Avaluació de l’eficàcia del tractament amb la fracció vascular estromal en pacients amb artrosi de genoll: un estudi experimental amb grup control
No full textL’artrosi de genoll és una patologia freqüent que causa dolor, tumefacció i disminució
de funcionalitat degut, entre d’altres, a l’aprimament del cartílag. Tractaments basats
en cèl·lules estromals mesenquimals, com la fracció vascular estromal (SVF), són
cada vegada més emprats pel seu potencial generador de cartílag però encara manca
evidència per a confirmar la seva eficàcia.
A fi de conèixer l’eficàcia de la SVF en l’artrosi de genoll s’ha realitzat una revisió
sistemàtica dels assajos clínics publicats que l’empren i un estudi experimental amb
grup control d’un producte estandarditzat de SVF amb avaluació clínica, funcional i
radiològica, mitjançant escala visual analògica, escala KOOS (Knee Injury and
Osteoarthritis Outcome Score), escala SF-36 i classificació MOCART (Magnetic
Resonance Observation of Cartilage Repair Tissue). S’han comparat les variables a
estudi prèvies al tractament, al mes, als 6 mesos i a l’any després del tractament.
S’han recollit els efectes adversos derivats del tractament.
A la revisió s’han inclòs 9 articles d’entre 266, incloent 239 pacients (274 genolls), amb
un seguiment de 6 a 24 mesos. Tots els estudis evidencien una milloria del dolor i de
la funcionalitat i 4 d’ells també descriuen millories radiològiques. No obstant, els
biaixos metodològics i la utilització de preparats no estandarditzats permeten
concloure que són necessaris més estudis.
A l’estudi experimental s´han inclòs 47 pacients al grup experimental (18 dones i 29
homes, amb edat mitjana 58.51±11.20 anys) i 27 pacients del grup control (15 dones i
12 homes, amb edat mitjana 55.21±11.47 anys). 67 genolls han estat analitzats al grup
experimental enfront a 33 al grup control, essent grups de característiques
demogràfiques i clíniques comparables. Com a diferència rellevant, el grup control
parteix d’una millor situació radiològica inicial, amb valors a l’escala MOCART
superiors respecte el grup experimental (85.91±36.56 vs 69.54±31.75, p=0.024).
Hi ha diferències estadísticament significatives tant en el dolor en repòs com en
activitat i en totes les subescales de l’escala KOOS previ al tractament respecte tots
els altres moments avaluats post tractament (p<0.001). La qualitat de vida a l’any
també mostra canvis significatius en el seu recompte global (p=0.021). No s’observen
canvis significatius en els valors del MOCART en el grup experimental (70±40 vs
65±40, p=0.748) mentre que en el grup control s’evidencia la disminució significativa
tant els valors del MOCART total (80±60 vs 75±70, p=0.001) com els del MOCART en
el compartiment femorotibial extern (35±25 vs 35±30, p=0.017). Destaca la diferència
estadísticament significativa respecte l’evolució radiològica entre el grup experimental i
el grup control (p=0.046).
El 26.9% dels pacients ha reportat efectes adversos al tractament, tots de caràcter lleu
i en la majoria dels casos resolts espontàniament.
Aquest projecte demostra que el tractament amb SVF és segur i eficaç per a millorar el
dolor, la funcionalitat i la qualitat de vida dels pacients afectes d’artrosi de genoll. S’ha
evidenciat una millora de les característiques radiològiques en part dels subjectes
analitzats. No obstant, es requereixen estudis randomitzats i amb una major mostra
per a identificar quin subgrup de pacients pot tenir una major resposta regenerativa al
tractament

Below are detailed the variables in the files and the units when necessary: Species: refers to the experimental species used. Each .txt file refers to one of the three seagrass species studied; i.e., Posidonia oceanica, Cymodocea nodosa and Zostera noltii. Time: Categorical variable referring to the sampling time for each experiment. Days: Elapsed number of days from the beginning of the experiment. Cage or replicate: Number relating to the experimental replicates. Replicates can be understood as controlled plots / patches of the different seagrass species where urchins were manipulated. Sea_urchin_density_ind_5625cm2: Number of sea urchins in experimental plots. Canopy_height_cm: Leaves length in cm measured within experimental plots. Shoot_density_shoot_m2: Measured as the number of shoots per square meter and referred to experimental plots. Integrating mathematical modeling into synthetic biology education: a comprehensive approach through webinars and syllabus development
No full text[EN] Mathematical modeling is a fundamental aspect of synthetic biology, enabling precise design and analysis of biological systems. To enhance students' understanding of this critical topic, we developed a series of webinars aimed at teaching mathematical modeling to iGEM teams. These webinars were initially created to maintain student engagement during a period of restricted lab access but quickly demonstrated their value as an effective educational tool. The success of these webinars highlighted the suitability of mathematical modeling as a topic well-suited to both onsite and online learning environments. Recognizing this, we expanded the content into a comprehensive syllabus for undergraduate courses in synthetic biology at the Universitat Polit & egrave;cnica de Valencia in Spain and Universidad de las Fuerzas Armadas-ESPE in Ecuador. The course now serves as a core component of synthetic biology education, offering students a robust framework for understanding and applying mathematical models. It includes a series of lectures, practical exercises, and case studies, all designed to deepen students' knowledge and skills in this essential area. To support educators and students, we have also developed a deck of slides and example scripts that provide practical examples and reinforce the concepts taught in the course. This manuscript presents the development, implementation, and impact of these educational initiatives, demonstrating how mathematical modeling can be effectively integrated into synthetic biology curricula to prepare students for real-world challenges in the field.The author(s) declare financial support was received for the research, authorship, and/or publication of this article.
