The interaction of electric fields with vesicles and cells: a study on electrodeformation and electroporation
The application of electric pulses to cells or vesicles induces complex responses. When electric field shocks are applied, a lipid membrane may become porated, which is a phenomenon known as electroporation. The lipid membrane may also deform under electromechanical and electrohydrodynamic forces, applied via electric fields,which is a phenomenon known as electrodeformation. Electroporation is widely employed in both biological research and clinical applications including areas such as drug and gene delivery, protein insertion, cancer therapy, and other processes where access to the cytoplasm is desired. Electrodeformation, on the other hand, can be harnessed as a means to probe the membrane's properties, and to detect pathological changes in cells. Despite its broad applicability, electroporation still suffers from low delivery efficiency and cell viability, in part due to a lack of a fundamental understanding of the mechanisms involved in this technique. Meanwhile, the electrodeformation phenomenon only received attention in the past decade, and therefore, a significant body of data only became available in the recent years. This is mainly due to the development of high-performance optical imaging systems which gave the opportunity to capture the dynamics associated with electrodeformation. In this work, we have designed and implemented experiments to study the electrodeformation of giant unilamellar vesicles, and to investigate the complex transport mechanisms involved in electroporation-mediated molecular delivery. In addition, we quantified the molecular uptake and cellular viability in electroporation experiments. First, we characterized the prolate deformations of giant unilamellar vesicles under strong DC electric fields. The vesicles exhibited prolate elongations along the direction of the electric field. In some cases, the aspect ratio of a deformed vesicle exceeded 10, representing a novel strong-deformation regime never before explored. Second, we studied the spatial and temporal transport of molecules via fluorescence microscopy, at the single cell level. The experimental results demonstrated that electrophoresis, not diffusion, is the dominant mode of of delivery during the pulse. Furthermore, we found that an electrokinetic mechanism known as field-amplified sample stacking (FASS) mediated an inverse correlation between delivery and the extracellular electrical conductivity. Following our accomplishment in the second task, we investigated a two-stage pulsing electroporation protocol that delivers separate pulses to porate the membrane and to electrokinetically-mediate species transport across the cell membrane. We found that while both delivery and viability are linearly dependent with respect to the duration of the second pulses, they both show a strong dependence on field strength. Moreover, a critical regime for maximized delivery and viability is achieved. This dissertation contributes to the electroporation and electrodeformation fields by generating explanations to several experimental observations based on physical mechanisms which were not previously available. In addition, the experimental tools for quantifying delivery and viability may help develop and improve the efficacy of electroporation protocols.Ph. Experimental investigation of two-phase flow phenomena in horizontal convective in-tube boiling system
The objective of this study was to investigate experimentally the thermal and hydrodynamic behavior, and dynamic instability in two-phase flow associated with refrigerant-11 in a uniformly heated horizontal in-tube convective boiling system.The experimental set-up used in this study was designed and built by the author. The test section was placed horizontally, and the stainless steel heater tubes were heated uniformly by a DC power source.Two major parts of experimental work were conducted: (1) Two-phase flow stratification due to gravity effects, and consequently its thermal and hydrodynamic behavior in a single horizontal tube under steady state conditions, was determined by measuring the top and bottom wall temperatures. A dimensionless number was found for the prediction of the occurrence of different flow patterns (Chapter 5). (2) Two-phase flow dynamic instabilities in a single tube configuration were found to bear similarities to vertical upflow systems. It was concluded however, that the entire unstable region was shifted to the right, compared to vertical flow, on the steady state characteristic plane (Chapter 6). Another important part of the work done was the application of dimensional analysis towards the steady state characteristic curves conventionally plotted in a P-G plane (Chapter 7). An infinite number of curves corresponding to different heat inputs could be converted to a single curve in a new coordinate system represented by two dimensionless numbers. PI theorem was used to perform this analysis.</p

