    The interaction of electric fields with vesicles and cells: a study on electrodeformation and electroporation

    The application of electric pulses to cells or vesicles induces complex responses. When electric field shocks are applied, a lipid membrane may become porated, which is a phenomenon known as electroporation. The lipid membrane may also deform under electromechanical and electrohydrodynamic forces, applied via electric fields,which is a phenomenon known as electrodeformation. Electroporation is widely employed in both biological research and clinical applications including areas such as drug and gene delivery, protein insertion, cancer therapy, and other processes where access to the cytoplasm is desired. Electrodeformation, on the other hand, can be harnessed as a means to probe the membrane's properties, and to detect pathological changes in cells. Despite its broad applicability, electroporation still suffers from low delivery efficiency and cell viability, in part due to a lack of a fundamental understanding of the mechanisms involved in this technique. Meanwhile, the electrodeformation phenomenon only received attention in the past decade, and therefore, a significant body of data only became available in the recent years. This is mainly due to the development of high-performance optical imaging systems which gave the opportunity to capture the dynamics associated with electrodeformation. In this work, we have designed and implemented experiments to study the electrodeformation of giant unilamellar vesicles, and to investigate the complex transport mechanisms involved in electroporation-mediated molecular delivery. In addition, we quantified the molecular uptake and cellular viability in electroporation experiments. First, we characterized the prolate deformations of giant unilamellar vesicles under strong DC electric fields. The vesicles exhibited prolate elongations along the direction of the electric field. In some cases, the aspect ratio of a deformed vesicle exceeded 10, representing a novel strong-deformation regime never before explored. Second, we studied the spatial and temporal transport of molecules via fluorescence microscopy, at the single cell level. The experimental results demonstrated that electrophoresis, not diffusion, is the dominant mode of of delivery during the pulse. Furthermore, we found that an electrokinetic mechanism known as field-amplified sample stacking (FASS) mediated an inverse correlation between delivery and the extracellular electrical conductivity. Following our accomplishment in the second task, we investigated a two-stage pulsing electroporation protocol that delivers separate pulses to porate the membrane and to electrokinetically-mediate species transport across the cell membrane. We found that while both delivery and viability are linearly dependent with respect to the duration of the second pulses, they both show a strong dependence on field strength. Moreover, a critical regime for maximized delivery and viability is achieved. This dissertation contributes to the electroporation and electrodeformation fields by generating explanations to several experimental observations based on physical mechanisms which were not previously available. In addition, the experimental tools for quantifying delivery and viability may help develop and improve the efficacy of electroporation protocols.Ph.     Novel Degree-Based Topological Descriptors of Fenofibrate Using M-Polynomial

    Chemical graph theory is currently expanding the use of topological indices to numerically encode chemical structure. The prediction of the characteristics provided by the chemical structure of the molecule is a key feature of these topological indices. The concepts from graph theory are presented in a brief discussion of one of its many applications to chemistry, namely, the use of topological indices in quantitative structure-activity relationship (QSAR) studies and quantitative structure-property relationship (QSPR) studies. This study uses the M-polynomial approach, a newly discovered technique, to determine the topological indices of the medication fenofibrate. With the use of degree-based topological indices, we additionally construct a few novel degree based topological descriptors of fenofibrate structure using M-polynomial. When using M-polynomials in place of degree-based indices, the computation of the topological indices can be completed relatively quickly. The topological indices are also plotted. Using M-polynomial, we compute novel formulas for the modified first Zagreb index, modified second Zagreb index, first and second hyper Zagreb indices, SK index, SK1 index, SK2 index, modified Albertson index, redefined first Zagreb index, and degree-based topological indices

    From Rabi'a to Ibn al-Farid : towards some paradigms of the Sufi conception of love

    This thesis aims to investigate the significance of Divine Love in the Islamic tradition with reference to Sufis who used the medium of Arabic to communicate their ideas. Divine Love means the mutual love between God and man. It is commonly accepted that the Sufis were the forerunners in writing about Divine Love. However, there is a relative paucity of literature regarding the details of their conceptions of Love. Therefore, this attempt can be considered as one of the first of its kind in this field. The first chapter will attempt to define the nature of love from various perspectives, such as, psychology, Islamic philosophy and theology. The roots of Divine Love in relation to human love will be explored in the context of the ideas that were prevalent amongst the Sufi authors regarded as authorities; for example, al-Qushayri, al-Hujwiri and al-Kalabadhi. The second chapter investigates the origins Of Sufism with a view to establishing the role that Divine Love played in this. The etymological derivations of the term Sufi will be referred to as well as some early Sufi writings. It is an undeniable fact that the Qur'an and tladith are the bedrocks of the Islamic religion, and all Muslims seek to justify their ideas with reference to them. This was especially true for the Sufis. The third and fourth chapters will, therefore, focus on the concept of Divine Love in the textual sources of Islam in order to determine the role that these played in the development of the Sufi conceptions of love in general. Having highlighted the origins and general context of Divine Love, the following five chapters will focus on selected Sufis whose contributions can be regarded as significant, original and representative of the Sufi tradition. The exclusive characteristics of each Sufi's concept of love will be analysed and an attempt will be made to present them as a paradigm of Sufi love. The paradigms of love of the following Sufis will be presented: Rabi`a, al-I-Jallaj, al-Ghazäli, Ibn 'Arabi and Ibn al-F5ricl. The concluding chapter will provide a synthesis of these paradigms of love by putting forward a framework which identifies the key stages in the development of the Sufi paradigm of love. Regarding the scope and limitations of this study, it should not be seen as a historical or biographical research. The historical analysis and information are presented merely to give some insight into the social and historical context of the Sufi's time. The primary aim of this research is to establish the different paradigms of love of the Sufis referred to above, by analysing the exclusive motives specific to each paradigm. Finally, this research is by no means a comprehensive account of the Sufi paradigms of love. The development of the thesis showed that this research can be approached from many different angles. For example, the concept of love is closely associated with the concept of ma`rifia, and the concept of God. The relationship of the two with the concept of love is a possible area of further research. In addition, the historical information available on each individual Sufi provides ample material for a detailed study of the concept of love in that particular Sufis paradigm. This work provides a general framework for further studies

Penelitian ini bertujuan untuk (1) menganalisis networking aktor pemerintah, pihak swasta, dan masyarakat; (2) menganalisis kapasitas kontrol aktor pemerintah; (3) menganalisis penyatuan sumber daya aktor pemerintah dan pihak swasta; dan (4) merumuskan model governance berbasis elektronik dalam pelayanan pertanahan di Kota Makassar. Unit analisis adalah (1) Kantor Pertanahan Kota Makassar (aktor pemerintah); (2) Pejabat Pembuat Akta Tanah (pihak swasta); dan (3) masyarakat Kota Makassar. Penelitian ini menggunakan pendekatan kualitatif, jenis fenomenologis, dan analisis model interaktif Miles dan Huberman. Hasil penelitian menunjukkan bahwa networking antar aktor pada Kantor Pertanahan dan Pejabat Pembuat Akta Tanah sebagai mitra kerja bersifat intermittent coordination; dan networking Kantor Pertanahan denqan masyarakat bersifat sociotechnical. Kapasitas kontrol aktor pemerintah telah diserahkan sebagian kepada Pejabat Pembuat Akta Tanah, khususnya kewenangan pembuatan akta autentik. Starting from the Sawerigading twin brothers who wanted to have transportation at sea, and the manufacture of the phinisi boat started from the wreck of the Sawerigading boat which was stranded in three areas, namely Ara, Lemo-lemo and Bira. in the Land of Beru.
