    Can nocodazole be used to synchronize cell cycle?

    Objective: Cell cycle for somatic cells is composed of interphase including G1, S and G2, and the M phase. In vitro cultured cells usually exist at various cell cycle phases even though they originated from one mother cell. To analyze specific variations of some proteins, genes or other cellular components it is essential to keep the cells at or bring them to the same cell cycle phase at the maximum level; this is called cell synchronization. Thus, we analyzed the use of benzimidazole derivative nocodazole, a microtubular depolymerizating agent, for cell synchronization. Material and Methods: To synchronize in vitro cultured Vero cells 0.3 mu M nocodazole was used. When untreated cells were considered the control group, the experiment group consisted of cells that were treated with nocodazole for 6, 12 or 18 hours. In the recovery group, following nocodazole treatment, cells were washed and cultured in nocodazole-free medium for 6, 12, 24 or 48 hours. After culture periods, cells were fixed and microtubules (after anti alpha+beta tubulin antibody, FITC) and nuclei (7-AAD) were labeled with fluorescent material. Slides were observed with a confocal microscope and the results were statistically analyzed. Results: Nocodazole kept the cells at mitosis, time dependently. Disrupted microtubules were seen in the cells particularly during mitosis. When nocodazole was removed from the culture medium, cells completed their mitosis and resumed to the interphase stage. At 12 hours of recovery, almost all cells (99%) were in the interphase. This result indicates that nocodazole synchronizes Vero cells. Besides, we also observed that nocodazole induced apoptosis in Vero cells. Conclusion: We noted that nocodazole administration at the appropriate time and dose might be used successfully to synchronize the cells in culture

    Highly Efficient Adsorption of Rh(III) from Chloride Containing Solutions by Triazine Polyamine Polymer

    Adsorption of Rh(III) ions from the aqueous phase containing chloride ions was investigated batchwise using 1,3,5-triazine pentaethylenehexamine (TAPEHA) resin, which is highly resistant to strong acidic conditions. The effects of pH, temperature, initial concentration, and contact time on adsorption were examined. Rh(III) adsorption is favorable from the aqueous phase containing 3.0 and 0.1 M HCl. In both acidic cases, Langmuir is best fitting isotherm equation to system. The kinetics of Rh(III) adsorption on TAPEHA polymer was found to obey the pseudo-second-order kinetic equation, at both cases. Langmuir monolayer adsorption capacities were calculated as 327.03 and 113.76 mg/g in 3.0 and 0.1 M HCl, respectively. Experimental maximum adsorption capacities were measured as 198 and 100 mg/g in 3.0 and 0.1 M HCl, respectively. Due to different Rh(III) complexes with chlorine depending on acid concentrations, adsorption of Rh(III) from solutions containing 3.0 and 0.1 M HCl occurred via two different mechanisms; namely, ion exchange and surface complexation. The reuse of TAPEHA was also studied by column procedure, and the adsorption capacity of TAPEHA was not changed by using it five times. Adsorbed Rh(III) ions onto TAPEHA were completely eluted with thiourea 3% (w/v) in 1.0 M HCl. These findings showed that TAPEHA has a high resistance to acidic solutions and a higher Rh(III) uptake capacity than commercial adsorbents. Hence it can be used for rhodium recycling

Yazar, romanları dışında fikir eserleri, gazeteciliği, köşe yazarlığı ve polemikleriyle de ünlüdür. Peyami Safa, Batılıların otodidakt dediği, kendi kendini yetiştirmiş nadir şahsiyetlerdendir. Kırk üç yıl gibi uzun denecek bir süre boyunca yazmıştır. Edebî değeri olmayan eserlerini Server Bedi takma adı ile yayımlamıştır. Yazarın eserlerinde tahliller önemli yer tutar. Zıt kavramları eserlerinde ustalıkla işlemiştir. Peyami Safa?nın hayatı, eserleri ve sanatı daha önce çok sayıda araştırmacı tarafından incelenmiş, fakat yazarın hayatında büyük yeri olan ve eserlerinde de kendisini yoğun olarak hissettiren hastalık konusu işlenmemiştir. Bu sebeple tarafımızdan araştırma konusu olarak seçilmiştir. Küçük yaşlardan itibaren hastalıklarla boğuşan Peyami Safa, başta Dokuzuncu Hariciye Koğuşu olmak üzere bütün eserlerinde bu durumu yansıtmaya çalışmıştır.     Mapping the Discipline of the Olympic Games An Author-Cocitation Analysis

