    Aprovechamiento de almidón de amaranto (A. hypochondriacus) como almidón resistente

    En México hay un incremento notable en los problemas de salud de la población que van desde problemas de malnutrición, problemas cardiovasculares y gastrointestinales debido a los malos hábitos de alimentación y la escasa disponibilidad de alimentos saludables. El almidón resistente (AR) es la fracción del almidón que no es hidrolizado en el intestino delgado entre los 120 minutos después de ser consumido, pero puede ser fermentado en el colón, por lo que al ser adicionado en los alimentos disminuye el índice glucémico (IG) de éstos, además existen evidencias de poseer capacidad prebiótica. La semilla de amaranto está compuesta principalmente de almidón, siendo únicamente de 0.48- 0.65% almidón resistente. La mayoría de las investigaciones se basan en la extracción de proteína de la semilla de amaranto para la obtención de péptidos bioactivos. En este trabajo se estudió el uso del residuo de la extracción de proteínas y lípidos el cual deja disponible el 63.6% de almidón de amaranto para darle un valor agregado al convertirlo a AR y evaluar su capacidad prebiótica. El extracto de almidón de la semilla fue sometido a hidrólisis ácida con ácido clorhídrico al 3.4% realizando una solución al 40% de sólidos de almidón, las condiciones de reacción fueron de 4 horas y 50 °C, lo que permitió obtener un almidón con un grado de hidrólisis de 39.27%, posteriormente se llevaron a cabo dos reacciones de modificación: succinatación y fosfatación. En el primer método se empleó como agente succinatante anhídrido octenil succínico (OSA) al 3% y condiciones de reacción de 30°C, 6 horas de reacción y pH 7.5 logrando obtener un contenido de almidón resistente de 46.69%. En el método de fosfatación se obtuvo un 56.67% de almidón resistente empleando tripolifosfato de sodio (TPS) al 0.4% y condiciones de reacción de 40 °C, pH 6.5 y 1 hora de reacción. El método de obtención de almidón utilizado en esta investigación causó una modificación en los gránulos de almidón, en las micrografías obtenidas no se observa la estructura morfológica característica, además el extracto de almidón presentó una baja capacidad de absorción (10.92%) y solubilización en agua (22.05%), características que no cambiaron al ser sometidos a los procesos de modificación por de succinatación, el cual presento un índice de absorción de agua del 7% y de solubilidad en agua de 9.20% y fosfatación el cual presento un índice de absorción de agua de 6.68% un índice de solubilidad en agua de 3.12%, mientras que en el proceso El almidón modificado por fosfatación bajo las condiciones en las que se obtiene una mayor conversión de almidón nativo a almidón resistente no presentó características prebióticas al ser empleado como fuente de carbono empleando como microorganismos Lactobacillus casei Shirota, Lactobacillus rhamnosus GG., Lactobacillus delbruekii, Lactobacillus fermentum, Pediococcus pentosaceus, Bifidobacterium longum y Lactococcus lactis subsp. cremoris NCFB-712 y Lactobacillus casei Shirota y Pediococcus pentosaceus

    Síntesis de oligosacáridos fucosilados en medio convencional y no-convencional empleando la α-L-fucosidasa de Thermotoga maritima

