Role and regulation of miR-483 in cancer

The hsa-mir-483 locus is located at chromosome 11p15.5 within intron 2 of the IGF2 locus. Because of its location, de-regulated in Wilms’ tumor and other neoplasia, I hypothesized that this microRNA had a potential role in tumors. By analyzing 19 Wilms’ tumors, I proved that miR-483-3p is indeed over-expressed in 100% of the cases and a co-regulation with the over-expression of IGF2 was found. However, several other types of common adult cancers exhibit high or even extremely high levels of miR-483-3p expression without IGF2 over-expression. Indeed, independently from IGF2, the expression of the miR-483-3p could also be induced by the oncoprotein β-catenin through a novel interaction with the basic Helix-Loop-Helix protein upstream stimulatory transcription factor 1 (USF1). I also show that β-catenin itself is a target of miR-483-3p, triggering a negative regulative loop that becomes ineffective in cells harbouring activating mutations of β-catenin pathway. The potential oncogenic role of miR-483-3p was supported by the findings that its ectopic expression protects cells from apoptosis and, conversely, its inhibition increase the level of apoptosis. To understand the mechanisms of its action, I investigated potential gene targets. Among these, an important pro-apoptotic protein, Puma, were inhibited by miR-483-3p. My results indicate that miR-483-3p functions as an anti-apoptotic oncogene, coordinately over-expressed with IGF2 in Wilms’ tumors or induced by β-catenin activation in other tumor types

miR-200c sensitizes breast cancer cells to doxorubicin treatment by decreasing TrkB and Bmi1 expression.

Acquired resistance to classical chemotherapeutics is a major obstacle in cancer treatment. Doxorubicin is frequently used in breast cancer therapy either as single-agent or in combination with other drugs like docetaxel and cyclophosphamide. All these chemotherapies have in common that they are administered sequentially and often result in chemoresistance. Here, we mimicked this pulse therapy of breast cancer patients in an in vitro cell culture model, where the epithelial breast cancer cell line BT474 was sequentially treated with doxorubicin for several treatment cycles. In consequence, we obtained chemoresistant cells displaying a mesenchymal-like phenotype with decreased levels of miR-200c. To investigate the involvement of miR-200c in resistance formation, we inhibited and overexpressed miR-200c in different cell lines. Thereby, the cells were rendered more resistant or susceptible to doxorubicin treatment. Moreover, the receptor tyrosine kinase TrkB and the transcriptional repressor Bmi1 were identified as miR-200c targets mediating the drug resistance. Hence, we provide a mechanism of acquired resistance to doxorubicin that is caused by the loss of miR-200c. Along with this, our study demonstrates the complex network of microRNA mediated chemoresistance highlighting the challenges in cancer therapy and the importance of novel microRNA-modulating anticancer agents

    No full text
Islamic high schools have existed in the Chicagoland area for a number of years. There has been no study studying the impact of the schools on the lives of graduates. The purpose of this study was to describe, analyze, and reflect on lived experiences of graduates of Islamic high schools in the Chicagoland area. The aim of this study was to give a voice to students who lived the experience of attending and graduating from an Islamic high school in the Chicagoland area. This qualitative phenomenological study focused on the experiences of six graduates from three Islamic high schools in the Chicagoland area of the U.S. The graduates were asked about their general experiences and also asked about some of the challenges students faced in college in terms of identity, drugs, alcohol, pre-marital physical relationships, and Islamophobia. Based on the analysis of the responses of the graduates revealed four themes to include life-long lessons learned; life-long relationships developed; students who felt academically prepared, but socially unprepared; and schools that overprotected the students. Based on the results, recommendations were provided and legal, political, internal, and external implications of the study assessed.|Keywords: Islamic schools, graduates, dissertationProQuest Traditional Publishing Optio

Towards Optimal Deployment of a Sensor Network in a 3D Indoor Environment for the Mobility of People with Disabilities (Short Paper)

    No full text
Mobility of people with disabilities is one of the most important challenges for their social integration. There have been significant effort to develop assistive technologies to guide the PWD during their mobility in recent years. However, these technologies have limitations when it comes to the navigation and guidance of these people through accessible routes. This is specifically problematic in indoor environments where detection, location and tracking of people, and other dynamic objects that may limit the mobility of these people, are very challenging. Thus, many researches have leveraged the use of sensors to track users and dynamic objects in indoor environments. However, in most of the described methods, the sensors are manually deployed. Due to the complexity of indoor environments, the diversity of sensors and their sensing models, as well as the diversity of the profiles of people with disabilities and their needs during their mobility, the optimal deployment of a sensor network is a challenging task. There exist several optimization methods to maximize coverage and minimize the number of sensors while maintaining the minimum connectivity between the sensor nodes in a network. Most of the current sensor network optimization methods oversimplify the environment and do not consider the complexity of 3D indoor environments. In this paper, we propose a novel 3D local optimization algorithm based on a geometric spatial data structure that takes into account some of these complexities for the purpose of helping PWD in their mobility in 3D indoor environments such as shopping centers, museums and other public buildings
