Going Beyond Counting First Authors in Author Co-citation Analysis
The present study examines one of the fundamental aspects of author co-citation analysis (ACA) - the way co-citation
counts are defined. Co-citation counting provides the data on which all subsequent statistical analyses and mappings
are based, and we compare ACA results based on two different types of co-citation counting - the traditional type that
only counts the first one among a cited work's authors on the one hand and a non-traditional type that takes into
account the first 5 authors of a cited work on the other hand. Results indicate that the picture produced through this non-traditional author co-citation counting contains more coherent author groups and is therefore considerably clearer. However, this picture represents fewer specialties in the research field being studied than that produced through the traditional first-author co-citation counting when the same number of top-ranked authors is selected and analyzed. Variations on the Author
“Variations on the Author” discusses two of Eduardo Coutinho’s recent films (Um Dia na Vida, from 2010, and Últimas Conversas, posthumously released in 2015) and their contribution to the general question of documentary authorship. The director’s filmography is characterized by a consistent yet self-effacing form of authorial self-inscription: Coutinho often features as an interviewer that rather than express opinions propels discourses; an interviewer that is good at listening. This mode of self-inscription characterizes him as an author who is not expressive but who is nonetheless markedly present on the screen. In Um Dia na Vida, however, Coutinho is completely absent form the image, while Últimas Conversas, on the contrary, includes a confessional prologue that moves the director from the margins to the center of his films. Appropriate Similarity Measures for Author Cocitation Analysis
We provide a number of new insights into the methodological discussion about author cocitation analysis. We first argue that the use of the Pearson correlation for measuring the similarity between authors’ cocitation profiles is not very satisfactory. We then discuss what kind of similarity measures may be used as an alternative to the Pearson correlation. We consider three similarity measures in particular. One is the well-known cosine. The other two similarity measures have not been used before in the bibliometric literature. Dispelling the Myths Behind First-author Citation Counts
We conducted a full-scale evaluative citation analysis study of scholars in the XML research field to explore just how different from each other author rankings resulting from different citation counting methods actually are, and to demonstrate the capability of emerging data and tools on the Web in supporting more realistic citation counting methods. Our results contest some common arguments for the continued
use of first-author citation counts in the evaluation of scholars, such as high correlations between author rankings by first-author citation counts and other citation
counting methods, and high costs of using more realistic citation counting methods that are not well-supported by the ISI databases. Mechanisms of targeted anti-tumor activity of dianthin-panitumumab conjugates in combination with the glycosylated triterpenoid SO1861