In particular, there are some new M5- and D3-branes among our solutions which have AdS limits and contain a cosmological singularity as well. We also find a time-dependent version of the dyonic membrane configuration in 11-dimensions by applying a Lunin–Maldacena deformation to our new M5-brane solution

Theoretical and experimental analysis of unsteady forced convection in circular ducts
The analysis of forced convective heat transfer with laminar and/or turbulent flow subjected to periodic variations of the fluid inlet temperature usually leads to a nonclassical Sturm-Liouville type eigenvalue problem for which no known solution is presently available. This work reports the findings of two complementary studies (theoretical and experimental, and theoretical and empirical), respectively, on unsteady and steady forced convection of a Newtonian fluid flowing inside the thermal entry region of a circular duct, being subjected to different boundary and inlet conditions. A theoretical approach based on ideas associated with the generalized integral transform technique was developed to alleviate the need for the solution of complex time-dependent eigenvalue problems, while the difficulties associated with other methods overcome through consideration of time-independent auxiliary eigenvalue problems, resulting in a transformed infinite set of coupled, first-order, linear differential equations in the complex domain.In the second phase, accurate computational procedures were developed for obtaining benchmark analytical results of a hybrid nature. For practical purposes, consideration was given to cases of: (a) unsteady thermally developing forced convection with laminar and turbulent air flow inside a circular duct subjected to periodic variations of the fluid inlet temperature while imposing a fifth-kind boundary condition at the internal fluid-solid interface, and (b) thermally developing steady forced convection with turbulent air flow inside a circular duct subjected to a uniform fluid inlet temperature distribution with the first-kind boundary condition imposed at the internal fluid-solid interface. Benchmark results for quantities of practical relevance were obtained in terms of the axial variation in amplitudes and phase lags of centerline and bulk fluid temperatures and wall heat flux, in addition to local and thermally developed Nusselt numbers for both periodic and steady state situations. Through consideration of practical values for, respectively, the modified Biot number, Bi, fluid-to-wall heat capacitance ratio, , and Reynolds Number, Re, the thermal response of the system and convergence characteristics of the hybrid analytical-numerical models, here advanced, were critically examined, while the limitations associated with the decoupled system discussed within the realms of the variable system parameters and convergence criteria.In the final phase, an experimental apparatus was designed and built (by the author) and used to provide validation data for the theoretical studies, representing the "first set" of systematically evaluated benchmark experimental results for the circular duct geometry, particularly applicable to unsteady forced convection with the fifth-kind boundary condition and periodic inlet fluid temperature variations. Based on the cases considered, simplified analytic formulas are proposed for accurately predicting the variation in phase lags and amplitudes of wall heat flux, fluid centerline and bulk temperatures along the thermal entrance region. Further, the analysis here advanced can easily be applied to other flow passage geometries through transformation.</p
WANDA OR MEHMED SADIK (CHAYKA) PASHA'S ANECDOTAL MEMOIRS ABOUT TURKEY
Souvenirs Anecdotiques sur la Turquie (1820-1870), a book about important events in Ottoman history of the 19th century, has not attracted the attention of researchers for a long time. An assessment of the book had not been conducted because the identity of its publisher " Wanda" could not be verified. In DATE It was found out that the original book, entitled Turetskiye Anekdoti, was written by Michael Czajkowski in DATE in Russian. During his late life the book was translated into French by his daughter Karolina Suchodolska who used the pseudonym "Wanda." Starting from the comments and estimations about the book and its author by researchers who utilized its French translation, this article deals with the original author of the book, Michael Czajkowski's journey to Istanbul. In Istanbul the events and stories developed on which the book was based and included later developments following his conversion to Islam and employment by the Ottoman Empire. In addition this article addresses the period in which he wrote his memoirs after moving to Kiev, his conversion into Orthodoxy in his late life, the value of his memoirs as a source for Ottoman history of 19th century, their comparison with Turetskiye Anekdoti, and the originality of his Turetskiye Anekdoti as an example of anecdotal writing
