    Latar Belakang: Sungkai (Peronema canescens Jack) sering disebut sebagai jati sabrang, ki sabrang, kurus sungkai, atau sekai, termasuk ke dalam famili Verbenaceae. Berdasarkan penelitian sebelumnya dilaporkan beberapa kandungan senyawa metabolit sekunder pada daun Sungkai (Peronema canescens Jack) yaitu flavonoid, alkaloid, saponin, tanin, steroid dan fenolik Senyawa flavonoid, saponin, alkaloid dan fenol memiliki aktivitas antiinflamasi. Dimana kandungan metabolit sekunder, tanin dan flavonoid tersebut memiliki aktivitas sebagai antioksidan sehingga dapat mencegah kerusakan akibat stres oksidatif. Daun sungkai juga mengandung mineral zinkum. Tujuan: Penelitian ini bertujuan untuk menentukan kadar kandungan zinkum pada daun sungkai segar (Peronema canescens Jack). Metode: Sampel daun sungkai didestruksi kering, kemudian dilakukan analisis kuantitatif zinkum dilakukan dengan menggunakan metode spektrofotometri serapan atom (AAS) yaitu pada panjang gelombang 213,86 nm. Keuntungan dari metode ini adalah dapat menentukan kadar mineral tanpa dipengaruhi oleh keberadaan mineral yang lain dan mineral dalam jumlah kecil.     Background: Jackfruit (Artocarpus heterophyllus), a plant belonging to the Moraceae family, is one of the fruits that has good benefits for humans. Jackfruit leaf parts can be used as natural antioxidants because they contain secondary metabolites. DPPH (1,1-diphenyl-2-picrylhydrazyl) is often used to assess the antioxidant activity of several extracts or natural ingredients so that it can evaluate the antioxidant potential of reducing free radicals. Objective: This study aims to determine the antioxidant activity of an ethanol extract of jackfruit (Artocarus heterophyllus) leaves using the DPPH method. Method: Testing of the antioxidant activity of the ethanol extract of jackfruit leaves was carried out using the DPPH method. Results: Jackfruit leaf ethanol extract has an IC50 antioxidant activity of 57.1552 with a regression equation of Y = 0.7466x + 7.3279 and a correlation coefficient of R2 = 0,9779.     Background: Antioxidants are compounds the body needs to stabilize and prevent damage from free radicals. Picria fel-terrae Lour. is a plant that has antioxidant activity to prevent degenerative diseases. Objective: Determine the secondary metabolites, total phenolic and flavonoid contens, and antioxidant activity of the ethanol extract of Picria fel-terrae Lour. Herbs using the CUPRAC and FRAP methods. Method: Examination of simplicia characteristics includes macroscopic, microscopic, water content, water soluble extract content, ethanol soluble extract content, ash content, and acid insoluble ash content. Phytochemical screening includes examination of alkaloids, flavonoids, glycosides, tannins, saponins, and steroids/triterpenoids by thin layer chromatography. Determination of total phenolic content by Folin-Ciocalteu reagent and total flavonoids by AlCl3 reagent. The extract was tested for antioxidant activity using the CUPRAC (Cupric Ion Reducing Antioxidant Capacity) method and the FRAP (Ferric Reducing Antioxidant Power) method with UV-Vis spectrophotometry at a wavelength of 450 nm and 700 nm. Result: The simplicia characterization results were 7.99% water content and 9.99% extract, 22.04% water soluble simplicia, 14.93% ethanol soluble simplicia, 5.43% total ash content and 2.14% extract, insoluble ash content acid 1.32% and extract 0.41%. The results of the phytochemical screening by thin layer chromatography positively contained alkaloids, saponins, flavonoids, tannins, glycosides, and steroids/triterpenoids. The results for determining the total phenolic content were 50.8944 mg GAE/g and the total flavonoids were 19.8007 mg QE/g. The results of antioxidant activity with the CUPRAC method were very strong with an IC50 value of 31.9463 μg/ml while the results of the FRAP method were in a strong category, namely an IC50 value of 62.5253 μg/ml.     