    Function of basic amino acids for the synergistic cytotoxicity of ribosome-inactivating proteins and triterpene saponins

    Die synergistische Zytotoxizität zwischen Ribosomen-inaktivierenden Proteinen und Triterpensaponinen beruht auf dem Triterpensaponin-vermittelten endosomal escape von Typ I RIPs. Typ I RIPs wirken zytotoxisch, indem sie einen spezifischen Adeninrest von der rRNA abspalten. Die Abspaltung hat das Erliegen der Proteinsynthese und den Zelltod zur Folge. Damit Typ I RIPs ihre Toxizität ausüben können, müssen sie aus den Endo- und Lysosomen freigesetzt werden, andernfalls werden sie proteolytisch abgebaut. Spezifische Triterpensaponine bewirken dabei, dass Typ I RIPs dem endolysosomalen Abbau entkommen, indem sie in das Zytosol freigesetzt werden. Bisherige Untersuchungen deuteten darauf hin, dass Triterpensaponine pH-abhängig an Typ I RIPs binden und dadurch die endolysosomale Freisetzung induzieren. Die essenzielle Glucuronsäure an C-3 der Triterpensaponine, negativ geladen in Endo- und Lysosomen, könnte dabei mit basischen Aminosäuren der Typ I RIPs, positiv geladen in Endo- und Lysosomen, interagieren. Voraussetzung für die weiteren Untersuchungen dieser Arbeit war das Erbringen des Nachweises der Bindung von SO1861 und HisDianthin, der bisher nur am Beispiel von HisSaporin-3 erbracht wurde. Massenspektrometrische Aufnahmen unter nativen Bedingungen zeigten bei pH 6,24 für SO1861 und HisDianthin eine Assoziation im stöchiometrischen Verhältnis von 1:1. Der Einfluss basischer Aminosäuren auf die bestätigte Interaktion zwischen SO1861 und HisDianthin wurde an 19 HisDianthin-Mutanten, bei denen Lysin- und Arginin-Reste durch Alanin ersetzt wurden, anhand ihrer synergistischen Zytotoxizität untersucht. Für die ausgewählten Lysin-Reste konnte keine Beteiligung an der synergistischen Zytotoxizität nachgewiesen werden. Einzig die synergistische Zytotoxizität von HisDianthin Arg24Ala war im Vergleich zu dem nativen HisDianthin reduziert. Die aufgetretene Reduktion der Zytotoxizität war nicht zelllinienabhängig. Allerdings war sie auch ohne Anwesenheit von SO1861 reduziert, bei gleichzeitig unbeeinträchtigter N-Glykosidaseaktivität. Die Markierung von HisDianthin Arg24Ala und HisDianthin mit Fluoreszenzfarbtsoff erlaubte via Durchflusszytometrie, die verminderte Zytotoxizität auf eine verringerte Endozytose zurückzuführen. Im Rahmen dieser Arbeit konnte somit gezeigt werden, dass die untersuchten Arginin- und Lysin-Reste der Typ I RIPs nicht hauptverantwortlich für die Interaktion mit SO1861 zu sein scheinen. Um aufzuklären, auf welcher Interaktion die Bindung zwischen Typ I RIPs und Triterpensaponinen, die vermutlich dem endosomal escape zu Grunde liegt, beruht, müssen weitere Experimente durchgeführt werden. Um die Funktion von Arg24 an weiteren Typ I RIPs validieren zu können, wurden aus den Samen der Gypsophila vaccaria (L.) Sm. die RIP-Isoformen Sapovaccarin-S1 und -S2 isoliert und sequenziert. Das Isoformengemisch wies eine für Typ I RIPs typische Proteinsequenz auf. Die N-Glykosidaseaktivität und die Thermostabilität waren mit denen von isolierten Typ I RIPs vergleichbar. Die Untersuchung der Funktion der Aminosäure Arg24 an den Typ I RIPs HisSapovaccarin-S1 und HisAgrostin bestätigte, dass auch hier eine Reduktion der Zytotoxizität zu verzeichnen war. Die durchgeführten Untersuchungen deuten darauf hin, dass die Funktion von Arginin an Position 24, die am HisDianthin aufgezeigt werden konnte, sich auch auf die Endozytose von HisAgrostin und HisSapovaccarin-S1, wenn nicht sogar auf Typ I RIPs im Allgemeinen übertragen lässt.The synergistic cytotoxicity between ribosome-inactivating proteins and triterpene saponins is due to the endosomal escape of type I RIPs induced by triterpene saponins. Type I RIPs are toxins that cleave off a specific adenine residue from rRNA. As a result, protein synthesis is inhibited, which is followed by cell death. To avoid degrading and to exert their cytotoxicity, type I RIPs must be released from endosomes/lysosomes. Specific triterpen saponins act as endosomal escape enhancers by releasing the toxin into the cytosol. Previous studies have suggested that endolysosomal release is triggered by pH-dependent binding of triterpene saponins to type I RIPs. The negatively charged essential glucuronic acid at position C-3 of triterpene saponins could interact in endo-/lysosomes with positively charged