Drivers of pigment behavior during fermentation in rosé wines
La couleur est essentielle pour la perception de la qualité des vins rosés et constitue un élément clé de l'acte d'achat des consommateurs, du fait de leur conditionnement en bouteilles de verre transparent. Ce projet vise à comprendre les mécanismes impliqués dans l'évolution de la couleur lors de la vinification des vins rosés en analysant l'impact des procédés sur les pigments afin de mieux les maîtriser. Les premiers résultats obtenus ont révélé une grande diversité de couleur des vins rosés qui s'explique par leurs compositions phénoliques très différentes. La couleur des vins rosés clairs est principalement due à des pigments dérivés résultant des réactions des anthocyanes avec les acides phénols et l'acide pyruvique, un métabolite de la levure. Au contraire, la couleur des vins rosés foncés est due, d'une part, aux anthocyanes et aux flavanols pour la composante rouge a*, en lien avec l'extraction, d'autre part, pour la composante jaune b *, à des produits d'oxydation découlant de la réactivité des anthocyanes et de l'éthanal. Cette première approche a permis d'identifier les pigments impliqués dans la couleur des vins rosés afin de pouvoir ensuite les étudier lors de la fermentation alcoolique.Cette thèse se concentre sur une étape primordiale de la vinification réputée pour avoir un impact fort sur la couleur, la fermentation alcoolique. Dans les conditions expérimentales choisies, le pH, la température ainsi que la concentration en sulfite lors de la fermentation alcoolique n'ont eu aucun impact significatif sur la composition pigmentaire finale des vins fermentés en laboratoire. Cependant, les mécanismes impliqués lors de la fermentation alcoolique ont été fortement impactés par la matière première (cépage). De plus, l'adsorption sélective de certains pigments sur les lies a accentué cet effet cépage. En effet, une part non négligeable de la couleur des vins rosés a été adsorbée sur les parois de levures mettant en lumière l'importance de ce phénomène dans la vinification des vins rosés. Une dernière approche non ciblée a permis de valider le protocole de fermentation alcoolique en laboratoire et de confirmer les hypothèses précédentes de la prédominance de l'effet cépage sur les phénomènes impliqués lors de la fermentation. De plus, l'approche non ciblée a permis de révéler une composition particulière du Cinsault avec la présence en grande quantité de pigments polymérisés. Ces résultats ont permis de rendre compte de la diversité de couleur des vins rosés et de progresser dans la compréhension des mécanismes impliqués lors de la fermentation alcoolique.Color is essential for the quality of rosé wines perception and is a key element in the consumer's purchasing act, due to their packaging in transparent glass bottles. This project aims to understand the mechanisms involved in the evolution of color during the vinification of rosé wines by analyzing the impact of processes on pigments in order to better control them.The first results obtained revealed a great diversity in the color of rosé wines, which can be explained by their very different phenolic compositions. The color of light rosé wines is mainly due to pigments derived from the reactions of anthocyanins with phenolic acids and pyruvic acid, a yeast metabolite. On the contrary, the color of dark rosé wines is due, to anthocyanins and flavanols for the red component a*, in connection with extraction, and for the yellow component b*, to oxidation products resulting from the reactivity of anthocyanins and ethanal. This first approach allowed to identify the pigments involved in the color of rosé wines in order to study them during the alcoholic fermentation. This thesis focuses on a key stage of the wine-making process, the alcoholic fermentation, which is known to have a strong impact on color. Under the experimental conditions chosen, pH, temperature and sulfite concentration during alcoholic fermentation had no significant impact on the final pigment composition of wines fermented in the laboratory. However, the mechanisms involved in alcoholic fermentation were strongly impacted by the raw material (grape variety). Moreover, the selective adsorption of certain pigments on the lees accentuated this varietal effect. Indeed, a significant part of the color of the rosé wines was adsorbed on the yeast walls, highlighting the importance of this phenomenon in the vinification of rosé wines. A final non-targeted approach allowed us to validate the alcoholic fermentation protocol in the laboratory and to confirm the previous hypotheses of the predominance of the grape variety effect on the phenomena involved during fermentation. In addition, the non-targeted approach revealed a particular composition of Cinsault with the presence of large quantities of polymerized pigments.These results allowed to account for the diversity of color of rosé wines and to progress in the understanding of the mechanisms involved during alcoholic fermentation
