    Indagini archeologiche e territoriali a Pian della Tirena (2012-2013)

    L'intervento ha inteso presentare i risultati delle ricerche archeologiche e territoriali condotte a Pian della Tirena, a Nocera Terinese (Cz), un promontorio su cui si sviluppa, in età ellenistica, un abitato brezio, cui segue un insediamento di età romana, forse riconducibile alla colonia di Tempsa, fondata nel 194 a.C. Le indagini hanno documentato, per la prima volta, la presenza di assi stradali disposti ortogonalmente e caratterizzati da una serie di fasi, che vanno dalla fine del IV sec. a.C. al II sec. a.C. La ripresa delle ricognizioni nel territorio, invece, hanno confermato la presenza di una serie di nuclei rurali sparsi, relativi soprattutto alla fase brezia, che occupano le fasce collinari interne. Le ricerche sono maturate nell'ambito del PRIN 2009 "Centri fortificati, insediamenti e processi ambientali nella Calabria tirrenica tra secondo e primo millennio a.C. Un'indagine interdisciplinare"

    Developing novel non-hydroxamate histone deacetylase inhibitors: the chelidamic warhead.

    we reported two novel series of histone deacetylase inhibitors bearing the pyridine-2,6-dicarboxylate moiety as a zinc binding group. Tested on U937 leukemia cell line at 50 μM, compounds 4a, 4c and 4d showed cell cycle block in S phase and apoptotic induction up to 50%, whereas compound 6h was able to give granulocytic differentiation up to 40%. From these results, the chelidamic scaffold will be further investigated to find more potent compound

    Measurement of extended half-life recombinant FVIII molecules: In vitro and ex vivo evidence of relevant assay discrepancies

    Background: Extended half-life recombinant FVIII products (EHL-rFVIIIs) have been engineered to improve the pharmacokinetic profile of FVIII, enabling better hemostatic protection with a reduced number of injections in persons with hemophilia. Previous studies showed several discrepancies in FVIII activity (FVIII:C) measurements for EHL-rFVIIIs comparing one-stage clotting assay (OSA) and chromogenic assay (CSA), although a systematic investigation of this phenomenon is still lacking. Objective: Evaluation of the accuracy and precision of measurement of all available EHL-rFVIIIs with 5 different assays both in vitro and ex vivo. Methods: Damoctocog alfa pegol, rurioctocog alfa pegol, turoctocog alfa pegol, and efmoroctocog alfa were tested with 3 OSA types: (1) aPTT-based commercial reagents with colloidal silica (Synthasil, Werfen-IL); (2) ellagic acid, Synthafax (Werfen-IL); and (3) OSA calibrated with each EHL-rFVIII product and colloidal silica. Measurements were also carried out with 2 different commercially available CSA reagents (Coamatic Factor VIII, Chromogenix-Werfen) and Trinichrom FVIII (Tcoag-Stago). A Bland-Altman analysis was performed to compare all assays. Results: The simple OSA showed significant discrepancies between the expected and measured EHL-rFVIII concentrations as CSA methods, whereas the calibrated OSA assay was accurate and precise in determining the activity of all EHL-rFVIIIs in the in vitro setting. Comparable results were found using ex vivo plasma samples. Conclusion: In this study, only OSA with a calibration curve constructed with each EHL-rFVIII product showed acceptable accuracy and precision in EHL-rFVIIIs measurements

    Fluorescent Light Energy (FLE) Generated through Red LED Light and a Natural Photoconverter Gel as a New, Non-Invasive Approach for Facial Age Control: A Pilot Study

    This study, for the first time, evaluated the safety and efficacy of a new natural-based topical gel containing a spirulina extract. This photoconverter gel generates fluorescent light energy (FLE) via a red LED light device, which is proven to be effective for age control of facial skin. This was a one-centre, observational, uncontrolled pilot trial. Eight healthy female subjects aged 35 to 65 years old, with Fitzpatrick skin types II–V were recruited. The duration of the study was five treatment sessions of one treatment every seven days, with a final follow-up at one month after the last treatment session. The images and the related data were acquired with the SONY® Mod. DSCRX10M3, the Canfield VISIA Facial Imaging System®, and QUANTIFICARE 3D® analysis. Patient compliance was excellent (100%) and the treatment was described as warm and pleasant by the patients. After 30 days, VISIA parameters such as wrinkles, texture, red areas, and Trueskin Age® had improved. The safety and efficacy of the FLE treatment assessed in this trial were achieved for overall rejuvenation of facial skin, focusing on wrinkles evaluated via the specific VISIA algorithms

    The main phenolic compounds responsible for the antioxidant capacity of sweet cherry (Prunus avium L.) pulp