basic amino acid residues of type I RIPs. A prerequisite for further investigations of this research was the demonstration of the binding of SO1861 and Hisdianthin, which has so far only been demonstrated using the type I RIP Hissaporin-3. Mass spectrometry under native conditions revealed a binding of SO1861 and Hisdianthin at pH 6.24 with a stoichiometric ratio of 1:1. The effect of basic amino acid residues on the interaction confirmed above between SO1861 and Hisdianthin were examined for the synergistic cytotoxicity of 19 Hisdianthin mutants. Lysine and arginine residues were substituted by alanine. The selected lysine residues did not contribute to synergistic cytotoxicity. Only Hisdianthin Arg24Ala reduced the synergistic cytotoxicity when compared to native Hisdianthin. The reduction in cytotoxicity was not cell line specific. However, cytotoxicity was reduced even without the presence of SO1861, whereas N-glycosidase activity was not affected. Hisdianthin Arg24Ala and Hisdianthin labelling allowed flow cytometry to attribute the reduction in cytotoxicity to decreased endocytosis. In this research, it has been shown that the studied arginine and lysine residues of type I RIPs do not seem to be primarily responsible for the interaction with SO1861. The interaction between type I RIPs and triterpene saponins, which presumably underlies the endosomal escape, could not be clarified further. To validate the effect of Arg24 on other type I RIPs, the RIP isoforms sapovaccarin-S1 and -S2 were isolated from the seeds of Gypsophila vaccaria (L.) Sm. and their protein sequence determined. The isoform mixture exhibited a typical type I RIP protein sequence, N-glycosidase activity and thermal stability comparable to other isolated type I RIPs. Examination of the effect of Arg24Ala in the type I RIPs Hissapovaccarin-S1 and Hisagrostin confirmed that the same mutation caused reduced cytotoxicity. This research suggests that the function of arginine at position 24, first shown in the example of Hisdianthin, can be transferred to the endocytosis of Hissapovaccarin-S1 and Hisagrostin, and possibly to the endocytosis of type I RIPs in general

    Cancer is among the leading diseases and causes for deaths worldwide. Even with a rise of treatment options in the last century, cancer therapy can still be limited and insufficient due to side-effects and drug resistances. Therefore, novel approaches have to be explored to overcome these limitations. In recent years, the development and application of antibody-drug conjugates, a combination of a highly cytotoxic payload with a target specific monoclonal antibody, have been receiving increasing attention. The majority of employed payloads in these antibody-drug conjugates can be considered as natural products or derivatives of these. Natural products make an important source for the development of novel anti-cancer drugs. Recently, microcystins, cyanobacterial cyclic heptapeptides, have been discussed as leads for the development of anti-cancer drug substances. Microcystins exhibit a high target toxicity by inhibiting eukaryotic serine/threonine protein phosphatases 1 and 2A. Unlike many cytotoxins that can cross cell membranes by passive diffusion, they depend on active uptake via organic anion transporting polypeptides 1B1 or 1B3, which are especially expressed by liver cells. This leads to a hepatotoxicity of microcystins and might limit the direct use of natural occuring microcystins as payloads. The aim of this thesis was to find optimized microcystins for use as payloads in antibodydrug conjugates. To maximize effectivity in targeted cells while reducing side-effects through uncontrolled uptake, these derivatives should display a high protein phosphatase inhibition potential but an overall low uptake by their transporters. As both phosphatase inhibition and transportability strongly depend on the structure of the individual microcystin, so-called clickable microcystins were modified with chemically diverse small molecules using click chemistry and tested for their bioactivity. A workflow for synthesis of new microcystins derivatives, structural confirmation, isolation, and quantification was developed. Two different kinds of click reactions, coppercatalyzed and strain-promoted, were used for synthesis. Different chromatographic approaches were chosen depending on the chromatographic issue, including solid-phase extraction, preparative, semi preparative, and analytical HPLC. Also, different detection methods depending on the analytical problem were used, e.g., mass spectrometry and tandem mass spectrometry for structural confirmation and an evaporative light-scattering detector for structure-independent quantification. 