    La leche humana es el alimento ideal en los primeros meses de vida del infante y su composición está adaptada a sus necesidades nutricionales. En la leche humana se encuentran oligosacáridos fucosilados, los cuales tienen diversos efectos benéficos en la salud de los infantes. Debido a las ventajas anteriormente mencionas, el objetivo de este trabajo fue aumentar el rendimiento de síntesis de oligosacáridos fucosilados, evaluando los parámetros de pH, temperatura y actividad de agua (aw) en la actividad hidrolítica y de transglicosilación de la α-L-fucosidasa de Thermotoga maritima. La fucosidasa presentó un intervalo de pH óptimo de 6-8 para la actividad hidrolítica y de 7-10 para la actividad de transglicosilación. Además, al aumentar la temperatura se observó un incremento tanto en la actividad hidrolítica como de transglicosilación, obteniéndose la mayor actividad enzimática a 95 °C. Asimismo, la productividad más alta en la síntesis de oligosacáridos fucosilados (3.54 mM/h) se registró a pH 8 y 95 °C empleando pNP-fucosa (3.5 mM) como sustrato donador y lactosa (438 mM) como sustrato aceptor. Posteriormente se evalúo el efecto de la aw, la cual se disminuyó mediante la adición de dimetil sulfóxido (DMSO), acetona y acetonitrilo. En el medio de reacción con acetona (aw 0.97, 0.95 0.93) y acetonitrilo (aw 0.96, 0.93 y 0.91) se obtuvo una actividad hidrolítica mayor al 100% respecto al ensayo control (medio acuoso), mientras que con DMSO a awDMSO >acetonitrilo (0.51>0.42>0.18 mM/h). La adición de acetona y DMSO favoreció la actividad de transglicosilación sobre la actividad hidrolítica, esto se comprobó al obtenerse una tasa de transglicosilación/hidrólisis (rS/H) de 1.21 y 1.43, respectivamente. Por el contrario, en el medio con acetonitrilo la actividad hidrolítica prevalece (rS/H 0.59), no obstante, con todos los solventes se obtuvo una mayor rS/H respecto al ensayo control (0.39). Adicionalmente, se evalúo el efecto de la temperatura en la estructura de la α-L-fucosidasa de T. maritima mediante simulación de dinámica molecular (DM). En las DM realizadas a 60-90 °C no se observaron cambios significativos en los valores del promedio de RMSD (RMSDprom) indicando la estabilidad de la enzima en ese intervalo de temperaturas, sin embargo, a 95 °C se registró un incremento significativo en el RMSDprom y promedio de RMSF (RMSFprom) reflejando cambios conformacionales importantes en la estructura de la enzima. Además, los resultados obtenidos indican que la temperatura contribuye a los cambios en la estructura secundaria de la enzima, ocasionando una disminución en el contenido de α-hélices con el subsecuente aumento de conformación irregular, lo que explicaría la pérdida de actividad enzimática a temperaturas elevadas (200 °C). Asimismo, la temperatura influye en la estructura secundaria del sitio activo de la enzima, el cual puede adoptar diferentes conformaciones: abierta, intermedia o cerrada. Finalmente, con la técnica de acoplamiento molecular se observaron diferencias en la interacción de la α-L-fucosidasa con los ligandos L-fucosa y pNP-Fuc, reflejándose en una menor energía libre de unión y una mayor afinidad de la enzima por el sustrato conforme se incrementó la temperatura.Human milk is the ideal food during the first months in the new-born life with his composition adapted to their nutritional needs. Fucosylated oligosaccharides are found in human milk and have diverse beneficial effects on the health of infants. In this work, the effect of pH, temperature and water activity (aw) on both the hydrolytic and transglycosylation activity on the synthesis of fucosylated oligosaccharides was evaluated by using the α-L-fucosidase from Thermotoga maritima. The fucosidase show an optimum pH range of 6-8 for hydrolytic activity and 7-10 for transglycosylation activity. In addition, when temperature was raised, an increase on both the hydrolytic and transglycosylation activities was observed, obtaining the highest enzymatic activity at 95 ° C. Additionally, the highest productivity in the synthesis of fucosylated oligosaccharides (3.54 mM/h) was obtained at pH 8 and 95 ° C by using pNP-fucose (3.5 mM) as donor substrate and lactose (438 mM) as acceptor substrate. Subsequently, the effect of aw was evaluated; aw was decreased by the addition of dimethyl sulfoxide (DMSO), acetone and acetonitrile. In the reaction medium with acetone (aw 0.97, 0.95 0.93) and acetonitrile (aw 0.96, 0.93 and 0.91), a hydrolytic activity greater than 100% was obtained with respect to the control (water media), while with DMSO at aw DMSO> acetonitrile (0.51> 0.42> 0.18 mM/h). With the addition of DMSO and acetone, transglycosylation activity was favoured over the hydrolytic one, this by obtaining a transglycosylation/hydrolysis rate (rS/H) of 1.21 and 1.43, respectively; on the contrary, in the medium with acetonitrile, the hydrolytic activity prevailed (rS/H 0.59), however, all solvents allowed to obtain a higher rS/H compared to the control (0.39). Additionally, the effect of temperature on the structure of the α-L-fucosidase of T. maritima was evaluated by molecular dynamics simulation (DM). In the DM at 60-90 °C, no significant changes were observed in the average RMSD (RMSDavg)values, indicating the stability of the enzyme in that range of temperatures. However, at 95 °C there was a significant increase on both, the RMSDavg and average RMSF (RMSFavg) values, which reflect important conformational changes in the enzyme structure. Furthermore, results indicate that temperature contributes to changes in the secondary structure of the enzyme, causing a decrease in the content of α-helices with the subsequent increased in irregular conformation, which would explain the loss of enzymatic activity at high temperatures (200 °C). Likewise, the temperature influences the secondary structure of the active site of the enzyme, which can adopt different conformations: open, intermediate or closed. Finally, with the molecular docking technique, differences were observed in the interaction of α-L-fucosidase with the ligand L-fucose and pNP-Fuc, as the temperature increased a lower free binding energy and higher affinity of the enzyme for the corresponding substrate was observed

