Design of fluorescent false substrates (FFS) to study vesicular nucleotide transporter and Sialin
Le concept de composés appelés FFS, pour faux substrats fluorescents (ou FFN pour les neurotransmetteurs), a été introduit par le groupe du Dr Sames qui a conçu et optimisé des molécules fluorescentes reconnues comme substrats par le Transporteur Vésiculaire de Monoamine (VMAT2). Ces composés, une fois accumulés dans des vésicules synaptiques, ont permis la visualisation de l’activité neuronale monoaminergique en temps réel. Ce concept a servi d’hypothèse pour ce travail de thèse. En effet, nous avons conçu, synthétisé et évalué la potentialité de deux familles chimiques comme substrats fluorescents de transporteurs de la famille SLC17 : VNUT (transporteur vésiculaire de nucléotides) et la Sialine (transporteur d’acide sialique). Les molécules fluorescentes synthétisées devront être capables de remplacer/mimer les substrats naturels de VNUT et de la Sialine (respectivement l’ATP et le Neu-5-Ac) et de couvrir une large gamme de longueurs d’ondes d’émission pour nous adapter aux problématiques biologiques. L’élaboration de tels composés a été effectuée par conception rationnelle en synergie avec les résultats des tests d’inhibition et de transport et à partir de touches issues du criblage virtuel. Deux composés ont été plus particulièrement étudiés. D’une part, l’ATP a été modifiée en remplaçant le motif adénine par un fluorophore (éthéno, coumarines, quinoléines…). D’autre part, une touche virtuelle (Fmoc-Lys(Cbz)-OH) issue d’un vHTS sur la Sialine a été sélectionnée pour sa structure facilement modifiable, qui a permis l’incorporation aisée de motifs fluorescents. Une cinquantaine de composés a donc été synthétisée et évaluée pour leur capacité d’inhibition ou de substrat sur les cibles sélectionnées. Les résultats prometteurs de plusieurs composés en tant qu’inhibiteur laissent de nombreuses perspectives pour la compréhension de la machinerie vésiculaire.The concept of compounds called FFSs for false fluorescent substrates (or FFNs for neurotransmitters) was introduced by the group of Dr Sames who designed and optimized fluorescent molecules recognized as substrates by the Vesicular Monoamine Transporter (VMAT2). Once accumulated in synaptic vesicules, these compounds have allowed visualization of the monoaminergic neuronal activity in real time. This concept has been used as hypothesis for this thesis work. Indeed, we have designed, synthesized and evaluated the potential of two chemical families as fluorescent substrates of SLC17 transporter family: VNUT (Vesicular Nucleotide Transporter) and Sialin (sialic acid transporter). The synthesized fluorescent molecules must be able to replace/mimic the natural substrates of VNUT and Sialin ( ATP and Neu-5-Ac, respectively) and to cover a wide range of emission wavelengths to deal with biological problems. The development of such compounds was performed by rational design in synergy with the results of inhibition and transport tests and by virtual screening. Two compounds have been specifically studied. On one hand, ATP was modified by replacing the adenine pattern by a fluorophore (etheno, coumarins, quinolines…). On the other hand, a virtual hit (Fmoc-Lys(Cbz)-OH), derived from vHTS targeted to Sialin, was selected for the ease to incorporate fluorescent patterns. Therefore, fifty compounds were synthesized and evaluated for their inhibitory or substrate ability on selected targets. Going Beyond Counting First Authors in Author Co-citation Analysis
The present study examines one of the fundamental aspects of author co-citation analysis (ACA) - the way co-citation
counts are defined. Co-citation counting provides the data on which all subsequent statistical analyses and mappings
are based, and we compare ACA results based on two different types of co-citation counting - the traditional type that
only counts the first one among a cited work's authors on the one hand and a non-traditional type that takes into
account the first 5 authors of a cited work on the other hand. Results indicate that the picture produced through this non-traditional author co-citation counting contains more coherent author groups and is therefore considerably clearer. However, this picture represents fewer specialties in the research field being studied than that produced through the traditional first-author co-citation counting when the same number of top-ranked authors is selected and analyzed. Reasons for these effects are discussed
Synthesis of fluorescent probes targeting VGLUTs for diagnosis and pharmacological studies