46 new microcystin derivatives were obtained and characterized through this workflow. In a second step, the influence of the small molecules on the bioactivity profile of microcystins was evaluated: an already established cytotoxicity assay was used to test for transportability, and a protein phosphatase 1 inhibition assay was established to test for target toxicity. The inhibition assay was complemented by a pharmacophore modeling approach. The results revealed that target inhibition and transportability of microcystins can independently be influenced by the physicochemical properties especially of the residue located in position 2 of the microcystin. The selectivity of the two transporters was evaluated to understand similarities and differences in substrates. Moreover, a ratio of transportability over inhibition potential was calculated to find optimized microcystins with high target toxicity but overall low transportability. It was shown that derivatization with small carboxylic acids or amino acids resulted in microcystins with a favorable ratio of inhibition to transportability, making these derivatives potentially suitable for application as payloads in antibody-drug conjugates.Krebs gehört weltweit zu den häufigsten Erkrankungen und Todesursachen. Obwohl sich die Behandlungsmöglichkeiten im letzten Jahrhundert verbessert haben, ist Krebstherapie trotzdem oft durch Nebenwirkungen und Tumor-assoziierte Arzneimittelresistenzen eingeschränkt und unzureichend. Um diese Einschränkungen zu überwinden, werden neue Ansätze verfolgt. In den letzten Jahren hat die Entwicklung und Anwendung von Antikörper-Wirkstoff-Konjugaten, einer Kombination aus einem hochgradig zytotoxischen Wirkstoff und einem zielspezifischen monoklonalen Antikörper, zunehmend an Bedeutung gewonnen. Die meisten der in diesen Antikörper-Wirkstoff-Konjugaten verwendeten Wirkstoffe sind Derivate von Naturstoffen, welche im Allgemeinen eine wichtige Quelle für die Entwicklung neuer Krebsmedikamente sind. Microcystine, eine Klasse zyklischer Heptapeptide aus Cyanobakterien, werden seit einigen Jahren als Leitstrukturen für die Entwicklung von Krebsmedikamenten diskutiert. Microcystine wirken stark toxisch durch Hemmung der eukaryotischen Serin/Threonin-Proteinphosphatasen 1 und 2A. Im Gegensatz zu vielen anderen Toxinen können sie die Zellmembran nicht durch passive Diffusion überwinden und sind daher auf einen aktiven Transport durch die Organo-Anion-Transporter 1B1 oder 1B3 angewiesen. Diese Transportproteine werden insbesondere von Leberzellen exprimiert, weshalb Microcystine hepatotoxisch wirken können. Diese Hepatotoxizität schränkt die direkte Verwendung von natürlich vorkommenden Microcystinen als Wirkstoffe in Antikörper-Wirkstoff-Konjugaten ein. Das Ziel dieser Arbeit war es, optimierte Microcystine für die Verwendung als Wirkstoffe in Antikörper-Wirkstoff-Konjugaten zu finden. Um die Wirksamkeit in den Zielzellen zu maximieren und gleichzeitig die Nebenwirkungen durch unkontrollierte Aufnahme zu reduzieren, sollten solche Microcystine ein hohes Inhibitionspotenzial für Proteinphosphatasen, aber eine insgesamt geringe Aufnahme durch ihre Transporter aufweisen. Da sowohl die Phosphatasehemmung als auch die Transportierbarkeit stark von der Struktur des einzelnen Microcystins abhängen, wurden sogenannte clickbare Microcystine mit chemisch unterschiedlichen niedermolekularen Molekülen mittels Click- Chemie modifiziert und auf ihre Bioaktivität getestet. Es wurde ein Arbeitsablauf für die Synthese neuer Microcystin-Derivate, deren Strukturbestätigung, Isolierung und Quantifizierung entwickelt. Für die Synthese wurden zwei Arten von Clickreaktionen verwendet: kupferkatalysierte und spannungsunterstützte. Je nach chromatographischem Problem wurden verschiedene chromatographische Methoden verwendet, unter anderem Festphasenextraktion, präparative, semipräparative und analytische HPLC. Außerdem wurden abhängig von der analytischen Fragestellung unterschiedliche Nachweismethoden eingesetzt, z.B. Massenspektrometrie und Tandem-Massenspektrometrie zur Strukturbestätigung und ein