    Microsatellites markers for Coffea arabica L.

    Os marcadores microssatélites (SSR) são desejáveis para estudos genéticos e seleção assistida por marcadores. A limitada disponibilidade destes marcadores para Coffea arabica, uma das principais culturas em importância sócio-econômica do mundo, justifica novos esforços para o desenvolvimento de marcadores SSR. Portanto, o objetivo deste trabalho foi desenvolver, caracterizar e avaliar a utilização de marcadores SSR para estudos da diversidade genética em Coffea. Duas bibliotecas genômicas enriquecidas para as sequências repetidas (GT) 15 e (AGG) 10 foram construídas, utilizando-se o DNA genômico do genótipo Bourbon Amarelo UFV 570. Um total de 756 clones positivos, com seqüências de boa qualidade, foi utilizado para a localização de 287 SSR e o desenho de 96 pares de primers SSR. Aproximadamente 15% dos clones foram redundantes. As repetições de dinucletídeos foram mais freqüentes que os tri- e tetranucleotídeos, sendo as classes AG (32,8%), AC (12,9%), AAG (9,8%) e AGG (5,6%) as mais abundantes na porção analisada do genoma de C. arabica. Em média, foi encontrado um SSR a cada 1,45kb. Do total de 96 primers SSR desenhados e sintetizados, 90 (94%) foram validados como marcadores SSR úteis para estudos genéticos em C. arabica, sendo 21 (23%) polimórficos entre os cafeeiros Híbrido de Timor UFV 445-46 e Catuaí UFV 2143-235. Trinta e três primers SSR foram analisados em 24 acessos de Coffea importantes para o programa de melhoramento da UFV/EPAMIG. O número médio de alelos por primer foi de 5,1 e os valores médios de PIC foram de 0,56 para C. canephora e de 0,22 para C. arabica, variarando de 0,08 a 0,79 entre todos os acessos avaliados. Neste trabalho, também, foi comparada a eficiência de 18 marcadores SSR genômicos e 17 EST-SSR para o estudo da diversidade genética em Coffea. O número médio de alelos/primer foi de 5,1 e 5,3 para os marcadores EST-SSR e SSR genômico, respectivamente. Os marcadores SSR genômicos apresentaram maior número de alelos exclusivos e foram mais polimórficos dentro e entre os grupos genéticos, quando comparado com os EST-SSR. Os SSR genômicos apresentaram também maiores coeficientes de dissimilaridade genética e maior número de alelos discriminantes e foram eficientes em distinguir todos os acessos estudados. Os resultados demonstraram o potencial dos marcadores SSR genômicos na detecção do nível de polimorfismo, dos alelos exclusivos e da dissimilaridade genética, viitornando-os úteis para estudos de diversidade genética em Coffea, principalmente em C. arabica que apresenta pequena variabilidade genética. Este trabalho aumentou o número de marcadores SSR disponíveis para estudos genéticos em C. arabica e espécies relacionadas do gênero Coffea importantes para o melhoramento genético.The molecular markers microsatellites (SSR) are desirable for genetic study and in molecular marker assisted selection for genetic improvement. The limited availability of this marker for Coffea arabica, which is one of the principal crops in socio economic activity in the world, justified new efforts for the development of SSR markers. Therefore, the objective of this work was to develop, characterize and evaluate the utilization of the SSR markers, for the study of genetic diversity in Coffea. Two enriched genomic libraries for the repetitive sequences (GT) 15 and (AGG) 10 , using genomic DNA from Bourbon Amarelo (UFV 570) genotype, were constructed. A total of 756 positive clones with a sequence of good quality were utilized for the localization of 287 SSR and to design 96 SSR primer pairs. Approximately 15% of those clones were redundant. The repeated sequence of dinucleotides were more frequent than trinucleotides and tetranucleotides, being as classes AG (32.8%), AC (12.9%), AAG (9.8%) and AGG (5.6%) were more abundant in the portion of the C. arabica genome analyzed. In average one SSR was found in each 1.45kb. From a total of 96 SSR primers designed and synthesized, 90 (94%) were tested as SSR marker useful for genetic study in C. arabica, being 21 (23%) polymorphic among progenitors of Híbrido de Timor UFV 445-46 and Catuaí UFV 2143-235. Thirty three SSR primers were tested in 24 accessions of Coffea which are important for the breeding program of UFV/EPAMIG. The average number of alleles per primer was 5.1 and the average value of PIC were 0.56 for C. canephora and 0.22 for C. arabica, within range of 0.08 up to 0.79 among all the accessions evaluated. In this work the efficiency of 18 genomic-SSR markers and 17 EST-SSR markers for genetic diversity study in Coffea, were also compared. The number of alleles/primer was 5.1 and 5.3 for EST-SSR markers and genomic-SSR markers, respectively. Those markers of genomic-SSR produced a higher number of exclusive alleles and were more polymorphic within and among the genetic groups, when compared with EST-SSR markers. The genomic- SSR markers also showed higher genetic dissimilarity and higher number of discriminate alleles, and were efficient to distinguish all the accessions studied. The result obtained demonstrated the potencial of genomic-SSR markers in detecting the level of polymorphism, the exclusive alleles, and the genetic dissimilarity, that make these markers usefull for genetic ixdiversity study in Coffea, mainly in C. arabica which present low genetic variability.     Going Beyond Counting First Authors in Author Co-citation Analysis

