Differential modulation after feeding in different salinities and response to ABA and extracellular Ca++ of APN activity in hepatopancreas of the intertidal euryhaline crab Neohelice granulata
Modulation of aminopeptidase N (APN) activity in the digestive tract by various factors would be important to adjust digestive and absorptive processes under different physiological and (or) environmental conditions. We studied the postprandial responses at different salinities and the effect of abscisic acid (ABA) and extracellular Ca2+ on APN activity in the hepatopancreas (the main site for nutrient digestion and absorption) of the model species Neohelice granulata (Dana, 1851). Enzyme activity was determined at different times (0, 24, 48, and 72 h) after feeding in crabs acclimated either to 35 psu (osmoconformation) or 10 psu (hyper-regulation). APN activity increased around 50% at 24 h after feeding at 35 psu, whereas no changes occurred at 10 psu. Enzyme activity was also assayed in the presence of ABA (1 × 10–4 mol・L–1) or extracellular Ca2+ (1 × 10–4 mol・L–1), showing increments of 60% and 56%, respectively. The results suggest a role of APN in postprandial adjustments and its modulation by different chemical messengers by direct effect on the hepatopancreas. Moreover, to our knowledge, this work is the first to show the effect of ABA on a digestive enzyme in the digestive tract of an animal.Fil: Michiels, Maria Soledad. Phenotypic flexibility in response to environmental salinity in the euryhaline crab Neohelice granulata from the mudflat and the saltmarsh of a SW coastal lagoon
This study constitutes a first attempt to investigate intraspecific differences in osmoregulatory capacity and digestive and metabolic responses at the biochemical level in relation to hyper- and hypo-regulation in a single species of estuarine crab inhabiting contrasting habitats within a same intertidal area. We compared hemolymph osmolality, key digestive enzymes, glycemia and energy reserves in Neohelice granulata (Dana in Proc Acad Nat Sci Philadelphia 5:247–254, 1851) from the mudflat and saltmarsh of Mar Chiquita coastal lagoon (37320 /37450 S- 57190 /57260 W) under a wide range of salinities (6–60 psu). Individuals from both sites exhibited high and similar osmoregulatory capacity, but while in individuals from mudflat low and high salinities affected lipase activity in hepatopancreas and triglycerides in muscle, in crabs from saltmarsh, high salinities affected glycogen in anterior gills. Low salinity differentially affected free glucose in anterior gills. The results suggest the occurrence of intraspecific distinct digestive and metabolic adjustments in relation to osmoregulatory responses and habitat.Fil: Pinoni, Silvina Andrea. The influence of habitat on metabolic and digestive parameters in an intertidal crab from a SW atlantic coastal lagoon
The hepatopancreas of decapod crustaceans is an organ which can act as indicator for digestive and metabolic parameters under different physiological and / or environmental conditions. However, biochemical studies on digestive and metabolic parameters of the hepatopancreas of euryhaline burrowing crabs such as Neohelice granulata from habitats with different diet compositions are still scarce. In the wild, adult males of N. granulata from Mar Chiquita Coastal Lagoon (Argentina) in mudflat habitat have diets with higher lipid and protein content than crabs from the saltmarsh, suggesting that diets could be an important factor influencing hepatopancreas activities. We tested this hypothesis here by exposing adult male crabs to a similar experimental diet and comparing hepatopancreas parameters for lipid components and protein metabolism between males from these two habitat types at different times (up to three months). At month 3, we noticed a decrease of the triglyceride concentration and lipase activity and an increase of protein concentration in crabs from the mudflat. In contrast, triglycerides and protein concentration did not change in crabs from the saltmarsh, while lipase activity decreased and levamisole insensitive AP increased at month 3. The results indicate that digestive and metabolic parameters in the hepatopancreas of crabs from habitats varying in diet content remain different, even if crabs are subsequently fed by a similar experimental diet. This suggests that specific intrinsic regulations of these hepatopancreas parameters could operate differently in each habitat and could not be changed by recent diet conditions.Fil: Méndez, Eugenia. Lipase activity sensitive to dopamine, Glucagon and cyclic amp in the Hepatopancreas of the Euryhaline burrowing crab Neohelice Granulata (Dana, 1851) (Decapoda, Grapsidae)
