    Pyocins are narrow-spectrum bacteriocins synthesized by most isolates of Pseudomonas aeruginosa and are presumed to play a role in niche establishment and protection in mixed populations. P. aeruginosa pyocins can be subdivided into insoluble R and F pyocins and soluble S pyocins. The majority of S-type pyocins (AP41, S1, S2, and S3) cause cell death by DNA cleavage due to an endonuclease C-terminal domain, while pyocins S4 and S5 have tRNAse and pore-forming activities, respectively. Pyocin-producing strains are protected from their own toxin by an immunity protein encoded by a gene usually located downstream the pyocin gene. Pyocin production can be induced by agents that cause DNA damage such as UV irradiation and mitomycin C. In this study, we show that the new small RNA SPA0021, which is transcribed within the pathogenicity island PAPI-1 of P. aeruginosa strain PA14, influences positively the expression of pyocin S3 whose genetic locus comprises two structural genes, pyoS3A and pyoS3I; encoding the killing (S3A) and the immunity (S3I) proteins, respectively. Interestingly, the deletion of SPA0021 gene resulted in increased sensitivity to UV irradiation and to the fluorochinolone antibiotic ciprofloxacin. The degree of UV sensitivity displayed by the PA14 strain lacking SPA0021 was comparable to that of a strain deleted for pyoS3A pyoS3I. These results suggested an involvement of pyocin S3 in DNA damage repair and a regulatory role of SPA0021 on this function

    Objectives: This clinical report describes a single-tooth implant restoration in a fresh extraction socket in the anterior maxilla, with immediate provisionalization and CAD-CAM zirconia final prosthesis. Methods and Materials: Due to a traumatic injury, an esthetically concerned 40 year-old female patient was treated with an atraumatic extraction of a maxillary central incisor, which was immediately substituted with a single implant. Immediate provisionalization was performed using a screw-retained resin crown. Two months after surgery, the patient was fitted with a final all-ceramic single crown cemented on a CAD-CAM customized zirconia abutment. Results: Healing was predictable, with the implant neither showing mobility nor presenting pain. The zirconia crown was undamaged. The patient was pleased with treatment time and esthetic result. Discussion: Implant treatment with immediate provisionalization offers several advantages: optimal esthetics is achievable as bone and soft tissue architecture is maintained; patients are provided with a fixed temporary restoration at the time of surgery; treatment time is shortened, as second stage surgery is eliminated. Furthermore, the fresh extraction socket is a metabolically active region with good potential for bone repair and successful implant placement.