Die stets ansteigende Zahl an Krebserkrankungen mit einhergehendem tödlichem Verlauf erfordert die stetige Weiter- und Neuentwicklung von entsprechenden Therapien, welche Tumorspezifisch agieren müssen. Vor allem schwerwiegende Nebeneffekte, die häufig bei den gängigen Radio- und Chemotherapien auftreten, gilt es zu minimieren. Der Aufbau von Antikörper-Wirkstoff-Konjugaten (ADC) vereint hier das zielgerichtete Wirken durch den Antikörper mit der Toxizität der Wirkstoffkomponente und so sind bereits acht dieser ADCs durch die Food and Drug Administration zugelassen worden. In dieser Arbeit wurde deswegen ein ADC aus Panitumumab, einem vollständig humanen IgG2-Antikörper mit geringem immunogenem Potential, und Dianthin, einem hoch toxischen pflanzlichen und enzymatisch wirkenden Protein, reproduzierbar mit einem Wirkstoff-zu-Antikörper-Verhältnis (DAR) von ~1 und einer molekularen Masse von ~180 kDa erzeugt. Dabei wurde die Bindungsaffinität von Dianthin-Panitumumab (DPan) zum Zielrezeptor EGFR, welcher vorwiegend von Krebszellen in einem Übermaß exprimiert wird, unvermindert gegenüber Panitumumab mit einem KD-Wert von 1.5 × 10^-10 M bestimmt. Die enzymatische Aktivität von DPan im Vergleich zu Dianthin war aufgrund der Kopplung um ~50 % von 312.2 auf 153.3 pmol(Adenin)/pmol(Protein)/h vermindert, jedoch bewies das ADC DPan in vitro ein enormes selektives und zielzellspezifisch zytotoxisches Potential, wobei in Kombination mit dem glykosylierten Triterpenoid SO1861 als Endosomal Escape Enhancer eine bemerkenswerte mittlere Effektivität von 0.00022 / 0.00006 nM (IncuCyte-Echtzeitmessung / MTT-Assay) erreicht wurde. Das zytotoxische Potential der Kombinationsbehandlung konnte weiterführend in einem 3D-Sphäroid-Versuch unter realitätsnahen Tumorbedingungen nachgewiesen werden. Ebenso konnte in dieser Arbeit erstmals gezeigt werden, dass DPan weiterhin und vergleichbar zu Panitumumab in der Lage ist, die Antikörperabhängige zellvermittelte Zytotoxizität (ADCC) über PBMCs und Monozyten auszulösen, um somit einen additiven antitumoralen Effekt zu erzeugen. Das in vitro hervorragend zytotoxisch wirkende ADC DPan zeigte dagegen in vivo an tumortragenden Mäusen eine im Vergleich zu Panitumumab leicht verminderte antitumorale Wirkung, wobei in Kombination mit SO1861 die Wirkung signifikant gesteigert wurde. Die eingesetzte Dosis mit 3.3 µg lag dabei weit unter der höchst verträglichen Dosis ohne schwerwiegende Nebeneffekte mit 126 µg. Untersuchungen der Biodistribution von Radionuklid (99mTc) markierten DPan und Panitumumab zeigten indes eine unterschiedliche Kinetik. Beide Proteine zeigten bezüglich der Konzentration im Tumor erst am Ende des untersuchten Zeitraums von 18 h die höchste Konzentration , wobei Panitumumab dominierend im Tumor akkumulierte. Die allgemeine Biodistribution im Körper betrachtend zeigte Panitumumab eine vermehrte Plasmapräsenz im Blut-Pool, wohingegen DPan stark in der Leber und Milz akkumulierte. Ferner konnte gezeigt werden, dass Dianthin bereits im Blutplasma geringfügig aus dem ADC abgespalten wird, vermutlich weil hier ein spaltbarer SPDP-Linker verwendet wurde. Da DPan in Kombination mit SO1861 aber in vitro ein hervorragendes antitumorales Potential zeigte, welches in vivo nicht gänzlich zum Tragen kam, sollte trotzdem in zukünftigen Arbeiten an der Optimierung der In-vivo-Effizienz von DPan gearbeitet werden.The ever-increasing number of cancers with a fatal outcome requires the constant further and new development of appropriate therapies, which have to act tumor-specific. Above all, serious side effects, which frequently occur with current radio- and chemotherapies, must be minimized. The development of antibody-drug conjugates (ADCs) combines the targeted action of the antibody with the toxicity of the drug component, and eight of these ADCs have already been approved by the Food and Drug Administration. In this work, an ADC was therefore generated from panitumumab, a fully human IgG2 antibody with low immunogenic potential, and dianthin, a highly toxic plant and enzymatically active protein, reproducibly with a drug to antibody ratio (DAR) of ~1 and a molecular mass of ~180 kDa. The binding affinity of dianthin-panitumumab (DPan) to the target receptor EGFR, which is predominantly expressed by cancer cells in excess, was determined undiminished compared to panitumumab with a KD value of 1.5 × 10^-10 M. The