Impact of organic viticulture on the genesis of varietal thiols in Côtes de Gascogne wines
Les thiols variétaux (3-sulfanylhexan-1-ol, 3SH et son acétate, 3SHA) sont des arômes puissants qui donnent des notes de pamplemousse et de fruits de la passion à certains vins. Ces notes aromatiques et leurs puissances font la typicité des vins des Côtes de Gascogne. Dans cette appellation, l'évolution des pratiques vers la viticulture biologique met en évidence des verrous techniques quant à la production de ces arômes. Ce travail vise à comprendre l'effet du mode de culture sur la production de ces thiols dans les vins des deux cépages emblématiques de la région, le Colombard et le Gros Manseng. Pour cela, l'effet du cuivre a été étudié au travers des différentes étapes de la vinification, de la parcelle au vin fini.Le suivi d'un réseau de parcelles au sein de l'appellation nous a permis de caractériser précisément le potentiel aromatique de type thiol de ces cépages, permettant ainsi de les classer parmi les plus concentrés en précurseurs de thiols, avec le Sauvignon blanc. Ces données d'observation sur les trois millésimes ont également montré un effet négatif de la pratique biologique sur la concentration en précurseurs de thiols dans les raisins des deux variétés, le Gros Manseng semblant être le plus impacté. L'étude au chai n'a pas révélé de comportement particulier des matrices biologiques par rapport aux conventionnelles. Quelle que soit le mode de conduite et le type de cépage, l'extraction des précurseurs de thiols est inférieure à celle du Sauvignon blanc, ce qui soulève la nécessité d'optimiser les étapes d'extraction pour ces deux cépages. Le cuivre n'est pas affecté par les étapes pré-fermentaires alors qu'il diminue de manière significative au cours de la fermentation alcoolique. Au cours de cette étape, nous avons identifié plusieurs effets du cuivre sur le développement des thiols variétaux. Tout d'abord, un effet oxydatif important, quel que soit le cépage. Cette oxydation diminue considérablement la fraction des thiols réduits (odorants) dans le vin fini. Ensuite, et uniquement pour le Gros Manseng, il y a une augmentation de la production de thiols totaux au cours de la fermentation. Cependant, cette augmentation ne permet pas de compenser les pertes dues à l'oxydation liée au cuivre. Enfin, nous avons identifié un nouvel intermédiaire S-conjugué et N-acétylé dans l'interconversion des précurseurs durant la fermentation, le N-acétyl-S-(1-hydroxyhexan-3-yl)-cystéine (NAC3SH). Ce composé produit uniquement par la levure en présence de cuivre lors de la fermentation complète notre connaissance de cette voie de dégradation et la rapproche un peu plus de la voie de détoxification des xénobiotiques. Le NAC3SH, lorsqu'il est produit par la levure, joue également le rôle de précurseur et permet la libération de 3SH et 3SHA en matrice synthétique. Similairement aux autres précurseurs, la révélation du NAC3SH en 3SH semble être diastéréoisomère dépendante, l'isomère (R,S) étant converti plus efficacement que le (R,R) par la levure X5.Varietal thiols (3-sulfanylhexan-1-ol, 3SH and its acetate, 3SHA) are powerful aromas that give grapefruit and passion fruit notes to certain wines. These aromatic notes and their power make the typicality of Côtes de Gascogne wines. In this appellation, the evolution of practices towards organic viticulture has highlighted technical obstacles to the production of these aromas. This work aims to understand the effect of the growing method on the production of these thiols in the wines of two emblematic grape varieties of the region, Colombard and Gros Manseng. To do this, the effect of copper was studied through the different stages of the wine-making process, from the plot to the finished wine.The monitoring of a network of plots within the appellation enabled us to characterize precisely the thiol-type aromatic potential of these grape varieties, thus enabling us to classify them among the most concentrated in thiol precursors, along with Sauvignon blanc. These observation data from the three vintages also showed a negative effect of organic practice on the concentration of thiol precursors in the grapes of both varieties, with Gros Manseng appearing to be the most impacted. The cellar study did not reveal any particular behavior of the