Therapist\u27s experience of psychotherapy relationship through the lens of attachment theory
V pričujočem delu se avtorica loteva raziskovanja terapevtovega doživljanja terapevtskega odnosa in sicer skozi prizmo navezanosti. Teoretični uvod tako predstavlja podrobno predstavitev teorije navezanosti od Bowlbyja naprej. Avtorica pregleda zgodovino koncepta kot tudi njegov razvoj in etiologijo. Drugi del teoretičnega uvoda je sestavljen iz predstavitve koncepta terapevtskega delovnega odnosa in njegove implikacije za terapevtsko delo. Avtorica se znova poglobi v razumevanje koncepta in skuša slediti njegovemu razvoju. Delo nato preide na raziskovalni del.
Avtorico je zanimalo, kako terapevti doživljajo terapevtski odnos skozi lečo navezanosti. Za ta namen je terapevtski odnos razdelila na tri faze – fazo prednavezanosti, ki jo zaznamuje vzpostavljanje kontakta in učenje pravil terapevtskega dela, sledi ji faza oblikovanja navezanosti, kjer je dinamika že utečena, klienti pa opažajo prve spremembe v svojem življenju, in zadnja faza, torej faza navezanosti, ki jo oblikuje visoka stopnja zaupanja, jasna komunikacija med klientom in terapevtom in dobra delovna aliansa. Avtorica je za namen naloge intervjuvala osem terapevtov in podatke, pridobljene v teh intervjujih analizirala s pomočjo kvalitativne metode fenomenološke raziskave. Ugotovitve so pokazale, da se doživljanje terapevtov v veliki meri sklada z do sedaj aktualno teorijo. V fazi prednavezanosti so terapevti poročali o tem, da se odnos šele oblikuje, zaupanje vzpostavlja, komunikacija še usklajuje. V fazi oblikovanja navezanosti terapevti poročajo o naraščajočem zaupanju in opažanju sprememb in v zadnji fazi poročajo o močni navezanosti in z rastjo odnosa vedno bolj enakovreden odnos med terapevtom in klientom. To je tudi obdobje, ko klient in terapevt načrtujeta zaključek.
Raziskovalno delo je avtorici ponudilo vpogled v globoko doživljanje terapevtskih odnosov s strani terapevta, spoznala je izzive, s katerimi se terapevti srečujejo pri svojem delu, pa tudi dobrobiti terapevtskega dela. V svojem delu je avtorica izpostavila tudi pomanjkanje informacij s področja raziskovanja terapevtske perspektive.In the current master thesis, the author is exploring a therapists’ perception of psychotherapy process through the lens of attachment. The theoretical part consists of precise description of ontogeny of the attachment theory from his beginner, John Bowlby. The author follows the history and etiology of the mentioned concept. The other part of the master thesis consists of presentation of the working alliance concept and its indication for therapeutic work. Author tries to get an overall understanding about the concept, its origins and current implications.
In the current master thesis, the author was interested in therapists’ perception of psychotherapy relationship through the attachment perspective. To enquire about it she divided the attachment process on three main stages – the preattachment stage, characterized by first contact and establishment of communication, second stage, namely attachment in the making stage, where the trust and therapy framework is already established and clients can see first changes, and last, third phase that is the attachment phase, where therapist and clients have high level of trust, good communication and good working alliance. Author interviewed eight therapists and analyzed the information from the interviews via phenomenological research.
Results have shown that the way therapists from the research see their relationships folds well with the current theory from the field. In the preattachment stage the therapist spoke about the way the relationship has yet to be built, the trust has yet to be gained and communication is still on shaky grounds. In the second, attachment in the making phase, therapists observe deeper trust, clients start seeing changes in their lives and in the last, attachment phase they describe the relationship as of one of strong attachment. With continuation of the relationship the relationship itself become more of an equal one and is, as such, slowly transcending into the conclusion phase.