Lichtstreudetektor zur strukturunabhängigen Quantifizierung. Auf diese Weise wurden 46 neue Microcystin-Derivate gewonnen und charakterisiert. Im zweiten Schritt wurde der Einfluss der eingebrachten niedermolekularen Moleküle auf das Bioaktivitätsprofil der Microcystine bewertet: Ein bereits etablierter Zytotoxizitätstest wurde zur Prüfung der Transportierbarkeit und ein Proteinphosphatase-1-Inhibitionstest zur Prüfung der Hemmung des Zielenzyms eingesetzt. Dies wurde durch ein Pharmakophor-Modell unterstützt. Die Ergebnisse zeigen, dass die Hemmung des Zielenzyms und die Transportierbarkeit von Microcystinen unabhängig voneinander durch die physikochemischen Eigenschaften, insbesondere durch den Rest in Position 2 des Microcystins, beeinflusst werden können. Die Selektivität der beiden Transporter wurde bewertet, um Ähnlichkeiten und Unterschiede hinsichtlich ihrer Substratpräferenzen zu identifizieren. Außerdem wurde ein Verhältnis von Transportierbarkeit zu Hemmungspotenzial berechnet, um optimierte Microcystine mit hoher Toxizität gegenüber dem Zielenzym, aber insgesamt geringer Transportierbarkeit zu finden. Es konnte gezeigt werden, dass die Derivatisierung mit kleinen Carbonsäuren oder Aminosäuren zu Microcystinen mit einem günstigen Verhältnis von Hemmung zu Transportierbarkeit führt.     Spaltbare Peptid-Triterpensaponin-Konjugate für einen verbesserten Gentransfer

    Gentherapien stellen vielversprechende neue Therapieoptionen für eine Vielzahl von Indikationen dar. Trotz mehrerer zugelassener Medikamente, die auf viralen Gentransfer-Vektoren basieren, bleibt das volle Potenzial gentherapeutischer Ansätze bislang ungenutzt. Dies liegt zum einen an der hohen Immunogenität und aufwändigen Herstellung viraler Vektoren, zum anderen an der geringen Effizienz nicht-viraler Gentransfersysteme, die vor allem auf den Einschluss der verwendeten Nanopartikel in Endosomen und den damit verbundenen Abbau zurückzuführen ist. SO1861, ein aus Saponaria officinalis L. isoliertes Triterpensaponin, kann die Freisetzung aus den Endosomen fördern und wird daher als Transfektionsverstärker eingesetzt. Die Saponin-unterstützte Transfektion wird als Sapofektion bezeichnet und beinhaltet die parallele Verabreichung des Saponins und der Nanopartikel. In vitro funktioniert dies problemlos, in vivo erweist sich die Synchronisation der beiden Komponenten jedoch als schwierig. Ziel der vorliegenden Arbeit war daher die Entwicklung und Optimierung eines nicht-viralen Gentransfersystems, das die zu transfizierende DNA und das transfektionsverstärkende Saponin in einem Partikel enthält und intravenös verabreicht werden kann. Durch die Integration eines zielgerichteten Liganden in den zu entwickelnden Nanopartikel sollte zusätzlich die zielgerichtete Wirkung des Gentransfersystems sichergestellt werden. Um dieses Forschungsziel zu erreichen, wurde SO1861 mit Hilfe eines pH-sensitiven Hydrazon-Linkers kovalent an ein Poly-L-Lysin-basiertes Peptidgerüst konjugiert. Die hergestellten SO1861-funktionalisierten Peptide wurden zur Komplexierung von DNA durch elektrostatische Wechselwirkung verwendet. Die resultierenden Nanoplexe wurden in vitro auf ihre Transfektionseffizienz und Verträglichkeit hin untersucht. In allen untersuchten Zelllinien transfizierten die Nanoplexe mit konjugiertem SO1861 deutlich effektiver als nicht-funktionalisierte Nanoplexe unter Zugabe von freiem SO1861 zum Transfektionsmedium. Die Transfektion mit den SO1861 enthaltenden Nanoplexen beeinflusste die Zellviabilität nicht. Zielgerichtete, konjugiertes SO1861 enthaltende Nanoplexe wurden durch Integration eines zielgerichteten Peptids hergestellt und in vivo in einem Allograft-Tumormodell in Mäusen getestet. Unter Verwendung eines Selbstmordgen-Vektors, der für das zytotoxische Protein Saporin kodiert, wurde bei der Behandlung mit zielgerichteten, SO1861 enthaltenden Nanoplexen im Vergleich zur Vehikelkontrolle ein verlangsamtes Tumorwachstum und eine verbesserte Überlebensrate beobachtet. Die intravenöse Injektion der SO1861 enthaltenden Nanoplexe war dabei gut verträglich.Gene therapies represent promising new therapeutic options for a variety of indications. Despite several approved drugs based on viral gene delivery vectors, the full potential of gene therapy approaches remains unexploited. This is due to the high immunogenicity and complex production of viral vectors as well as the low efficacy