    La presenza femminile sulla scena pubblica dall’età dei lumi allo Stato unitario se è poco conosciuta, nondimeno è significativa per numero e spessore di personalità. Alle donne si è voluto dedicare, nell’anno in cui è ricorso il 150° dell’Unità italiana, un convegno di studio – i cui atti sono raccolti in questo volume – capace di cogliere le diverse sfaccettature del loro protagonismo. Le donne hanno discusso, parlato e scritto e si sono impegnate in prima persona nella stagione del Risorgimento italiano, in un clima europeo di primi pensieri “emancipazionisti” cresciuti sui modelli inglesi e francesi e rafforzati dalla pubblicazione nel 1848 del Manifesto di Seneca Falls. Le donne bolognesi sono state protagoniste di primo piano nei loro salotti, nei circoli, nei teatri, sul lavoro del dibattito politico e culturale che ha preparato e realizzato l’Unità italiana. Altrettanto forte è stato il loro impegno in campo socio-educativo concretizzato attraverso realizzazioni pratiche. Tutte operarono in una società in via di lento, ma costante mutamento nelle mode e nei costumi.Bolognese women were leading actors in their living rooms, clubs, theaters, on the work of political and cultural debate that has prepared and implemented the Italian unification.     In recent years, techniques involving the use oforganisms to remove or neutralize pollutants from contaminatedsites have attracted great attention. The aim of bioremediationis to use naturally occurring organisms to degradedangerous substances to less toxic or non toxic molecules.The gram-negative bacterium Pandoraea pnomenusa strainB-356 (Pp) has been found to be able to transform a persistentclass of organic pollutant compounds, namely the biphenyland polychlorinated biphenyls (PCBs). A key enzyme in thePCB catabolic pathway is NAD-dependent cis-2,3-dihydrobiphenyl-2,3-diol dehydrogenase (BphB), for whichthe crystal structure from Pp has been crystallized in apo-,NAD-bound and biphenyldiol-/NAD-bound forms. The substratebinding loop structure has not been completely resolvedto date in the former two bound states. Here we report theresults of the first extensive molecular dynamics simulationson the three different states of PpBphB. This allowed an indepth characterization of the mechanism of ligand uptake andbinding, including unraveling of the gating mechanism. Oursimulations give a deep insight into several dynamic featuresof the enzyme that were not captured by crystal structure

    Evolution of Macromolecular Docking Techniques: The Case Study of Nickel and Iron Metabolism in Pathogenic Bacteria

    The interaction between macromolecules is a fundamental aspect of most biological processes. The computational techniques used to study protein-protein and protein-nucleic acid interactions have evolved in the last few years because of the development of new algorithms that allow the a priori incorporation, in the docking process, of experimentally derived information, together with the possibility of accounting for the flexibility of the interacting molecules. Here we review the results and the evolution of the techniques used to study the interaction between metallo-proteins and DNA operators, all involved in the nickel and iron metabolism of pathogenic bacteria, focusing in particular on Helicobacter pylori (Hp). In the first part of the article we discuss the methods used to calculate the structure of complexes of proteins involved in the activation of the nickel-dependent enzyme urease. In the second part of the article, we concentrate on two applications of protein-DNA docking conducted on the transcription factors HpFur (ferric uptake regulator) and HpNikR (nickel regulator). In both cases we discuss the technical expedients used to take into account the conformational variability of the multi-domain proteins involved in the calculations