    The present study examines one of the fundamental aspects of author co-citation analysis (ACA) - the way co-citation counts are defined. Co-citation counting provides the data on which all subsequent statistical analyses and mappings are based, and we compare ACA results based on two different types of co-citation counting - the traditional type that only counts the first one among a cited work's authors on the one hand and a non-traditional type that takes into account the first 5 authors of a cited work on the other hand. Results indicate that the picture produced through this non-traditional author co-citation counting contains more coherent author groups and is therefore considerably clearer. However, this picture represents fewer specialties in the research field being studied than that produced through the traditional first-author co-citation counting when the same number of top-ranked authors is selected and analyzed. Reasons for these effects are discussed

    Variations on the Author

    “Variations on the Author” discusses two of Eduardo Coutinho’s recent films (Um Dia na Vida, from 2010, and Últimas Conversas, posthumously released in 2015) and their contribution to the general question of documentary authorship. The director’s filmography is characterized by a consistent yet self-effacing form of authorial self-inscription: Coutinho often features as an interviewer that rather than express opinions propels discourses; an interviewer that is good at listening. This mode of self-inscription characterizes him as an author who is not expressive but who is nonetheless markedly present on the screen. In Um Dia na Vida, however, Coutinho is completely absent form the image, while Últimas Conversas, on the contrary, includes a confessional prologue that moves the director from the margins to the center of his films. This article examines the ways in which these works stand out in the filmography of a director who offers new insights into the notion of cinematic authorship

    Appropriate Similarity Measures for Author Cocitation Analysis

    We provide a number of new insights into the methodological discussion about author cocitation analysis. We first argue that the use of the Pearson correlation for measuring the similarity between authors’ cocitation profiles is not very satisfactory. We then discuss what kind of similarity measures may be used as an alternative to the Pearson correlation. We consider three similarity measures in particular. One is the well-known cosine. The other two similarity measures have not been used before in the bibliometric literature.     Dispelling the Myths Behind First-author Citation Counts

    We conducted a full-scale evaluative citation analysis study of scholars in the XML research field to explore just how different from each other author rankings resulting from different citation counting methods actually are, and to demonstrate the capability of emerging data and tools on the Web in supporting more realistic citation counting methods. Our results contest some common arguments for the continued use of first-author citation counts in the evaluation of scholars, such as high correlations between author rankings by first-author citation counts and other citation counting methods, and high costs of using more realistic citation counting methods that are not well-supported by the ISI databases. It is argued that increasingly available digital full text research papers make it possible for citation analysis studies to go beyond what the ISI databases have directly supported and to employ more sophisticated methods

    We have done our best to complete the author checklist relating to the use of animals in the hut study. Note that the objective for the hut study was to evaluate the IRS treatment applications for residual efficacy against Anopheles mosquitoes, including the local An. coluzzii mosquito population. Cows were only used to attract mosquitoes into the huts and no tests were carried out directly on the cows. The author checklist is intended for use with studies where experiments are carried out on animals, which is why we have had such difficulty in completing this for the hut study, as many of the questions do not relate to how the cows were used