organic matrices compared to the conventional ones. Regardless of the management method and the type of grape variety, the extraction of thiol precursors was lower than that of Sauvignon blanc, which raised the need to optimize the extraction steps for these two grape varieties. Copper was not affected by the pre-fermentation stages, whereas it decreased significantly during alcoholic fermentation. During this stage, we identified several effects of copper on the development of varietal thiols. Firstly, there was a significant oxidative effect, regardless of the grape variety. This oxidation considerably reduced the fraction of reduced (odorous) thiols in the finished wine. Secondly, and only for Gros Manseng, there was an increase in total thiol production during fermentation. However, this increase did not compensate for the losses due to copper-related oxidation. Finally, we identified a new S-conjugated and N-acetylated intermediate in the interconversion of precursors during fermentation, N-acetyl-S-(1-hydroxyhexan-3-yl)-cysteine (NAC3SH). This compound produced only by the yeast in the presence of copper during fermentation completed our knowledge of this degradation pathway and brought it a little closer to the detoxification pathway of xenobiotics. NAC3SH, when produced by yeast, also acted as a precursor and allowed the release of 3SH and 3SHA in synthetic matrix. Similar to the other precursors the release of NAC3SH to 3SH appeared to be diastereomer dependent, with the (R,S) isomer being converted more efficiently than the (R,R) by X5 yeast
Study of the combined effects of nitrogen and temperature on the synthesis of aromas in alcoholic fermentation in oenology
Aujourd'hui, la demande est orientée vers des vins disposant de profils sensoriels fruités et floraux. Afin d'être en adéquation avec les attentes des consommateurs, il est nécessaire de développer des stratégies de contrôle du procédé de vinification pour obtenir les propriétés organoleptiques souhaitées. L'objectif général de ce travail est donc d'étudier les effets individuels et combinés de deux paramètres clés de la fermentation sur le métabolisme de la levure et la synthèse des arômes fermentaires : l'azote et la température. La première partie de ce travail de thèse a été consacrée à l'étude de l'impact du timing d'ajout d'azote en cours de fermentation sur la synthèse des composés d'arôme par la levure. En ce sens, une étude mélangeant modélisation statistique et analyse de l'expression des gènes notamment impliqués dans la synthèse des arômes par la levure a permis de déterminer le timing optimal de l'ajout d'azote en cours de fermentation pour maximiser la teneur en arômes positifs (principalement les esters d'acétate). Ensuite, une évaluation des effets combinés de l'azote et de la température sur le métabolisme levurien a été effectuée grâce à un outil de suivi en ligne des arômes fermentaires. Une température idéale de synthèse a été identifiée pour chaque composé suivi. Enfin, l'examen de l'impact des ajouts d'azote et de la température sur les cinétiques fermentaires et aromatiques pour un pilotage de la fermentation a été étendue à l'étude de profils anisothermes de température. Des profils comprenant une baisse de température ont été identifiés comme les plus favorables à la synthèse et l'accumulation des esters d'éthyles dans le liquide.Today, the demand is oriented towards wines with fruity and floral sensory profiles. In order to meet consumer expectations, it is necessary to develop strategies to control the winemaking process to obtain the desired organoleptic properties. The general objective of this work is therefore to study the individual and combined effects of two key fermentation parameters on yeast metabolism and fermentation aroma synthesis: nitrogen and temperature. The first part of this thesis was devoted to studying the impact of the nitrogen addition timing during fermentation on the synthesis of aroma compounds by the yeast. In this sense, a study combining statistical modeling and analysis of the expression of genes involved in the aroma synthesis by yeast allowed us to determine the optimal timing of the nitrogen addition nitrogen during fermentation in order to maximize the content of positive aromas (mainly acetate esters). Then, an evaluation of the combined effects of nitrogen and temperature on yeast metabolism was carried out thanks to an online monitoring tool for fermentative aromas. An ideal synthesis temperature was identified for each compound monitored. Finally, the examination of the impact of nitrogen addition and temperature on fermentation and aroma kinetics for fermentation control was extended to the study of anisothermal temperature profiles. Study of sterol requirements of oenological yeasts (Saccharomyces Cerevisiae)