Background: One of the plants containing flavonoids is the poguntano plant (Picria fel-terrae Lour.). P. fel-terrae Lour extract. efficacious as diuretic, antipyretic, hepatoprotective, cardioprotective, antidiabetic, antioxidant (due to high content of flavonoids and phenols), anti-inflammatory, anthelmintic activity, and analgesic. Quercetin is a flavonol found in fruits and vegetables which is a food component that has been shown to be beneficial to health. Quercetin is one of the most powerful antioxidants among polyphenols. One of the quercetin derivatives that has various pharmacological activities is rutin. Rutin is a basic ingredient of drugs belonging to the class of flavonoid compounds which are very widely used in the pharmaceutical field. Objective: The aim of this research is to identify and determine the content of poguntano (Picria fel-terrae Lour.) ethanol extract using the UPLC method. Method: This research includes making poguntano ethanol extract using the maceration method and characterization of simplicia and extracts which includes macroscopic, microscopic examination, determination of water content, water soluble essence content, ethanol soluble essence content, total ash content, acid insoluble ash content, phytochemical screening , qualitative tests, measurements with UPLC and method validation. Results: The yield of Poguntano ethanol extract was obtained at a yield of 11% with water content, water soluble essence content, ethanol soluble essence content, total ash content, simplicia acid insoluble ash content respectively equal to 7.99%; 22.05%; 14.93%; 5.43%; 1.32% and water content, total ash content, extract acid insoluble ash content respectively 9.32%; 2.14%; 0.41%. The results of identification using UPLC contained the flavonoid quercetin with a retention time of 3.359 using a mobile phase of acetonitrile: 0.1% formic acid with a ratio of 60:40, flow rate of 5 μL/minute and wavelength of 254 nm. The weight of the quercetin extract compound detected was 16.621195 mg/g extract. Validation test results obtained % recovery 99.26548; Standard Deviation 1.91536; LOD 9.82913; and LOQ 32.76376 Conclusion: The results of this study indicate that the ethanol extract of poguntano (Picria fel-terrae Lour.) is thought to contain flavonoid compounds in the flavonol group, namely quercetin, which is indicated by a retention time of 3.359 and a weight of the quercetin extract compound of 16.621195 mg/g extract.121 PagesSkripsi Sarjan

    Background: Kluwih (Artocarpus camansi (Parkinson) Fosberg) from the Moraceae tribe is one of the plants that grows in Indonesia and can be used in traditional medicine. Kluwih leaves contain phytochemical compounds such as alkaloids, tannins, steroids, flavonoids, triterpenoids, glycosides, anthraquinones, and GABA (Gamma Amino Butyl Acid). The flavonoid compound content is known to have antioxidant, anti-inflammatory, and antiviral activity. Antioxidant activity testing can be done using the DPPH method. DPPH (1,1 diphenyl-2-picryhydrazyl) is a DPPH radical absorption method that is simple, easy, and uses a small sample in a short time. In the DPPH test, the parameter used to measure antioxidant activity is the IC50. Objective: This study aims to determine the antioxidant activity of the ethanol extract of kluwih leaves (Artocarpus camansi) using the DPPH method. Method: Testing of the antioxidant activity of the ethanol extract of kluwih leaves was carried out using the DPPH (1,1 diphenyl-2-picryhydrazyl) method using a UV-Vis spectrophotometer. Results: Kluwih leaf ethanol extract has an IC50 antioxidant activity of 19.6982 μg/ml with a regression equation of y = 1.3176x + 24.0456 and a correlation coefficient of r = 0.8757.     