    Substantial Fatigue Similarity of a New Small-Scale Test Rig to Actual Wheel-Rail System

    The substantial similarity of fatigue mechanism in a new test rig for rolling contact fatigue (RCF) has been investigated. A new reduced-scale test rig is designed to perform controlled RCF tests in wheel-rail materials. The fatigue mechanism of the rig is evaluated in this study using a combined finite element-fatigue prediction approach. The influences of loading conditions on fatigue crack initiation have been studied. Furthermore, the effects of some artificial defects (squat-shape) on fatigue lives are examined. To simulate the vehicle-track interaction by means of the test rig, a threedimensional finite element (FE) model is built up. The nonlinear material behaviour of the rail steel is modelled in the contact interface. The results of FE simulations are combined with the critical plane concept to determine the material points with the greatest possibility of fatigue failure. Based on the stress-strain responses, by employing of previously postulated criteria for fatigue crack initiation (plastic shakedown and ratchetting), fatigue life analysis is carried out. The results are reported for various loading conditions and different defect sizes. Afterward, the cyclic mechanism of the test rig is evaluated from the operational viewpoint. The results of fatigue life predictions are compared with the expected number of cycles of the test rig by its cyclic nature. Finally, the estimative duration of the experiments until fatigue crack initiation is roughly determined.Structural EngineeringCivil Engineering and Geoscience

    Wind Turbine Generator Condition Monitoring via the Generator Control Loop

    This thesis focuses on the development of condition monitoring techniques for application in wind turbines, particularly for offshore wind turbine driven doubly fed induction generators. The work describes the significant development of a physical condition monitoring Test Rig and its MATLAB Simulink model to represent modern variable speed wind turbine and the innovation and application of the rotor side control signals for the generator fault detection. Work has been carried out to develop a physical condition monitoring Test Rig from open loop control, with a wound rotor induction generator, into closed loop control with a doubly fed induction generator. This included designing and building the rotor side converter, installing the back-to-back converter and other new instrumentation. Moreover, the MATLAB Simulink model of the Test Rig has been developed to represent the closed loop control, with more detailed information on the Rig components and instrumentation and has been validated against the physical system in the time and frequency domains. A fault detection technique has been proposed by the author based on frequency analysis of the rotor-side control signals, namely; d-rotor current error, q-rotor current error and q-rotor current, for wind turbine generator fault detection. This technique has been investigated for rotor electrical asymmetry on the physical Test Rig and its MATLAB Simulink model at different fixed and variable speed conditions. The sensitivity of the each proposed signal has been studied under different operating conditions. Measured and simulated results are presented, a comparison with the results from using stator current and total power has been addressed and the improvement in condition monitoring detection performance has been demonstrated in comparison with previous methods, looking at current, power and vibration analysis

    Concrete ice abrasion rig and wear measurements

    The wear of concrete material due to ice movement is a challenge for offshore and coastal structures. Concrete surfaces exposed to moving ice are subjected to wear at various rates depending on concrete and ice properties. At NTNU, Department of Structural Engineering, concrete ice abrasion phenomena are studied both theoretically and experimentally. This paper describes the accelerated concrete ice abrasion rig which was comprehensively improved recently.     RIG-I functions as a dual effector against HBV

    Host innate recognition triggers key immune responses for viral elimination. The sensing mechanism of hepatitis B virus (HBV), a DNA virus, and the subsequent downstream signaling events remain to be fully clarified. Here we found that type III but not type I interferons are predominantly induced in human primary hepatocytes in response to HBV infection, through retinoic acid-inducible gene-I (RIG-I)-mediated sensing of the 5'-ε region of HBV pregenomic RNA. In addition, RIG-I could also counteract the interaction of HBV polymerase (P protein) with the 5'-ε region in an RNA-binding dependent manner, which consistently suppressed viral replication. Liposome-mediated delivery and vector-based expression of this ε region-derived RNA in liver abolished the HBV replication in human hepatocyte-chimeric mice. These findings identify an innate recognition mechanism by which RIG-I dually functions as an HBV sensor activating innate signaling and to counteract viral polymerase in human hepatocytes.Supplemental materials are available on the publisher's website

    Dynamic Spin Rig Upgraded With a Five- Axis-Controlled Three-Magnetic-Bearing Support System With Forward Excitation