Le glutamate est le neurotransmetteur excitateur majeur du système nerveux. Avant sa libération par exocytose, le glutamate est accumulé dans les vésicules synaptiques par les transporteurs vésiculaires glutamates (VGLUT1-3). LesVGLUTs sont des acteurs majeurs de la transmission glutamatergique et leurs implications dans les pathologies neurologiques et pathologiques a été bien établie au cours des 10 dernières années. Ils représentent des cibles thérapeutiques clés pouvant être utilisés spécifiquement pour les études physiopathologiques ou le diagnostic des maladies impliquant le système glutamatergique. Toutefois malgré leur importance notoire la pharmacologie de ces transporteurs reste très pauvre et limite les investigations. Les modulateurs sélectifs développés à ce jour se limitent à quelques analogues du substrat naturel et les colorants azoïques comme le Bleu Trypan. Notre projet porte sur le développement d'outils pharmacologiques fluorescents sélectifs et affins des VGLUTs. Ces modulateurs seront utilisés pour mieux comprendre l'implication des VGLUTs dans les maladies neurodégénératives mais également dans les études sur la compréhension de la neurotransmission glutamatergique, les études physiopathologiques ou le diagnostic de maladies impliquant le système glutamatergique. Pour atteindre ces objectifs nous avons développé des bioisostères fluorescents du Bleu Trypan en pharmacomodulant la tête polaire naphtyle par des fluorophores coumarines et quinoléines. L'activité de ces composés a été mesurée in vitro sur vésicules synaptiques purifiées à partir de cerveaux de rats.Vesicular Glutamate Transporters (VGLUTs) allow the loading of presynapic glutamate vesicles and thus play a critical role in glutamatergic synaptic transmission. The implication of VGLUTs in several major neuropathologies as Alzheimer or Parkinson's disease have characterized during the last decades. Moreover, VGLUTs represent the best biomarker for glutamatergic neuropathologies and a key biological target for their diagnostic or treatment. Despite the pivotal role of VGLUTs, their pharmacology appears quite limited. Known competitive inhibitors are restricted to some dyes, such as Trypan Blue and glutamate analogues. The lack of pharmacological tools has heavily hampered VGLUT investigations. To better understand VGLUT implications, we proposed to synthesize specific VGLUT inhibitors with high selectivity and improved pharmacological properties. In particular, we focused our attention in the design of fluorescent analogues. Indeed, fluorescent compounds mimicking glutamate and targeting VGLUTs could be used as imaging biomarkers to study glutamatergic neurotransmission in physio-pathological context. We thus selected fluororescent scaffolds, such as coumarin and quinoline to perform scaffold hoping of Trypan Blue naphtyl moiety. Variations on the Author
“Variations on the Author” discusses two of Eduardo Coutinho’s recent films (Um Dia na Vida, from 2010, and Últimas Conversas, posthumously released in 2015) and their contribution to the general question of documentary authorship. The director’s filmography is characterized by a consistent yet self-effacing form of authorial self-inscription: Coutinho often features as an interviewer that rather than express opinions propels discourses; an interviewer that is good at listening. This mode of self-inscription characterizes him as an author who is not expressive but who is nonetheless markedly present on the screen. In Um Dia na Vida, however, Coutinho is completely absent form the image, while Últimas Conversas, on the contrary, includes a confessional prologue that moves the director from the margins to the center of his films. This article examines the ways in which these works stand out in the filmography of a director who offers new insights into the notion of cinematic authorship
Appropriate Similarity Measures for Author Cocitation Analysis
We provide a number of new insights into the methodological discussion about author cocitation analysis. We first argue that the use of the Pearson correlation for measuring the similarity between authors’ cocitation profiles is not very satisfactory. We then discuss what kind of similarity measures may be used as an alternative to the Pearson correlation. We consider three similarity measures in particular. One is the well-known cosine. The other two similarity measures have not been used before in the bibliometric literature. Molécules photoisomérisables : conception, synthèse, caractérisation et application pharmacologique
