Differential stability of 2'F-ANA*RNA and ANA*RNA hybrid duplexes: roles of structure, pseudohydrogen bonding, hydration, ion uptake and flexibility

14 pags., 7 figs., 3 tabs.Hybrids of RNA with arabinonucleic acids 2′F-ANA and ANA have very similar structures but strikingly different thermal stabilities. We now present a thorough study combining NMR and other biophysical methods together with state-of-the-art theoretical calculations on a fully modified 10-mer hybrid duplex. Comparison between the solution structure of 2′F-ANA•RNA and ANA•RNA hybrids indicates that the increased binding affinity of 2′F-ANA is related to several subtle differences, most importantly a favorable pseudohydrogen bond (2′F-purine H8) which contrasts with unfavorable 2′-OH-nucleobase steric interactions in the case of ANA. While both 2′F-ANA and ANA strands maintained conformations in the southern/eastern sugar pucker range, the 2′F-ANA strand's structure was more compatible with the A-like structure of a hybrid duplex. No dramatic differences are found in terms of relative hydration for the two hybrids, but the ANA•RNA duplex showed lower uptake of counterions than its 2′F-ANA•RNA counterpart. Finally, while the two hybrid duplexes are of similar rigidities, 2′F-ANA single strands may be more suitably preorganized for duplex formation. Thus the dramatically increased stability of 2′F-ANA•RNA and ANA•RNA duplexes is caused by differencesin at least four areas, of which structure and pseudohydrogen bonding are the most important. © The Author(s) 2010. V empiričnem delu smo opravljali intervju z direktorjem Afriških naložb o Senegalu na področju politike, zdravstva, vodne oskrbe, izobraževanja, kmetijstva in turizma. Rezultate smo analizirali z metodo kodiranja in jih prikazali z njeno zadnjo fazo, tj. z formuliranjem teorije. In the master\u27s thesis we studied the country of Senegal with an emphasis on natural and social data. In the theoretical part, we presented the natural-geographical and socio-geographical characteristics of Senegal and highlighted the most common environmental problems. In addition, we determined the economic cooperation between Senegal and Slovenia and found that no major projects are underway between the two countries, with the exception of the project for ensuring self-sufficiency, in cooperation with the International Center for Eco remediation and the Most Institute - Connecting Europe and Africa. In the empirical part, we conducted an interview with the director for African Investment of Senegal in the fields of politics, healthcare, water supply, education, agriculture and tourism. The results were analyzed by the coding method and presented with its last phase - the formulation of the theory. Senegal has a favorable geographical location, as it lies at the crossroads of major traffic routes. Because it is a flat country, most people are engaged in agriculture, which is the most important economic activity, but agricultural productivity has deteriorated in recent years due to desertification and deforestation. In addition to these, the results highlighted climate change, which is causing landscape change, which is reflected in rising sea levels and the degradation of ecosystems. Half of the total population is poor, starving and lacks basic health care. The country has a predominantly young population, who do not have equal opportunities for education due to poor access to educational institutions. The lack of adequate road and railway transport in the country is also reflected in poorer tourism development. Based on the detailed knowledge of the country of Senegal, its potentials and needs, we designed two projects as an opportunity for cooperation between Slovenia and Senegal in the field of sustainable tourism development, provision of quality education and local self-sufficiency

In the herewith thesis I explore the challenges parents face while raising their children during the period of adolescence and how they cope with them. I have also been interested in exploring the changes this period brings along, the role of social and professional support, the emotional burdens or stress experienced by parents, and the way they cooperate with the child’s mother or father. In the theoretical introduction, I first defined adolescence, presented puberty, and described the physical, cognitive, psychosocial, and emotional changes. I then discussed parenting, described different parenting styles, the parent-child relationship, and the conflicts that may arise. Special attention was given to the impact of new technologies and social media. Lastly, I introduced the role of social work as well. In the empirical section, I presented the findings of a qualitative study based on eight interviews. I found that the relationship between parents and their children changes during adolescence. Parents observe an increased need for freedom and independence in their adolescents, which requires them to adapt and seek balance between control and trust. Key challenges include setting boundaries, managing technology use, and adjusting to behavioural and emotional changes in their children. Parents experience different levels of stress and emotional strain, which they try to manage through self-reflection, conversations, physical activity, and support from their partner or close ones. Most parents did not seek professional help, but their experiences show that social support is an important factor when it comes to easing distress. In the conclusion, I present findings and suggestions for practice, theory, and further research, particularly in the area of support provided to parents by school counselling services and social work