    The NASA Glenn Research Center Dynamic Spin Rig is used for experimental evaluation of vibration analysis methods and dynamic characteristics for rotating systems. Measurements are made while rotors are spun and vibrated in a vacuum chamber. The rig has been upgraded with a new active magnetic bearing rotor support and excitation system. This design is expected to provide operational improvements over the existing rig. The rig will be able to be operated in either the old or new configuration. In the old configuration, two ball bearings support the vertical shaft of the rig, with the test article located between the bearings. Because the bearings operate in a vacuum, lubrication is limited to grease. This limits bearing life and speed. In addition, the old configuration employs two voice-coil electromagnetic shakers to apply oscillatory axial forces or transverse moments to the rotor shaft through a thrust bearing. The excitation amplitudes that can be imparted to the test article with this system are not adequate for components that are highly damped. It is expected that the new design will overcome these limitations

    Scaling strategy of a new experimental rig for wheel-rail contact

    A new small–scale test rig developed for rolling contact fatigue (RCF) investigations in wheel–rail material. This paper presents the scaling strategy of the rig based on dimensional analysis and mechanical modelling. The new experimental rig is indeed a spinning frame structure with multiple wheel components over a fixed rail-track ring, capable of simulating continuous wheelrail contact in a laboratory scale. This paper describes the dimensional design of the rig, to derive its overall scaling strategy and to determine the key elements’ specifications. Finite element (FE) modelling is used to simulate the mechanical behavior of the rig with two sample scale factors of 1/5 and 1/7. The results of FE models are compared with the actual railway system to observe the effectiveness of the chosen scales.     RIG-I triggers a signaling-abortive anti-SARS-CoV-2 defense in human lung cells

    Efficient immune responses against viral infection are determined by sufficient activation of nucleic acid sensor-mediated innate immunity(1,2). Coronavirus disease 2019, caused by severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2), remains an ongoing global pandemic. It is an urgent challenge to clarify the innate recognition mechanism to control this virus. Here we show that retinoic acid-inducible gene-I (RIG-I) sufficiently restrains SARS-CoV-2 replication in human lung cells in a type I/III interferon (IFN)-independent manner. RIG-I recognizes the 3 ' untranslated region of the SARS-CoV-2 RNA genome via the helicase domains, but not the C-terminal domain. This new mode of RIG-I recognition does not stimulate its ATPase, thereby aborting the activation of the conventional mitochondrial antiviral-signaling protein-dependent pathways, which is in accordance with lack of cytokine induction. Nevertheless, the interaction of RIG-I with the viral genome directly abrogates viral RNA-dependent RNA polymerase mediation of the first step of replication. Consistently, genetic ablation of RIG-I allows lung cells to produce viral particles that expressed the viral spike protein. By contrast, the anti-SARS-CoV-2 activity was restored by all-trans retinoic acid treatment through upregulation of RIG-I protein expression in primary lung cells derived from patients with chronic obstructive pulmonary disease. Thus, our findings demonstrate the distinctive role of RIG-I as a restraining factor in the early phase of SARS-CoV-2 infection in human lung cells

    The essential role of a ubiquitin ligase, Riplet, in RIG-I-dependent antiviral innate immune responses

    RNA virus infection is recognized by the RIG-I-like receptors RIG-I and MDA5, which induce antiviral responses including the production of type I inter- ferons (IFNs) and proinflammatory cytokines. RIG-I is regulated by Lys63-linked polyubiquitination, and three E3 ubiquitin ligases, RNF125, TRIM25, and Riplet, are reported to target RIG-I for ubiquitination. To examine the importance of Riplet in vivo, we generated Riplet-deficient mice. Fibroblasts, macro- phages, and conventional dendritic cells from Riplet- deficient animals were defective for the production of IFN and other cytokines in response to infection with several RNA viruses. However, Riplet was dispens- able for the production of IFN in response to B-DNA and DNA virus infection. Riplet deficiency abolished RIG-I activation during RNA virus infection, and the mutant mice were more susceptible to vesicular stomatitis virus infection than wild-type mice. These data indicate that Riplet is essential for regulating RIG-I-mediated innate immune response against RNA virus infection in vivo