Les composés photoisomérisables ou « photoswitchs » sont capables de changer de structure lors d'une illumination à différentes longueurs d'onde. L'azobenzène est l'une des molécules les plus répandues dans les applications biologiques en raison de sa capacité à s'isomériser entre les formes trans et cis. Sous lumière UV, cette molécule subit une isomérisation de la forme trans thermodynamiquement plus stable à la forme cis métastable. L'approche « azologisation » consiste à introduire un groupement azobenzène isostère dans la structure centrale du composé de référence. Les récepteurs métabotropiques du glutamate (mGluR) font partie des RCPG de classe C qui se caractérisent par un grand domaine amino-terminal replié en deux lobes reliés par une charnière flexible, nommée VFT (Venus FlyTrap) où se trouve le site de liaison. Il est situé près de la charnière où deux sites chlorure ont été identifiés dans des sites adjacents du domaine de liaison. Notre groupe a découvert le LSP4-2022, un agoniste mGlu4R puissant et sélectif, qui contient une partie de type glutamate et une partie distale contenant un carboxylate qui fournit l'affinité et la sélectivité et est proposé pour se lier à l'un ou l'autre des sites chlorure. Afin de développer des outils photopharmacologiques pour sonder si le LSP4-2022 est capable de se lier aux deux sites chlorure de mGluR4, nous avons conçu un photoswitch en remplaçant la partie acide phénoxyacétique par le 5-(phényldiazényl)-1H-tétrazole, capable de fournir la charge négative nécessaire. Cependant, l'obtention de l'intermédiaire clé, le 4-((1H-tétrazol-5-yl)diazényl)benzaldéhyde, a échoué malgré diverses approches en raison de l'instabilité du diazényltétrazole en milieu acide. Pour contourner ce problème, nous nous sommes tournés vers le 2,6-difluoro-4-(phényldiazényl)phénol, dont le pKa est inférieur à 7, et qui pourrait également fournir des charges négatives dans le tampon PBS (pH = 7,4). Bien que la puissance de cet analogue du LSP4-2022 soit très réduite par rapport au LSP4-2022, ce composé reste un agoniste du récepteur mGlu4. Des études ont montré que l'azobenzène substitué en ortho par des atomes de fluor peut augmenter efficacement la stabilité thermique de la forme cis. Ainsi, nous avons synthétisé 3 types de 4-phénylazophénol comportant différents atomes de fluor dans différentes positions. Le spectre UV-Vis de l'unité la plus prometteuse a montré une courbe typique d'isomérisation réversible dans le toluène, mais pas dans le tampon PBS, montrant que la tautomérisation est favorisée dans un solvant polaire. Pour surmonter ce problème, nous avons alors déplacé le groupe hydroxyle en position meta de l'azobenzène. La synthèse des dérivés meta-(phénylazo)phénol s'est avérée beaucoup plus difficile que prévu. Après de nombreuses tentatives, nous avons finalement réussi à synthétiser une série de dérivés possédant 0 à 4 atomes de fluor autour de la liaison azoïque. Comme prévu, en déplaçant l'hydroxyle de la position para à la position meta tout en gardant les deux atomes de fluor de chaque côté de l'hydroxyle a considérablement amélioré la stabilité thermique de l'isomère Z dans le tampon PBS sans photofatigue. Parmi ces composés, nous avons constaté que plusieurs composés présentaient d'excellentes propriétés photoswitchs à la fois dans le DMSO et dans le tampon. De plus, la forme trans peut être isomérisée en cis avec une lumière à 490 nm offrant un plus grand avantage pour les applications biologiques.Pour profiter des propriétés photophysiques des dérivés du 2,6-difluoro-3-(phényldiazényl)phénol, nous les avons ensuite couplés à la partie glutamate du LSP4-2022 pour obtenir des ligands finaux afin de développer des outils photopharmacologiques et sonder si ces analogues du LSP4-2022 sont capables de se lier aux deux sites chlorure du mGlu4R. Cependant, ces dérivés présentent une activité agoniste modeste sur le récepteur mGlu4 sans différence claire entre les isomères trans et cis.Photoswitches are moieties able to change their structure upon irradiation with different wavelengths of light. Azobenzene is one of the most prevalent molecules in biological applications due to its ability to isomerize between the trans and the cis isomers. When irradiated with UV light, this molecule undergoes isomerization from the thermodynamically more stable trans form to a metastable cis form. "Azologization" approach consists in introducing an isosteric azobenzene moiety into the core structure of the reference compound. Metabotropic glutamate receptors (mGluR) are members of the class C GPCRs that are characterized by a large amino-terminal domain folded in two lobes linked by a flexible hinge, named VFT (Venus FlyTrap) where the binding site is found. It is