Illegal Waste Disposal - Case Study of Pollution of the Ljubljana Marshes

This graduation thesis deals with waste situated on Ljubljana Marshes as a part of eco crime and consequently represents the problem of pollution of the Marshes itself. The purpouse of this thesis is to study illegal waste disposal in the area of Ljubljana Marshes. However, the aim is to present the topic as worrying and problematic. Due to the interest for actual state of the problem, a survey was carried out where questions on the subject were sent to institutions. An interview was also conducted and the data obtained were later analysed. The thesis contains detailed analysis of waste definitions and how waste is being handled here in Slovenia. Also the topic of organized ecological crime is mentioned, and it is stated that this kind of crime goes beyond borders of our country. At the same time, the thesis also provides an overview of the EU regulatory framework and the waste framework directive, as well as a review of the descriptions of the regulation in Slovenia. The Ljubljana Marshes are defined and classified in the category. The origin and ways of financing the landscape park are also included. There were several conclusions, as well as solutions. It is true that we can say the legislation is still a bit cumbersome and therefore it makes even harder to solve problems related to waste. The results of the research have shown, among other things, that it is primarily important to raise people´s awareness. The fact is that their own contribution is a major part of the progress for conservation of the Ljubljana Marshes. The truth however is that environmental problems have grown global and therefore they require interstate integration

Sedimentary record of Holocene transgression and tectonic activity in the Gulf of Trieste

The rapid post-LGM sea-level rise resulted in fast migrations and changes of sedimentary environments in present-day shallow continental shelves. In the northern Adriatic the last transgression submerged the vast alluvial plain of the Po river and its Apennine, Alpine and Dinaric confluences. The Gulf of Trieste represents the northeasternmost part of the Adriatic Sea, where the Holocene change from alluvial to shallow marine sedimentary environments was already observed in sedimentary cores. As high-resolution geophysical methods have been increasingly applied for investigation of seabed sediments in the last few decades, we wanted to use them to study the Holocene transgression in the Gulf of Trieste. We used these methods, mostly the sub-bottom sonar, to correlate the acoustic and sedimentary facies of various Late Quaternary deposits. We also used high-resolution acustic and seismic profiling of seabed sediments of the Gulf of Trieste to study the tectonic activity, to determine the thickness of the Holocene marine sediments and to determine the sound velocity through Late Quaternary deposits. Our results show that in the Gulf of Trieste, terrestrial, transgressive and marine sediments can be differentiated on the basis of their acoustic facies. The sedimentary environment of terrestrial sediments can be determined from geophysical and sedimentological data. Dating results show that fluvial sedimentation in the study area persisted at least until the Younger Dryas and that deposition of transgressive sediments took place in the Early Holocene. While we did not find direct indicators of tectonic activity, we did consider Late Pleistocene fluvial channels as geomorphic indicators of tectonic activity. These indicators do not correlate with the measured vertical motion rates on the Slovenian coastline, which indicates that the recent vertical motion is not expressed in the sedimentary record. Instead, the recent vertical movement rates may result from surface deformation above a locked thrust. The thickness of Holocene marine sediments in the Gulf of Trieste is variable, but generally the thickness decreases away from the coast, where the thickest successions are located. Holocene marine sediments in the Slovenian part of the Gulf of Trieste are usually thicker than 5 meters. Sound velocity in thin (up to 20 meters thick) Late Quaternary successions predominantly depends on the sediment grain-size. Additionally, the presence of gas greatly influences the sound velocity
