    Neuroprotection antiinflammatoire et antiischémique par modulation du protéasome

    L'inhibition du protéasome a un effet anti-inflammatoire en bloquant l'activation de NF-êB. Dans ce travail, nous avons étudié cette propriété anti-inflammatoire dans deux modèles animaux de maladies neurologiques: la sclérose en plaques et l'AVC. Nous avons utilisé le ritonavir, un antiprotéase du VIH et qui a des effets de modulation sur le protéasome. Ainsi l'administration du ritonavir a permis d'inhiber l'EAE modèle animal de SEP. D'autre part, un effet anti-ischémique cérébral a été obtenu ches les animaux ayant subi une occlusion de l'artère cérébrale moyenne, modèle animal d'AVC. Ces effets neuroprotecteurs ont été corrélés à une inhibition de la réponse inflammatoire chez les animaux traités. Ces résultats ouvrent des nouvelles perspectives thérapeutiques dans les maladies inflammatoires basées sur la modulation du protéasomeThe inhibition of the proteasome has an anti-inflammatory effect by blocking the activation of NF-êB. In this work, we studied this anti-inflammatory property in two animal models of neurological diseases: the multiple sclerosis and stroke. We used the ritonavir, an antiprotease of the HIV and which has effects of modulation on the proteasome. So the administration of the ritonavir allowed to inhibit the animal model EAE of MS. On the other hand, an anti-ischemic effect was obtained in animal model of stroke. These neuroprotective effects were correlated with an inhibition of the inflammatory response. These results open new therapeutic perspectives in the inflammatory diseases based on the modulation of the proteasome

    OTX2 (orthodenticle homeobox 2) est un facteur de transcription impliqué dans de nombreux processus développementaux tels que la mise en place de l'axe antéropostérieur et le développement du système nerveux central. Ce facteur est également retrouvé après la naissance dans certaines structures adultes dans le but de maintenir leur intégrité et leur bon fonctionnement. Ainsi, en raison de sa pléiotropie de fonctions, OTX2 est impliqué dans plusieurs pathologies.Dans le cadre de ma thèse, je m'intéresse à deux de ces pathologies : la dégénérescence maculaire liée à l'âge (DMLA) qui est une dégénérescence de la rétine, et le médulloblastome, qui est un cancer pédiatrique du cervelet.Notre laboratoire a mis en évidence qu'OTX2 est une cible thérapeutique intéressante pour ces deux pathologies qui manquent encore de thérapies efficaces et sûres. Dans le modèle murin, une augmentation du niveau d'OTX2 dans l'épithélium pigmenté de la rétine (RPE) permet de prévenir la dégénérescence des photorécepteurs caractéristique de la DMLA. À l'inverse, dans le cadre du médulloblastome, une diminution du niveau d'expression d'OTX2 contribue au contrôle de la tumorigénèse ainsi qu'à l'amélioration de la survie.Malheureusement, à ce jour, aucun régulateur chimique d'OTX2 n'a été décrit et ne peut donc être utilisé dans ces deux pathologies.Ainsi, dans la première partie de ma thèse, nous avons entrepris d'identifier, respectivement, des régulateurs positifs et négatifs d'OTX2 dans le RPE et les cellules de médulloblastome. Afin de sélectionner de telles molécules, j'ai réalisé un crible pharmacologique de 1200 composés par RT-qPCR qui m'a permis d'identifier deux composés inhibiteurs.Dans la seconde partie de ma thèse, ces deux composés ont été plus amplement analysés dans les cellules de médulloblastome afin d'étudier leur impact sur les capacités tumorigènes de ces cellules cancéreuses.Ce projet m'a ainsi permis d'identifier deux régulateurs négatifs d'OTX2. L'étude de leurs effets in vitro et in vivo m'a permis de mettre en évidence que ces molécules ont un impact sur la tumorigénèse des médulloblastomes et constituent ainsi des thérapies intéressantes pour ces tumeurs.OTX2 (orthodenticle homeobox 2) is a transcription factor involved in numerous developmental processes such as the anteroposterior axis establishment and the central nervous system development. Notably, it is expressed postnatally in the adult in several organs and acts to maintain their proper functioning. On account of its pleiotropy in terms of function, OTX2 is involved in a large set of pathologies.As part of my thesis, I focused on two of them : Age-related Macular Degeneration (AMD), which is caused by degeneration of the retina, and medulloblastoma, which is a pediatric cancer in the cerebellum; both of which still lack efficient and safe therapies as of today.Our lab showed that OTX2 can be an interesting therapeutic target for both pathologies. Indeed, in a murine model that enhancing OTX2 expression in the retinal pigment epithelium (RPE) prevents photoreceptors degeneration, a phenotype characteristic of the AMD. On the other hand, in the case of medulloblastoma, reducing OTX2 levels in the cerebellum improves tumorigenesis control and survival.Nevertheless, no chemical regulators of OTX2 that could counteract these pathologies have been described.Therefore, in the first part of my thesis, I began identifying positive and negative regulators of OTX2 in RPE and medulloblastoma cells, respectively. To select such molecules, I carried out a pharmacological screening of up to 1200 compounds by means of RTqPCR, which allowed to identify 2 negative regulators of OTX2. In the second part of my thesis, I analysed both compounds in medulloblastoma cells to study their impact on the tumorigenic abilities of these cancerous cells.Overall, this project allowed to identify two negative regulators of OTX2, which in vitro and in vivo studies showcased an impact on the tumorigenic ability of medulloblastoma cells. Thus, these compounds constitute potential therapeutic targets for this cancer

