    Tissue culture aiming at mutation breeding in banana (Musa spp.)

    Os métodos tradicionais de melhoramento de plantas apresentam certas dificuldades para algumas culturas, principalmente as de propagação vegetativa, como é o caso das bananeiras e plátanos. Os cultivares são normalmente constituídos de híbridos interespecíficos, sendo comum a heterozigozidade a poliploidia e a esterilidade na maioria dos clones, sendo que esta última cria frequentemente, uma barreira insuperável para os cruzamentos. Devido à propagação vegetativa, estas culturas possuem uma homogeneidade genética muito grande nas plantações, tornando-a vulnerável a uma série de doenças, dentre as quais destaca-se a Fusariose, causada pelo Fusarium oxysporum f. sp. cubense, a qual impossibilita o plantio comercial de diversos cultivares, como é o caso da bananeira cultivar Maçã (grupo AAB), de ampla aceitação por todo o Brasil. Uma vez que existe um grande potencial para a biotecnologia nesta cultura, o presente trabalho buscou adaptar diferentes metodologias de cultura de tecidos visando trabalhos de indução de mutações em Musa. Foram extraídos meristemas de bananeira dos cultivares Maçã, Nanica, Nanicão e Grand Naine (clone GN-60), os quais foram inoculados em meio modificado de MURASHIGE & SKOOG (1962). Após multiplicação in vitro as plantas obtidas foram utilizadas para a realização de diversos experimentos. Foram testados cinco diferentes tipos de extração de ápices caulinares, a serem utilizados a partir de plantas in vitro para reinício da micropropagação sendo constatado que os ápices caulinares dos quais foram retiradas as baínhas foliares e aqueles dos quais foi eliminada a dominância apical tenderam a apresentar maior número de brotações. Comparou-se os diferentes níveis de brotação obtidos em meio de cultura semi-sólido com os obtidos em meio liquido com e sem agitação, concluindo-se que o líquido sob agitação tendeu a estimular uma maior brotação que o semi-sólido, e ambos superaram o meio líquido sem agitação. Foi ainda avaliada a menor concentração de BAP indutora da maior taxa de brotação in vitro, para o cultivar Maçã, entre as concentrações de 0; 2,5; 5,0; 7,5; 10,0; 12,5; 15,0; 17,5 e 20,0 mg/l. A menor concentração que permitiu a obtenção de um maior número de brotos situou-se ao redor de 5,0 mg/l de BAP. Para um enraizamento mais eficiente em plântulas de bananeira comparou-se os reguladores de crescimento IAA, IBA e NAA em diferentes concentrações. Observou-se que na concentração mais baixa de NAA e IBA (0,1 mg/l) o enraizamento foi mais adequado. Estudou-se a radiossensitividade de ápices caulinares do cultivar Maçã às doses de 0, 20, 40, 60, 80, e 100 Gy. Observou-se que a dose de 20 Gy provocou um leve estímulo a multiplicação clonal do cultivar, enquanto que a que causou 50% de letal idade (LD5), esteve por volta de 40 Gy, já as doses de 80 e 100 Gy não permitiram a recuperação de brotações. Os explantes irradiados foram subcultivados por 4 ciclos vegetativos (M1V4), este número de gerações foi considerado satisfatório para o isolamento de setores mutados, uma vez que nesta geração, in vitro, foram observadas algumas alterações fenotípicas para variegação foliar, a partir de explantes irradiados nas doses de 40 e 60 Gy. As alterações fenotípicas, no entanto, tornaram-se mais perceptíveis em casa de vegetação, onde a partir de 2070 plantas observadas quantificou-se as alterações mais significativas. Foram obtidas plantas com folhas variegadas, plantas com maior teor de antocianina e plantas com alterações para o formato e arquitetura foliar e de planta. Após o isolamento do Fusarium oxysporum f. sp. cubense a partir de plantas de campo infestadas, foi realizado o seu cultivo em meio BDA, obtendo-se após 15 dias uma maior produção de esporos, a partir dos quais foram confeccionadas suspensões para inoculação em casa de vegetação. Realizou-se a inoculação de 2765 plantas, de bananeira "Maçã", originadas de ápices caulinares irradiados e micropropagados in vitro por 4 gerações vegetativas. À inoculação foi realizada através de 50 ml de suspensão contendo 5 x 104 esporos/ml do Fusarium. Embora as respostas tenham sido diferenciadas de planta à planta, não foram observadas plantas resistentes ao fungo. O sistema utilizado permitiu uma triagem rápida e eficiente das plantas susceptíveis. Seguindo metodologia descrita por NOVAR et al. (1989), induziu-se a formação de estruturas embriogênicas sobre tecidos do rizoma e pseudocaule de plantas pertencentes aos cultivares Maçã, Grand Naine (clone GN-60) e Nanicão. Para esta finalidade utilizou-se o regulador de crescimento Dicamba, cuja melhor concentração, variou em função do cultivar testado. Destas estruturas obtidas não se originaram plantas diretamente, mas o seu subcultivo em meio líquido, permitiu o surgimento de novas estruturas