    Exploiting long-range atomic ordering for the investigation of strain relaxation in lattice-mismatched epitaxy - A case study on CUPtB-type ordered (GaIn)P on GaAs

    The great potential of exploiting growth-induced long-range atomic ordering for investigating strain relaxation in lattice-mismatched epitaxy is demonstrated by transmission electron microscopy (TEM) experiments performed on a CuPtB-type ordered (GaIn)P layer grown on top of a GaAs substrate. Due to the larger unit cell of the ordered structure compared to the zincblende structure of the disordered alloy some of the 60degrees misfit dislocations are no longer perfect dislocations, but are connected to antiphase boundaries (APBs) which were produced during the layer growth by dislocation glide. From the contours of these APBs in (110) cross-section TEM images, we are able to determine post-growth the layer thicknesses at which the individual 60degrees misfit dislocations were formed durin- the layer growth. This information is hardly available from any other technique. Apart from the problem of investigating a sufficiently large number of dislocations by TEM, this allows us to reconstruct the complete path of the strain relaxation. A striking feature revealed by the APBs is the simultaneous formation of many misfit dislocations with identical Burgers vectors at a layer thickness of about 12 times the critical thickness, indicating that dislocation multiplication took place during the layer growth.     Case StudyExtreme thermal piping displacements were thought to be the root cause for the observed failures at a large refinery, namely 9 mechanical seal failures in 3 months. Optical Metrology methods were used to identify failure modes of critical service vacuum bottoms pumps. The testing involved the use of both photogrammetry and digital image correlation to show how the large thermal displacements of the piping contributed to the mechanical seal failures. A high speed optical meteorology rotodynamic study was also performed to observe the effects of the piping displacements on the operation of the pump

    Vegetacija pohojenih habitatov v Prekmurju (SV Slovenija)

    The work deals with the vegetation growing along and on roads, footpaths and wheel tracks in the Prekmurje region. The author concludes that the communities appearing in these habitats can be classified into several syntaxa. The most characteristic associations are classified into the class Polygono-Poëtea annuae. On less trampled sites one can find also the communities that are otherwise partly adapted to trampling, however, they are classified into other syntaxonomic units, for example into the alliance Sisymbrion or Malvion (both from the class Stellarietea mediae) or into the alliance Plantagini-Prunellion from the class Molinio-Arrhentheretea. The following plant communities were established Matricarietum discoideae-recutitae Jarolímek et al. 1997 (Malvion neglectae), Polygono arenastri-Lepidietum ruderalis Mucina 1993 (Sisymbrion) and Poëtum annuae Felföldy 1942 (Polygono-Poëtea annuae) and Prunello vulgaris-Ranunculetum repentis Winterhoff 1963 (Molinio- Arrhenatheretea).Delo obravnava vegetacijo na cestah in ob njih, ob poteh in kolovozih v Prekmurju. Avtor ugotavlja, da lahko združbe, ki se pojavljajo na teh habitatih, uvrstimo v več sintaksonov. Najbolj značilne asociacije uvrščamo v razred Polygono-Poëtea annuae. Na manj pohojenih rastiščih pa najdemo tudi združbe, ki so sicer delno prilagojene na teptanje, vendar pa jih uvrščamo v druge sintaksonomske enote, na primer zvezi Sisymbrion ali Malvion (obe iz razreda Stellarietea mediae) ali v zvezo Plantagini-Prunellion iz razreda Molinio-Arrhenatheretea. Ugotovljene so bile naslednje rastlinske združbe Matricarietum discoideae-recutitae Jarolímek et al. 1997 (Malvion neglectae), Polygono arenastri-Lepidietum ruderalis Mucina 1993 (Sisymbrion), Poëtum annuae Felföldy 1942 (Polygono-Poëtea annuae) in Prunello vulgaris-Ranunculetum repentis Winterhoff 1963 (Molinio-Arrhenatheretea)

SUCHT. 2025;71(5):301-306.Zusammenfassung: Ziel: Angesichts der Herausforderungen der Komorbidität von Substanzkonsum- und Posttraumatischen Belastungsstörungen (PTBS) und der Empfehlungen aktueller Leitlinien wurde untersucht, wie viele Kliniken für Psychiatrie und Psychotherapie in Deutschland ein entsprechendes Therapieangebot („Trauma und Sucht“) vorhalten. Methodik: Dazu wurde ein Fragebogen konstruiert, als SoSci Survey online eingestellt und die Mitgliedskliniken via Fachgesellschaften um Bearbeitung gebeten. Wegen des geringen Rücklaufs (ca. 10 %) wurde eine Nacherhebung per Zufallsauswahl vorgenommen, die drei weitere Klinken mit einem Therapie Angebot Trauma und Sucht erbrachte. Ergebnisse: 15 der 41 antwortenden Kliniken gaben an, ein solches Schwerpunktangebot vorzuhalten, in fast allen (14) Fällen stationär, bei (knapp) der Hälfte auch teilstationär und/oder ambulant, in 6 Kliniken als eigenständiger Bereich. Nach regelhafter Voranmeldung/Indikationsstellung wird die Therapie meist ohne zuvor längere Abstinenzzeiten angeboten, beinhaltet Interventionen zur Stabilisierung, Psychoedukation und Traumakonfrontation sowie suchtspezifische Interventionen (in 9 Kliniken integriert). Die Dauer der Behandlung variiert zwischen unter 4 und über 6 Wochen. Schlussfolgerungen: Diese nicht repräsentative Untersuchung zeigt, dass eine recht kleine Zahl von Kliniken für Psychiatrie und Psychotherapie in Deutschland spezifische Therapieangebote Trauma und Sucht vorhält, diese dann aber an den aktuellen Leitlinienempfehlungen bei unterschiedlicher Organisation und Dauer orientiert zu sein scheinen.Aim: In view of the challenges enfacing the comorbidity of substance use and post-traumatic stress disorder (PTSD) as well as the recommendations of current guidelines, we investigated how many clinics for psychiatry and psychotherapy in Germany offer a corresponding treatment program (“Trauma and Addiction”). Methods: For this purpose, a questionnaire was constructed, posted online as a SoSci Survey and the clinics were asked to complete it via psychiatric societies. Due to the low response rate (approx. 10 %), a follow-up survey was carried out by random selection, which yielded three additional clinics with a trauma and addiction therapy service. Results: 15 of the 41 responding clinics stated that they offered such a specialized service, in almost all (14) cases on an inpatient basis, in nearby half of them also on a day-care and / or outpatient basis, and in 6 clinics as an independent department. After regular pre-registration/indication, therapy is usually offered without longer periods of abstinence and includes interventions for stabilization, psychoeducation and trauma confrontation as well as addiction-specific interventions (integrated in 9 clinics). The duration of treatment varies between less than 4 and more than 6 weeks. Conclusions: This non-representative study shows that a relatively small number of clinics for psychiatry and psychotherapy in Germany provide specific trauma and addiction treatment services, but that these appear to be oriented towards the current guideline recommendations with varying organization and duration