    La alimentación es uno de los factores que más condiciona la salud de los individuos, ejerciendo un papel primordial sobre el desarrollo físico y el crecimiento, la reproducción y el rendimiento físico e intelectual. Actualmente la obesidad es uno de los problemas más importantes de salud pública en México. Coexistiendo paradójicamente con la desnutrición, una epidemia de sobrepeso y obesidad crece y afecta a la mayor parte del mundo, esto debido a que la dieta suele contener una cantidad excesiva de carbohidratos, específicamente los productos afines a maíz y trigo, los cuales son deficientes en lisina (Lys) y triptófano (Trp). El objetivo del presente trabajo es la extracción de la proteína de la semilla amaranto seguida de los lípidos que la componen, para lo que se realizó una hidrólisis enzimática empleando la proteasa de Bacillus licheninformis, al residuo obtenido se le realizó la extracción de aceite por el método de soxhlet usando hexano como solvente. La proteína extraída se agregó en diferentes proporciones a pan de caja elaborado con harina de trigo para evaluar el aumento en el contenido proteico, el perfil de textura y la aceptabilidad por parte de los consumidores. Al aceite se le cuantificó la capacidad antioxidante y se midió el índice de acidez para conocer si el pre-tratamiento enzimático tiene algún efecto en ello. Al realizar una hidrólisis enzimática de 4 horas usando la proteasa en una concentración de 3.6U/g se obtuvo un rendimiento de extracción de proteínas de 84.4%; este extracto acuoso de proteína se empleó como el agua necesaria para la elaboración del pan, se adicionaron diversas proporciones de proteína hidrolizada:agua (0.5:1, 1:1) de proteína lo máximo que se puede adicionar al pan de caja para que sea aceptado por el consumidor, con un aumento hasta del 5% en el contenido proteico respecto al pan control. Con la extracción del aceite a partir de harina pre-tratada por 4 horas empleando la proteasa a una concentración tanto del 2.4U/g como del 0.036 tiene una extracción de grasa del 20% en comparación con el control que es del 9%. El aceite obtenido mediante soxhlet de la harina pre-tratada durante 4 horas con 3.6U/g de proteasa presenta un índice de acidez de 0.12, sin embargo el aceite obtenido a partir de la harina pre-tratada por 1 hora tanto con una concentración de enzima de 2.4 U/g y 3.6U/g muestra la mayor capacidad antioxidante (58%). El aceite obtenido de la harina pre-tratada es de mejor calidad y se obtiene un mayor rendimiento de extracción del mismo.Food is one of the factors that affect the health of individuals, exerting a major role on the physical development and growth, reproduction and the physical and intellectual performance. Obesity is currently one of the most important public health problems in Mexico. Paradoxically coexisting with under nutrition, an epidemic of overweight and obesity is growing and affects most of the world that is because diet usually contains an excessive amount of carbohydrates, specifically related do maize and wheat products which are deficient in lysine (Lys) and tryptophan (Trp). The aim of this work is the extraction of the amaranth seed protein followed by lipids, for which an enzymatic hydrolysis is carried out using protease from Bacillus licheniformis, the residue obtained from this hydrolysis was subjected to soxhlet extraction using hexane as solvent. The protein hidrolisate protein was added to wheat bread in different proportions to evaluate the increase in protein content, texture profile and acceptability. Antioxidant capacity and acid measure from the oil was quantified to determine whether enzymatic pre-treatment has any effect on it. The hydrolysis using protease 3.6 U/g at 4 hours 84.4% yield of protein was reached; this aqueous protein hidrolisate was used as the water needed for making bread, different proportions protein hidrolisate/water were added (0.5:1 and 1:1). The most of protein hidrolisate:water that can be added to bread to be accepted is 0.5:1 which means an increase of 5% in protein content. However the oil extracted from the pre-treated flour for 4 hour with 2.4 U/g and 3.6 U/g of protease have a fat removal of 20% compared to the control 9%. The oil obtained from the pre-treated flour for 4 hours using protease at a concentration of 3.6 U/g has and acid value of 0.12, however the oil obtained from flour pre-treated for 1 hour with an enzyme concentration of 2.4 U/g and 3.6 U/g showed the highest antioxidant capacity (58%). The oil obtained from the pre-treated flour is of better quality and higher yields of extraction