of non-viral gene delivery vehicles that is in particular attributable to endosomal entrapment and degradation of the nanoparticles used. SO1861, a naturally occurring triterpenoid saponin isolated from Saponaria officinalis L., promotes endosomal escape and is therefore used as a transfection enhancer. The saponin-enhanced transfection was termed sapofection and involves the parallel administration of the endosomal-escape enhancing saponin and the DNA-containing nanoparticles. Parallel administration is not a problem in vitro, but the synchronization of the two components is a challenge in vivo. The aim of the present work was therefore to develop and optimize a non-viral gene delivery vehicle that contains the DNA to be transfected and the transfectionenhancing saponin SO1861 in one particle that can be administered intravenously. The integration of a targeting ligand into the gene delivery vehicle to be developed shall ensure the targeted delivery of the nanoparticles. To achieve this research goal, SO1861 was covalently conjugated to a poly-L-lysinebased peptide scaffold using a pH-sensitive hydrazone linker. The prepared SO1861-functionalized peptides were used to complex DNA by electrostatic interaction. The resulting nanoplexes were evaluated for in vitro transfection efficiency and tolerability. In a variety of cell lines, the nanoplexes with conjugated SO1861 transfected significantly more effectively than non-functionalized nanoplexes with supplementation of free SO1861 in the transfection medium. The transfection with SO1861-nanoplexes was well tolerated. Targeted nanoplexes containing covalently conjugated SO1861 were prepared by incorporation of a targeting peptide and tested in vivo in an allograft tumor model in mice. Using a suicide gene vector encoding the cytotoxic protein saporin, treatment with targeted SO1861-containing nanoplexes was observed to slow tumor growth and improve survival compared to vehicle control.     Going Beyond Counting First Authors in Author Co-citation Analysis

    The present study examines one of the fundamental aspects of author co-citation analysis (ACA) - the way co-citation counts are defined. Co-citation counting provides the data on which all subsequent statistical analyses and mappings are based, and we compare ACA results based on two different types of co-citation counting - the traditional type that only counts the first one among a cited work's authors on the one hand and a non-traditional type that takes into account the first 5 authors of a cited work on the other hand. Results indicate that the picture produced through this non-traditional author co-citation counting contains more coherent author groups and is therefore considerably clearer. However, this picture represents fewer specialties in the research field being studied than that produced through the traditional first-author co-citation counting when the same number of top-ranked authors is selected and analyzed.     “Variations on the Author” discusses two of Eduardo Coutinho’s recent films (Um Dia na Vida, from 2010, and Últimas Conversas, posthumously released in 2015) and their contribution to the general question of documentary authorship. The director’s filmography is characterized by a consistent yet self-effacing form of authorial self-inscription: Coutinho often features as an interviewer that rather than express opinions propels discourses; an interviewer that is good at listening. This mode of self-inscription characterizes him as an author who is not expressive but who is nonetheless markedly present on the screen. In Um Dia na Vida, however, Coutinho is completely absent form the image, while Últimas Conversas, on the contrary, includes a confessional prologue that moves the director from the margins to the center of his films. This article examines the ways in which these works stand out in the filmography of a director who offers new insights into the notion of cinematic authorship

    Appropriate Similarity Measures for Author Cocitation Analysis

    We provide a number of new insights into the methodological discussion about author cocitation analysis. We first argue that the use of the Pearson correlation for measuring the similarity between authors’ cocitation profiles is not very satisfactory. We then discuss what kind of similarity measures may be used as an alternative to the Pearson correlation. We consider three similarity measures in particular. One is the well-known cosine. The other two similarity measures have not been used before in the bibliometric literature.     Das Potenzial der überkritischen Fluidchromatographie für die Trennung von therapeutischen Peptiden unter Berücksichtigung aktueller pharmazeutischer Richtlinien