enzymatic activity of DPan compared to dianthin was reduced by ~50% from 312.2 to 153.3 pmol(adenine)/pmol(protein)/ due to coupling. But the ADC DPan demonstrated an enormous selective and target cell specific cytotoxic potential in vitro. In combination with the glycosylated triterpenoid SO1861 as an endosomal escape enhancer, a remarkable mean effectiveness of 0.00022 / 0.00006 nM (IncuCyte real-time measurement / MTT assay) was achieved. The cytotoxic potential of the combination treatment was further demonstrated in a 3D-spheroid experiment under realistic tumor conditions. In this work it could also be shown for the first time that DPan is still able to induce antibody-dependent cell-mediated cytotoxicity (ADCC) via PBMCs and monocytes in a manner comparable to panitumumab, thus producing an additive anti-tumor effect. In contrast, the in vitro outstanding cytotoxicity of ADC DPan showed in vivo in tumor-bearing mice a slightly decreased antitumor effect compared to panitumumab, whereas in combination with SO1861 the effect was significantly increased. The applied dose of 3.3 µg was far below the highest tolerated dose of 126 µg and no serious side effects were observed. Investigations of the biodistribution of radionuclide (99mTc) labeled DPan and Panitumumab showed different kinetics. Both proteins showed the highest concentration in the tumor only at the end of the investigated period of 18 h, with panitumumab accumulating predominantly in the tumor. Regarding the general biodistribution in the body, panitumumab showed an increased plasma presence in the blood pool, whereas DPan accumulated strongly in the liver and spleen. Furthermore, it was shown that dianthin is already slightly cleaved from ADC in the blood plasma, presumably because a cleavable SPDP linker was used. However, since DPan in combination with SO1861 showed an excellent antitumor potential in vitro, which did not fully materialize in vivo, future work should nevertheless focus on optimizing the in vivo efficiency of DPan
Immunmodulatorische, anti-inflammatorische und antineoplastische Wirkungen der Naturstoffe aus Ganoderma lucidum, Tremella aurantia lba und Sargassum graminifolium
In vorliegender Arbeit wurden die biologische Aktivität einiger Fraktionen und gereinigter Stoffe aus Heilkräuter untersucht. GLP20 und JRP sind jeweils Polysaccharid-Fraktionen aus G. lucidum, bzw. T. aurantia lba. GLPss58 und JRP-SS sind die sulfatierten Fraktionen jeweils aus GLP20 und JRP. Sarg-Z ist Polysaccharide-Fraktion aus S. graminifolium. GLP20 und JRP-SS konnten den L-selektin-, Komplementssystem- oder Zytokin-vermittelte Entzündungsprozess herunterregulieren. Außer dem Anti-inflammatorischen Effekt zeigten GLPss58 und JRP-SS ein hohes Potential die Blutgerinnung verhindern zu können, insbesondere über den intrinsischen Weg. GLPss58 und JRP-SS zeigten auch einen Antitumor-Effekt, indem sie die Migration und Invation von Tumorzellen inhibieren und Wnt/ß-catenin-Signalweg in Melanomazellen aktivieren. Sarg-Z wies eine Multifunktion bei der Immunmodulation auf, sowohl bei der Aktivierung von Makrophagen und Lymphozyten, als auch bei der Regulierung von Entzündungsprozessen
Formation of protease inhibitors through template-assisted fragment-ligation and in-situ amidation
Fragment-basierte Ansätze zur Entwicklung neuer Arzneistoffe erfreuen sich immer größerer Beliebtheit. Dies liegt darin begründet, dass bereits durch den Einsatz und die Kombination von verhältnismäßig kleinen Fragment-Bibliotheken ein großer chemischer Raum abgedeckt werden kann. Als Startpunkt dient dabei meist ein Primärfragment, das durch geschicktes Anhängen weiterer funktioneller Gruppen oder durch Verknüpfung (Ligation) mit weiteren Fragmenten so erweitert wird, dass neue Interaktionspunkte mit der jeweiligen Zielstruktur entstehen. Hierdurch ist die Bildung hochaffiner Liganden möglich.
