interest in subsurface habitats, little is known about ecosystems beneath arid landscapes, where surface water input is limited by the low annual precipitation. This study compares granitic subsurface environments beneath arid and humid surface ecosystems, highlighting the link between surface climate and subsurface biodiversity. DNA was extracted from granitic subsurface rocks recovered from two endmember sites along a north–south climate gradient in Chile’s Coastal Cordillera. Microbial communities inhabiting down to 55 m deep subsurface rocks were characterized using 16S rRNA amplicon and shotgun metagenomic sequencing. We identified an abundant and potentially active subsurface community below both climates dominated by heterotrophic bacteria, including Pseudarthrobacter, Janthinobacterium, and Pseudomonas. However, rare taxa affiliated with common chemolithoautrophs, e.g. Thiobacillus, Sulfuriferula, and Sulfuricurvum, were only observed in the arid subsurface, indicating increased oligotrophic conditions and reliance on inorganic electron donors in the deeper subsurface of the desert. Functional analysis revealed sulphur, hydrogen, and carbon monoxide as potential inorganic electron donors. These findings expand the current understanding of microbial life in the subsurface of granite rocks showing the influence of surface climate on nutrient conditions in the deeper subsurface, providing new insights into the extent and functional capacity of terrestrial subsurface habitats and their role in global biogeochemical processes

    Le mot 'moderne' est associé, la plupart du temps, de façon positive, par la société, à des choses et des personnes qui ont excellé dans des domaines divers et qui ont fait des découvertes nouvelles pour les communautés dont ils sont issus. le concept de 'modernité' repose quant à lui sur une vision plus subjective. que signifie cette notion ? cette thèse, dont le sujet est : 'modernité et progrès dans les sociétés latino-américaines du XXème siècle: l'exemple de doña Barbara de Romulo Gallegos' va se centrer sur cette œuvre vénézuélienne contemporaine, du XXème siècle, dont le personnage principal est une femme cruelle, corrompue et avide de richesse. ce travail de recherche de doctorat portera sur les concepts d'évolution, de modernité et de progrès d'un point de vue ethnologique, politique, littéraire et territorial en grande partie, et couvrant les thèmes dépeints dans le roman de Gallegos parmi d'autres œuvres liées au dit-thème. les concepts de société, de monde développé, des 'Amériques', de civilisations primitives etc. seront évidemment abordés lors de ce travail ainsi que l'auteur Romulo Gallegos. en conclusion de mon mémoire de master, 2 portant sur le monde 'moderne' et primitif, j'étais arrivé à la question: 'la notion de confort dans sa pluralité, ne nous a-t'-elle pas éloigné des racines primitives que sont les autres hommes et de l'essence de la vie?' c'est bel et bien la conclusion à laquelle, arrive le protagoniste de los pasos perdidos, de l'auteur Alejo Carpentier, roman que j'avais pris pour sujet de ce sujet de master 1 et 2. désormais, pour cette thèse, mes recherches vont se centrer sur les significations et leurs applications, pour les sociétés latino-américaines, des termes suivants : l'évolution, le progrès et la ou les modernité(s) focalisant ainsi mon travail sur l'œuvre de doña Barbara, exemple à part entière de confusion, d'anarchie, de despotisme, et d'injustice ; tant de termes qui représentent nos sociétés actuelles.The word "modern" is associated, mostly in a positive way, by society, things and people who have excelled in various fields and have made new discoveries for the communities from which they come. The concept of "modernity" lies however on a more subjective vision. What does it mean? This thesis, whose subject is: "The challenge of Progress in the Latino-American societies of the twentieth century: the case of Doña Barbara from Romulo Gallegos", will focus on this Venezuelan contemporary novel from the twentieth century, whose main character is a cruel-corrupted and greedy woman. This doctorate research will focus on the concepts of evolution, modernity and progress from an ethnological point of view, as also political, literary and territorial aspects, largely covering then the themes portrayed in the novel by Gallegos among others novels related to this PhD’s main theme. The concepts of society, developed world, primitive "Americas" and so on, will obviously be addressed during this work, as well as the presentation of the author: Romulo Gallegos. In conclusion of my previous Master's thesis, based on the contrasts of both the "modern" and “primitive” world, I came to end the work with the following question: “The concept of comfort in its plurality did it not withdrawn us away from the primitive roots and human beings that represent our life’s true essence?” This is indeed the conclusion, to which the protagonist arrives to, in Los pasos perdidos by Cuban author Alejo Carpentier, a novel that I had work on with dissimilar subjects for my Master 1 and 2. Now, for this PhD’s thesis, my research will focus on the meanings and applications for Latino-American societies, on the following terms: evolution, progress and modernity and I’ll start the research with the novel: Doña Barbara, as the perfect example of confusion, anarchy, despotism, and injustice; in fact so many words that are very representative of our current societies’ working mode

