Going Beyond Counting First Authors in Author Co-citation Analysis
The present study examines one of the fundamental aspects of author co-citation analysis (ACA) - the way co-citation
counts are defined. Co-citation counting provides the data on which all subsequent statistical analyses and mappings
are based, and we compare ACA results based on two different types of co-citation counting - the traditional type that
only counts the first one among a cited work's authors on the one hand and a non-traditional type that takes into
account the first 5 authors of a cited work on the other hand. Results indicate that the picture produced through this non-traditional author co-citation counting contains more coherent author groups and is therefore considerably clearer. However, this picture represents fewer specialties in the research field being studied than that produced through the traditional first-author co-citation counting when the same number of top-ranked authors is selected and analyzed. Reasons for these effects are discussed
In vitro study of sorghum grain proteins digestibility
Le grain du sorgho (Sorghum bicolor L. Moench) est considéré comme une culture résiliente au changement climatique et un aliment énergétique, malgré ces avantages, il est encore sous-utilisé dans l'alimentation humaine en raison de sa qualité nutritionnelle relativement inférieure à celle des autres céréales principales (blé, maïs et riz). Les protéines de stockage des grains de sorgho (kafirines), qui représentent 60 % des protéines totales, forment de gros agrégats hydrophobes, une tendance encore exacerbée par la complexation avec des composés phénoliques (tanins) pendant la cuisson. De plus, ces protéines de stockage sont regroupées dans des organites cellulaires micrométriques, les corps protéiques. Ces caractéristiques macromoléculaires et physiques contribuent à la faible digestibilité des kafirines par les protéases gastro-intestinales. Dans ce contexte, la germination des graines a été examinée comme un bioprocédé prometteur pour améliorer la digestibilité des protéines. L'objectif principal de cette thèse était d'étudier l'effet du temps de germination des graines sur les propriétés biochimiques et nutritionnelles du sorgho. La digestibilité des protéines a été évaluée à l'aide du protocole INFOGEST référencé, en considérant les compartiments gastrique et gastro-duodénal. De plus, nous avons combiné des approches protéomiques, transcriptomiques et microscopiques multi-échelles pour identifier les événements moléculaires et physiques se produisant lors de la germination. L'étude a été réalisée sur des grains de deux génotypes de sorgho avec (IS30400, T+) et sans tanins (Macia, T-) qui ont été germé à 16 °C/60 % d'humidité relative et collectés à huit stades de germination, à partir de l'imbibition d'eau à 100 % des graines présentant une émergence de radicules de ±3 mm de long. L'analyse des acides aminés des grains a révélé de modestes modifications lors de la germination à l'exception de la cystéine qui a augmenté de manière significative. De manière constante, les groupes sulfhydryles ont augmenté lors de la germination. L'extraction séquentielle des kafirines et leur analyse ultérieure par chromatographie d'exclusion stérique ont révélé une diminution légère mais significative des kafirines réticulées, sans changements notables dans la solubilité des différentes classes de kafirines. L'analyse par microcopie électronique à balayage et par microcopie optique de grains non germés et germés a révélé des altérations dans la cohésion de la matrice protéique et dans la distribution des corps protéiques tandis que les granules d'amidon apparaissaient dégradés. Ces modifications structurelles étaient imputables à des enzymes nouvellement synthétisées (protéases et amylases). Ces dernières ont été identifiées par des analyses transcriptomiques et protéomiques et nous avons montré que la dégradation moléculaire de la kafirine coïncidait avec l'accumulation de protéases spécifiques à la cystéine. Aussi et en lien avec l'évolution de la cystéine libre, nous avons identifié la synthèse d'oxydoréductases. Nos résultats apportent de nouvelles informations et mettent en lumière l’impact de la germination des grains sur la digestibilité des protéines in vitro par les protéases gastro-intestinales. Dans le cas du génotype T-, nous avons constaté que la germination améliorait la digestibilité des protéines, tandis que la cuisson n'avait aucun impact notable. Pour le génotype T+, le gain de digestibilité apporté par la germination est significatif mais insuffisant pour compenser l'impact très néfaste de la cuisson. Cette étude fournit pour la première fois des informations cohérentes sur les acteurs moléculaires et cellulaires de la germination qui contribuent à la digestibilité in vitro des kafirines par les protéases gastro-intestinales humaines et ouvre la voie à une utilisation plus large du sorgho dans l'alimentation humaine.Although considered as a climate change resilient crop and a high-energy food, sorghum grain (Sorghum bicolor L. Moench) is still underutilized in human diet due to its relatively lower nutritional quality compared to the other main cereals (Wheat, maize and rice). Sorghum seed storage proteins (kafirins) that account for 60% of total protein form large hydrophobic aggregates, a propensity further exacerbated by complexation with phenolic compound (tannins) during cooking. Furthermore, these storage proteins are packed in micrometric cell organelles, the protein bodies. These macromolecular and physical features contribute to lower the digestibility of kafirin by the gastrointestinal proteases. In this context, seed germination was examined as a promising bio-process to improve protein digestibility.The main objective of this PhD thesis was to investigate the effect of seed germination time on sorghum biochemical and nutritional properties. Protein digestibility was assessed using the referenced INFOGEST protocol, considering the gastric and gastro-duodenal compartments. In addition, we combined multi-scale proteomic, transcriptomic and microscopy approaches to decipher the molecular and physical events occurring during germination. The study was carried out on seeds from two sorghum genotypes with (IS30400, T+) and without tannins (Macia, T-) which were germinated at 16 °C/60 % relative humidity and collected at eight stages of germination, from the water imbibition to 100 % of the seeds showing radicle emergence of ±3 mm long. The amino acid analysis of seeds revealed modest modifications upon germination with the exception of cysteine which increased significantly. Consistently, flour free sulfhydryl groups increased upon germination. Sequential extraction of the kafirin and