In der vorliegenden Arbeit wird die Amidierung als irreversible Form der Ligation zweier Fragmente näher untersucht. Die Arbeit gliedert sich dabei in zwei Teile. Im ersten Teil wurde ein Ligationsassay entwickelt, bei der die Amidierung Templat-unterstützt im Stil einer Proteintarget-geleiteten Synthese durchgeführt wird. Nach vorheriger Bindung der beiden Fragmente begünstigt dabei das Templat bzw. der Templat-Effekt die Reaktion der beiden Fragmente, wenn sich deren komplementäre funktionelle Gruppen in ausreichender Nähe zueinander befinden. Als Zielprotein wurde dabei Faktor Xa eingesetzt, an dem die Ligationsreaktion eines S1-bindenden Amin-Fragments, mit einem aktivierten Carbonsäure-Fragment Templat-unterstützt, durchgeführt werden sollte. Das Produkt dieser Reaktion ist ein nanomolar-aktiver Faktor Xa-Inhibitor. Die Templat-unterstützte Amidierung wurde dabei durch Umsetzung unterschiedlich reaktiver Carbonsäurederivate (Aktivester) nachgewiesen, indem die Reaktion in An- und Abwesenheit des
Proteintemplats untersucht wurde. Da im Zuge der Reaktion aus zwei millimolar-aktiven Fragmenten ein nanomolar-aktives Ligationsprodukt gebildet wird, konnte zur Detektion der Produktbildung ein Enzymaktivitätsassay verwendet werden. Dieser ermöglichte aufgrund der hohen Affinitätsunterschiede der Ausgangsfragmente im Vergleich zum
Fragment-Kombinationsprodukt bereits den Nachweis kleinster Produktmengen. Für den Phenyl- und den Trifluorethylester der Carbonsäure konnte dabei eine selektiv-templierte Reaktion nachgewiesen werden, d. h. die templierte Reaktion läuft ohne gleichzeitige
Reaktion durch nicht gebundene Fragmente ab (Hintergrundreaktion). Die Menge des gebildeten Produkts konnte nachfolgend mittels HPLC-QTOF-MS quantifiziert werden. Ebenfalls konnte durch die Untersuchung der Faktor Xa-Kristallstruktur mit dem co-
kristallisierten Fragment-Ligationsprodukt der Reaktionsmechanismus der Reaktion auf molekularer Ebene aufgeklärt werden. Abschließend konnten Reaktionsbedingungen
gefunden werden, die auch im Mikrotiterplatten-Maßstab eine erfolgreiche Umsetzung von Carbonsäuren zum entsprechenden Aktivester zulassen.
Im zweiten Teil wurde ein in-situ Screening gegen die NS2B-NS3-Protease des West-Nil-Virus durchgeführt. Dabei sollte eine aus 1615 Carbonsäure Fragmenten
bestehende Substanz-Bibliothek mit einem Amin-Fragment verknüpft werden. Das verwendete Amin stellte hierbei ein bekannter S1-Binder der WNV-Protease dar. Zur Ligation der Fragmente mussten zunächst geeignete Reaktionsbedingungen gefunden werden, die eine möglichst effiziente Umsetzung der Carbonsäuren zum entsprechenden Amid
gewährleisten. Hierbei stellten sich die Aktivierung mittels HBTU/DIPEA in einem DMF/DMSO-Gemisch und die Aktivierung mittels DCC/DMAP in Dioxan als geeignet heraus. Nachfolgend wurde das Protease-Screening durchgeführt. Zur Detektion wurde dabei ein Fluoreszenz-basierter Enzymaktivitätsassay eingesetzt. Unterstützend wurden in-silico Methoden verwendet. Nach der Validierung der erhaltenen Ergebnisse konnten durch geeignete Hit-Selektionsschritte drei Fragment-Ligationspaare ausgewählt werden, die in größerem Maßstab nachsynthetisiert wurden und anschließend näher untersucht wurden. Für die aussichtsreichste Fragment-Kombination CA3-Amid konnte dabei ein IC50 -Wert von 2,66 µM ermittelt werden.Fragment-based drug discovery approaches are getting popular more and more. This is due to fact that despite using and combining relatively small fragment-libraries a broad chemical space can be covered. In most cases a primary fragment is used as a starting point which consecutively gets expanded by further functional groups or through the
ligation with another fragment. Thereby additional interactions with the target structure can be formed that can finally lead to the formation of highly active ligands.
