    Going Beyond Counting First Authors in Author Co-citation Analysis

    The present study examines one of the fundamental aspects of author co-citation analysis (ACA) - the way co-citation counts are defined. Co-citation counting provides the data on which all subsequent statistical analyses and mappings are based, and we compare ACA results based on two different types of co-citation counting - the traditional type that only counts the first one among a cited work's authors on the one hand and a non-traditional type that takes into account the first 5 authors of a cited work on the other hand. Results indicate that the picture produced through this non-traditional author co-citation counting contains more coherent author groups and is therefore considerably clearer. However, this picture represents fewer specialties in the research field being studied than that produced through the traditional first-author co-citation counting when the same number of top-ranked authors is selected and analyzed.     “Variations on the Author” discusses two of Eduardo Coutinho’s recent films (Um Dia na Vida, from 2010, and Últimas Conversas, posthumously released in 2015) and their contribution to the general question of documentary authorship. The director’s filmography is characterized by a consistent yet self-effacing form of authorial self-inscription: Coutinho often features as an interviewer that rather than express opinions propels discourses; an interviewer that is good at listening. This mode of self-inscription characterizes him as an author who is not expressive but who is nonetheless markedly present on the screen. In Um Dia na Vida, however, Coutinho is completely absent form the image, while Últimas Conversas, on the contrary, includes a confessional prologue that moves the director from the margins to the center of his films. This article examines the ways in which these works stand out in the filmography of a director who offers new insights into the notion of cinematic authorship

    Appropriate Similarity Measures for Author Cocitation Analysis

    We provide a number of new insights into the methodological discussion about author cocitation analysis. We first argue that the use of the Pearson correlation for measuring the similarity between authors’ cocitation profiles is not very satisfactory. We then discuss what kind of similarity measures may be used as an alternative to the Pearson correlation. We consider three similarity measures in particular. One is the well-known cosine. The other two similarity measures have not been used before in the bibliometric literature.     Dispelling the Myths Behind First-author Citation Counts

    We conducted a full-scale evaluative citation analysis study of scholars in the XML research field to explore just how different from each other author rankings resulting from different citation counting methods actually are, and to demonstrate the capability of emerging data and tools on the Web in supporting more realistic citation counting methods. Our results contest some common arguments for the continued use of first-author citation counts in the evaluation of scholars, such as high correlations between author rankings by first-author citation counts and other citation counting methods, and high costs of using more realistic citation counting methods that are not well-supported by the ISI databases. It is argued that increasingly available digital full text research papers make it possible for citation analysis studies to go beyond what the ISI databases have directly supported and to employ more sophisticated methods

