    What the distribution of sperm size can tell about the stability of spermatogenesis in hybrid frogs Pelophylax esculentus

    Interspecies hybrid frogs Pelophylax esculentus and one of its parental species Pelophylax ridibundus inhabit the Siversky Donets center of diversity of water frogs in Eastern Ukraine. These frogs can crossbreed and form progeny in population systems which are called hemiclonal (HPS). Such systems have their own exceptional features which make them interesting for studying. The Lower Dobrytskiy Pond, which is situated in the National Nature Park “Homilshansky lisy” and is a part of Siversky Donets river basin, is on focus. Current work is devoted to the combination of two methods of spermatogenesis investigation. First, using the method of Ag-staining we observed high variability of meiotic chromosomal plates in testes of 24 adult male water frogs P. esculentus (2n=26). Only one male had 100% of full meiotic plates with no aneuploid plates. A significant amount of studied males (21/24) produced aneuploid chromosomal plates (4-68% of the total amount of meiotic plates). This may lead to a decrease in their fertility or even to their entire sterility. Also, we have not observed any chromosomal meiotic plates in two of 24 males. Some males (8/24) even produced meiotic chromosomal plates with 26 bivalents (i.e. 4n germ cells) which may testify about the ability to produce diploid sperm. Further, the lengths of urinary sperm cells’ heads were measured. Finally, we performed an analysis of both meiotic chromosomes in testes and the distribution of sizes of urinary sperm cells’ heads of hybrid water frogs Pelophylax esculentus from Siversky Donets basin to find out if there is a link between these two features. No difference in sperm heads lengths was found between males producing moderate and low amounts of sperm. Based on the data of meiotic plates all males were assigned into five categories via PCA (principal component analysis). A significant difference in sperm heads lengths was found within the category I (males with mostly full meiotic plates). The analysed data shows that each male from the studied population has his own unique features. No direct link between sperm cells size and features of meiotic chromosomes in testes was found

    Acute phlegmonous appendicitis in rare variant of appendix diverticulum (case report)

    Background. Appendicitis-like diverticulitis is considered a rare disease and is usually the subject of clinical reports. Clinical manifestations of this pathology is similar to those of appendicitis, which is why a preoperative diagnosis is made quite rarely. When carrying out the study, we considered a clinical case of acute phlegmonous appendicitis with a rare variant of the appendix wall diverticulum.Material and methods. To assess clinical cases, a retrospective analysis of medical documentation (medical records of patients with diverticulitis in particular) was used. The assessment of clinical symptoms was accompanied by using general clinical, histological and microbiological methods.Results. Patient I., 62 years old, was diagnosed with acute appendicitis. An urgent appendectomy was carried out, during which the vermiform process was removed. It was located retrocecally, in the adhesive process and loose infiltrate. The length of the process is 7–8 cm, its diameter is 1–1,5 cm. 6 diverticula up to 0,7 cm in size are visible. When the process is cut, the pyoid contents protrudes from its lumen. In addition, the appendix removed during appendectomy was sent for pathological study. A standard histopathological study was carried out with the step-like sectioning and hematoxylin and eosin staining.Conclusion. From the results of a pathological study, it was found that the layers of the vermiform process wall with diverticula are diffusely infiltrated by polymorphonuclear leukocytes. The mucous membrane is anesthetized and had ulcerative defects. The patient was diagnosed with phlegmonous appendicitis, periappendicitis, diverticulitis.Vermiform process diverticulitis is not a common pathology, however a timely diagnosis can cause quite severe complications

    A state of titanium oxide and oxynitride coatings on L316 steel has been studied before and after their contact with model biological fluids. Electrokinetic investigation in 1 mmol potassium chloride showed significant (more than 10 times) fall of magnitude of electrostatic potential of thin (200–300 nm) titanium films at pH changing in the range of 5–9 units during 2 h. Nevertheless, zeta-potential of all samples had negative charge under pH > 6.5. Long-term (5 weeks) contact of samples with simulated body fluid (SBF) promoted steel corrosion and titanium oxide and oxynitride films dissolution. On the other hand, sodium and chloride ions precipitation and sodium chloride crystals formation occurred on the samples. Of positive fact is an absence of calcification of tested artificial surfaces in conditions of long-term being in SBF solution. It is supposed decreasing hazard of fast thrombosis and loss of materials functional properties. According to in vitro experiment conducted, prospective biocompatibility of materials tested before and after their contact with SBF lines up following manner: Ti–O–N (1/3) > Ti–O–N (1/1), TiO2 > Steel. It may be explained by: 1) the corrosion-preventive properties of thin titanium oxide and oxynitride films;2) a store of surface negative charge for Ti–O–N (1/3) film; 3) minor augmentation of mass and thickness of titanium films connected with speed of mineralization processes on the interface of solution/solid body. At the same time, initial (before SBF contact) differences of samples wettability became equal. Modifying effect of model biological fluids on physicochemical characteristics of materials tested (roughness enhancement, a reduction or reversion of surface negative potential, sharp augmentation of surface hydrofilicity) should took into account under titanium oxide and oxynitride films formation and a forecast of their optimal biological properties as the materials for cardiovascular stents

    Invasibility of Common Plant Community Types of the Middle Urals

    This paper specifies the invasibility of common plant community types in the natural habitats of the Middle Urals. Invasibility was defined as the vulnerability of a community to alien plant species invasions, regardless of the conditions in which the community existed. We analyzed 749 vegetation relevés made in natural bogs, floodplains, rocky grasslands, meadows, and forest communities. We surveyed urban and non-urban habitats (30–40 km from the city). Invasibility was calculated in two different ways based on two parameters: the number and proportion of alien species in the relevé. These invasibility parameters are widely applicable and comparable, scale-independent, measurable, and reliable, based on data that do not require the destruction of ecosystems or biota and are based on available data. Based on the invasibility degree, we identified three community-type groups with two subgroups in one group. In ascending order of invasibility degree, the community types were arranged as follows: (1) bogs with and without trees; (2a) coniferous forests; grass communities in floodplains; (2b) deciduous forests, coniferous plantations, floodplain communities with woody plants; and (3) dry meadows and rocky grasslands. Obtained results of the assessment of different plant community invasibility may be used to understand patterns of alien plant distribution in local habitats and the reasons for the different vulnerability of communities to plant invasions

    Widespread distribution of HLA-DR-expressing cells in macroscopically undiseased intima of the human aorta : a possible role in surveillance and maintenance of vascular homeostasis

    The architectonics and cell composition of the human large arteries are not sufficiently understood. The present study is the first to undertake an analysis of the distribution and quantities of HLA-DR-expressing cells in grossly undiseased human intima using immunohistochemical and immunofluorescent analysis, complemented by the advantages of confocal microscopy. The study revealed a widespread distribution of HLA-DR-expressing cells throughout the intimal space where the cells were integrated into continuous networks via long cell processes. Numbers of HLA-DR+ cells were found to be significantly larger in the middle third of the intima than in the superficial and deep intimal portions. We speculate that a widespread distribution of HLA-DR-expressing cells in the intima of normal human aorta might play a role in the surveillance and maintenance of vascular homeostasis