Les stérols font partie du lipidome des levures et sont essentiels pour leur croissance, leur métabolisme et leur viabilité, en particulier vis-à-vis de conditions de stress rencontrées par la levure pendant la fermentation alcoolique en œnologie. L'ergostérol est le stérol levurien, synthétisé en présence d'oxygène, tandis que les phytostérols (présents dans les particules solides des moûts de raisin) sont des stérols assimilés par les levures Saccharomyces cerevisiae lors de la fermentation œnologique (en anaérobiose). Cependant, une clarification excessive des moûts de raisin entraîne une perte de la quantité de phytostérols, ce qui peut conduire à des fermentations languissantes ou des arrêts de fermentation. L'impact physiologique de l'assimilation des phytostérols par rapport à l'ergostérol et l'influence du type de stérol sur les paramètres de la cinétique de fermentation sont peu connus. Cette thèse a eu pour objectif mieux comprendre le rôle des stérols et étudier l'impact du type de stérol sur la fermentation œnologique. Dans un premier temps, nous avons comparé l'impact de l'ergostérol par rapport aux phytostérols sur les paramètres fermentaires et sur la synthèse des métabolites clés de la fermentation pour 27 souches S. cerevisiae dans deux conditions de stress retrouvées en œnologie : carence en stérols ou stress osmotique (excès de sucres). L'ergostérol est le stérol permettant un meilleur maintien de la viabilité en fin de fermentation et, par conséquent, de finir la fermentation plus tôt, tandis que les phytostérols ont conduit à une réduction des quantités d'acétate et glycérol dans les deux conditions testées. Une diversité phénotypique a été mise en évidence, indiquant que les souches S. cerevisiae ne résistent pas de la même manière aux conditions de stress. L'évaluation de l'impact de la dose et du type de stérol sur la fermentation œnologique de 10 souches de S. cerevisiae a révélé que les différences entre ergostérol et phytostérols sur les performances fermentaires des souches sont plus importantes quand les stérols sont le nutriment limitant. L'effet du timing d'ajout de stérols, évalué pour 2 souches S. cerevisiae dans un moût synthétique carencé en stérols, a montré une meilleure efficacité en début de fermentation par rapport à un ajout en phase stationnaire : fermentation plus rapide, augmentation de la production d'arômes fermentaires. Enfin, des analyses transcriptomiques ont démontré que l'aptitude de certaines souches à résister à une carence en stérols était liée à leur capacité à développer une réponse ”génétique” spécifique au type de stérol (ergostérol ou phytostérols). Enfin, la dernière partie de cette étude a permis de valider en moût de raisin naturel les résultats obtenus en moût synthétique.Sterols are part of the yeast lipidome and are essential for their growth, metabolism and viability, particularly under stress conditions encountered by the yeast during wine alcoholic fermentation. Ergosterol is the yeast sterol, synthesized in the presence of oxygen, while phytosterols (present in the solid particles of grape musts) are sterols assimilated by Saccharomyces cerevisiae yeasts during oenological fermentation (under anaerobic conditions). However, excessive clarification of grape musts results in a loss of phytosterols, leading to sluggish or stuck fermentations. Little is known about the physiological impact of phytosterols uptake compared to ergosterol and the influence of sterol type on fermentation kinetics parameters. The objective of this thesis was to better understand the role of sterols and to study the impact of sterol type on oenological fermentation. First, we compared the impact of ergosterol versus phytosterols on fermentation parameters and on the synthesis of key fermentation metabolites for 27 S. cerevisiae strains under two stress conditions found in enology: sterol deficiency or osmotic stress (excess of sugars). Ergosterol is the sterol that allows a better maintenance of viability at the end of fermentation and, consequently, to finish the fermentation earlier, while phytosterols led to a reduction of acetate and glycerol quantities in the two conditions tested. Phenotypic diversity was revealed, indicating that S. cerevisiae strains are not equally resistant to stress conditions. Evaluation of the impact of sterol dose and type on the enological fermentation of 10 S. cerevisiae strains revealed that the differences between ergosterol and phytosterols on the fermentative performance of the strains were greater when sterols were the limiting nutrient. The effect of the timing of sterol addition, evaluated for two S. cerevisiae strains in a synthetic must deficient in sterols, showed a better efficiency at the beginning of fermentation compared to an addition in the stationary phase: faster fermentation, increase in the production of fermentative aromas. Finally, transcriptomic analyses showed that the ability of certain strains to resist sterol deficiency was linked to their capacity to develop a "genetic" response specific to the type of sterol (ergosterol or phytosterols). Optimization of acetaldehyde synthesis and aromatic profile of distillation wines through alcoholic fermentation