We studied the biochemical characteristics and modulation by dopamine, glucagon and cAMP of lipase activity in hepatopancreas of the euryhaline crab Neohelice granulata (Dana, 1851), considered to be an emergent model for biochemical, physiological and ecological research. Lipase activity was maximum at pH 8.5; it exhibited Michaelis-Menten kinetics (apparent Km = 0.018 mM), was highest at 37°C but appeared to be cold- and heat-tolerant, since it was high at 4°C and at 45°C. Lipase activity was enhanced upon incubation of hepatopancreas with 10?4 M dopamine (about 100%), 2 × 10?3 M glucagon (about 250%) and 10?4 M cAMP (about 150%) suggesting a role of these chemical messengers in mechanisms of regulation of lipolytic activities and its direct effect on the hepatopancreas. The concomitant decrease in triglycerides content upon dopamine and cAMP treatment suggests a link between enhanced lipase activity by these messengers and triglycerides catabolism.Fil: Lopez Mañanes, Alejandra Antonia. Biochemical characteristics and modulation by external and internal factors of aminopeptidase-N activity in the hepatopancreas of a euryhaline burrowing crab
Strikingly, in spite of its physiological importance, information about occurrence, biochemical characteristics and mechanisms of regulation of aminopeptidase-N (APN) in the hepatopancreas of intertidal euryhaline crabs is still lacking. In this work, we determined the occurrence, biochemical characteristics, response to environmental salinity and dopamine of APN in the hepatopancreas of the euryhaline crab Neohelice granulata (Dana 1851) from the open mudflat of Mar Chiquita coastal lagoon (Buenos Aires province, Argentina). APN activity was maximal at pH and temperature range of 7.6–9.0 and 37–45 °C, respectively. APN activity exhibited Michaelis–Menten kinetics (apparent Km = 0.19 ± 0.10 mM) (pH 7.6, 37 °C) and appeared to be sensitive to bestatin (I 50 = 15 mM) and EDTA (I 50 = 9 mM). In crabs acclimated to 10 psu (hyper-regulation conditions) and 37 psu (hypo-regulation conditions), APN activity was about 45 and 160 % higher, respectively, than in 35 psu (osmoconformation). APN activity in the hepatopancreas was stimulated in vitro (about 137 %) by 10−4 M dopamine. Higher dopamine concentrations produced a similar extent of increase. The responses of APN activity to salinity and dopamine in vitro suggest the role of APN in digestive adjustments upon hyper and hypo-regulatory conditions and its modulation via direct mechanisms on hepatopancreas by dopamine.Fil: Michiels, Maria Soledad. Analyses of body condition and digestive/metabolic parameters of Odontesthes argentinensis (Atherinopsidae) from Mar Chiquita Coastal Lagoon (Argentina) during different phases of ovarian development
This study analyses body condition and digestive/metabolic parameters of Odontesthesargentinensis from Mar Chiquita Coastal Lagoon (MChCL) during different ovary maturity phases (II: developing; III: spawning capable; IIIas: actively spawning subphase). Silverside individuals in phases II and III maintained their body condition, while it improved in actively spawning subphase. Total weight and Fulton’s condition factor increase in 70% and 50%, respectively. No changes were observed during the ovary maturation phases in hepatosomatic index and intestinal coefficient. Maltase activity in the intestine remained unchanged, while Aminopeptidase-N was higher, about 300%, in the spawning capable phase (III). Alkaline phosphatase in the intestine was unchanged pointing out the maintenance of intestinal homeostasis. Liver glycogen concentration decreased, about 40%, in the actively spawning subphase (IIIas), while muscle glycogen abruptly decreased in phase III and recovered in subphase IIIas. In intestinal fat, triglycerides concentration was lower 70% in subphase IIIas, while no changes occurred in liver and muscle. Muscle protein concentration decreased about 50% in actively spawning subphase. The findings shed light on the remarkable adaptability of this population of O. argentinensis from MChCL to support its gonadal maturation and active spawning maintaining and even improving its physical condition.Fil: Méndez, Eugenia. Trypsin and N-aminopeptidase (APN) activities in the hepatopancreas of an intertidal euryhaline crab: Biochemical characteristics and differential modulation by histamine and salinity
