The author focuses on the question of prostitution in the medieval Cracow in the light of edited-by- himself treaty by John Falkenberg Iudicium de aedificandis domis pro meretricibus. Writing his treaty Falkenberg was trying, at the request of the Municipal Council, answer the following questions: do laws established by people allow the existence of prostitutes, and: can the ruler of the city or councillors build brothels, put prostitutes there and then profit from such business. The text contains a short review of the problem of prostitution in Cracow of that time as well as the explanati of a context in which Falkenberg’s work was written. The author analyses the legal action of the Polish delegation against the Satira by Dominican friar John Falkenberg, which appeared at the Council of Constance (1414–1418). In his work, Falkenberg called for the extermination of the Polish nation as well as the king Wladyslaw Jagiello. He announced that those who would achieve this aim would attain salvation. The Polish delegation, especially Paul Włodkowic and archbishop Nicolas Trąba, made efforts aimed at the recognition of the Dominican’s work as heretical. Falkenberg was arrested and his work was condemned by the Commission of Faith, however it was not recognized as heretical. The new pope Martin V, in turn, did not want the verdict of the commission to become a resolution of the Council. In response, Poles, despite the threat of anathema, at the last session of the Council, appealed against the Satira to the next council, which undermined the authority of the pope. In the article, the author shows the factors which prevented Poles from achieving complete success in the Falkenberg case. He addresses the question whether the determination of the Polish envoys in the defence of honour of their ruler and state in the final days of the council brought positive results in the years after the council. The author gives consideration to the question whether the actions of Polish diplomacy against ‘Satira’ combined with other steps on the international forum contributed to a certain extent to creating Jagiello’s image as the last apostle of the barbarian Europe

Kundenloyalität in Dienstleistungsbeziehungen: untersucht am Beispiel der Deutschen Bahn AG
Warum sind Reisekunden der DB AG spezielle Kunden, deren Loyalität vergleichsweise wenig belastbar und besonders leicht zu verletzen ist? Was unterminiert ihre Leidenschaft für das Bahnfahren? Warum ist selbst für den Quasimonopolisten DB AG die Förderung einer uneingeschränkten Loyalität seiner Reisekunden relevant? Was charakterisiert die Bindung des Loyalitätstypus alter Art? Und was kennzeichnet die Entwicklung und spezifische Funktionsweise der Bindung des Loyalitätstypus neuer Art?
Mit diesen Fragen greift die Autorin das Schnittstellenthema ‚Kundenloyalität in Dienstleistungsbeziehungen‘ auf, das viele Disziplinen bewegt, jedoch bislang in erster Linie quantitativ und aus Marketingsicht beforscht wurde. Carolyn Koch-Falkenberg fragt danach, wann und warum sich Kunden emotional an ein Unternehmen binden und diesem loyal sind, in welcher Form ihre Loyalität zu Tage tritt, was diese konterkariert und welche Folgen die Art ihrer Loyalitätsform nach sich zieht.
Im Mittelpunkt steht damit eine spezifische Form der Bindungsorientierung, welche die Autorin explorativ mittels qualitativer Methoden konsequent aus der Subjektperspektive der Kunden am Beispiel der Dienstleistungsbeziehung zwischen Reisekunden und dem Unternehmen DB AG sozialwissenschaftlich untersucht.Why are travel customers of DB AG special customers with a comparatively less resilient loyalty which is particularly easy to be violated? What undermines their passion for going by train? Why is even the promotion of the unlimited loyalty of travel customers relevant to the quasimonopolist DB AG? What characterizes the attachment of the old fashioned loyalty type? And what characterizes the development and specific functioning of the binding of the ‚new‘ loyalty type?
The author seizes the interdisciplinary topic of the interface theme 'customer loyalty in service relationships', which ocupy many science disciplines, but has so far been primarily researched quantitatively from a marketing perspective. Carolyn Koch-Falkenberg asks when and why clients are emotionally attached to their loyality for a company, how their loyalty is revealed, how it counteracts and illustrates the consequences of their natural loyalty form.
The focus is on a specific form of attachment orientation. The author is using socially and scientifically qualitative methods in order to research consistently the subject perspective of the customer at the example of the service relationship between travel customers and the company DB AG

Cordia curassavica (JACQ.) ROEM. & SCHULT: Influência de fatores ambientais no crescimento e na produção de metabólitos