The research work enabled the author to see the depth of the therapists’ experiencing of the relationships they make with their clients, she get to know the challenges of the therapeutic relationships and also the benefits of it. As a conclusion, the author exposes the lack of information available on research done of exploring the therapists’ perspective
A secure and efficient inventory management system for disasters
Over the last three decades, disasters worldwide claimed more than 3 million lives and adversely affected the lives of at least 1 billion people (Noji, 1997). Regarding the threats posed by these disasters, emergency disaster management has emerged as a vital tool to reduce the harm and alleviate the suffering these disasters can cause to their victims. A significant task of planners involved in emergency disaster management is planning for and satisfying the vital needs of the people located in emergency shelters such as the Superdome in New Orleans. This thesis proposes a novel and comprehensive framework for the development of a humanitarian emergency inventory management system based on the real-time tracking of emergency supplies and demands through the integration of emerging technologies such as Radio Frequency Identification Devices (RFID) for commodity tracking and logistics. The novelty of this thesis is that, for the first time in the emergency inventory management field, the proposed approach combines an offline planning strategy with online control techniques in a unified framework. Within this framework, the offline planning problem is solved by the stochastic humanitarian inventory management approach, whereas the online modeling strategies include the application of neural network-based functional approximation, simultaneous perturbation stochastic approximation (SPSA), and continuous time model predictive control (CMPC) techniques. Unlike previous studies, the flexibility of the proposed inventory management and control model allows the application of the developed mathematical model to extreme events making online real-time tracking possible. Realistic case studies built using information available from past disasters are used to examine the differences in inventory strategies for different types of disasters based on the impact area and duration of the extreme event. The proposed methodology is also capable of representing and understanding real-life cases where uncertainty and limitations on the inventory levels and flow of supplies can be modeled by introducing different levels of stochasticity and real-life constraints. The overall findings of this thesis have pointed out that the proposed integrated framework can be efficiently used for emergency inventory planning and inventory control during disaster relief operations without ignoring the real-world uncertainties, fluctuations, and constraints of disaster conditions.Ph. Skrining Pertumbuhan Jamur Endofit dari Tumbuhan Zingiberaceae pada Medium Mengandung Pestisida dan Kemampuan Menghasilkan Biosurfaktan
Penelitian tentang skrining pertumbuhan jamur endofit dari tumbuhan Zingiberaceae pada medium mengandung pestisida dan kemampuan menghasilkan biosurfaktan telah dilakukan. Dari 31 isolat jamur endofit asal tumbuhan Zingiberaceae yang telah diujikan dan didapatkan semua isolat jamur endofit mampu tumbuh pada medium Bushnell- Haas Agar (BHA) yang mengandung pestisida curacron berbahan aktif profenofos dan pestisida berbahan aktif matador lamda sihalotrin. Isolat JRE 1A asal endofit tumbuhan Hedychium coronarium merupakan isolat yang memiliki pertumbuhan lebih baik. Indeks emulsifikasi (%IE24) tertinggi pada medium 0,25% matador sebesar 35,45%. Oil Spreading tertinggi pada medium 0,25% matador dengan diameter zona bening sebesar 40,4 mm. Konsentrasi biosurfaktan yang dihasilkan tertinggi pada hari ke-10 sebesar 181,857 ppm pada medium 0,5% matador.Screening the growth of endophytic fungi from Zingiberaceae on a medium containing pesticides and the ability to produce biosurfactants was conducted. A total of 31 isolates of endophytic fungi originated from the Zingiberaceae was tested and all of the fungal isolates were able to grow on Bushnell-Haas Agar (BHA) medium containing curacron pesticide with active ingredient profenofos and pesticide with active ingredient matador lamda cyhalothrin. JRE 1A isolate from endophytic Hedychium coronarium was the isolate that had the best growth. The highest emulsification index test (%IE24) was at 0.25% matador medium at 35.45%. The highest Oil Spreading test was on 0.25% matador medium with a clear zone diameter of 40.4 mm. Keragaman dan Potensi Bakteri Pendegradasi Plastik yang Diisolasi dari Sampah Plastik Tempat Pembuangan Akhir (TPA) Terjun Medan Marelan
The difficulty of degrading plastic waste in nature causes plastic waste to
accumulate and become a problem in the environment, especially in landfill sites. This
study aims to determine the diversity and potential of plastic degrading bacteria
isolated from Terjun landfill Medan Marelan. Bacteria were isolated from four types
of plastic waste namely Low Density Polyethylene (LDPE), Low Linear Density
Polyethylene (LLDPE), High Density Polyethylene (HDPE), Polypropylene (PP) from
Terjun Medan Marelan landfill. Bacteria were screened for their ability to grow on
minimal medium containing 0.5% plastic powder. The ability of bacteria to degrade
LDPE and LLDPE on agar media was determined based on plastic weight reduction
analysis, Scanning Electron Microscope (SEM) and Fourier Transform Infra Red
(FTIR) observations. Isolation results obtained as many as 24 isolates with different
morphologies. Total of 5 isolates is L6, LL4, LL6, and H5 were able to grow on media
containing plastic powder. The results of molecular identification of known type of
bacteria in isolates L6, LL4, and LL6 Bacillus paramycoides, while in isolate H5
Sphingobacterium changzhouense. The ability of five types of bacteria in degrading
LDPE and LLDPE sheets, it is known that all bacteria can reduce the weight of plastic
sheets in incubation time for 30 days by 8,4% on LDPE and 5,0% on LLDPE.

