    The Role of miRNAs in Downregulation of PTEN for Glioblastoma Multiforme

    In this paper we show, in silico, the role of miRNAs in the post-transcriptional regulation of the onco suppressor gene PTEN by means of a (stochastic) competitive model including interactions with miRNAs and other concurrent genes. The model also covers protein formation and the mechanism going from the protein back to the miRNAs. The numerical simulations confirm the biologically observed situation that the increase of miRNAs concentration within the cell results in the lower expression of PTEN, thus suggesting severe implications in brain tumorigenesis

    Simplified mitral valve modeling for prospective clinical application of left ventricular fluid dynamics

    The fluid dynamics inside the left ventricle of the human heart is considered a potential indicator of long term cardiovascular outcome. In this respect, numerical simulations can play an important role for integrating existing technology to reproduce flow details and even conditions associated to virtual therapeutic solutions. Nevertheless, numerical models encounter serious practical difficulties in describing the interaction between flow and surrounding tissues due to the limited information inherently available in real clinical applications. This study presents a computational method for the fluid dynamics inside the left ventricle designed to be efficiently integrated in clinical scenarios. It includes an original model of the mitral valve dynamics, which describes an asymptotic behavior for tissues with no elastic stiffness other than the constrain of the geometry obtained from medical imaging; in particular, the model provides an asymptotic description without requiring details of tissue properties that may not be measurable in vivo. The advantages of this model with respect to a valveless orifice and its limitations with respect to a complete tissue modeling are verified. Its performances are then analyzed in details to ensure a correct interpretation of results. It represents a potential option when information about tissue mechanical properties is insufficient for the implementations of a full fluid-structure interaction approach

    Stochastic modelling of PTEN regulation in brain tumors: a model for Glioblastoma Multiforme

    This work is the outcome of the partnership between the mathema- tical group of Department DISBEF and the biochemical group of Department DISB of the University of Urbino Carlo Bo"in order to better understand some crucial aspects of brain cancer oncogenesis. Throughout our collabora- tion we discovered that biochemists are mainly attracted to the instantaneous behaviour of the whole cell, while mathematicians are mostly interested in the evolution along time of small and dierent parts of it. This collaboration has thus been very challenging. Starting from [23, 24, 25], we introduce a compe- titive stochastic model for post-transcriptional regulation of PTEN, including interactions with the miRNA and concurrent genes. Our model also covers protein formation and the backward mechanism going from the protein back to the miRNA. The numerical simulations show that the model reproduces the expected dynamics of normal glial cells. Moreover, the introduction of transla- tional and transcriptional delays oers some interesting insights for the PTEN low expression as observed in brain tumor cells

    A sensitivity analysis of a mathematical model for the synergistic interplay of amyloid beta and tau on the dynamics of Alzheimer’s disease

    We propose a mathematical model for the onset and progression of Alzheimer’s disease based on transport and diffusion equations. We treat brain neurons as a continuous medium and structure them by their degree of mal- functioning. Three different mechanisms are assumed to be relevant for the temporal evolution of the disease: i) diffusion and agglomeration of soluble Amyloid beta, ii) effects of phosphorylated tau protein and iii) neuron-to- neuron prion-like transmission of the disease. We model these processes by a system of Smoluchowski equations for the Amyloid beta concentration, an evolution equation for the dynamics of tau protein and a kinetic-type transport equation for the distribution function of the degree of malfunctioning of neurons. The latter equation contains an integral term describing the random onset of the disease as a jump process localized in particularly sensitive areas of the brain. We are particularly interested in investigating the effects of the synergistic interplay of Amyloid beta and tau on the dynamics of Alzheimer’s disease. The output of our numerical simulations, al- though in 2D with an over-simplified geometry, is in good qualitative agreement with clinical findings concerning both the disease distribution in the brain, which varies from early to advanced stages, and the effects of tau on the dynamics of the disease

    Going Beyond Counting First Authors in Author Co-citation Analysis

    The present study examines one of the fundamental aspects of author co-citation analysis (ACA) - the way co-citation counts are defined. Co-citation counting provides the data on which all subsequent statistical analyses and mappings are based, and we compare ACA results based on two different types of co-citation counting - the traditional type that only counts the first one among a cited work's authors on the one hand and a non-traditional type that takes into account the first 5 authors of a cited work on the other hand. Results indicate that the picture produced through this non-traditional author co-citation counting contains more coherent author groups and is therefore considerably clearer. However, this picture represents fewer specialties in the research field being studied than that produced through the traditional first-author co-citation counting when the same number of top-ranked authors is selected and analyzed. Reasons for these effects are discussed

    “Variations on the Author” discusses two of Eduardo Coutinho’s recent films (Um Dia na Vida, from 2010, and Últimas Conversas, posthumously released in 2015) and their contribution to the general question of documentary authorship. The director’s filmography is characterized by a consistent yet self-effacing form of authorial self-inscription: Coutinho often features as an interviewer that rather than express opinions propels discourses; an interviewer that is good at listening. This mode of self-inscription characterizes him as an author who is not expressive but who is nonetheless markedly present on the screen. In Um Dia na Vida, however, Coutinho is completely absent form the image, while Últimas Conversas, on the contrary, includes a confessional prologue that moves the director from the margins to the center of his films. This article examines the ways in which these works stand out in the filmography of a director who offers new insights into the notion of cinematic authorship

    Appropriate Similarity Measures for Author Cocitation Analysis

    We provide a number of new insights into the methodological discussion about author cocitation analysis. We first argue that the use of the Pearson correlation for measuring the similarity between authors’ cocitation profiles is not very satisfactory. We then discuss what kind of similarity measures may be used as an alternative to the Pearson correlation. We consider three similarity measures in particular. One is the well-known cosine. The other two similarity measures have not been used before in the bibliometric literature.     Dispelling the Myths Behind First-author Citation Counts

    We conducted a full-scale evaluative citation analysis study of scholars in the XML research field to explore just how different from each other author rankings resulting from different citation counting methods actually are, and to demonstrate the capability of emerging data and tools on the Web in supporting more realistic citation counting methods. Our results contest some common arguments for the continued use of first-author citation counts in the evaluation of scholars, such as high correlations between author rankings by first-author citation counts and other citation counting methods, and high costs of using more realistic citation counting methods that are not well-supported by the ISI databases. It is argued that increasingly available digital full text research papers make it possible for citation analysis studies to go beyond what the ISI databases have directly supported and to employ more sophisticated methods