their subsequent analysis by size-exclusion chromatography in adapted solvents revealed a slight but significant decrease in kafirin cross-linking, without notable changes in the solubility of the different kafirin classes. Scanning electron and light microcopy analysis of ungerminated and germinated seeds revealed alterations in the protein matrix cohesiveness and in the of the protein bodies distribution while starch granules appeared pitted. These structural modifications were accountable for by newly synthesized enzymes (proteases and amylases). The latter were identified by transcriptomic and proteomic analyses and we showed that the kafirin molecular breakdown coincided with the accumulation of specific cysteine proteases. Also and in link with the change in free cysteine we identified the synthesis of oxidoreductases.Our findings bring new information and shed a light on how seed germination impacted the in vitro protein digestibility by gastrointestinal proteases. In the case of the T- genotype we found that germination improved protein digestibility, while cooking as no noticeable impact. For the T+ genotype, the digestibility gain brought by germination was significant but not enough to compensate for the highly detrimental impact of cooking.This study provides for the first time coherent information about the molecular and cellular actors of germination that help for the in vitro digestibility of kafirin by human gastrointestinal proteases and paves the way for a wider use of sorghum in the human diet
Variations on the Author
“Variations on the Author” discusses two of Eduardo Coutinho’s recent films (Um Dia na Vida, from 2010, and Últimas Conversas, posthumously released in 2015) and their contribution to the general question of documentary authorship. The director’s filmography is characterized by a consistent yet self-effacing form of authorial self-inscription: Coutinho often features as an interviewer that rather than express opinions propels discourses; an interviewer that is good at listening. This mode of self-inscription characterizes him as an author who is not expressive but who is nonetheless markedly present on the screen. In Um Dia na Vida, however, Coutinho is completely absent form the image, while Últimas Conversas, on the contrary, includes a confessional prologue that moves the director from the margins to the center of his films. This article examines the ways in which these works stand out in the filmography of a director who offers new insights into the notion of cinematic authorship
La déficience en cytidine désaminase est-elle un marqueur de résistance ou de sensibilité à un traitement anti-tumoral ?
National audienceLe syndrome de Bloom (BS) est une maladie génétique rare qui prédispose à tous types de cancers, qui résulte de la mutation des deux copies du gène BLM. L’équipe a montré que (1) l’absence d’une protéine BLM fonctionnelle était associée à la chute d’expression de la cytidine désaminase (CDA), une enzyme impliquée dans la fabrication des briques constituant l’ADN, et (2) la déficience en CDA entrainait certaines anomalies cellulaires connues pour prédisposer au développement de cancers. Dans le cadre de mon stage post-doctoral, mon projet de recherche avait pour objectif de déterminer si la déficience en CDA pouvait être impliquée dans certains processus de cancérogenèse dans la population générale. Notre étude a révélé pour la première fois que l’expression de CDA est perdue dans une grande proportion de tumeurs (~60 %), principalement en raison de modifications épigénétiques du gène CDA ; ainsi, des tumeurs qui étaient classiquement définies comme appartenant à un même groupe sur la base de critères histologiques, cellulaires et moléculaires, pouvaient en fait présenter des propriétés différentes selon leur statut d’expression de CDA. Ainsi, le statut d’expression de CDA permet de définir deux nouveaux sous-groupes : les tumeurs déficientes et les tumeurs proficientes en CDA. Nos résultats indiquent que l’analyse par immunohistochimie du niveau d’expression initial de CDA dans les tumeurs pourrait avoir des implications importantes dans le choix du traitement à proposer aux patients. Nous avons ainsi montré que l’aminoflavone, agent anti-cancéreux qui a atteint une phase II en essai clinique, affectait spécifiquement la croissance des cellules tumorales déficientes en CDA, sans aucun effet sur les cellules tumorales exprimant CDA. Ainsi, le niveau d’expression de CDA pourrait être utilisé comme un nouveau marqueur pour guider la thérapie anticancéreuse
Appropriate Similarity Measures for Author Cocitation Analysis
We provide a number of new insights into the methodological discussion about author cocitation analysis. We first argue that the use of the Pearson correlation for measuring the similarity between authors’ cocitation profiles is not very satisfactory. We then discuss what kind of similarity measures may be used as an alternative to the Pearson correlation. We consider three similarity measures in particular. One is the well-known cosine. The other two similarity measures have not been used before in the bibliometric literature. Dispelling the Myths Behind First-author Citation Counts
We conducted a full-scale evaluative citation analysis study of scholars in the XML research field to explore just how different from each other author rankings resulting from different citation counting methods actually are, and to demonstrate the capability of emerging data and tools on the Web in supporting more realistic citation counting methods. Our results contest some common arguments for the continued
use of first-author citation counts in the evaluation of scholars, such as high correlations between author rankings by first-author citation counts and other citation
counting methods, and high costs of using more realistic citation counting methods that are not well-supported by the ISI databases. It is argued that increasingly available digital full text research papers make it possible for citation analysis studies to go beyond what the ISI databases have directly supported and to employ more

We have done our best to complete the author checklist relating to the use of animals in the hut study. Note that the objective for the hut study was to evaluate the IRS treatment applications for residual efficacy against Anopheles mosquitoes, including the local An. coluzzii mosquito population. Cows were only used to attract mosquitoes into the huts and no tests were carried out directly on the cows. The author checklist is intended for use with studies where experiments are carried out on animals, which is why we have had such difficulty in completing this for the hut study, as many of the questions do not relate to how the cows were used
