    Novel Nuclear Export Signal-Interacting Protein, Nesi, Critical for the Assembly of Hepatitis Delta Virus

    The process of host factor-mediated nucleocytoplasmic transport is critical for diverse cellular events in eukaryotes and the life cycle of viruses. We have previously identified a chromosome region maintenance 1- independent nuclear export signal (NES) at the C terminus of the large form of hepatitis delta antigen (HDAg), designated NES(HDAg- L) that is required for the assembly of hepatitis delta virus (HDV) (C.-H. Lee et al. , J. Biol. Chem. 276:8142-8148, 2001). To look for interacting proteins of the NES(HDAg-L), yeast two-hybrid screening was applied using the GAL4- binding domain fused to the NES(HDAg-L) as bait. Among the positive clones , one encodes a protein, designated NESI [NES (HDAg-L) interacting protein ] that specifically interacted with the wild-type NES(HDAg-L) but not with the export/ package-defective HDAg-L mutant, NES*(HDAg-L), in which Pro- 205 has been replaced by Ala. Northern blot analysis revealed NESI as the gene product of a 1.9-kb endogenous mRNA transcript that is present predominantly in human liver tissue. NESI consists of 467 amino acid residues and bears a putative actin-binding site and a bipartite nuclear localization signal. Specific interaction between HDAg-L and NESI was further confirmed by coimmunoprecipitation and immunofluorescence staining . Overexpression of antisense NESI RNAs inhibited the expression of NESI and abolished HDAg-L-mediated nuclear export and assembly of HDV genomic RNA. These data indicate a critical role of NESI in the assembly of HDV through interaction with HDAg-L

New variational principles in L^{infty} are introduced, allowing for the description of the yield set in the context of a simplified model of polycrystal plasticity.     “Heaven” of Data Deficient Species: The Conservation Status of the Endemic Amphibian Fauna of Vietnam

    Predicting the true status of Data Deficient (DD) species is a prominent theme in recent conservation biology, but there still is much debate regarding the conservation approach that should be used for DD taxa and no definitive conclusions are yet available. We review and analyse the current data available on the conservation status of amphibians in Vietnam, with an emphasis on the DD species. We also compare Vietnamese DD frequency of occurrence with other regions of the world, examine the extent of the range of taxa divided by Red List status, and explore the protection attributes of the taxa based on their inclusion within protected areas of Vietnam. We documented that the analysis of amphibians in Southeast Asia, and especially in Vietnam, substantially agrees with patterns highlighted by previous global research, and confirms the risk that several DD species may silently go extinct without their actual risk ever being recognized. Importantly, our study showed that fine-scale analyses are essential to highlight the potential drivers of extinction risk for the DD species of amphibians. A crucial next step for conservation policies in Vietnam (and in surrounding countries) is developing and implementing species-specific studies targeted at addressing each species’ drivers of extinction and determining science-based strategies for minimizing their extinction risk

    Gradient integrability and rigidity results for two-phase conductivities in two dimensions

    This paper deals with higher gradient integrability for σ-harmonic functions u with discontinuous coefficients σ, i.e. weak solutions of div(σ∫u)=0 in dimension two. When σ is assumed to be symmetric, then the optimal integrability exponent of the gradient field is known thanks to the work of Astala and Leonetti and Nesi. When only the ellipticity is fixed and σ is otherwise unconstrained, the optimal exponent is established, in the strongest possible way of the existence of so-called exact solutions, via the exhibition of optimal microgeometries. We focus also on two-phase conductivities, i.e., conductivities assuming only two matrix values, σ1 and σ2, and study the higher integrability of the corresponding gradient field |∫u| for this special but very significant class. The gradient field and its integrability clearly depend on the geometry, i.e., on the phases arrangement described by the sets Ei=σ-1(σi). We find the optimal integrability exponent of the gradient field corresponding to any pair { σ1,σ2} of elliptic matrices, i.e., the worst among all possible microgeometries. We also treat the unconstrained case when an arbitrary but finite number of phases are present. © 2013 Elsevier Masson SAS. All rights reserved