    Going Beyond Counting First Authors in Author Co-citation Analysis

    The present study examines one of the fundamental aspects of author co-citation analysis (ACA) - the way co-citation counts are defined. Co-citation counting provides the data on which all subsequent statistical analyses and mappings are based, and we compare ACA results based on two different types of co-citation counting - the traditional type that only counts the first one among a cited work's authors on the one hand and a non-traditional type that takes into account the first 5 authors of a cited work on the other hand. Results indicate that the picture produced through this non-traditional author co-citation counting contains more coherent author groups and is therefore considerably clearer. However, this picture represents fewer specialties in the research field being studied than that produced through the traditional first-author co-citation counting when the same number of top-ranked authors is selected and analyzed. Reasons for these effects are discussed

    Neurofibromatosis disease modelling : development of a test study and applications for the discovery of active molecules

    La neurofibromatose de type 1 (NF1) représente la maladie génétique autosomale dominante la plus fréquente en France, après la mucovisidose, avec une incidence de 1 individu sur 3500. Il s'agit d'une pathologie multi-systémique présentant un tableau clinique varié. Parmi la pléthore de symptôme, sont comptées des manifestations neurocutanées telles que les taches « café-au-lait » (zone d'hyperpigmentation localisée) et les neurofibromes (tumeurs bénignes de la gaine périphérique de la myéline) mais également des défauts osseux et des troubles cognitifs. La pénétrance de NF1 est complète mais la manifestation et la sévérité des symptômes peuvent varier d'un individu à l'autre. Le gène NF1 responsable de la maladie, localisé sur le chromosome 17, est un gène suppresseur de tumeur qui code pour la neurofibromine. Dans le but de développer un modèle cellulaire humain pertinent pour l'étude des défauts osseux associés à NF1, nous avons utilisé des cellules souches induites à la pluripotence porteuse de la mutation causale NF1 (hiPS-NF1). Dans ces travaux, nous avons montré qu'une perte d'expression de la neurofibromine dans les ostéoblastes dérivés de hiPS-NF1 reproduisait le phénotype d'ostéogénèse réduite et qu'il était possible d'améliorer la capacité des hiPS-NF1 à se différencier en ostéoblaste à l'aide de molécules pharmacologiques. Toujours dans le but de proposer un modèle cellulaire humain le plus relevant possible, nous avons développé des lignées isogéniques avec la technologie CRISPR/Cas 9 afin d'étudier l'impact d'une perte partielle ou totale de l'expression de la neurofibromine sur le phénotype osseux. En parallèle, afin de pouvoir étudier un autre phénotype associé à NF1, un protocole de différenciation de cellules de Schwann sous culture définie en utilisant des facteurs de croissance et des molécules de signalisation, a partir des hiPS a été développé. Des cellules de Schwann-like ont été obtenues en 30 jours en engagement la différenciation des cellules souches vers la crête neurale afin d'induire l'émergence de précurseurs de cellules de Schwann par l'action de molécules telles que le récepteur de type I du TGF-ß (SB431542), l'hereguline ß1, l'IGF1, le FGF2 et un activateur de WNT3a (CHIR99021). L'analyse par q-PCR montre une augmentation des marqueurs de différents stades de différenciation de la cellule de Schwann : crête neurale (SOX10, ERBB3), cellules de Schwann précurseurs (MPZ, CAD19) et cellules de Schwann immatures (S100) au bout de 30 jours de différenciation. Ces résultats ont été complétés par l'analyse protéique des cellules différenciées et par la mise en co-culture de ces cellules avec des motoneurones différenciés à partir d'hiPS. L'ensemble de ce travail a permis de valider la pertinence de l'utilisation des cellules souches pluripotentes porteuses de mutation dans la modélisation de pathologie génétique. Permettant à plus long terme la recherche de molécules actives par des approches de de criblage pharmacologique ou par des approches de thérapie cellulaire.Neurofibromatosis type 1 (NF1) is an autosomal genetic disease with an incidence of 1 in 3,500 individuals. This is a multi-systemic disorder with a plethora of various symptoms. Among with we find neurocutaneous manifestations such as "café-au-lait" spots (zone of localized hyperpigmentation) and neurofibromas (benign tumors of the peripheral sheath of myelin), but also bone defects and cognitive disorders. The penetrance of NF1 is complete but the manifestation and severity of symptoms may vary from one individual to another. The NF1 gene responsible for the disease, located on chromosome 17, is a tumor suppressor gene that encodes neurofibromin. In order to develop a relevant human cellular model for the study of bone defects associated with NF1, we used pluripotency-induced stem cells that carry the NF1 causal mutation (hiPS-NF1). In this work, we have shown that loss of neurofibromin expression in osteoblasts derived from hiPS-NF1 reproduces the reduced osteogenesis phenotype. We have also shown that pharmacological molecules can improve the ability of hiPS-NF1 to differentiate in osteoblast. In order to propose the most relevant human cell model, we have developed isogenic lines with CRISPR / Cas 9 technology to study the impact of a partial or total loss of neurofibromin on the bone phenotype. Simultaneously, a Schwann cells differentiation protocol from hiPS was developed under culture defined using growth factors and signaling molecules. Schwann-like cells were obtained in 30 days by the use of molecules such as the type I receptor TGF-β (SB431542), heregulin β1, IGF1, FGF2 and activator of WNT3a (CHIR99021). The q-PCR analysis shows an increase in Schwann cell markers: neural crest (SOX10, ERBB3), precursor Schwann cells (MPZ, CAD19) and immature Schwann cells (S100). These results were confirmed by protein analysis of the differentiated cells and by the co-culture analysis of these cells with differentiated motoneurons from hiPS. All of this work validated the relevance of pluripotent stem cells in the modeling of genetic pathology