    Going Beyond Counting First Authors in Author Co-citation Analysis

    The present study examines one of the fundamental aspects of author co-citation analysis (ACA) - the way co-citation counts are defined. Co-citation counting provides the data on which all subsequent statistical analyses and mappings are based, and we compare ACA results based on two different types of co-citation counting - the traditional type that only counts the first one among a cited work's authors on the one hand and a non-traditional type that takes into account the first 5 authors of a cited work on the other hand. Results indicate that the picture produced through this non-traditional author co-citation counting contains more coherent author groups and is therefore considerably clearer. However, this picture represents fewer specialties in the research field being studied than that produced through the traditional first-author co-citation counting when the same number of top-ranked authors is selected and analyzed. Reasons for these effects are discussed

    “Variations on the Author” discusses two of Eduardo Coutinho’s recent films (Um Dia na Vida, from 2010, and Últimas Conversas, posthumously released in 2015) and their contribution to the general question of documentary authorship. The director’s filmography is characterized by a consistent yet self-effacing form of authorial self-inscription: Coutinho often features as an interviewer that rather than express opinions propels discourses; an interviewer that is good at listening. This mode of self-inscription characterizes him as an author who is not expressive but who is nonetheless markedly present on the screen. In Um Dia na Vida, however, Coutinho is completely absent form the image, while Últimas Conversas, on the contrary, includes a confessional prologue that moves the director from the margins to the center of his films. This article examines the ways in which these works stand out in the filmography of a director who offers new insights into the notion of cinematic authorship

    Appropriate Similarity Measures for Author Cocitation Analysis

    We provide a number of new insights into the methodological discussion about author cocitation analysis. We first argue that the use of the Pearson correlation for measuring the similarity between authors’ cocitation profiles is not very satisfactory. We then discuss what kind of similarity measures may be used as an alternative to the Pearson correlation. We consider three similarity measures in particular. One is the well-known cosine. The other two similarity measures have not been used before in the bibliometric literature.     Dispelling the Myths Behind First-author Citation Counts

    We conducted a full-scale evaluative citation analysis study of scholars in the XML research field to explore just how different from each other author rankings resulting from different citation counting methods actually are, and to demonstrate the capability of emerging data and tools on the Web in supporting more realistic citation counting methods. Our results contest some common arguments for the continued use of first-author citation counts in the evaluation of scholars, such as high correlations between author rankings by first-author citation counts and other citation counting methods, and high costs of using more realistic citation counting methods that are not well-supported by the ISI databases. It is argued that increasingly available digital full text research papers make it possible for citation analysis studies to go beyond what the ISI databases have directly supported and to employ more sophisticated methods

    We have done our best to complete the author checklist relating to the use of animals in the hut study. Note that the objective for the hut study was to evaluate the IRS treatment applications for residual efficacy against Anopheles mosquitoes, including the local An. coluzzii mosquito population. Cows were only used to attract mosquitoes into the huts and no tests were carried out directly on the cows. The author checklist is intended for use with studies where experiments are carried out on animals, which is why we have had such difficulty in completing this for the hut study, as many of the questions do not relate to how the cows were used