que parecem possuir boa capacitação embriogênica, uma vez que apresentaram emissão de radículas e início de emissão de parte aérea. Não foi possível a recuperação de plantas por embriogênese somática até o momento. A partir do subcultivo das estruturas embriogênicas previamente descritas foram obtidas suspensões celulares, as quais mostraram-se apropriadas para o isolamento de protoplastos.Conventional plant breeding methods for have shown difficulties for some crops, principally for the vegetative propagated ones such as bananas and plantains. The cultivars normally consist of interspecific hybrids and heterozygosity and polyploidy are commonly seen in most of the clones. Its assexual behaviour is often a serious limitation to cross-breeding. Due to the vegetative propagation, those cultivars show high genetic homogeneity and this makes them vulnerable to serious diseases such as Fusarium wilt caused by Fusarium oxysporum f.sp. cubense which hampers commercial production in the cultivars with the AAB genome, as "Maçã", of large acceptance in Brazil. As there exists a great potential of biotechnology to this crop, the present work was carried out to adapt different tissue culture methodologies to Musa species, aiming at future mutation breeding programs. Meristems were extracted from the cultivars Maçã, Nanica, Nanicâo and Grand Nain (clone GN-60), and were inoculated on the modified MURASHIGE & SKOOG (1962) medium. After in vitro propagation, these plants were used for several experiments. Shoot tip apices were extracted in five different ways. After in vitro propagation it was found that those without sheath and apical dominance tended to produce a larger number of buds. Different levels of bud formation were observed on semi-solid medium culture when compared with agitated or static liquid ones. The agitated liquid medium tended to increase the number of buds more than the semi-solid one which was better than static liquid one. The growth regulator BAP was used in several concentrations (0, 2.5, 5.0, 7.5, 10.0, 12.5, 15.0, 17.5 and 20.0 mg/l) for bud formation of the shoot tip apices. The lowest concentration which permitted a higher number of cluster formation was around 5.0 mg/l for "Maçã". For efficient rooting of in vitro plants, the auxins IAA, IBA and NAA with different concentrations were tested. Lower concentrations of NAA or IBA (0.1 mg/l) or IAA (0.5 mg/l) showed high efficiency, permitting rooted plants formation in two weeks. Shoot tip apices radiosensitivity was carried out in "Maçã". Doses of 0, 20, 40, 60, 80 and 100 Gy were used. Twenty Gy showed low stimulus of bud formation when compared with the control. Fourty Gy was found to be the dose that permitted the survival of 50 % of plants (LD50). Higher doses as 80 and 100 Gy did not permit bud formation. Irradiated explants were subcultivated for four vegetative generations (M1V4). This procedure was found to be sufficient for isolation of the mutated sectors, because it was possible to observe in this generation some phenotypic changes of leaf variegation at the doses of 40 and 60 Gy. Phenotypic changes, meanwhile, were more perceptible in green house condition where they were quantified in 2070 plants. Plants with leaf variegation, concentrated anthocyanin, disturbed leaf shape and changed plant and leaf architecture were observed. After isolation of Fusarium oxysporum f. sp. cubense from infested plants in the field, its cultivation was carried out in BDA medium. After 15 days, higher production of spores was obtained and they were used to prepare suspention for inoculation in green house. The inoculation was carried out to 2765 plants of "Maçã", originated from irradiated shoot tip apices that were micropropagated in vitro for 4 vegetative generations. The inoculation was made. Fifty ml of suspention containning 5 x 104 spores/ml of Fusarium was inoculated to each plant. Although the differentiated responses, no resistant plant was found. The utilized system permitted a quick and efficient screening of the susceptible ones. Following the methodology described by NOVAK et al. (1989), embryogenic structure formation was induced on corn and pseudostem tissues of the cultivars Maçã, Grand Nain (clone GN-60) and Nanicão. Dicamba, a growth regulator, was used for this objective. The best concentration was changed in the function of tested cultivar. These structures did not originate plants directly, but after liquid subcultivation, permitted the appearance of new structures which seems to have a good embryogenic capacity. They permitted root emission and initiation stem formation, but it was not possible to regenerate plants by somatic embryogenesis until the present. From 1iquid subculture of embryogenic structure, cell suspension was obtained and this showed to be appropriate for protoplast isolation