Background: Silver nanoparticles of coffee fruit skin extract (coffea canephora) are formulated into a good gel preparation because the gel can well deliver silver nanoparticles to the skin. Objective: To know the formulation and physical evaluation of Nanoparticle Gel preparation. Method: Distilled water, HPMC, methylparaben, silver nanoparticles, propylparaben, propylene glycol. Tests on gel preparations include organoleptic testing, viscosity, spreadability, adhesion, and pH of the preparation. Results: From the results of the research that has been carried out, it is obtained that the silver nanoparticle gel preparation is brownish yellow due to the addition of extracts, does not have a distinctive odor, has a qualified spreadability of 5-7 cm, has a qualified adhesion> 4 seconds, has a viscosity value of 2900.835-4700.588, has a pH value of 6.8-7.25.     BACKGROUND: Poguntano herb extract has good efficacy for health, poguntano extract has diuretic effect, antipyretic, hepatoprotective, cardioprotective activity, antidiabetic, etc. Apigenin is compounds of the flavone group of flavonoids. which is a compound of the polyphenol group that is widely distributed in plants. UPLC method is used to check the levels of apigenin in poguntano herb extract. OBJECTIVE: The purpose of this study was to examine and determine the levels of apigenin in the ethanol extract of Herba poguntano with UPLC method and to determine the UPLC method and Conditions used to meet the validation requirements. METHOD: Quantitative analysis using UPLC method used acetonitrile mobile phase:formic acid 0.1% (35: 65) with flow rate 0.44 mL / min, temperature 26 SICHERAHC, wavelength 254 nm, injection volume 10 sicherahl, and column C18 (octadecyl silica). RESULTS: The results of the study with UPLC conditions provide good results, where the value of tailings factor 1 and theoretical plate 20248. On the chromatogram results of herb extract poguntano showed that there is apigenin compounds with levels of 5.95897. Then from the calibration obtained regression equation for apigenin obtained results are Y= 61.162 x+1327.2 with a correlation coefficient of 0.998.The results of validation parameters obtained are 99.2059% for % recovery, and 1,7529% for Relative Standard Deviation (RSD), and 0,6790 mg/g extract for LOD, 2,0578 mg/g extract for LOQ from apigenin raw. CONCLUSION: Levels of apigenin compounds in the ethanol extract of the herb poguntano obtained is 5.95897 mg / g extract.     Background: Sungkai (Peronema canescens Jack) is one of the plant that is used as medicine, especially of its leaves because it is proven containing secondary metabolite compounds including the alkaloid group, flavonoids, tannins, saponins, glycosides, and steroids/triterpenoids that have antibacterial activity. Based on previous research, it is known that ethanol extract of sungkai leaf may inhibit the growth of some bacteria. Staphylococcus epidermidis and Propionibacterium acnes are gram-positive bacteria that is known to be able causing skin infections, one of which is acne. Purpose: This study aims to determine the secondary metabolites found in simplicia and ethanol extract of Sungkai leaves and to test the antibacterial activity of the ethanol extract of Sungkai leaves against Staphylococcus epidermidis and Propionibacterium acnes bacteria. Methods: This research was conducted using an experimental method started by extraction using two methods, namely the reflux method, and then the phytochemical screening test, characterization, and testing the antibacterial activity of the ethanol extract of sungkai leaves against Staphylococcus epidermidis and Propionibacterium acnes with the agar diffusion method using paper discs. In this test, various concentrations of ethanol extract of Sungkai leaves were used, clindamycin as positive control, aquades as negative control, and DMSO solution as solvent control. Results: The simplicia characterization result obtained 9.992% water content, 10.286% water-soluble extract content, 14.505% ethanol-soluble extract content, 3.268% total ash content, and 0.274% acid-insoluble ash content. The chemical compounds contained in simplicia and ethanol extract of Sungkai leaves are alkaloids, flavonoids, glycosides, saponins, tannins, steroids/triterpenoids. Sungkai leaf ethanol extract has antibacterial activity against Staphylococcus epidermidis and Propionibacterium acnes with Minimum Inhibitory Concentration (MIC) for both bacteria at the concentration of 0.7812 mg/ml with an inhibition zone diameter of 6.93 mm for Propionibacterium acnes and 6.10 mm for Staphylococcus epidermidis. The effective concentration of ethanol extract of sungkai leaves on Propionibacterium acnes and Staphylococcus epidermidis was at a concentration of 100 mg/ml with an inhibition zone diameter of 15.2 mm for Propionibacterium acnes and 14.13 mm for Staphylococcus epidermidis.     