In the presented work, the use of the amidation reaction is investigated to link two fragments irreversibly. The work is divided into two parts. In the first part a ligation assay was developed that uses the template-assisted amidation in a “target-guided-synthesis” reaction. Here, the protein template accelerates the reaction of two fragments if both are bound to the template and if the complementary functional group of the two fragments are in the right orientation to each other. Factor Xa was thereby used as a test system. This factor Xa-template was subsequently used to establish a method to link an S1-
binding amine fragment to an activated carboxylic acid fragment. The reaction of the two fragments, here, leads to the formation of a nanomolar-active factor Xa inhibitor. To
investigate the occurrence of a template-assisted reaction several active esters were
synthesized, covering a broad spectrum of reactivity. These synthesized esters were subsequently tested to react in a template-assisted manner with the amine fragment in presence of the protein template. The reaction in abscence of the protein-template was used as a control. Since in the course of the ligation reaction two millimolar-active fragments are linked together to form a nanomolar-active ligation product, the formation of the product can be detected highly sensitively through the use of an enzyme activity assay. In the case of the phenyl- and trifluoroethyl-ester a selective templated reaction was
demonstrated without the formation of the ligation product through unbound fragments (background reaction). In later experiments the amount of the formed product was
quantified via HPLC-QTOF-MS. Furthermore using the crystal structure with the co-crystallized ligation product the reaction mechanism of the templated amidation reaction have been clarified. Finally, reaction conditions were found that allow the formation of active esters from carboxylic acids in microtiter plates.
In the second part an in-situ screening against the NS2B-NS3 protease of the west-nile virus was conducted. Firstly, a library containing 1615 carboxylic acid fragments were supposed to be linked to an amine fragment. In this case the used amine fragment was a known S1-binding ligand of the WNV protease. In the starting experiments several reaction conditions were investigated to find an efficient way to convert the carboxylic acids to the respective amides. The activation via HBTU/DIPEA in a DMF/DMSO-
mixture and the activition with DCC/DMAP in dioxane were found to be effective and were subsequently used for the protease screening. To detect inhibiting amides an enzyme
activity assay was performed using a fluorogenic substrate. As a support in-silico methods were used. After the validation of the identified screening hits using several hit selection approaches, three different hit-structure groups were chosen. One compound representing each group was synthesized in a larger scale and further investigated. For the best
fragment-combination CA3-Amid, an IC50 -value of 2.66 µM was found
Addictiveness and Attractiveness of Tobacco Additives: Analytical and Toxicological Characterisation
“The only way to avoid a smoking-related risk is not to smoke” (Baker, 2006).
Tobacco smoke consists of a multitude of harmful substances many of them known carcinogens. In addition, the tobacco alkaloid nicotine is a highly addictive compound, thus attempts to quit smoking in most cases fail. Therefore the best way to prevent smoking related diseases is to prevent people from getting started to smoke. The revision of the European Tobacco Product Directive 2014/40/EU (EU, 2014) is targeted on this approach. Concerns raised that tobacco additives could be used to facilitate the addictiveness and attractiveness of the product. And by that means to attract especially young people and entrap them to become addicted smokers. Under Article 6 of the relaunched Directive manufacturers and importers are required to submit data that confirm that the ingredients used do not
• increase the toxicity or the addictive potency of the product,
• result in a characteristic flavour,
• or facilitate inhalation or nicotine uptake.
To provide answers to this complex question, of whether tobacco additives can potentially enhance the effect of the inherently harmful and addictive consumer product tobacco, an on-line coupled oxidative pyrolysis technique was established and coupled to gas chromatography-mass spectrometry (GC/MS). By applying this instrumental tool, 19 different tobacco additives like raw cane sugar, liquorice or cocoa as well as additive-free tobacco were screened for their thermal decomposition products after pyrolysis. In the present work 72 different compounds with toxic potential were detected. Among these compounds, vinyl acrylate, fumaronitrile, methacrylic anhydride, isobutyric anhydride and 3-buten-2-ol were exclusively detectable upon pyrolysis of tobacco additives. By contrast, these toxicants revealed undetectable in tobacco smoke itself. The most frequently formed pyrolysis products were formaldehyde and acetaldehyde. A semi-quantitative approach used to look into the formation of polycyclic aromatic hydrocarbons during pyrolysis of cocoa and/or tobacco could demonstrate that the addition of cocoa to tobacco is unlikely to significantly alter the relative amounts of these highly carcinogenic compounds in the smoke of cigarettes refined with this flavour.