    In vivo and in vitro methodology for mutagenesis of chrysanthemum (Dendranthema grandiflora Tzvelev) with use of gamma ray and ethyl methanesulfonate (EMS) aiming genetic improvement

    O presente trabalho teve como objetivos principais: desenvolvimento e aplicação de metodologias in vivo e in vitro na indução de mutação em crisântemo (Dendranthema grandiflora Tzvelev), com o uso de raios-gama e o mutagênico químico metanossulfonato de etila (EMS); comparação das metodologias in vitro e in vivo quanto à freqüência de mutantes e o tamanho do setor mutado. Visou-se a obtenção de mutantes para a coloração das flores no cultivar mutante 'Ingrid' de cor rosa escuro, obtido a partir do cultivar original 'Repin Rosa'. Os experimentos foram realizados no Laboratório de Radiogenética do CENA em Piracicaba - SP e na empresa Van Shaik da Cooperativa Holambra em Holambra - SP. Para a irradiação de plantas utilizou-se a fonte de 60Co do CENA. EMS foi utilizado para o tratamento in vitro de pedicelos de botões florais imaturos visando-se a indução de gemas adventícias e a regeneração de plantas. Realizou-se um teste preliminar de sensitividade, escolhendo-se a concentração em função da redução de 50% no número de plantas regeneradas por pedicelo. Após o tratamento com a concentração escolhida (0,075 M 1:45 horas) obteve-se uma freqüência de 14% de mutantes sendo que destes houve uma ocorrência de uma alta freqüência (11,8%) de mutantes só1idos para a cor da inflorescência. Num experimento preliminar, determinou-se que a dose de 22,5 Gy de raios-gama reduzia cerca de 50% a altura das plantas da geração M1V1 obtida por meio de plantas crescidas in vitro. Plantas in vitro foram irradiadas com esta dose e o avanço de gerações foi realizado utilizando a técnica das podas repetidas, por meio do cultivo de internódios in vitro. Mudas enraizadas foram irradiadas in vivo com 20 Gy de raios-gama, avançando-se in vivo as gerações também por meio das podas repetidas. A freqüência de mutantes foi maior in vitro (13,8%) do que in vivo (7,1%) e em ambos os casos na geração M1V3 obteve-se a maior porcentagem de quimeras periclinais. Avaliou-se a freqüência e o tipo de quimeras em plantas da geração M1V2 proveniente de 6 gemas axilares obtidas ao longo de ramos M1V1 originados após a irradiação de plantas enraizadas in vivo com 20 Gy de raios-gama. O número de quimeras periclinais foi baixo (0,4%) e não foram observadas diferenças significativas no número e tipos de quimeras observadas nas plantas originadas das 6 diferentes posições. Dentre os diferentes tipos de mutantes obtidos nos vários experimentos, foram selecionados os de cor chá-rosa, vinho, bronze e variegado que foram avaliados em ensaios de produção. O mutante variegado mostrou instabilidade para a coloração, os outros, com exceção do chá-rosa, mostraram potencial para liberação como novos cultivares, porque além do interesse pela nova coloração de pétalas, mantiveram em geral as boas características agronômicas e comerciais do cultivar original. Estes mutantes estão sendo multiplicados pelo produtor para verificação de sua aceitação pelos consumidores.The present work had as major objectives: the development and application of in vitro methodologies for mutation induction in chrysanthemum (Dendranthema grandiflora Tzvelev), using gamma ray and the chemical mutagen ethyl methanesulfonate (EMS); the development and application of in vivo methodology for mutation induction using gamma ray; comparison of in vitro and in vivo methodologies for frequency of mutant induction and the size of mutated sector. The attainment of mutants for flower color in the mutant cv 'Ingrid' with dark pink color, originally obtained from cv 'Repin Rosa', was aimed. The experiments were carried out at the Laboratory of Radiation Genetic of CENA in Piracicaba-SP and in the company Van Shaik of the Holambra Cooperative in Holambra-SP. A source of 60Co at CENA was used to irradiate the plants. EMS was used to mutate pedicels of immature floral buds in vitro, aiming to produce adventitious buds and plant regeneration. A preliminary test of sensitivity determined the concentration that reduced the number of plants regenereted from pedicel in 50%. The chosen concentration (0.075 M EMS for 1:45 hours) induced 14% of mutants, with a high frequency (11.8%) of solid mutants for flower color. In a preliminary experiment, it was determined that a dose of 22.5 Gy of gamma ray reduced the height in about 50% of the M1V1 plants. In vitro plants were irradiated with this dose and generations were advanced using cutting back technique by culturing stem segments. Rooted cuttings were irradiated in vivo with 20 Gy of gamma ray, advancing generations by cutting back. The frequency of mutants was larger in vitro (13.8%) than in vivo (7.1 %), and in both the cases, the generation M1V1 a got the largest percentage of periclinal chimeras. The frequency and type of chimeras were evaluated in plants of generation M1V2, proceeding from six axillary buds originated from branches of rooted plants in vivo M1V1 after irradiation with 20 Gy of gamma ray. The number of periclinal chimeras was low and no significant differences were observed in number and type of chimeras. Among the different types of mutants obtained in the experiments, the "tea-rose", "wine", "bronze" and "variegated" were selected and evaluated in production trial. The variegated mutant showed instability for color. With the exception of tea-rose, all mutants had shown potential as new cultivars. The mutants have kept the good agronomic and commercial features besides their new colors. These mutants are being multiplied by the producer for market test with consumers

