    Touching Freud's dog: H.D.'s tactile poetics

    "Do not touch me", Frau Emmy warns Freud in 1889. "Do not touch", Freud echoes in 1933. This time, he is referring to his pet chow, Yofi, warning H.D. that "she snaps - she is very difficult with strangers". Examining the prohibition in light of work by Jacques Derrida and Jean-Luc Nancy, this article charts the withdrawal that always interrupts touch. Despite Freud's taboo, however, H.D.'s writing seeks to make contact in strange and unnerving ways. Developing Julia Kristeva's account of the semiotic, this paper proposes a literature of touch. Reading H.D.'s poems, alongside Tribute to Freud, and her letters, the author demonstrates that H.D.'s poetics are always haunted by the very (im)possibility of contact

    Provenance-based trust for grid computing: Position Paper

    Current evolutions of Internet technology such as Web Services, ebXML, peer-to-peer and Grid computing all point to the development of large-scale open networks of diverse computing systems interacting with one another to perform tasks. Grid systems (and Web Services) are exemplary in this respect and are perhaps some of the first large-scale open computing systems to see widespread use - making them an important testing ground for problems in trust management which are likely to arise. From this perspective, today's grid architectures suffer from limitations, such as lack of a mechanism to trace results and lack of infrastructure to build up trust networks. These are important concerns in open grids, in which "community resources" are owned and managed by multiple stakeholders, and are dynamically organised in virtual organisations. Provenance enables users to trace how a particular result has been arrived at by identifying the individual services and the aggregation of services that produced such a particular output. Against this background, we present a research agenda to design, conceive and implement an industrial-strength open provenance architecture for grid systems. We motivate its use with three complex grid applications, namely aerospace engineering, organ transplant management and bioinformatics. Industrial-strength provenance support includes a scalable and secure architecture, an open proposal for standardising the protocols and data structures, a set of tools for configuring and using the provenance architecture, an open source reference implementation, and a deployment and validation in industrial context. The provision of such facilities will enrich grid capabilities by including new functionalities required for solving complex problems such as provenance data to provide complete audit trails of process execution and third-party analysis and auditing. As a result, we anticipate that a larger uptake of grid technology is likely to occur, since unprecedented possibilities will be offered to users and will give them a competitive edge

    "This article reviews the origins, development and impact of the European Employment Strategy against the backdrop of the employment performances and policies of the EU, from the early 1990s up to the mid-term review of the Lisbon Strategy in 2005. It recalls how the issue of employment was put at the centre of the policy debate in the 1990s and of the discussion about the right balance between economic and social policy integration at EU level. While the EU-wide emphasis on employment initiated in the mid-1990s is likely to have had some positive effects, in particular in terms of a return to employment growth and an increase in participation and employment rates, many major challenges remain or have developed in the meantime. The experience of the EES shows that there is room for a European strategy to promote growth and jobs."     The regions of the infectious bursal disease virus (IBDV) genome with regulatory function are not known. In the present study, progressively deleted lengths of the 5' noncoding region of segment A were constructed in pGL3 vectors having SV40 enhancer or promoter, and a luciferase (LUC) reporter gene. Transient transfections of the constructs made in a promoter-less pGL3-Enhancer vector when transfected in Vero cells and the lysates assayed for LUC expression, allowed the localization of maximal activity to the 32-nucleotide stretch (precursor polyprotein ORF positions -131 to -100), which is highly conserved at the 5' end of both genome segments. This fragment, when evaluated in parallel in an enhancer-less pGL3-Promoter vector demonstrated no activity. To determine if this region is recognized by IBDV replicative proteins, we engineered modifications in an enhancer-less pGL3-Promoter vector where the terminal 32-bp fragment, the full-length noncoding region, or the noncoding region with the 32-bp fragment deleted was positioned in either the plus-sense or the minus-sense orientation immediately downstream of the SV40 promoter and upstream of the LUC gene. Transfections of these constructs in IBDV-infected and uninfected Vero cells resulted in the endogenous generation of recombinant viral-LUC RNAs containing the 5' terminal viral RNA sequences in either the plus-sense or the minus-sense orientation. LUC assays of the infected cell lysates showed up-regulated expression of LUC only with constructs containing the 32-bp fragment in the minus-sense orientation. Deletion of this 32-bp fragment abolished such LUC expression. We therefore conclude that the 5'-terminal 32 base pairs of genomic segment A contain a major promoter element in IBDV.     Sexual dimorphism of hepatic epigenetic marks and machinery in offspring of obese and diabetic mothers fed a control diet during periconceptional/gestation/lactation period

    National audienceEarly nutritional events may have an influence on later life health mainly through epigenetic processes.1 In our two- generation mice model, providing obese and diabetic mice with a control diet during the periconceptional/gestation/ lactation period led to a pronounced sex-specific shift from susceptibility to resistance to a high-fat diet (HFD) in the female offspring only.2,3 The aim of this study was to detect sex-specific differences in the expression of candidate genes and epigenetic marks and machinery in the liver of both sexes and both generations. As a key organ for lipid processing and detoxification, liver plays a major role in conditions of chronic lipid oversupply. According to the sex, female (F) or male (M), the generation, first (F1) or second (F2), and diet types, CD or HFD, mice were divided into eight groups (F-F1-CD, F-F1-HFD, M-F1-CD, M-F1-HFD, F-F2-CD, F-F2-HFD, M-F2-CD and M-F2-HFD). Body weight, blood glucose level and blood cholesterol levels were measured. Liver morphology was identified by hematoxylin–eosin staining and oil red O staining. Hepatosteatosis was found to be more common in all HFD groups with adaptation of the liver phenotype in F2 females but not in males, in parallel with obesity and cholesterol levels.4 Global DNA methyla- tion and histone modifications were investigated by LUMA and Western blot analysis, respectively. Interestingly, although no significant difference was found within groups, global DNA methylation level was significantly negatively correlated to steatosis percentage. Using RT-qPCR, sexual dimorphism was observed for the gene expression of 12 genes encoding enzymes of the epigenetic machinery. These marks may help us to understand the sex-specific epigenetic mechanisms of the underlying sex-specific responses to HFD and improve the early life nutritional environment in a sex-specific manner.     Risk based inspection of flood defence dams: An application to grass revetments

    In The Netherlands, inspection and maintenance are essential for maintaining stringent flood protection standards. Flood defences are assessed every 12 years to ensure they meet their risk-based safety standards,which are given as legally-binding maximum failure probabilities. Between assessments, flood defence managers are subject to risk-maintenance requirements: they must ensure that the failure probability of the defence does not increase in excess of its safety standard. However, there is no prescribed methodology to meet this requirement. In the study presented here, we developed a method which enables flood defence managers to derive inspection strategies using visual inspection data that will allow them to meet their risk-maintenance requirements.We applied the method to the assessment of grass revetments on the outer slope of the Oesterdam, one of the dams of the Dutch Delta Works. The application illustrates how – using visual inspection data, degradation information, and a failure mechanism model – inspection intervals can be derived that will ensure the risk-maintenance requirements are met.Accepted Author ManuscriptHydraulic Structures and Flood Ris