(Boraginaceae) tem como sinonímia científica Cordia verbenacea DC. e é conhecida popularmente como erva baleeira. Popularmente utilizada para tratamento de reumatismo, inflamação e promover a cicatrização. É uma espécie ocorrente no Domínio da Mata Atlântica e vegeta preferencialmente em terrenos arenosos e ensolarados, como restingas, dunas e praias. As principais substâncias presentes na C. curassavica com ação antiinflamatória são a artemetina e o a-humuleno, porém sabe-se que além dessas existem outras substâncias antiinflamatórias presentes nesta planta, como o ácido rosmarínico, que é um metabólito secundário comum na família Boraginaceae, mais facilmente encontrado em extratos alcoólicos do que no óleo essencial. O ambiente de crescimento da planta pode influenciar na composição de seus compostos químicos de uso medicinal e na quantidade de material vegetal. Este estudo procurou verificar a influência de três intensidades de luz (100%, 70% e 20% da luz solar) e de substratos com dois níveis de fertilidade (areia de duna e um substrato composto de solo argiloso, areia de rio e composto orgânico termofílico) no crescimento, composição e quantidade de compostos químicos em C. curassavica. Os resultados indicaram que maior intensidade de luz: (1) aumentou a massa seca da raiz e diminuiu a área foliar; (2) aumentou o rendimento relativo dos extratos éter de petróleo e etanólico e o rendimento relativo total; (3) aumentou ou diminuiu, dependendo da substância, apenas duas substâncias das nove detectadas na cromatografia de camada delgada; (4) aumentou a atividade biológica (avaliada pela toxicidade dos extratos etanólicos para larvas de Artemia salina). O substrato mais fértil: (1) aumentou a massa seca total e a área foliar e proporcionou maior valor da taxa de crescimento relativo para as plantas; (2) aumentou o rendimento relativo dos extratos éter de petróleo e etanólico e o rendimento relativo total; (3) não influenciou a intensidade ou número de metabólitos detectados em cromatografia de camada delgada (CCD), no extrato etanólico, nem interferiu na atividade biológica desse extrato. Face ao exposto, este estudo mostra que a alta intensidade de luz solar aumenta a produção de metabólitos em C. curassavica, mas não a biomassa de folhas. Enquanto que solos de maior fertilidade aumenta a produção de metabólitos e da biomassa de folhas - órgão utilizado para extração de metabólitos neste estudo
Investigação fitoquímica de Zollernia ilicifolia (Brongniart) Vogel (Fabaceae): contribuição ao controle de qualidade de espinheira-santa (Maytenus spp.)

Programa de Pós-Graduação em Farmácia.Maytenus ilicifolia é popularmente conhecida como espinheira-santa e utilizada no tratamento de problemas gástricos. Devido ao formato espinhoso das folhas, é facilmente confundida com outras duas espécies, Zollernia ilicifolia e Sorocea bonplandii. Os objetivos deste trabalho foram identificar e caracterizar estruturalmente substâncias de Zollernia ilicifolia para aplicação no controle cromatográfico de qualidade de produtos à base de espinheira-santa. Extratos diclorometânicos destas 3 espécies foram comparados cromatograficamente com vistas à identificação de fitomarcadores para Z. ilicifolia, sendo identificados três potenciais marcadores: ZI1, ZI2 e ZI3. Através de métodos cromatográficos e espectroscópicos, ZI2 foi isolado e identificado como o triterpeno betulina. Amostras destas 3 espécies foram coletadas mensalmente, de março/2000 a março/2001, e seus extratos diclorometânicos foram monitorados para verificar se os marcadores propostos se encontravam de forma constante ao longo do ano. O monitoramento dos extratos de Z. ilicifolia demonstrou que as substâncias ZI1 e ZI2 foram encontradas nos extratos de todos os meses. A análise densitométrica dos extratos desta espécie em software ScionImageâ demonstrou que o material vegetal dos meses de maio/2000 e janeiro/2001 apresentou maiores concentrações de ZI2 (betulina). O monitoramento dos extratos de M. ilicifolia demonstrou que os marcadores propostos anteriormente por outros autores, friedelina e friedelanol, também estavam presentes em todos os extratos. A análise cromatográfica de 10 amostras comerciais à base de espinheira-santa com vistas à detecção dos marcadores de M. ilicifolia, Z. ilicifolia e S. bonplandii demonstrou adulterações nas amostras. Em 5 das 10 amostras foi detectada a presença de M. ilicifolia; destas, 1 apresentou Z. ilicifolia e outras 2, S. bonplandii. Das 5 amostras restantes, 3 apresentaram S. bonplandii, 1 apresentou Z. ilicifolia e outra, tanto Z. ilicifolia como S. bonplandii. Estes resultados evidenciam a baixa qualidade dos produtos do mercado, bem como a aplicabilidade da betulina como marcador de Z. ilicifolia e dos marcadores propostos para S. bonplandii no controle de qualidade dos fitoterápicos à base de espinheira-santa
Super-resolution microscopy reveals that mammalian mitochondrial nucleoids have a uniform size and frequently contain a single copy of mtDNA.
Mammalian mtDNA is packaged in DNA-protein complexes denoted mitochondrial nucleoids. The organization of the nucleoid is a very fundamental question in mitochondrial biology and will determine tissue segregation and transmission of mtDNA. We have used a combination of stimulated emission depletion microscopy, enabling a resolution well below the diffraction barrier, and molecular biology to study nucleoids in a panel of mammalian tissue culture cells. We report that the nucleoids labeled with antibodies against DNA, mitochondrial transcription factor A (TFAM), or incorporated BrdU, have a defined, uniform mean size of ∼100 nm in mammals. Interestingly, the nucleoid frequently contains only a single copy of mtDNA (average ∼1.4 mtDNA molecules per nucleoid). Furthermore, we show by molecular modeling and volume calculations that TFAM is a main constituent of the nucleoid, besides mtDNA. These fundamental insights into the organization of mtDNA have broad implications for understanding mitochondrial dysfunction in disease and aging.</jats:p