Depuis plus de 250 ans, la Maison Hennessy élabore des cognacs d'exception au cœur de sa région d'origine, la Charente. Dans l'exigence constante de la satisfaction des consommateurs, les œnologues, les distillateurs, le maître assembleur et les membres de la filière R&D/Qualité de la maison Hennessy sont impliqués de façon récurrente dans une problématique cruciale : la maîtrise de l'acétaldéhyde, aussi appelé éthanal, dans les eaux-de-vie nouvelles. L'éthanal représente un réel enjeu qualitatif dans le processus d'élaboration du Cognac : lorsqu'il est présent en excès dans les vins, il est responsable du caractère « éthéré » qui déprécie la qualité des eaux-de-vie. En effet, l'acétaldéhyde, au cours du vieillissement en forte présence d'alcool va tendre vers un équilibre chimique avec sa forme acétalisée appelée acétal (1,1-Diéthoxyéthane), qui apportera alors une odeur d'alcool à brûler, qui ne s'accommode pas avec la gourmandise, la puissance et le fruité recherchés. En œnologie, l'acétaldéhyde est un produit intermédiaire de transformation, principalement produit par la levure à un stade charnière entre deux mécanismes biochimiques : la glycolyse et la fermentation alcoolique. Ainsi, l'objectif de cette thèse est de maîtriser et minimiser la teneur résiduelle en acétaldéhyde dans les vins destinés à la distillation tout en préservant le potentiel aromatique des vins. Pour répondre à la problématique, de nouvelles approches de suivi en ligne de l'acétaldéhyde pendant la fermentation ont été mises en œuvre. Grâce à cet outil, la réalisation de bilans matière complets pendant la fermentation a permis de dissocier la production/consommation biologique de l'acétaldéhyde par la levure de son évaporation physique, en lien avec la température. D'un point de vue biologique, les températures de fermentation élevées favorisent une production importante d'acétaldéhyde en fin de phase de croissance. Par ailleurs, elles entrainent aussi des vitesses de consommation de la molécule par la levure plus élevées pendant la phase stationnaire ce qui conduit à des teneurs résiduelles en acétaldéhyde plus faible dans les vins finis. De plus, la vitesse de pertes en lien avec l'évaporation physique de l'acétaldéhyde augmente avec la température de fermentation. En phase stationnaire, la diminution de l'acétaldéhyde est principalement liée à la consommation de la levure même si l'effet physique moindre de l'évaporation n'est pas négligeable. Lorsque la température augmente, les vitesses de consommation et d'évaporation augmentent mais l'effet biologique supplante toujours l'effet physique. Ceci a pu notamment être corroboré avec l'utilisation de profils de température anisothermes. La part de consommation de l'acétaldéhyde par la levure étant significativement plus importante que la part d'évaporation de la molécule, des liens métaboliques ont pu être mis en évidence entre l'acétaldéhyde et des marqueurs du métabolisme tels que les acides organiques. La gestion de l'azote assimilable (quantité et qualité) semble aussi être un levier important pour maîtriser la teneur en acétaldéhyde dans les vins destinés à la distillation. En lien avec les fortes turbidités des moûts vinifiés en Cognac, l'excès lipidique a également fait l'objet d'études. La forte teneur en lipides, non limitante, montre des effets sur la modulation du profil aromatique des vins en favorisant la synthèse des alcools supérieurs tout en inhibant la synthèse des esters d'acétate sans influencer la production des esters d'éthyle. Les résultats obtenus, grâce à des approches pluridisciplinaires, permettent ainsi de proposer des stratégies de pilotage pour la mise en œuvre des fermentations alcooliques en orientant le profil aromatique tout en maitrisant les teneurs résiduelles en acétaldéhyde, sans oublier l'importance du choix de la souche de levure dans la résultante aromatique des vins finis.From more than 250 years, Hennessy has been producing exceptional cognacs in the heart of its region of origin, the