Defesa : Palotina, 27/02/2023Inclui referências: p. 36-39Resumo: O complexo de enfezamento, transmitido pela cigarrinha do milho (Dalbulus maidis), tem se tornado uma das principais doenças do milho nas últimas safras no Brasil e na América Latina. A resistência genética é uma das melhores formas de controle e prevenção às doenças de plantas, principalmente devido a sua eficiência e baixo impacto ambiental, pela redução na utilização de produtos fitossanitários. Este trabalho buscou identificar fontes de resistência ao complexo dos enfezamentos por meio da avaliação da incidência e severidade e seus efeitos nas variáveis agronômicas em diferentes genótipos de milho (Zea mays L.). O experimento foi conduzido em duas safras (2020/21 e 2021/22) em Palotina e Entre Rios do Oeste - PR. Utilizou-se o delineamento em blocos aumentados, com 60 genótipos e 3 híbridos testemunhas, arranjadas em 4 blocos. As parcelas experimentais foram constituídas por quatro linhas com 5 m de comprimento e espaçamento de 0,9 m, com uma população aproximada de 55.500 plantas ha-1, sendo avaliadas as duas fileiras centrais. Dos 60 genótipos, foram selecionados 31 genótipos que tiveram bom desenvolvimento inicial de estande e dados produtivos completos para avaliações. Entre os estádios R2 e R3 realizaram-se as avaliações para incidência e severidade dos enfezamentos, seguindo as metodologias e protocolos propostos pela Embrapa. Foram avaliadas características agronômicas dos diferentes genótipos para comparações com os efeitos causados pelo complexo dos enfezamentos, e a tolerância de cada genótipo, sendo identificados os melhores indicadores produtivos para avaliação de tolerância de acordo com a correlação dos dados de incidência e severidade com os parâmetros produtivos. Os genótipos responderam diferentemente para a incidência e severidade dos enfezamentos. Análises de regressão e análise multivariada apresentaram resultados que comprovam o efeito do complexo de enfezamentos do milho para redução da produtividade, sendo possível classificar em níveis a tolerância e resistência dos genótipos avaliados. Os genótipos GNZ 7799 VIP3, Sempre 10A40 VIP3, DKB 360 PRO3, FS 575 PWU, NK Defender VIP3 e MG 540 PWU mostraram-se como os melhores genótipos para servirem de fontes de resistência ao complexo do enfezamento.Abstract: The stunting complex, transmitted by the corn leafhopper (Dalbulus maidis), has become one of the main corn diseases in recent harvests in Brazil and Latin America. Genetic resistance is one of the best ways to control and prevent plant diseases, mainly due to its efficiency and low environmental impact, due to the reduction in the use of phytosanitary products. This work sought to identify sources of resistance to the stunt complex through the assessment of incidence and severity and their effects on agronomic variables in different genotypes of maize (Zea mays L.). The experiment was treated in two seasons (2020/21 and 2021/22) in Palotina and Entre Rios do Oeste - PR. An augmented block design was used, with 60 genotypes and 3 control hybrids, arranged in 4 blocks. The experimental plots were distributed over four rows, 5 m long and spaced 0.9 m apart, with an approximate population of 55,500 plants ha-1, with the two inner central plots evaluated. Of the 60 genotypes, 31 genotypes were selected that had good initial stand development and complete yield data for estimation. Between stages R2 and R3, estimates of incidence and severity of stunting were carried out, following the methodologies and protocols proposed by Embrapa. Agronomic characteristics of the different genotypes were evaluated for comparisons with the effects caused by the stunting complex, and the tolerance of each genotype, being identified the best productive indicators for tolerance evaluation according to the influence of the incidence and severity data with the productive results. The genotypes responded differently for stunting incidence and severity. Regression analyzes and multivariate analysis showed results that prove the effect of the maize stunting complex to reduce productivity, making it possible to classify satisfactory levels of tolerance and resistance of the genotypes. Genotypes GNZ 7799 VIP3, Semper 10A40 VIP3, DKB 360 PRO3, FS 575 PWU, NK Defender VIP3 and MG 540 PWU are known as the best genotypes to serve as sources of resistance to stunting complex
