A new technological approach in diagnostic pathology: mass spectrometry imaging-based metabolomics for biomarker detection in urachal cancer
Neumann JM, Niehaus K, Neumann N, et al. A new technological approach in diagnostic pathology: mass spectrometry imaging-based metabolomics for biomarker detection in urachal cancer. Laboratory investigation. 2021.Urachal adenocarcinomas (UrC) are rare but aggressive. Despite being of profound therapeutic relevance, UrC cannot be differentiated by histomorphology alone from other adenocarcinomas of differential diagnostic importance. As no reliable tissue-based diagnostic biomarkers are available, we aimed to detect such by integrating mass-spectrometry imaging-based metabolomics and digital pathology, thus allowing for a multimodal approach on the basis of spatial information. To achieve this, a cohort of UrC (n=19) and colorectal adenocarcinomas (CRC, n=27) as the differential diagnosis of highest therapeutic relevance was created, tissue micro-arrays (TMAs) were constructed, and pathological data was recorded. Hematoxylin and eosin (H&E) stained tissue sections were scanned and annotated, enabling an automized discrimination of tumor and non-tumor areas after training of an adequate algorithm. Spectral information within tumor regions, obtained via matrix-assisted laser desorption/ionization (MALDI)-Orbitrap-mass spectrometry imaging (MSI), were subsequently extracted in an automated workflow. On this basis, metabolic differences between UrC and CRC were revealed using machine learning algorithms. As a result, the study demonstrated the feasibility of MALDI-MSI for the evaluation of FFPE tissue in UrC and CRC with the potential to combine spatial metabolomics data with annotated histopathological data from digitalized H&E slides. The detected Area under the curve (AUC) of 0.94 in general and 0.77 for the analyte taurine alone (diagnostic accuracy for taurine: 74%) makes the technology a promising tool in this differential diagnostic dilemma situation. Although the data has to be considered as a proof-of-concept study, it presents a new adoption of this technology that has not been used in this scenario in which reliable diagnostic biomarkers (such as immunohistochemical markers) are currently not available
Equivalent Results in Minimax Theory
In this paper we review known minimax results with applications ingame theory and show that these results are easy consequences of thefirst minimax result for a two person zero sum game with finite strategysets published by von Neumann in 1928: Among these results are thewell known minimax theorems of Wald, Ville and Kneser and their generalizationsdue to Kakutani, Ky-Fan, König, Neumann and Gwinner-Oettli. Actually it is shown that these results form an equivalent chainand this chain includes the strong separation result in finite dimensionalspaces between two disjoint closed convex sets of which one is compact. To show these implications the authors only use simple propertiesof compact sets and the well-known Weierstrass Lebesgue lemma.convex analysis;game theory;finite dimensional separation of convex sets;generalized convexity;minimax theory

Decision-making support systems: Theory & practice - eBooks and textbooks from bookboon.com
Pages : 148This collection of work flows from author Udo Richard Averweg’s curiosity and long experience in the Information Systems (IS) field of decision-making support systems. It is not only a description of data and methods, but a commentary on theoretical constructs in different contexts, with a broad set of snapshots from Udo’s ongoing participation in the IS field. This book is a timely review and look into the nature and content of decision-making styles and methods. It is a valuable contribution from an author with a strong mix of practitioner and academic work, both locally and internationally
Prospect theory and hedging risks
The prospect theory is one of the most popular decision-making theories. It is based on the S-shaped utility function, unlike the von Neumann and Morgenstern (NM) theory, which is based on the concave utility function. The S-shape brings in mathematical challenges: simple extensions and generalizations of NM theory into the prospect theory cannot be frequently achieved. For example, the nature of monotonicity of the indifference curve depends on the underlying mean. Price hedging decisions also become more complex within the prospect theory. We discuss these topics in detail and offer a general result concerning the sign of a covariance from which we then infer desired properties of the indifference curve and also justify hedging decisions within the prospect theory. We illustrate our general considerations with a thoroughly worked out example. --prospect theory,mean-variance model, indifference curve, price uncertainty, hedging
Emotional regulation and motivation to drink: gender, negative emotionality, behavioral undercontrol, and family history
Despite support for the effectiveness of alcohol use disorders (AUD) treatment programs and the positive impact of policy changes related to alcohol use, AUD remains a major public health concern in the United States. As part of an effort to encourage transdisciplinary research, the integration of objective biological measures for AUD risk and existing psychosocial-based risk measures (e.g., demographic variables, personality characteristics, comorbid psychological disorders) are emerging as important areas of inquiry, with implications for the prevention and treatment of AUD. Theories of alcohol use emphasize the fundamental role of emotional regulation in drinking behaviors, and multiple psychosocial factors have been identified which influence such motivations for alcohol use. Through three separate, laboratory-based experimental investigations, this dissertation aimed to gain an understanding of the physiological mechanisms underlying the relationship between psychosocial factors (gender, personality, family history) and one’s desire to use alcohol for emotional regulation. In particular, this dissertation focused on examining the applicability of heart rate variability (HRV), an established psychophysiological measure of peripheral and central modulation of emotional arousal, in studying individual differences in emotional regulation. Changes in HRV in response to experimental manipulation of emotion and adaptive responding were linked to gender and personality differences in motivations for alcohol use. Gender differences in emotional reactivity suggest distinctive pathways toward unhealthy use of alcohol in men and women; that is, the pharmacological effects of alcohol appear to directly promote alcohol use in men, whereas cognitive expectancies, such as expectation that alcohol can counteract negative emotions, may underlie alcohol use in women, particularly when they are prone to negative mood states. Further, personality constructs of negative emotionality were associated with physiological dysregulation of emotion, which was linked to tendency to use alcohol for emotional regulation, particularly to suppress negative emotion. As a future direction, identification of malleable biological markers and the translation of these findings into clinical practices may help to better identify individuals at risk and suggest a novel approach for preventing or intervening in the development of AUD, which may in turn contribute to population health.Ph.D.Includes bibliographical referencesby Tomoko Udo Schalle
Differential Taxation and Corporate Futures-Hedging
Using a two-moment decision model this paper analyzes corporate hedging behavior in the presence of unified and differential income taxation. We start with the well-known result that risk-taking may increase when income tax rates increase and, therefore, the incentive for hedging reduces. We demonstrate that pure hedging is differently affected by taxation than speculative hedging is. Analysing tax-sensitivity of the corporate hedge shows that a higher risk in the first place may reduce the tax-induced incentive to revise a futures position. --taxation,hedging,mean-variance model, unified and differential taxation,Roy preference function