Charente. With the constant demand for consumer satisfaction, the oenologists, distillers, master blender and members of the R&D/Quality department of Hennessy are constantly involved in a crucial issue: the control of acetaldehyde, also called ethanal, in new eaux-de-vie. Ethanal represents a real quality issue in the Cognac production process: when it is present in excess in the wines, it is responsible for the "ethereal" character that lowers the quality of the eaux-de-vie. Indeed, acetaldehyde, when aged in the presence of alcohol, will tend to reach a chemical equilibrium with its acetalized form called acetal (1,1-Diethoxyethane), bringing an odor of burning alcohol, not compatible with the delicacy, intensity and fruity notes sought. In oenology, acetaldehyde is an intermediate product of transformation, mainly produced by yeast at an intermediary stage between two crucial biochemical mechanisms: glycolysis and alcoholic fermentation. Thus, the objective of this thesis is to control and minimize the residual acetaldehyde content in wines intended for distillation, while preserving their aromatic potential. To address this issue, new approaches for on-line monitoring of acetaldehyde during fermentation have been implemented. Thanks to a new tool, the realization of complete mass balances during fermentation made it possible to dissociate the biological production/consumption of acetaldehyde by the yeast from its physical evaporation, which is linked to temperature. From a biological point of view, high fermentation temperatures favor a high production of acetaldehyde at the end of the growth phase. Moreover, they also lead to higher consumption rates of the molecule by the yeast during the stationary phase, which results in lower residual acetaldehyde levels in the finished wines. In addition, the rate of losses related to the physical evaporation of acetaldehyde increases with the fermentation temperature. In the stationary phase, the decrease in acetaldehyde is mainly related to the consumption from the yeast, even though a slight physical effect of evaporation is not negligible. When temperature increases, the consumption and evaporation rates increase, but the biological effect still overrides the physical effect. This could be corroborated with the use of anisothermal temperature profiles. As the consumption of acetaldehyde by yeast is significantly higher than the evaporation of the molecule, metabolic links between acetaldehyde and metabolic markers such as organic acids could be demonstrated. The management of assimilable nitrogen (quantity and quality) also seems to be an important lever to control the acetaldehyde content in wines intended for distillation. In connection with the high turbidity of musts vinified in Cognac, the lipid excess has also been studied. A high lipid content showed effects on the modulation of the aromatic profile of wines by promoting the synthesis of higher alcohols while inhibiting the synthesis of acetate esters, without influencing the production of ethyl esters. The results obtained, thanks to multidisciplinary approaches, make it possible to propose strategies for the implementation of alcoholic fermentations by orienting the aromatic profile while controlling the residual acetaldehyde content, without forgetting the importance of the choice of the yeast strain in the aromatic result of the finished wines
Modulation de la transition respiro-fermentaire chez Saccharomyces cerevisiae par l'oléate : analyse cinétique et métabolique en culture continue sur substrats mixtes
L'objectif de ce travail est d'étudier la transition respiro-fermentaire chez Saccharomyces cerevisiae et de déterminer l'étape ou les étapes contrôlant ce processus, à partir d'une approche originale de génie microbiologique basée sur l'utilisation de chémostat sur substrats mixtes. Dans un premier temps, l'analyse bibliographique a orienté notre travail vers l'étude du rôle du transport du carbone du cytosol vers la mitochondrie. Pour déterminer si ce transport exerce un contrôle sur la bascule métabolique, nous avons mis en place une stratégie consistant à introduire en chémostat sous limitation glucose en mode oxydatif une perturbation métabolique ciblée ( présence de co-substrat, modification génétique) et à analyser les conséquences de cette perturbation sur la bascule métabolique étudiée. L'oléate a été un des co-substrats testés dans ce travail car il est connu pour stimuler les enzymes impliquées dans le transport du carbone à travers la membrane mitochondriale. La transition respiro-fermentaire a donc été quantifiée en chémostat glucose seul et en chémostat glucose/oléate. En présence d'oléate, il a été observé un retard dans l'apparition de la bascule métabolique et une redirection du flux carboné de la voie fermentaire vers la voie oxydative. Ce résultat marquant met en évidence une diminution de la transition respiro-fermentaire en présence d'oléate et constitue un des rares succès reporté dans la littérature de réduction de l'effet Crabtree. L'action de l'oléate a été approfondie et discutée au cours de ce travail.\ud