    Der Einsatz ionisierender Strahlung ist wesentlicher Bestandteil der kurativen Therapie solider Tumore. Die letale Wirkung auf Tumorzellen basiert insbesondere auf der Induktion von DNA-Doppelstrangbrüchen. Dabei ist die Schonung des umliegenden Normalgewebes eine der größten Herausforderungen der Strahlentherapie und häufig kommt es zu starken, teilweise irreversiblen Nebenwirkungen. Oft wird die Bestrahlung zudem noch mit einer Chemotherapie kombiniert, was die Toxizität der Behandlung weiter erhöht. Daher ist die Suche nach alternativen zielgerichteten Therapien das Thema aktueller Forschung in der Radioonkologie. Plattenepithelkarzinome des Kopf- und Hals-Bereiches (Head and neck squamous cell carcinomas, HNSCC) sind eine genetisch komplexe, heterogene Gruppe bösartiger Erkrankungen und weltweit die siebthäufigste Krebsart. Neben jahrelangem starken Tabak- und Alkoholkonsum ist die Infektion mit Hochrisikostämmen des Humanen Papillomvirus (HPV) ein weiterer wichtiger onkogener Faktor. Die Strahlentherapie ist auch bei der Behandlung von HNSCC ein zentrales Element der multimodalen Therapie. Aufgrund der im Kopf-Hals-Bereich vorhanden Risikoorgane wie z. B. der Speicheldrüsen und Schluckmuskulatur, stellt die Bestrahlungsplanung jedoch eine besondere Herausforderung dar und Patient:innen mit Tumoren im Kopf-Hals-Bereich leiden häufig lebenslang unter den irreversiblen Nebenwirkungen der Bestrahlung. Bezüglich des Ansprechens auf die Strahlentherapie haben sich HPV-positive Tumore als besonders empfindlich erwiesen und diese Patient:innen besitzen eine dementsprechend gute Prognose. Die Mechanismen hinter der erhöhten Strahlenempfindlichkeit sind noch nicht vollständig geklärt, allerdings wurde für HPV-positive Tumore bereits auf zellulärer Ebene eine erhöhte Strahlenempfindlichkeit, basierend auf einem Defekt in der Doppelstrangbruchreparatur, nachgewiesen. Während das wichtigste Ziel für Patient:innen mit HPV-positiven HNSCC eine sichere Deintensivierung der Behandlung ist, besteht das Ziel für Patient:innen mit HPV-negativen Tumoren in einer Verbesserung des Überlebens. Vorarbeiten aus dem Labor für Strahlenbiologie zeigen zudem, dass sowohl die Inhibition der DNA-Reparatur als auch die Inhibition des G2-Zellzyklus-Kontrollpunktes HNSCC-Zelllinien radiosensibilisieren kann. Die Inhibition des Enzyms PARP durch Olaparib führt zu zusätzlichen Schäden in der S- und G2-Phase; die Inhibition der Kinase Wee1 durch Adavosertib zur Hemmung der Zellzyklus-Kontrollpunkte, was zu einem vorzeitigen Eintritt in die Mitose trotz bestehender Doppelstrangbrüche, sowie zur Induktion von massivem Replikationsstress führt. Vor diesem Hintergrund wurde in der vorliegenden Dissertation die kombinierte Inhibition von PARP und Wee1 an drei strahlenempfindlichen HPV-positiven (UD-SCC-2, UM-SCC-47, UPCI-SCC-154) und drei radioresistenten HPV-negativen (HSC4, SAS, UT-SCC-60A) Zelllinien untersucht, um die strahlentherapeutische Behandlung von Patient:innen mit HNSCC zu verbessern. Die Inhibition von Wee1 führte in den HPV-positiven Zellen zu einer effektiven Aufhebung des G2-Arrestes, in den HPV-negativen Linien war der Arrest sowie seine Inhibition nur schwach ausgeprägt. Die alleinige Inhibition von PARP sowie die Zugabe von Olaparib zu Adavosertib hatten wenig Einfluss auf den G2-Arrest. In den HPV-negativen Zellen wurde bei letzterem die Reduktion des Arrestes sogar vermindert. Eine Analyse des Replikationsstressmarkers γH2AX in der Durchflusszytometrie zeigte sowohl in HPV-positiven als auch in HPV-negativen Zelllinien einen starken Anstieg der γH2AX-Signalintensität in S- und G2-Phase-Zellen nach der Inhibition von Wee1 und PARP/Wee1. Bereits eine Einzelgabe der Inhibitoren radiosensibilisierte HPV-positive und HPV-negative Zelllinien im delayed-plating Koloniebildungstest, die wirksamste Radiosensibilisierung war allerdings bei dualer Inhibition erkennbar. Die Auszählung residueller strahleninduzierter 53BP1 Reparaturfoci, als Marker für Doppelstrangbrüche in der Immunfluoreszenz, konnte zeigen, dass die Inhibition von PARP/Wee1 in den HPV-negativen Zellen zu einer erhöhten Focizahl führte, wohingegen die Anzahl der Reparaturfoci in den HPV-positiven Zellen nicht anstieg. Allerdings zeigte sich hier eine Abnahme der Foci in der G2-Phase und eine Zunahme der Focizahl in der G1-Phase. Dies deutet darauf hin, dass die Zellen trotz nicht-reparierter Doppelstrangbrüche die Mitose durchschreiten, was mit einem besonderen Risiko letaler Chromosomenaberrationen und des mitotischen Zelltodes verbunden ist. Von besonderer Bedeutung für das Zellüberleben ist, dass sich in allen verwendeten HNSCC-Zelllinien die Anzahl der nur gering geschädigten Zellen, welche die größte Überlebenswahrscheinlichkeit besitzen, nach dualer Inhibition verringerte. Im Rahmen dieser Dissertation wird dargestellt, dass die duale Inhibition von PARP und Wee1 ein hochwirksamer Ansatz zur Radiosensibilisierung von HNSCC-Zelllinien ist. Zwar zeigten sich im Detail Unterschiede zwischen HPV-positiven und HPV-negativen HNSCC-Zelllinien, dennoch sollte dieser Ansatz bezüglich seines therapeutischen Nutzens weitergehend untersucht werden. Eine Radiosensibilisierung von HNSCC könnte die Effektivität der Strahlentherapie bei gleichzeitiger Reduktion der strahleninduzierten Nebenwirkungen ermöglichen

    We have done our best to complete the author checklist relating to the use of animals in the hut study. Note that the objective for the hut study was to evaluate the IRS treatment applications for residual efficacy against Anopheles mosquitoes, including the local An. coluzzii mosquito population. Cows were only used to attract mosquitoes into the huts and no tests were carried out directly on the cows. The author checklist is intended for use with studies where experiments are carried out on animals, which is why we have had such difficulty in completing this for the hut study, as many of the questions do not relate to how the cows were used