    Banana micropropagation, Maçã and Grand Naine cultivars, aiming low cost plantlet

    Objetivou-se desenvolver método que permitisse a obtenção de mudas de bananeira micropropagadas de baixo custo. Utilizaram-se as cultivares de bananeira Maçã e Grande Naine, analisando-se os custos de produção de mudas durante três épocas dos anos de 1999 a 2000, em dois ambientes distintos. Um dos ambientes foi a sala de crescimento, normalmente usada para micropropagação, com controle de iluminação e temperatura e o outro um telado sem tais controles, com iluminação e temperatura ambientes e com instalações mais simples e de menores custo. Foram efetuados os cálculos dos custos das mudas micropropagadas obtidas nos dois ambientes, levando-se em conta os seguintes fatores: total de mudas produzidas nas três épocas; produtos químicos usados para confecção de meios de cultura para multiplicação e enraizamento; energia elétrica. Calculou-se também o custo para implementação de sala de crescimento e telado, semelhantes aos usados nessa pesquisa. Concluiu-se que para os dois cultivares houve redução de custo das mudas produzidas em condição de telado. Atribuiu-se esse menor custo principalmente devido ao fato que em telado nao se necessita de energia elétrica, que é usada na iluminação e refrigeração da sala de crescimento. O custo de implementação de um telado é bem menor do que uma sala de crescimento. Tanto em condição de sala de crescimento como em telado o número de mudas produzidas nos dois cultivares foi menor do que em sala de crescimento mas para os dois cultivares a redução de custo de produção em telado foi próxima (81,1% para banana Grande Naine e 85,4% para banana Maçã). Entretanto, o menor número de mudas produzidas em telado pode exigir um espaço físico relativamente maior para a produção de um certo número de mudas necessário para plantio em época determinada e isto precisa ser considerado quando da aplicação desse método.The present work was carried out to establish the methodology to obtain plantlets of banana at low cost through micropropagation. The cultivars Maçã and Grand Naine were utilized, analyzing multiplication rates and production cost of plantlets during different three periods fro 1998 to 2000 in to distinct environrnents: one was growth room, normally used for micropropagation where temperature and light are controlled and another was screened house which is a simple and low cost structure with environrnental light and temperature. The cost of plantlets obtained in these two environrnents was calculated considering the following factors: total plantlets obtained in three periods in a year, costs of chemical products used to prepare media for multiplication and rooting and of electricity consumption. The costs of installation of temperature and light controlled growth room and screened house, similar to those used in the present work, were also calculated. It was concluded that production cost of plantlets of these two banana cultivars was lower in screened house than in growth room, which is attributed principally to no consumption of electricity that is higher in growth room to control both light and temperature. The cost of installation of screened house is much lower than that of growth room. The number of plantlets of the cultivar Grand Naine obtained both in growth room and screened house was higher than that of Maçã, although the reduction of production cos of plantlets in screened house of the both cultivars was almost the same (81,1 % for Grand Naine and 85,4% for Maçã). However, reduced number of plantlets produced in screened house can require relatively large physical space in order to supply them which are necessary to plant in a determined period. This should be taken in consideration when this methodology is applied