____________________________________________________________This work aims to study the respiro-fermentative transition in Saccharomcyces cerevisiae and to determine the controlling step(s) involved in this process, by using an original approach of microbiological engineering based on co-substrate chemostat cultivations. Based on the literature analysis, we focused our work on investigating the importance of the carbon transport from cytosol to michondria in the onset of the metabolic shift. To determine whether this transport exerts a control on the metabolic change, a strategy was implemented consisting in introducing, during oxidative glucose limited chemostat, a specific metabolic perturbation ( addition of co-substrate, genetic modification) and in analysing the consequences of such a perturbation on the studied metabolic event. The oleic acid was one of the co-substrates tested in this work; this fatty acid is indeed know to simulte enzymes implied in the carbon transport between the different cell compartments. The metabolic transition was then studied in sole glucose chemostat and in glucose/oleic acid chemostat. In the presence of oleic acid, a delay in the onset of the metabolic shift and a redirection of the carbon flux from the reductive pathway were observed. This important result shows a modulation in the respiro -fermentative transition in presence of oleic acid and constitutes one of the few success reported in the literature of reduction in the Crabtree effect. the action of the oleic acid was further investigated and discussed throughout this work
Going Beyond Counting First Authors in Author Co-citation Analysis
The present study examines one of the fundamental aspects of author co-citation analysis (ACA) - the way co-citation
counts are defined. Co-citation counting provides the data on which all subsequent statistical analyses and mappings
are based, and we compare ACA results based on two different types of co-citation counting - the traditional type that
only counts the first one among a cited work's authors on the one hand and a non-traditional type that takes into
account the first 5 authors of a cited work on the other hand. Results indicate that the picture produced through this non-traditional author co-citation counting contains more coherent author groups and is therefore considerably clearer. However, this picture represents fewer specialties in the research field being studied than that produced through the traditional first-author co-citation counting when the same number of top-ranked authors is selected and analyzed. Reasons for these effects are discussed
Variations on the Author
“Variations on the Author” discusses two of Eduardo Coutinho’s recent films (Um Dia na Vida, from 2010, and Últimas Conversas, posthumously released in 2015) and their contribution to the general question of documentary authorship. The director’s filmography is characterized by a consistent yet self-effacing form of authorial self-inscription: Coutinho often features as an interviewer that rather than express opinions propels discourses; an interviewer that is good at listening. This mode of self-inscription characterizes him as an author who is not expressive but who is nonetheless markedly present on the screen. In Um Dia na Vida, however, Coutinho is completely absent form the image, while Últimas Conversas, on the contrary, includes a confessional prologue that moves the director from the margins to the center of his films. This article examines the ways in which these works stand out in the filmography of a director who offers new insights into the notion of cinematic authorship
Appropriate Similarity Measures for Author Cocitation Analysis
We provide a number of new insights into the methodological discussion about author cocitation analysis. We first argue that the use of the Pearson correlation for measuring the similarity between authors’ cocitation profiles is not very satisfactory. We then discuss what kind of similarity measures may be used as an alternative to the Pearson correlation. We consider three similarity measures in particular. One is the well-known cosine. The other two similarity measures have not been used before in the bibliometric literature.