    Prospecting the first Chilean Martian simulants from the Atacama Desert for ISRU and potential applications

    Martian simulants are an essential part of in situ resource utilization (ISRU) technology development and testing, and aid in providing new astrobiology insights regarding Mars’s past habitability. This study proposes a new method involving average and standard deviation (ASD), complementary to figure of merit (FOM) simulant evaluation methods, for geochemical prospection and evaluation of simulant feedstock using portable XRF (pXRF) and whole-rock (ICP-OES) analyses. It also presents simulant modification calculations (SMC) that integrate multiple approaches used in simulant production and design, and provides theoretical mineral recipes (estimated mineral composition) and their estimated geochemical compositions for FOM evaluation of an intended target composition. These methods were used to prospect simulant material in volcanic-subvolcanic outcrops at the SW edge of the Atacama Desert in Chile, and to evaluate their potential for standard and enhanced simulant development. Through ASD (pXRF data) and FOM (ICP-OES data) evaluation, this resulted in the presentation of the first Chilean basic Martian bedrock simulant that can be used for biomining applications, which is tentatively labeled as Chilean Atacama Desert 1 (CAD-1). An Excel file with the applied prospection method and database is also available for simulant feedstock prospection (Supplementary section).     Virgilio Malvezzi fue autor italiano de gran éxito en la corte de Felipe IV a partir de la traducción de su biografía clásica Il Romulo (1629) por Francisco de Quevedo en 1632, pasando a convertirse en uno de los modelos de la prosa lacónica en castellano. Autores cortesanos como el mismo Quevedo o Diego de Saavedra Fajardo escribirán sus obras de carácter político en la prosa lacónica y concentrada que pone de moda Il Romulo, y su influencia se puede seguir incluso en autores secundarios como Mártir Rizo. Asimismo, en autores que comienzan a escribir a mediados o finales de la década de los años treinta del siglo XVII, cuyo ejemplo fundamental es Baltasar Gracián, que con sus tres obras menores (El héroe, El discreto y El político) muestra la influencia de ese nuevo estilo cortesano, si bien, desde un principio, con un grado de elaboración y refinamiento excepcional, que lo llevarán a convertirse en el ejemplo clásico del estilo lacónico.Virgilio Malvezzi was a greatly successful Italian author in the court of Filipe IV following the classic biographical translation of Il Romulo (1629) into Spanish by Francisco de Quevedo in 1632, becoming one of the models of laconic prose in Castilian. Court authors like Quevedo or Diego de Saavedra Fajardo wrote their political works in laconic prose in the fashion of Il Romulo, and its influence can be seen in secondary authors like Mártir Rizo as well as authors beginning to write by the middle to late 1630's. A fundamental example is Baltasar Gracián who, with his three minor works (El héroe, El discreto and El político), fully demonstrates the new courtesan style with a degree of elaborate and exceptional refinement and emerges as a classic example of the laconic style