    This work explores the potential of supercritical fluid chromatography for the analysis of peptide drugs. The regulatory aspects of peptides are more complex and challenging than for small drug molecules. The same applies to the analytical characterization of peptides. After technical innovations and the introduction of a new instrument generation, supercritical fluid chromatography nowadays is a suitable alternative to reversed-phase chromatography, which is considered the gold standard in peptide analysis. However, the scientific data on the analysis of peptides with supercritical fluid chromatography is limited. In the first part of this thesis, supercritical fluid chromatography is used to develop a method to separate the variety of cyclic and linear peptides characteristic for tyrothricin. A software-based design of experiments approach is utilized to optimize an analytical method that provides superior separation of cyclic peptides compared to the reversed-phase separation. Even isomeric peptide pairs are efficiently separated. Subsequently, the potential of a binary mixture of methanol and acetonitrile combined with additives as the modifier for the separations of other cyclic peptides is investigated. The proportion of the mixture is optimized to improve the resolution and control the elution order of the analytes. Paired with an aromatic stationary phase, this chromatographic parameter offers an effective method optimization opportunity. This knowledge is furthermore extended to larger and more hydrophilic peptides in the following experiment. A single method for the purity analysis of human insulin and six analogs is optimized. A crown ether is also investigated as a promising additive to control the elution order of the insulins in dependence to its concentration. In the last study, the applicability of an established chromatographic modeling software for the in-silico optimization of peptide separations in supercritical fluid chromatography is demonstrated. Optimizing a binary mixture of methanol and acetonitrile serves as an effective parameter of the model. Using such software tools offers enormous time savings and, at the same time, meets the current requirements for analytical method development according to the quality by design concept. These already published studies are finally put into the context of the life cycle concept of analytical methods that will soon be demanded by the ICH Q14 guideline "Analytical procedure development". Existing knowledge, in the form of scientific publications, is employed as the basis for a general risk assessment applicable to peptide separations with supercritical fluid chromatography. From this, a generic method development strategy is derived and presented. This way, the current state of science and technology is taken into account to define an efficient workflow. This is particularly relevant in early pharmaceutical development phases, from where only a small fraction of initial drug candidates reaches market maturity. At the same time, this approach enhances the drug knowledge in early development phases and ultimately serves patient safety.     We conducted a full-scale evaluative citation analysis study of scholars in the XML research field to explore just how different from each other author rankings resulting from different citation counting methods actually are, and to demonstrate the capability of emerging data and tools on the Web in supporting more realistic citation counting methods. Our results contest some common arguments for the continued use of first-author citation counts in the evaluation of scholars, such as high correlations between author rankings by first-author citation counts and other citation counting methods, and high costs of using more realistic citation counting methods that are not well-supported by the ISI databases. It is argued that increasingly available digital full text research papers make it possible for citation analysis studies to go beyond what the ISI databases have directly supported and to employ more sophisticated methods