plastic sheets using SEM found the attachment of bacterial colonies and changes in
the surface of plastic sheets such as holes and cracks. However, FTIR data did not

Isolation, Partial Purification and Characterization of Urease Enzyme from Red Rice Beans (Vigna Umbellata)
Urease (EC 3.5.1.5) adalah enzim yang menghidrolisis urea menjadi amonia dan karbon dioksida. Enzim urease memiliki banyak manfaat seperti pengolahan limbah industri, hemodialisis, diagnosis medis dan pengukuran kandungan urea dalam darah manusia. Kacang beras merah (Vigna umbellata) merupakan salah satu tanaman kacang-kacangan dari famili Fabaceae yang selama ini tidak dimanfaatkan secara optimal. Umumnya, kacang beras merah hanya dimanfaatkan sebagai makanan pelengkap bagi manusia dan pakan ternak. Tanpa disadari kacang beras merah mengandung aktivitas enzim urease yang cukup tinggi. Penelitian ini bertujuan untuk isolasi, pemurnian dan karakterisasi enzim urease dari biji kacang beras merah (Vigna umbellata). Tahapan isolasi dilakukan secara terpisah, yaitu dari kacang beras merah dengan kulit biji dan tanpa kulit biji. Aktivitas urease dari biji kacang beras merah tanpa kulit biji memiliki aktivitas enzim yang lebih tinggi dibandingkan dengan kulit biji. Kemudian ammonia yang dihasilkan juga lebih tinggi pada urease tanpa kulit biji (13 μg ammonium) dari pada urease dengan kulit biji (8,8 μg ammonium). Konsentrasi urease dari biji kacang beras merah adalah 7,238 mg/mL. Urease dari biji kacang beras merah tersusun atas empat polipeptida dengan berat molekul yaitu 52, 33, 22 dan 10 kDa. Selanjutnya diperoleh bahwa biji kacang beras merah tidak memiliki aktivitas antijamur terhadap strain jamur seperti Aspergillus niger, Aspergillus fumigatus, Microsporum canis, Fusarium oxysporum subsp. lini, Trichophyton rubrum, Candida glabrata dan Candida albicans.45 HalamanSkripsi Sarjan
Isolasi dan Skrining Bakteri Selulolitik dari Usus Belalang (Oxya Chinensis) dan Potensi Selulolitiknya
Grasshoppers are herbivorous insects or plant eaters which in large
quantities can cause losses to farmers. Many studies have been conducted on
cellulolytic microbes from herbivorous insects. This study aims to obtain bacterial
isolates from the guts of grasshoppers (Oxya chinensis), to determine the diversity
of cellulolytic bacteria and to determine their potential to produce cellulase
enzymes. There are were obtained 27 isolates from 3 parts of the grasshopper
intestine (foregut, midgut, hindgut). Cellulolytic bacteria are characterized by the
formation of clear zones on CMC agar media after being flooded with 0.1% Congo
red. All bacterial isolates have the ability to hydrolyze cellulose with a cellulolytic
index ranges from 0,12 to 1,23. Based on the molecular identification of potential
bacterial isolates indicated 5 different species, namely from the foregut section
found Bacillus wiedmanniii (OCF2), midgut section found Bacillus
marcorestinctum (OCM4), Bacillus halotolerans (OCM8) and hindgut section
found Paenibacillus zanthoxyli (OCH4), Bacillus hominis (OCH7). Bacillus
wiedmanniii (OCF2) had the highest cellulase enzyme activity of 0,5830 U/mL and

Keragaman Jamur pada Plastik Tempat Pembuangan Akhir (TPA) Terjun Medan Marelan dan Potensinya dalam Mendegradasi Sampah Plastik
Plastic waste, including Low-Density Polyethylene (LDPE), Linear-Low Density Polyethylene, High-Density Polyethylene (HDPE), and Polypropylene (PP), is predominantly found in landfill sites. This study aimed to investigate the diversity of fungi associated with plastic waste and their potential in plastic degradation. Plastic waste samples were collected from Terjun Landfill in Medan Marelan. Fungi were isolated directly from the plastic waste on Potato Dextrose Agar (PDA) medium and subsequently characterized using macroscopic and microscopic observations. Screening for plastic degradation potential was conducted by cultivating fungi on Mineral Salt Medium Agar (MSMA) supplemented with 0.5% (w/v) of plastic powder. Potential fungal isolates were molecularly identified using ITS-1F and ITS-4R primers. The plastic degradation potential was assessed by cultivating fungi on MSMA medium with 1x1 cm LDPE and LLDPE plastic sheets and in Mineral Salt Medium Broth (MSMB) containing plastic powder, both media supplemented with 