    Computational Approaches for Investigating Three-Dimensional Structures of RNA

    The functions of many complex RNA molecules, including ribozymes and ribosomes, are dependent on their folding into well defined 3-D structures. Intramolecular interactions such as hydrogen bonds have important roles in stabilizing functional tertiary structures of RNA. The 3-D structures of RNA are usually analyzed and annotated through exhaustive visual examination of models generated from X-ray diffraction data. The increasing volume of structural data and diversity of folds have resulted in the need to develop computational approaches to more efficiently and exhaustively analyze as well as annotate 3-D structures of RNA automatically. Approaches which use graph theory have proven to be effective when employed for automatic identification of interaction patterns and structural motifs present in RNA tertiary structures. Hydrogen bonded base interactions such as pairs, triples, quadruples, quintuples and the A-minor motifs are some of the targets which have been used to define the parameters for annotating the 3-D structure of an RNA molecule. In addition to annotation, graph theoretical concepts have also been implemented to represent RNA conformations. This has enabled the comparison of RNA folds and the prediction of novel folds

    Introducing the Iguana Genome Project

    The Iguana Genome Project is currently conducted in the frame of the Consortium for Iguana Genome (CIG). The CIG is coordinated and led by the University Tor Vergata (Rome, Italy) and at present is in partnership with the Malaysia Genome Institute, the Universiti Kebangsaan Malaysia (UKM), and the University of Leeds (UK). The project aims to provide a genomic perspective on the origins, evolution, and adaptation of Galápagos Iguanas, taking advantage of latest advances in genomics and transcriptomics. Several questions will be tackled such as the adaptation to diving, the origin and implications of depigmentation, the genetic base of ecological requirements, and physiological and behavioral responses to the environment. Additionally, the project uses a genomic approach to reconstruct the most recent demographic history of specific populations and address the role of hybridization in guiding the evolutionary processes in Galápagos species. Finally, the project also aims to provide reference genomes for the conservation of threatened species. The CIG has already produced genome data of Conolophus marthae (93X coverage), Conolophus pallidus (10X coverage), and Amblyrhynchus cristatus (40 X coverage). Conolophus subcristatus’ genome is currently being sequenced. RNAs have been processed for transcriptome analysis for all Galápagos species. The project aims to carry out the investigations within a phylogenetic framework, and so the sequencing of the genomes of other genera of Iguanidae (Cyclura and Ctenosaura) is currently programmed.     This thesis presents a comparative measurement for under voltage load shedding in power system. The technique for comparing the voltage profile for the particular bus is developed by using the Simulink. The voltage profiles for this condition: initial condition, some loads are shed; some generators are turned off, at stressed condition, and also in the event of line outage condition; are observed. From the observation, the condition of voltage level can be determined. Result shown that when the load shedding is applied, the voltage profile at any particular bus is better. This can avoid the voltage instability for the system that can lead to the event of voltage collapse