    “Variations on the Author” discusses two of Eduardo Coutinho’s recent films (Um Dia na Vida, from 2010, and Últimas Conversas, posthumously released in 2015) and their contribution to the general question of documentary authorship. The director’s filmography is characterized by a consistent yet self-effacing form of authorial self-inscription: Coutinho often features as an interviewer that rather than express opinions propels discourses; an interviewer that is good at listening. This mode of self-inscription characterizes him as an author who is not expressive but who is nonetheless markedly present on the screen. In Um Dia na Vida, however, Coutinho is completely absent form the image, while Últimas Conversas, on the contrary, includes a confessional prologue that moves the director from the margins to the center of his films. This article examines the ways in which these works stand out in the filmography of a director who offers new insights into the notion of cinematic authorship

    Appropriate Similarity Measures for Author Cocitation Analysis

    We provide a number of new insights into the methodological discussion about author cocitation analysis. We first argue that the use of the Pearson correlation for measuring the similarity between authors’ cocitation profiles is not very satisfactory. We then discuss what kind of similarity measures may be used as an alternative to the Pearson correlation. We consider three similarity measures in particular. One is the well-known cosine. The other two similarity measures have not been used before in the bibliometric literature.     Dispelling the Myths Behind First-author Citation Counts

    We conducted a full-scale evaluative citation analysis study of scholars in the XML research field to explore just how different from each other author rankings resulting from different citation counting methods actually are, and to demonstrate the capability of emerging data and tools on the Web in supporting more realistic citation counting methods. Our results contest some common arguments for the continued use of first-author citation counts in the evaluation of scholars, such as high correlations between author rankings by first-author citation counts and other citation counting methods, and high costs of using more realistic citation counting methods that are not well-supported by the ISI databases. It is argued that increasingly available digital full text research papers make it possible for citation analysis studies to go beyond what the ISI databases have directly supported and to employ more sophisticated methods

    We have done our best to complete the author checklist relating to the use of animals in the hut study. Note that the objective for the hut study was to evaluate the IRS treatment applications for residual efficacy against Anopheles mosquitoes, including the local An. coluzzii mosquito population. Cows were only used to attract mosquitoes into the huts and no tests were carried out directly on the cows. The author checklist is intended for use with studies where experiments are carried out on animals, which is why we have had such difficulty in completing this for the hut study, as many of the questions do not relate to how the cows were used