No studies are available about biochemical characteristics and modulation (i.e. by endogenous and/or environmental cues) of trypsin (a key digestive endoprotease) in hepatopancreas of intertidal euryhaline crabs neither on the possible concomitant modulation of key ectoproteases such as aminopeptidase-N (APN) involved in final steps of protein digestion. Furthermore, nothing is still known in decapods crustaceans about the role of histamine as primary chemical messenger for modulation of main components of digestive process (i.e. proteases). We determined biochemical characteristics and investigated the effect of histamine injections; of histamine in vitro and of acclimation of individuals to low and high salinity on trypsin and aminopeptidase-N (APN) activities in the hepatopancreas of the euryhaline crab Cyrtograpsus angulatus (Dana 1851). Trypsin activity was maximal at pH 7.4 and at 45 °C. APN activity increased from pH 6.6 to 7.6–9.0 and was maintained high at 37–45 °C. Both activities exhibited Michaelis-Menten kinetics (apparent Km: trypsin = 0.36 mM; APN = 0.07 mM). The injection of 10− 4 M histamine decreased trypsin activity (about 40%) in hepatopancreas while did not affect APN activity. Similarly, in vitro 10-4 M histamine decreased trypsin activity (about 52%) in hepatopancreas but not APN activity. Trypsin activity in the hepatopancreas was not affected by acclimation of crabs to low (10 psu) or high (40 psu) environmental salinity while APN activity was increased (about 200%) in 10 psu. The results show the differential modulation of trypsin and APN by distinct cues and point to histamine as modulator of intracellular trypsin by direct action on the hepatopancreas.Fil: Michiels, Maria Soledad. Going Beyond Counting First Authors in Author Co-citation Analysis
The present study examines one of the fundamental aspects of author co-citation analysis (ACA) - the way co-citation
counts are defined. Co-citation counting provides the data on which all subsequent statistical analyses and mappings
are based, and we compare ACA results based on two different types of co-citation counting - the traditional type that
only counts the first one among a cited work's authors on the one hand and a non-traditional type that takes into
account the first 5 authors of a cited work on the other hand. Results indicate that the picture produced through this non-traditional author co-citation counting contains more coherent author groups and is therefore considerably clearer. However, this picture represents fewer specialties in the research field being studied than that produced through the traditional first-author co-citation counting when the same number of top-ranked authors is selected and analyzed. Reasons for these effects are discussed
Variations on the Author
“Variations on the Author” discusses two of Eduardo Coutinho’s recent films (Um Dia na Vida, from 2010, and Últimas Conversas, posthumously released in 2015) and their contribution to the general question of documentary authorship. The director’s filmography is characterized by a consistent yet self-effacing form of authorial self-inscription: Coutinho often features as an interviewer that rather than express opinions propels discourses; an interviewer that is good at listening. This mode of self-inscription characterizes him as an author who is not expressive but who is nonetheless markedly present on the screen. In Um Dia na Vida, however, Coutinho is completely absent form the image, while Últimas Conversas, on the contrary, includes a confessional prologue that moves the director from the margins to the center of his films. This article examines the ways in which these works stand out in the filmography of a director who offers new insights into the notion of cinematic authorship