    Identification and characterization of two temperature-induced surface-associated proteins of Streptococcus suis with high homologies to members of the arginine deiminase system of Streptococcus pyogenes

    The present study was performed to identify stress-induced putative virulence proteins of Streptococcus suis. For this, protein expression patterns of streptococci grown at 32, 37, and 42°C were compared by one- and two-dimensional gel electrophoresis. Temperature shifts from 32 and 37 to 42°C induced expression of two cell wall-associated proteins with apparent molecular masses of approximately 47 and 53 kDa. Amino-terminal sequence analysis of the two proteins indicated homologies of the 47-kDa protein with an ornithine carbamoyltransferase (OCT) from Streptococcus pyogenes and of the 53-kDa protein with the streptococcal acid glycoprotein (SAGP) from S. pyogenes, an arginine deiminase (AD) recently proposed as a putative virulence factor. Cloning and sequencing the genes encoding the putative OCT and AD of S. suis, octS and adiS, respectively, revealed that they had 81.2 (octS) and 80.2% (adiS) identity with the respective genes of S. pyogenes. Both genes belong to the AD system, also found in other bacteria. Southern hybridization analysis demonstrated the presence of the adiS gene in all 42 serotype 2 and 9 S. suis strains tested. In 9 of these 42 strains, selected randomly, we confirmed expression of the AdiS protein, homologous to SAGP, by immunoblot analysis using a specific antiserum against the SAGP of S. pyogenes. In all strains AD activity was detected. Furthermore, by immunoelectron microscopy using the anti-S. pyogenes SAGP antiserum we were able to demonstrate that the AdiS protein is expressed on the streptococcal surface in association with the capsular polysaccharides but is not coexpressed with the

    Filopodia explore the environment, sensing soluble and mechanical cues during directional motility and tissue morphogenesis. How filopodia are initiated and spatially restricted to specific sites on the plasma membrane is still unclear. Here, we show that the membrane deforming and curvature sensing IRSp53 (Insulin Receptor Substrate of 53 kDa) protein slows down actin filament barbed end growth. This inhibition is relieved by CDC42 and counteracted by VASP, which also binds to IRSp53. The VASP: IRSp53 interaction is regulated by activated CDC42 and promotes high-density clustering of VASP, which is required for processive actin filament elongation. The interaction also mediates VASP recruitment to liposomes. In cells, IRSp53 and VASP accumulate at discrete foci at the leading edge, where filopodia are initiated. Genetic removal of IRSp53 impairs the formation of VASP foci, filopodia and chemotactic motility, while IRSp53 null mice display defective wound healing. Thus, IRSp53 dampens barbed end growth. CDC42 activation inhibits this activity and promotes IRSp53-dependent recruitment and clustering of VASP to drive actin assembly. These events result in spatial restriction of VASP filament elongation for initiation of filopodia during cell migration, invasion, and tissue repair

    Changes in faecal bacteria and metabolic parameters in foals during the first six weeks of life

    Many foals develop diarrhoea within the first two weeks of life which has been suggested to coincide with postpartum oestrus in their dams. To analyse the pathogenesis of this diarrhoea we have determined faecal bacteria in foals and their dams (n=30 each), and serum IGF-1 and gamma-globulins for 6 weeks after birth. In addition, effects of beta-carotene supplementation to mares (group 1: 1000 mg/day, n=15, group 2: control, n=15) on diarrhoea in foals were studied. Diarrhoea occurred in 92 and 79% of foals in groups 1 and 2, respectively, but was not correlated with oestrus in mares. Beta-carotene supplementation was without effect on foal diarrhoea. In mares, bacterial flora remained stable. The percentage of foals with cultures positive for E. coli was low at birth but increased within one day, the percentage positive for Enterococcus sp. was low for 10 days and for Streptococcus sp. and Staphylococcus sp. was low for 2-4 weeks. By 4 weeks of age, bacterial flora in foals resembled an adult pattern. Concentration of serum IGF-1 was low at birth (group 1: 149 +/- 11, group 2: 166 +/- 17ng/ml), increased after day 1 (day 7 group 1: 384 +/- 30, group 2: 372 +/- 36) but at no time differed between groups. Serum gamma-globulin concentration in foals was low before colostrum intake and highest on day 1 (p<0.001 over time). In conclusion, neonatal diarrhoea in foals does not coincide with postpartum oestrus in their dams but with changes in intestinal bacteria and is not influenced by beta-carotene supplementation given to mares