    Para possibilitar a utilização de técnicas de biologia celular e molecular em variedades nacionais de cana-de-açúcar foram selecionados calos a partir de explantes foliares que deram origem a suspensões celulares embriogênicas. Tais suspensões tiveram seu crescimento quantificado segundo parâmetros como número de células, peso fresco, peso seco e volume do total de células. Plantas foram regeneradas a partir de calos e suspensões celulares com até cinco meses de subcultivo. Protoplastos foram isolados a partir de calos e células em suspensão em diferentes soluções enzimáticas, métodos e idades de cultura, calos foram obtidos de protoplastos isolados de células em suspensão, mas não houve regeneração de plantasThis work was carried out to study callus induction, ceel suspension initiation and protoplast isolation and culture. Two cultivated sugarcane varieties were used namely SP 70-1143 and SP 79-1011. The following steps were accomplished: (1) Selection of the best culture conditions to promote callus formation and support the next steps. (2) Cell suspension culture were iniciated and cells selected which would had cells with proper morphogenic characteristics to be used in protoplast isolation and plant regeneration. The selection of the best technique was supported by parameters obtained from number of cells, fresh and dry weigth, and packed cell volume. The variable number of cells was the best expression to the cellular suspension growth. Different culture media were evaluated with emphasis on the 2, 4-D dosages requerid for long time duration of the cell culture. The MS culture medium with 1.5 to 3.0 mg / 1 of 2,4-D, 500 mg / 1 of hidrolysed casein and coconut milk (5 %) showed efficient. ( 3) Plant regeneration from cell suspensions were tested to establish the conditions for the use of biobalistic techniques. The MS culture medium was able to regenerate plants in a cellular suspension until the period of five months. (4) Protoplast isolation were studied using different enzymes with known methodologies and different culture age. The rates of isolation, colony and microcallus formation were recorded. The enzyme solution containing Cellulase RS (1 %), Pectolyase Y23 (0.2 %), EMS (5.0 mM) and CPW salts with manitol (13 %) as osmoticum yielded a good number of protoplasts. Calli derived from protoplasts were obtained in three differents media, being R2 the best followed by N6 and GM. ( 5) Plant regeneration from protoplasts.     Indução de mutação in vivo, no melhoramento de crisântemo (Dendranthema grandiflora Tzvelev) cv. Repin Rosa

