THE USE OF IRONY AS A FACE-SAVING STRATEGY IN THE MEMOIR "I'M GLAD MY MOM DIED" BY JENNETTE MCCURDY
This article seeks to examine how irony functions as a politeness strategy to preserve the protagonist's face in the best-seller memoir I'm Glad My Mom Died (2022) by Jennette McCurdy. To accomplish this, two excerpts are analyzed in light of the concept of irony stated by Hutcheon (1992, 1994), Spencer-Oatey’s (2005) rapport management framework, and the types of faces discussed by Culpeper (2011). The samples illustrate the use of the aforementioned figured speech as a way of maintaining the face of the main character’s face by preventing her from creating an inharmonious relationship with her mother. Therefore, irony is used as a tool to enhance the general storytelling experience by enabling the development of more complex characters and relationships between them. Finally, the research also indicates the value of utilizing literature as pragmatic data, as it allows a comprehensive analysis of linguistic concepts found that are seen in real-world situations as well. This article seeks to examine how irony functions as a politeness strategy to preserve the protagonist's face in the best-seller memoir I'm Glad My Mom Died (2022) by Jennette McCurdy. Sarcasm in I’m Glad My Mom Died by Jennette McCurdy and Its bahasa Indonesia translation
In the era of globalization translation has become more important. Humans have even created translation machines to connect people from around the world. However, translator machines are different from human living translators. The sense of one culture cannot be translated by machine. That is why translators are important. This research aims to find out the translation technique used to translate the sarcasm found in the memoir entitled I’m Glad My Mom Died (2022) by Jennette McCurdy. This research used John Dryden’s translation technique to analyse the translation of the sarcasm of the memoir. There are three translation techniques according to Dryden (in Munday, 2002); metaphrase, paraphrase, and imitation. This study also analyzed the types of sarcasm that are found in the memoir based on Camp (2012); like-prefixed, propositional, lexical, and illocutionary. The researchers of this study found 11 sarcasms from the memoir. The 11 data of sarcasm in the book align with the types of sarcasm outlined by Camp (2012). The researcher found four types of sarcasm in the memoir and each of the sarcasms were translated with different techniques. The use of different techniques for each sarcasm is to make it easier for the target audience to understand and to not lose the sarcasm essence from the memoir

Impact of child summertime obesity interventions on body mass index, and weight-related behaviours: a systematic review and meta-analysis protocol
IntroductionIn previous studies, it has been found that on average, children consistently gained weight during the summer months at an increased rate compared with the 9-month school year. This contributed to an increased prevalence of overweight and obesity in children. Several obesity-related interventions have occurred during or targeting the summer months. We propose to conduct a systematic review and meta-analysis of the impact of obesity prevention and treatment interventions for school-age children conducted during the summer or targeting the summer months when children are not in school on their body mass index (BMI), or weight-related behaviours. Methods and analysesA literature search will be conducted by the first author (JPM) using MEDLINE/PubMed, Cochrane Library, Scopus, CINAHL, PsycINFO, EMBASE and Proquest Dissertations and Theses databases from the date of inception to present. Studies must examine interventions that address the modification or promotion of weight-related behaviours (eg, dietary patterns, eating behaviours, physical activity (PA), sedentary behaviour or sleep) and target school-age children (ages 5–18). The primary outcomes will be changes from baseline to postintervention and/or the last available follow-up measurement in weight, BMI, BMI percentile, standardised BMI or per cent body fat. Secondary outcomes will include changes in dietary intake, PA, sedentary behaviour or sleep. Risk of bias will be assessed using the Cochrane risk of bias tool for randomised and non-randomised studies, as appropriate.Ethics and disseminationBecause this is a protocol for a systematic review, ethics approval will not be required. The findings will be disseminated via presentations at scientific conferences and published in a peer-reviewed journal. All amendments to the protocol will be documented and dated and reported in the PROSPERO trial registry.PROSPERO registration numberCRD42016041750</jats:sec
High basal activity of the PTPN22 gain-of-function variant blunts leukocyte responsiveness negatively affecting IL-10 production in ANCA vasculitis.
Consequences of expression of the protein tyrosine phosphatase nonreceptor 22 (PTPN22) gain-of-function variant were evaluated in leukocytes from patients with anti-neutrophil cytoplasmic autoantibody (ANCA) disease. The frequency of the gain-of-function allele within the Caucasian patient cohort was 22% (OR 1.45), compared to general American Caucasian population (16.5%, p = 0.03). Examination of the basal phosphatase activity of PTPN22 gain-of-function protein indicated persistently elevated activity in un-stimulated peripheral leukocytes, while basal activity was undetectable in leukocytes from patients without the gain-of-function variant. To examine consequences of persistently high PTPN22 activity, the activation status of ERK and p38 MAPK were analyzed. While moderate levels of activated ERK were observed in controls, it was undetectable in leukocytes expressing PTPN22 gain-of-function protein and instead p38MAPK was up-regulated. IL-10 transcription, reliant on the ERK pathway, was negatively affected. Over the course of disease, patients expressing variant PTPN22 did not show a spike in IL-10 transcription as they entered remission in contrast to controls, implying that environmentally triggered signals were blunted. Sustained activity of PTPN22, due to the gain-of-function mutation, acts as a dominant negative regulator of ERK activity leading to blunted cellular responsiveness to environmental stimuli and expression of protective cytokines
Revised 2017 international consensus on testing of ANCAs in granulomatosis with polyangiitis and microscopic polyangiitis
Anti-neutrophil cytoplasmic antibodies (ANCAs) are valuable laboratory markers used for the diagnosis of well-defined types of small-vessel vasculitis, including granulomatosis with polyangiitis (GPA) and microscopic polyangiitis (MPA). According to the 1999 international consensus on ANCA testing, indirect immunofluorescence (IIF) should be used to screen for ANCAs, and samples containing ANCAs should then be tested by immunoassays for proteinase 3 (PR3)-ANCAs and myeloperoxidase (MPO)-ANCAs. The distinction between PR3-ANCAs and MPO-ANCAs has important clinical and pathogenic implications. As dependable immunoassays for PR3-ANCAs and MPO-ANCAs have become broadly available, there is increasing international agreement that high-quality immunoassays are the preferred screening method for the diagnosis of ANCA-associated vasculitis. The present Consensus Statement proposes that high-quality immunoassays can be used as the primary screening method for patients suspected of having the ANCA-associated vaculitides GPA and MPA without the categorical need for IIF, and presents and discusses evidence to support this recommendation
The Oxford IgA nephropathy clinicopathological classification is valid for children as well as adults
To study the predictive value of biopsy lesions in IgA nephropathy in a range of patient ages we retrospectively analyzed the cohort that was used to derive a new classification system for IgA nephropathy. A total of 206 adults and 59 children with proteinuria over 0.5 g/24h/1.73 m(2) and an eGFR of stage-3 or better were followed for a median of 69 months. At the time of biopsy, compared with adults children had a more frequent history of macroscopic hematuria, lower adjusted blood pressure, and higher eGFR but similar proteinuria. Although their outcome was similar to that of adults, children had received more immunosuppressants and achieved a lower follow-up proteinuria. Renal biopsies were scored for variables identified by an iterative process as reproducible and independent of other lesions. Compared with adults, children had significantly more mesangial and endocapillary hypercellularity, and less segmental glomerulosclerosis and tubulointerstitial damage, the four variables previously identified to predict outcome independent of clinical assessment. Despite these differences, our study found that the cross-sectional correlation between pathology and proteinuria was similar in adults and children. The predictive value of each specific lesion on the rate of decline of renal function or renal survival in IgA nephropathy was not different between children and adults.