Background: Poguntano herb (Picria fel-terrae Lour.) is one of the species that often used by people in Berastagi, Karo, North Sumatra as medicine for various diseases. Poguntano herb contains of alkaloids, flavonoids, glycosides, saponins, steroids/triterpenoids, and tannins which have the potential as antibacterial. Objective: The purpose of this research was to determine the antibacterial activity of ethanol extract of poguntano herb (Picria fel-terrae Lour.) against Staphylococcus epidermidis and Propionibacterium acnes. Methods: This research included the manufacture of dried powder, characterization of dried powder, phytochemical screening, extraction by maceration method as well as testing of antibacterial activity against Staphylococcus epidermidis and Propionibacterium acnes with agar diffusion method using paper discs. Results: The results indicated that the dried powder of poguntano herb (Picria fel terrae Lour.) have water content of 7.9979%; water soluble extract content of 22.0498%; ethanol soluble extract content of 14.9367%; total ash content of 5.4317% and acid insoluble ash content 1.3248%. The ethanol extract of poguntano herb contained alkaloids, steroids/triterpenoids, flavonoids, glycosides, tannins, and saponins. Antibacterial activity test showed that ethanol extract of poguntano herb inhibited the growth of Propionibacterium acnes at the MIC 1.56 mg/ml with diameter of 8.13±0.05 mm and Staphylococcus epidermidis at the MIC 1.56 mg/ml with diameter of 7.06±0.05 mm. Conclusion: The ethanol extract of poguntano herb (Picria fel-terrae Lour.) has antibacterial activity against Staphylococcus epidermidis and Propionibacterium acnes.115 HalamanSkripsi Sarjan

    Background: Infectious diseases are a major problem especially in poor and developing countries including Indonesia. The most common cause is bacterial infections such as Klebsiella pneumonia and Staphylococcus aureus. Based on previous research, it is known that sungkai leaves contain secondary metabolite compounds such as alkaloids, steroids, phenolics, tannins, and saponins that have potential as antibacterials. Objective: This study aims to determine the content of chemical compounds contained in the simplicity and ethanol extract of sungkai leaves and test the antibacterial activity of ethanol extract of sungkai leaves against Klebsiella pneumonia and Staphylococcus aureus. Methods: This study was conducted with experimental methods including characterization of sungkai leaves simplicity, phytochemical screening test, extraction by reflux method using ethanol p.a and testing antibacterial activity using disc diffusion method with various concentrations. Results: The results of simplicity characterization obtained a moisture content of 9.99%, water soluble juice content of 10.28%, ethanol soluble juice content of 14.5%, total ash content of 3.829%, and acid insoluble ash content of 0.421%. Chemical compounds contained in simplicity and ethanol extracts of sungkai leaves are alkaloids, saponins, flavonoids, steroids, glycosides, and tannins. Ethanol extract of sungkai leaves has antibacterial activity against Klebsiella pneumonia and Staphylococcus aureus with Minimum Inhibitory Concentration against Klebsiella pneumonia at a concentration of 1.5625 mg/mL with an inhibition zone diameter of 6.4 mm and on Staphylococcus aureus bacteria at a concentration of 1.5625 mg/mL with an inhibition zone diameter of 6.97 mm. Conclusion: Ethanol extract of sungkai leaves (Peronema canescens Jack.) has antibacterial activity against Klebsiella pneumonia and Staphylococcus aureus.97 HalamanSkripsi Sarjan