    The antioxidant capacity of sweet cherry (Prunus avium L.) pulp extracts is strictly related to the phenolic content, starting from the fact that the higher content of phenolic compounds corresponds to the higher antioxidant indexes. This work aims to assess which compounds characterized three cultivars, namely Ferrovia, Sweetheart, and Lapins grown in Southern Italy and mainly influenced the antioxidant capacity of their extracts. HPLC-MS/ MS analyses were conducted to identify and quantify 17 flavonoids and 25 hydroxicinnamates derivatives. A significant influence of cultivar was revealed from one-way MANOVA (p < 0.05). Furthermore, the extracts were tested for their radical scavenging activity (DPPH and ABTS assays) and reducing power using the Folin-Ciocalteau method. Lapins and Sweetheart extracts, richer in phenolic compounds, returned the highest reducing power and radical scavenging capacity. Finally, a Factorial Analysis was applied to the collected data allowing reliable correlations between phenolics and antioxidant indexes

    Pyrrole-Based Hydroxamates and 2-Aminoanilides: Histone Deacetylase Inhibition and Cellular Activities

    (Chemical Equation Presented) Pyrrole-based HDAC inhibitors: Pyrrolyl-hydroxamates (3 a-g) and 2-aminoanilides (4 a-g) derived from the class II-selective histone deacetylase (HDAC) inhibitor MC1568 (1) were prepared and tested against human recombinant HDAC1, HDAC4, and HDAC6. Unlike compound 1, most of the tested compounds inhibited both HDAC1 and HDAC6, and were less potent or completely inactive against HDAC4. Consistent with their high HDAC1/HDAC6 inhibition, compounds 3 a and 3 b induced>20 % cell death in U937 cells at 1m. © 2009 Wiley-VCH Verlag GmbH & Co. KGaA

    The p.P1127S pathogenic variant lowers von Willebrand factor levels through higher affinity for the macrophagic scavenger receptor LRP1: Clinical phenotype and pathogenic mechanisms

    Background The index case is a 21-year-old Italian woman with a mild hemorrhagic syndrome and von Willebrand factor antigen (VWF:Ag) = 34.3 U/dl, VWF recombinant glycoprotein Ib (VWF:GpIbR) = 32.8 U/dl, and factor VIII (FVIII) = 55.3 IU/dl. Aims The aim of this study is to characterize from a genetic and biochemical standpoint this low VWF phenotype. Methods Coagulation and biochemical methods were used to study the structural and functional pattern of VWF multimers in the index case's plasma. Recombinant wild-type and p.P1127S VWF variants were produced using human embryonic kidney (HEK)-293 cells. In addition, genetic screening was carried out to detect single nucleotide variants of some scavenger VWF/FVIII receptor genes such as CLEC4M, STAB2, and ASGR2. Results Genetic investigation revealed that the index case inherited from her mother the heterozygous missense mutation c.3379C > T (VWF exon 25), causing the p.P1127S substitution in the VWF D ' D3 domain. The index case was also homozygous for the scavenger receptor ASGR2 c.-95 CC-genotype. Desmopressin normalized the VWF level of the patient, although its clearance was faster (t(1/2) = 6.7 h) than in normal subjects (t(1/2) = 12 +/- 0.7 h). FVIII-VWF interaction, A Disintegrin And Metalloprotease with ThromboSpondin type 1 motif-13 levels, ristocetin-induced-platelet-aggregation, and VWF multimeric pattern were normal. The p.P1127S variant was normally synthesized and secreted by HEK-293 cells, and molecular modeling predicts a conformational change showing higher affinity for the macrophagic scavenger receptor lipoprotein receptor-related protein 1 (LRP1), as also experimentally verified. Conclusions The p.P1127S variant may cause a low VWF phenotype, stemming from an increased VWF affinity for the scavenger receptor LRP1 and, consequently, an accelerated clearance of VWF

    Gourmet Table Salts: The Mineral Composition Showdown

    Table salts with their specialty flake size, textures, flavors, and colors can be considered a gastronomy niche food already increasing in demand worldwide. Being unrefined, they can contain trace elements potentially both healthy and toxic. In this study, 12 mineral elements (Al, Ca, Co, Cr, Cu, Fe, Hg, Mn, Ni, Pb, Se, and Zn) in 10 different salts commercially available in southern Italy namely, Atlantic grey, Baule volante, Guerande, Hawaiian pink, Hawaiian black, Himalayan pink, Maldon, Mozia, Persian blue, and smoked salts were analyzed by inductively coupled plasma mass spectrometry (ICP-MS) and thermal decomposition amalgamation-atomic absorption spectrophotometry (TDA-AAS). The concentration of mineral elements was variable according to the type of salt and its geographical origin. Co, Cr, Cu, Hg, Pb, and Se levels were tolerable and Al, Ca, Fe, Mn, Ni, and Zn ranged significantly among the samples. Persian Blue and Atlantic Grey salts showed elevated levels of Fe and Zn; their intake can be helpful in some specific conditions. Nevertheless, Ni levels were high in Persian Blue and Smoked salts. Pb exceeded the maximum level in all samples. Additional monitoring analyses of mineral contents in table salts are recommended for human health
