    Trabectedin for the treatment of soft tissue sarcomas

    Introduction: Trabectedin, a marine-derived DNA-binding antineoplastic agent, has been registered by the EMA and recently also by the FDA for the treatment of patients with advanced soft-tissue sarcoma (STS), a rare and heterogeneous disease. Areas covered: The antitumor activity of trabectedin is related both to direct effects on cancer cells, such as growth inhibition, cell death and differentiation, and indirect effects related to its anti-inflammatory and anti-angiogenic properties. Furthermore, trabectedin is the first compound that targets an oncogenic transcription factor with high selectivity in mixoid liposarcomas. This peculiar mechanism of action is the basis of its clinical development. The clinical pharmacology of trabectedin, the subsequent phase I, II and III trials are summarized and put into perspectives in this review. Expert opinion: Trabectedin is a relevant pleiotropic antitumoral agent within the complex scenario of the management of STS. It can be used in advanced STS, either after failure of anthracyclines and ifosfamide or in patients unfit for these drugs, especially when reaching a high-tumor control and a long-term benefit is a priority. Toxicity profile is acceptable and manageable with no reported cumulative toxicities. Therefore, trabectedin has become one relevant therapeutic option in metastatic STS, especially in selected histologies. © 2016 Informa UK Limited, trading as Taylor & Francis Group

    Orientador: Prof. Rui MaggiMonografia (graduação) - Universidade Federal do Paraná, Setor de Ciências Agrárias, Curso de Engenharia Industrial Madeireira.Resumo: Este trabatho se dedicou em estudar a viabilidade técnica da implantação de um sistema de georreferenciamento de empresas clientes da Marrari Automação Industrial. O estudo mostrou que a aplicação deste sistema poderá trazer benefícios â empresa no que tange oportunidades econômicas e economia de tempo e custo com rotas definidas em GPS. Custos iniciais foram levantados para a aquisição do programa que compila os dados (ArcGis). Porém, acredita-se que a longo prazo, os benefícios sejam maiores do que o ônus da aquisição do referido programa. A Marrari é uma empresa brasileira que existe há mais de vinte anos e tem crescido bastante no mercado. Com o avanço tecnológico particular, a Marrari vem abrindo o seu leque de serviços e soluções para atender, cada vez melhor, o cliente final que deseja se automatizar com qualidade. Este trabalho então surgiu para que, logisticamente, a empresa também ganhasse em qualidade na hora de se planejar para distribuir seus funcionários vis itando os clientes e mapeando seu campo de novas oportunidades. Logo, o banco de dados que no início era só acessado por um Software no computador, agora, com seu georreferenciamento, ganhou um apoio e ficou mais prático de situar as localizações das empresas por uma determinada região brasileira. No caso, foi feito um levantamento apenas de empresas localizadas, na região próxima a Rio Branco do Sul - PR, ao longo da Rodovia dos Minérios (Norte de Curitiba - Aproximadamente 38 km da Marrari ). Durante as visitas aos clientes foi gravado as coordenadas geográficas do posicionamento de cada Indústria no mapa para, futuramente com o auxílio de um Software (por exemplo: ArcGis), ter um banco de dados georreferenciados completo

    Going Beyond Counting First Authors in Author Co-citation Analysis

    The present study examines one of the fundamental aspects of author co-citation analysis (ACA) - the way co-citation counts are defined. Co-citation counting provides the data on which all subsequent statistical analyses and mappings are based, and we compare ACA results based on two different types of co-citation counting - the traditional type that only counts the first one among a cited work's authors on the one hand and a non-traditional type that takes into account the first 5 authors of a cited work on the other hand. Results indicate that the picture produced through this non-traditional author co-citation counting contains more coherent author groups and is therefore considerably clearer. However, this picture represents fewer specialties in the research field being studied than that produced through the traditional first-author co-citation counting when the same number of top-ranked authors is selected and analyzed.     Variations on the Author

    “Variations on the Author” discusses two of Eduardo Coutinho’s recent films (Um Dia na Vida, from 2010, and Últimas Conversas, posthumously released in 2015) and their contribution to the general question of documentary authorship. The director’s filmography is characterized by a consistent yet self-effacing form of authorial self-inscription: Coutinho often features as an interviewer that rather than express opinions propels discourses; an interviewer that is good at listening. This mode of self-inscription characterizes him as an author who is not expressive but who is nonetheless markedly present on the screen. In Um Dia na Vida, however, Coutinho is completely absent form the image, while Últimas Conversas, on the contrary, includes a confessional prologue that moves the director from the margins to the center of his films. This article examines the ways in which these works stand out in the filmography of a director who offers new insights into the notion of cinematic authorship

    Appropriate Similarity Measures for Author Cocitation Analysis

    We provide a number of new insights into the methodological discussion about author cocitation analysis. We first argue that the use of the Pearson correlation for measuring the similarity between authors’ cocitation profiles is not very satisfactory. We then discuss what kind of similarity measures may be used as an alternative to the Pearson correlation. We consider three similarity measures in particular. One is the well-known cosine. The other two similarity measures have not been used before in the bibliometric literature.     Multipeptide vaccination in cancer patients

    Since the identification of tumor associated antigens (TAA) in different tumor histotypes, many vaccination strategies have been investigated, including peptide-based vaccines. Results from the first decade of clinical experimentation, though demonstrating the feasibility and the good toxicity profile of this approach, provided evidence of clinical activity only in a minority of patients, despite inducing immunization in up to 50% of them. In this review, we discuss the different approaches recently developed in order to induce stronger peptide-induced immune-mediated tumor growth control, possibly translating into improved clinical response rates, with specific focus on multipeptide-based anti-cancer vaccines. This strategy offers many advantages, such as the possibility of bypassing tumor heterogeneity and selection of antigen (Ag)-negative clones escaping peptide-specific immune responses, or combining HLA class I- and class II-restricted epitopes, thus eliciting both CD4- and CD8-mediated immune recognition. Notably, advances in Ag discovery technologies permit further optimization of peptide selection, in terms of identification of tumor-specific and unique TAA as well as Ags derived from different tumor microenvironment cell components. With the ultimate goal of combining peptide selection with patient-specific immunogenic profile, peptide based anti-cancer vaccines remain a promising treatment for cancer patients, as attested by of pre-clinical and clinical studies