    Mutation induction was used with the objective of obtaining mutants for flower colour of chrysanthemum, cv. Repin (pink colour). Rooted cuttings were irradiated with 20 Gy of gamma rays and before the selection the cutting back method was used to advance the generations. The frequency of colour mutants observed was 5.8%. Among the mutants obtained the white and dark-pink-coloured ones were evaluated in yield trial and post-harvest. The results indicated that these mutants mantained the same agronomical characteristics showed by the control, with the exception of plant height in the white mutant that was shorter. Due to commercial interest of the producers, these mutants were multiplied and released as new cultivars. The white flower colour mutant was named Cristiane and the dark-pink, Ingrid. This was the first example of cultivars from an ornamental plant released by mutation breeding in Brazil.Objetivou-se adaptar e utilizar o método de indução de mutação por irradiação no melhoramento de crisântemo, para a obtenção de mutantes na coloração de pétalas da cultivar Repin Rosa. Mudas obtidas a partir de estacas enraizadas foram submetidas a irradiação com 20 Gy de raios-gama, aplicando-se a seguir o método de podas repetidas para avanço das gerações, antes da seleção dos mutantes. Observou-se uma freqüência de 5,80% de mutantes para coloração de pétalas. Dentre as várias colorações obtidas, os mutantes de cor branca e rosa-escuro foram selecionados, considerando-se a preferência do mercado consumidor, e incluídos em ensaios de produção e avaliação pós-colheita. Os resultados indicaram que, com exceção da altura de planta do mutante de cor branca, que foi menor, as demais características agronômicas foram semelhantes às da cultivar original. Estes mutantes foram multiplicados e lançados no comércio; o de cor branca foi denominado Cristiane, e o de cor rosa-escuro, Ingrid. Trata-se do primeiro exemplo de plantas ornamentais obtidas por indução de mutação no Brasil

    Going Beyond Counting First Authors in Author Co-citation Analysis

    The present study examines one of the fundamental aspects of author co-citation analysis (ACA) - the way co-citation counts are defined. Co-citation counting provides the data on which all subsequent statistical analyses and mappings are based, and we compare ACA results based on two different types of co-citation counting - the traditional type that only counts the first one among a cited work's authors on the one hand and a non-traditional type that takes into account the first 5 authors of a cited work on the other hand. Results indicate that the picture produced through this non-traditional author co-citation counting contains more coherent author groups and is therefore considerably clearer. However, this picture represents fewer specialties in the research field being studied than that produced through the traditional first-author co-citation counting when the same number of top-ranked authors is selected and analyzed.     “Variations on the Author” discusses two of Eduardo Coutinho’s recent films (Um Dia na Vida, from 2010, and Últimas Conversas, posthumously released in 2015) and their contribution to the general question of documentary authorship. The director’s filmography is characterized by a consistent yet self-effacing form of authorial self-inscription: Coutinho often features as an interviewer that rather than express opinions propels discourses; an interviewer that is good at listening. This mode of self-inscription characterizes him as an author who is not expressive but who is nonetheless markedly present on the screen. In Um Dia na Vida, however, Coutinho is completely absent form the image, while Últimas Conversas, on the contrary, includes a confessional prologue that moves the director from the margins to the center of his films. This article examines the ways in which these works stand out in the filmography of a director who offers new insights into the notion of cinematic authorship

    Appropriate Similarity Measures for Author Cocitation Analysis

    We provide a number of new insights into the methodological discussion about author cocitation analysis. We first argue that the use of the Pearson correlation for measuring the similarity between authors’ cocitation profiles is not very satisfactory. We then discuss what kind of similarity measures may be used as an alternative to the Pearson correlation. We consider three similarity measures in particular. One is the well-known cosine. The other two similarity measures have not been used before in the bibliometric literature.     Dispelling the Myths Behind First-author Citation Counts

    We conducted a full-scale evaluative citation analysis study of scholars in the XML research field to explore just how different from each other author rankings resulting from different citation counting methods actually are, and to demonstrate the capability of emerging data and tools on the Web in supporting more realistic citation counting methods. Our results contest some common arguments for the continued use of first-author citation counts in the evaluation of scholars, such as high correlations between author rankings by first-author citation counts and other citation counting methods, and high costs of using more realistic citation counting methods that are not well-supported by the ISI databases. It is argued that increasingly available digital full text research papers make it possible for citation analysis studies to go beyond what the ISI databases have directly supported and to employ more sophisticated methods