located near the hinge and two chloride binding sites have been identified in adjacent sites. Our group has discovered LSP4-2022, a potent and selective mGlu4R agonist, which contains a glutamate-like part and a distal part holding a carboxylate that provides affinity and selectivity and is proposed to bind to either chloride sites. In order to develop photopharmacological tools to probe whether LSP4-2022 is able to bind to both chloride sites of the mGluR4 binding domain, we initially designed a photoswitch compound by replacing the phenoxyacetic acid part with a 5-(phenyldiazenyl)-1H-tetrazole, to provide the negative charge that mimics the chloride ion. However, obtaining the key intermediate, 4-((1H-tetrazol-5-yl)diazenyl)benzaldehyde, failed despite various approaches due to the instability of the diazenyltetrazole in acidic medium. To circumvent this problem, we turned to 2,6-difluoro-4-(phenyldiazenyl)phenol, whose pKa is lower than 7, and could also provide negative charges in PBS buffer (pH= 7.4) to mimic the chloride ions. Although the potency of the LSP4-2022 analog is much reduced compared to LSP4-2022, this compound remains an mGlu4 receptor agonist. Previous studies have shown that ortho substituted azobenzene with fluorine can effectively increase the cis form thermal stability. Thus, we designed and synthesized three types of 4-phenylazophenol with different numbers and positions of fluorine atoms. Among them, the UV'Vis spectrum of the most promising unit showed a typical curve in toluene allowing the reversible photoswitching, whereas in PBS buffer, they overlap. We assume that tautomerization effect of the para-(phenylazo)phenol is very strong in a polar solvent. To overcome this problem, we then decided to move the hydroxyl group to the meta-position. The synthesis process of 3-phenylazophenol derivatives turned out to be much more difficult than expected. After many attempts, we finally succeeded to synthesize a series of meta-(phenylazo)phenol with 0 to 4 fluorine atoms around the azo bond. As expected, the change by moving the para hydroxyl to the meta position while keeping the two fluorine atoms on either side of the hydroxyl greatly enhanced thermal stability of the Z isomer in PBS solution without photofatigue. Among those compounds, we found that several compounds showed excellent photoswitchable properties both in DMSO and assay buffer. Moreover, the trans form can be switched to the cis isomer with 490 nm light that provides greater advantage for biological applications. To benefit from optimal photophysical properties of these photoswitching units, we next coupled the 2,6-difluoro-3-(phenyldiazenyl)phenol photoswitching units and the glutamate-like part of LSP4-2022 to get the final ligands in order to develop photopharmacological tools to probe whether LSP4-2022 is able to bind to both chloride sites of mGlu4R binding domain. However, these derivatives display modest mGlu4 receptor agonist activity with no clear difference between trans and cis isomers
Dispelling the Myths Behind First-author Citation Counts
We conducted a full-scale evaluative citation analysis study of scholars in the XML research field to explore just how different from each other author rankings resulting from different citation counting methods actually are, and to demonstrate the capability of emerging data and tools on the Web in supporting more realistic citation counting methods. Our results contest some common arguments for the continued
use of first-author citation counts in the evaluation of scholars, such as high correlations between author rankings by first-author citation counts and other citation
counting methods, and high costs of using more realistic citation counting methods that are not well-supported by the ISI databases. It is argued that increasingly available digital full text research papers make it possible for citation analysis studies to go beyond what the ISI databases have directly supported and to employ more

We have done our best to complete the author checklist relating to the use of animals in the hut study. Note that the objective for the hut study was to evaluate the IRS treatment applications for residual efficacy against Anopheles mosquitoes, including the local An. coluzzii mosquito population. Cows were only used to attract mosquitoes into the huts and no tests were carried out directly on the cows. The author checklist is intended for use with studies where experiments are carried out on animals, which is why we have had such difficulty in completing this for the hut study, as many of the questions do not relate to how the cows were used