Article 7 of the Directive regulates the prohibition of additives that create a characteristic flavour in cigarettes and in roll-your-own tobacco. It remains unclear how to reliably measure such properties. A powerful chemical approach based on headspace (HS) solid phase microextraction (SPME) coupled to GC/MS was elaborated to identify chemical markers for complex fruity flavours. These marker substances can now be further used to assign manufacturers’ data on tobacco additives that have been employed to flavour their products. Considered as an exemplary case, the chemical marker substances for strawberry flavour were identified applying this method. This work resulted in a list of 11 different chemical compounds that appropriately indicate the presence of strawberry flavour. Overall the established approach provides a fast analysis of characteristic flavours used in tobacco products. For further clarification additional analytical means including sensory tests would be mandatory.
Mentholated cigarettes are part of the regulation of characteristic flavouring of tobacco products. As menthol is biologically active the issue was to be addressed whether tiny amounts of this compound in cigarettes that remain below the sensory threshold would still be sufficient to induce a physiological response, that is, a cooling sensation in the bronchial epithelia, and thus may facilitate deeper inhalation of tobacco smoke in the lungs of consumers. To this end, analytical methods were to be developed that would allow reliable quantification of the menthol levels in cigarettes. An analytical GC/MS based method has thus been established and validated to quantify this compound in tobacco products. Based on this we found that only some mentholated cigarettes contained menthol levels high enough to create a characteristic flavour. On the other hand, menthol contents of American blend, additive-free and light cigarettes remained below this sensory threshold. In these cases, however, the issue had to be addressed whether or not even such sub-sensory levels of menthol would still be capable of activating menthol responsive cold receptors. For that reason, a bioassay has been established suited to determine the minimum menthol content needed to activate the transient receptor potential cation channel subfamily M (melastatin) member 8 (TRPM8) protein. The experimental data show that menthol levels in cigarettes above 4 µg/cigarette are well be able to activate the TRPM8 cold receptor although they would not necessarily fall under the tobacco regulation of characteristic flavourings. The non-mentholated cigarettes investigated in our studies, however, did not exceed this value of 4 µg/cigarette. Yet it is known from the literature that there are non-mentholated cigarettes on the market with menthol contents certainly high enough to activate this TRPM8 receptor. Such cigarettes might therefore facilitate deeper inhalation, an increased nicotine uptake during smoking and, ultimately, an amplified addictiveness of the respective product.
For the assessment of chiral compounds that might also be applied as tobacco additives (i.e., menthol), the distinction between the individual enantiomers would be crucial. Since enantiomers usually differ in their physiological properties, their ability to activate the TRPM8 receptor or their kind of flavour perception could be different as well. As a most welcome side effect enantioselective analysis would also allow to identify the origin of the respective compound, and to decide whether it would be natural or synthetic. For that reason, an enantioselective (Es) GC/MS method has been developed to enable the distinction between enantiomers in selected terpenoids present in strawberry flavourings. The established method would also be suitable to identify the enantiomeric composition of menthol in tobacco products. This latter aspect would be of particular interest if menthol addition to tobacco products will be restricted by law. Since menthol occurs naturally in the leaves of the tobacco plant, an Es-GC/MS analysis could serve as authenticity proof for the origin of this compound in cigarettes.„Der einzige Weg zur Vermeidung rauch-induzierter Risiken ist nicht zu rauchen“ (Baker, 2006).