0.5% glucose. The isolation yielded 24 different fungal isolates, and five isolates showed potential for plastic degradation based on their growth on MSMA medium: Fusarium solani (LDPE5), Botryosphaeria laricina (LLDPE10), Aspergillus fumigatus (HDPE1), Aspergillus flavus (HDPE3), and Aspergillus niger (PP5). Degradation tests on plastic sheets over 45 days revealed that Fusarium solani and Botryosphaeria laricina exhibited the highest degradation abilities, with degradation percentages of 20.83% and 6.49%, respectively. Scanning Electron Microscopy (SEM) images showed changes in the surface morphology of the plastic sheets, becoming rough and wrinkled. Skrining Aktivitas Kitinase Jamur Endofit Tumbuhan Zingiberaceae dan Kemampuannya Menghambat Fusarium oxysporum dan Ganoderma boninense
Endophytic fungi is a fungal community inhabiting a plant tissue without exhibiting any negative effect to its host. The study of chitinase activity from Zingberaceae endophytic fungi and their abilities in inhibiting Fusarium oxysporum and Ganoderma boninense has been conducted. Thirty endophytic fungal isolates were tested for its growth on chitin agar medium and two isolates namely JRE 1A and JRE 4B had chitinase activity. Chitinase activity of the both isolates were 4.15 and 4.76 U/mL, respectively. The optimum activity of chitinase produced by both isolates were detected at pH 6 to 7. The activity crude enzyme was increased by Mn2+ ion, while being inhibited by the presence of K+ and Ca2+ ions. The isolates JRE 1A and JRE 4B were able to inhibit Fusarium oxysporum and Ganoderma boninense on chitin agar. Inhibitory activity of JRE 1A and JRE 4B were higher against Fusarium oxysporum. Crude chitinase of both isolates precipitated with 70% ammonium sulphate also exhibited antifungal activities against Fusarium oxysporum and Ganoderma boninense.Jamur endofit adalah jamur yang tumbuh dalam jaringan tumbuhan tanpa menimbulkan efek negatif pada tanaman inangnya. Penelitian tentang aktivitas kitinase dari jamur endofit tumbuhan Zingiberaceae serta kemampuannya dalam menghambat Fusarium oxysporum dan Ganoderma boninense telah dilakukan. Sebanyak tiga puluh satu isolat jamur endofit telah diujikan pada medium kitin agar dan dua isolat yaitu JRE 1A dan JRE 4B memiliki aktivitas kitinase. Aktivitas kitinase kedua isolat secara berturut yaitu 4,15 U/mL dan 4,76 U/mL. Kitinase yang dihasilkan dari kedua isolat jamur bekerja optimum pada rentang pH antara 6 hingga 7. Aktivitas ekstrak kasar enzim dari kedua isolat ditingkatkan oleh ion logam Mn2+, namun dihambat oleh ion logam K+ dan Ca2+. Kedua isolat JRE 1A dan JRE 4B mampu menghambat pertumbuhan Fusarium oxysporum dan Ganoderma boninense pada medium kitin agar. Aktivitas penghambatan JRE 1A dan JRE 4B lebih tinggi terhadap Fusarium oxysporum. Potential of Terminal Area Soil Bacteria in Degrading used Oil
Environmental pollution caused by used oil is a serious problem due to its toxic
hydrocarbon content, which is difficult to break down naturally. This study aims to
examine the potential of bacteria isolated from the Amplas Terminal area to degrade
used oil. Samples were isolated from three locations contaminated with oil spills and
selected based on their ability to grow on mineral media with the addition of used oil
as a carbon source. Molecular identification of selected isolates was performed by 16S
rRNA gene analysis to determine the type of bacteria. The degradation ability of
selected bacteria was measured based on the reduction in oil residue weight and
bacterial growth. A total of 23 isolates with different morphologies were successfully
isolated, and four isolates (A3, A4, B8, and C4) showed the ability to grow on media
enriched with oil. Isolate A4 showed the highest used oil degradation ability (57.06%).
Molecular identification results showed that isolate A4 belonged to the species
Acinetobacter baumannii. The ability to degrade used oil components was determined
through Gas Chromatography-Mass Spectrometry (GC-MS) analysis. GC-MS analysis
clarified that most (>90%) of the components of used oil were degraded after 30 days
of treatment. The results of this study explain that bacteria isolated from contaminated
areas can be used as potential agents for the bioremediation of used oil contamination.54 PagesSkripsi Sarjan