Darüber hinaus handelt es sich bei dem Tabak-Alkaloid Nikotin um einen sehr starken Suchtstoff, wodurch die meisten Versuche mit dem Rauchen aufzuhören missglücken. Von daher ist der beste Weg den Folgen des Tabakkonsums vorzubeugen, den Einstieg zum Tabakkonsum vorzubeugen. Die Neufassung der Tabak-Produktrichtlinie 2014/40/EU (EU, 2014) stützt sich auf diesen Gedanken. Bedenken kamen auf, dass Tabakzusatzstoffe dazu verwendet werden könnten, eine suchtverstärkende Wirkung oder eine Attraktivitätssteigerung des Tabakprodukts zu erzielen. Auf diese Weise würden besonders Jugendliche dazu verführt werden, mit dem Rauchen anzufangen. Unter Artikel 6 der Richtlinie werden Hersteller und Importeure zur Bereitstellung von Daten verpflichtet, die zeigen, dass die verwendeten Zusatzstoffe nicht dazu beitragen,
• die Toxizität oder die Attraktivität des Produktes zu erhöhen,
• zu einem charakteristischen Aroma führen,
• oder die Inhalation sowie die Nikotinaufnahme zu erhöhen.
Zur Klärung der komplexen Fragestellung, ob Tabakzusatzstoffe potentiell dazu in der Lage sind, die Wirkung des an sich bereits gesundheitsschädlichen und suchterzeugenden Verbraucherprodukts Tabak zu steigern, wurde eine Methode zur oxidativen Pyrolyse, on-line gekoppelt an die Gaschromatographie mit anschließender Massenspektroskopie (GC/MS), entwickelt. Mit dieser Methode wurden 19 verschiedene Tabakzusatzstoffe wie Rohrohrzucker, Süßholz, Kakao, etc., sowie zusatzstofffreier Tabak hinsichtlich ihrer thermischen Zerfallsprodukte nach der Pyrolyse analysiert. In der vorliegenden Arbeit konnten 72 verschiedene Substanzen mit toxischem Potential detektiert werden. Darunter befanden sich Stoffe wie Vinylacrylate, Fumaronitril, Methacrylsäureanhydrid, Isobuttersäureanhydrid und 3-Buten-2-ol, die ausschließlich während der Pyrolyse von Tabakzusatzstoffen gebildet werden. Die am regelhaftesten durch Pyrolyse gebildeten Substanzen waren Formaldehyd und Acetaldehyd. Weiterhin wurde in einem semi-quantitativen Ansatz die Bildung von Polycyclischen Aromatischen Kohlenwasserstoffen nach der Pyrolyse von Kakao und/oder Tabak untersucht. Hierbei konnte gezeigt werden, dass die Zugabe von Kakao zu dem Tabak die relativen Mengen der untersuchten Verbindungen nicht beeinflusst.
Artikel 7 der Richtlinie reguliert das Verbot von Zusatzstoffen die zur Bildung von charakteristischen Aromen in Zigaretten und Tabak zum Selbstdrehen beitragen. Wie diese Eigenschaften gemessen werden sollen bleibt unklar. Aus diesem Anlass wurde im Rahmen der vorliegenden Arbeit eine Untersuchungsmethode entwickelt, die es ermöglichen soll, basierend auf Headspace (HS) Festphasen Microextraktion (SPME), gekoppelt an eine GC/MS-Analytik, chemische Marker Substanzen für komplexe fruchtige Aromen zu identifizieren. Diese chemischen Marker könnten dann zukünftig genutzt werden, um die von der Tabakindustrie übermittelten Daten zu den verwendeten Zusatzstoffen zu überprüfen und die Präsenz charakteristischer Aromen nachzuweisen. Mittels der etablierten Methode wurden exemplarisch die chemischen Marker für das Erdbeeraroma definiert. Dabei ergab sich eine Gruppe von 11 verschiedenen chemischen Verbindungen, die für die Verwendung als Erdbeeraroma-Marker geeignet sind. Dieser Ansatz bietet somit eine Option zur analytischen Prüfung der Anwesenheit von charakteristischen Aromen in Tabakerzeugnissen. Ergänzende Verfahren, beispielsweise sensorische Tests, könnten dann zur abschließenden Verifikation eingesetzt werden.
Menthol-Zigaretten fallen ebenfalls unter die Regulierung charakteristischer Aromen in Tabakerzeugnissen. Da es sich bei Menthol um eine biologisch aktive Substanz handelt, muss untersucht werden, ob auch Menthol-Gehalte unterhalb der sensorischen Wahrnehmungsgrenze ausreichend sind, um einen kühlenden Effekt in Bronchialzellen erzeugen zu können, welcher dann zur erleichterten Inhalation von Tabakrauch beitragen würde. Für diese Fragestellung werden analytische Methoden benötigt, die es erlauben, den Menthol-Gehalt in Zigaretten und Zigarettenrauch zu bestimmen. Im Rahmen dieser Arbeit wurde eine GC/MS-basierte Methode erarbeitet und validiert, um Menthol in Tabakprodukten zu quantifizieren. Bei einigen der Menthol-Zigaretten konnte ein ausreichend hoher Menthol-Gehalt nachgewiesen werden, der für die Ausprägung eines charakteristischen Aromas im Produkt maßgeblich war. Die Menthol-Gehalte der untersuchten „American Blend“-, zusatzstofffreien und „Light“-Zigaretten lagen dagegen unterhalb der Wahrnehmungsgrenze. Daher ergab sich die anschließende Fragestellung, ob ein solch geringer Menthol-Gehalt in Zigaretten ausreichend ist, um den Menthol-Kälterezeptor zu aktivieren und damit zur Sensorik eines kühlenden Effekts während des Rauchens beizutragen. Aus diesem Grund wurde im Rahmen dieser Arbeit ein Bioassay etabliert, der es erlaubt, die minimale Menge an Menthol zu bestimmen, die benötigt wird, um das Transient Receptor Potential Cation Channel Subfamily M (Melastatin) Member 8 (TRPM8) Protein zu aktivieren. Die daraus resultierenden Daten zeigen, dass Menthol-Gehalte ab 4 µg/Zigarette zwar nicht zwangsläufig unter die Definition der charakteristischen Aromen fallen, aber dennoch in der Lage sind, den TRPM8 zu aktivieren. Keine der hier untersuchten mentholfreien Zigaretten überschritt den Wert von 4 µg/Zigarette. Allerdings geht aus der Literatur hervor, dass angeblich mentholfreie Zigaretten auf dem Markt erhältlich sind, die ausreichend Menthol enthalten, um den TRPM8 zu aktivieren. Solche Zigaretten könnten ebenfalls die tiefere Inhalation des Tabakrauchs und damit die Nikotinaufnahme erleichtern.
Für die Beurteilung von chiralen Verbindungen, die als Tabakzusatzstoffe genutzt werden (z.B. Menthol), kann die Unterscheidung zwischen den Enantiomeren wichtig werden. Die individuellen Enantiomere einer chiralen Verbindung können sich prinzipiell in ihren physiologischen Eigenschaften, beispielsweise in ihrer Fähigkeit, den TRPM8 zu aktivieren, oder in ihrer Ausprägung einer speziellen Geruchsnote, unterscheiden. Zusätzlich eignet sich eine enantioselektive Analytik auch dafür, Schlüsse über die Herkunft (natürlich oder synthetisch?) einer chiralen Substanz zu ziehen. Aus diesem Grund wurde eine enantioselektive (Es) GC/MS-Methode entwickelt, die es ermöglicht, eine Unterscheidung zwischen den Enantiomeren einiger Terpene des Erdbeeraromas vorzunehmen. Diese Methode ließe sich dann auch zur Trennung der Enantiomeren von Menthol, das Tabakprodukten zugesetzt wird, verwenden. Eine solche Möglichkeit wäre von großem Nutzen, wenn Menthol als Zusatzstoff verboten wird, da die Verbindung auch natürlicherweise in den Blättern der Tabakpflanze vorkommt. Mithilfe der hier entwickelten Es-GC/MS-Methode könnten somit Rückschlüsse über die Herkunft des Menthols in Zigaretten gezogen werden