    The authors conducted an author cocitation analysis on prominent authors writing about the Olympics during the 1990s. Author cocitation is an established bibliometric technique that can be used to measure the relative similarities of topics written about by the cited authors. This enables a visual representation of the “intellectual space” of the discipline, in this case the Olympics, to be created for the period under review. So core and peripheral research areas are identified, along with their major contributors. The representation appears as a two-dimensional cluster-enhanced map. Subject expertise was then applied to the results to place labels on the generated clusters of authors and their topics

    Usage of new generation music streaming platforms such as Spotify and Apple Music has increased rapidly in the last years. Understanding the music preferences of the user base is valuable for these firms, translating into higher customer satisfaction. In this study, we develop and compare several statistical models to quantify the effects of the different factors on music popularity by using acoustic and artist-related features. We compare results from three countries to understand whether there are any cultural differences in how much each of these factors affects song popularity. To compare the results, we use weekly top 200 charts and songs’ acoustic features as data sources. In addition to acoustic features, we add acoustic similarity, genre, song recentness features into the dataset. We apply the Flexible Least Squares (FLS) method and developed Optimal Stepwise Linear Regression (SLR) methods to and observe time-varying regression coefficients. We also propose a regression tree-based heuristic algorithm to solve the SLR problem in a reasonable time. The FLS method tries to keep the differences of adjacent weeks’ coefficients as small as possible. On the other hand, the SLR does not control the variation between consecutive weeks’ coefficients however, it only allows a limited number of coefficient changes over time. Coefficients that we obtain from the FLS method show that we can keep track of the changes of the factors that affect music listening habits and associate real-life events with these changes. Besides, we propose for the SLR method, we can quickly achieve near-optimal solutions which give us clues about the time of the significant changes in the music taste in different countries. Finally, inferences we made with the study may help the music industry grow and contribute to the anthropological fields for discovering socio-cultural/political aspects of the music taste changes.Spotify ve Apple Music gibi yeni nesil m¨uzik akı¸s platformlarının kullanımı son yıllarda hızla artmı¸stır. Bu ¸calı¸smada, akustik ¨ozellikleri ve sanat¸cılar ile ilgili ¨ozellikleri kullanarak farklı fakt¨orlerin m¨uzik pop¨ularitesi ¨uzerindeki etkilerini ¨ol¸cmek i¸cin ¸ce¸sitli istatistiksel modeller geli¸stirilmi¸s ve kar¸sıla¸stırılmı¸stır.     Simulation of thermal plant optimization and hydraulic aspects of thermal distribution loops for large campuses

    Following an introduction, the author describes Texas A&M University and its utilities system. After that, the author presents how to construct simulation models for chilled water and heating hot water distribution systems. The simulation model was used in a $2.3 million Ross Street chilled water pipe replacement project at Texas A&M University. A second project conducted at the University of Texas at San Antonio was used as an example to demonstrate how to identify and design an optimal distribution system by using a simulation model. The author found that the minor losses of these closed loop thermal distribution systems are significantly higher than potable water distribution systems. In the second part of the report, the author presents the latest development of software called the Plant Optimization Program, which can simulate cogeneration plant operation, estimate its operation cost and provide optimized operation suggestions. The author also developed detailed simulation models for a gas turbine and heat recovery steam generator and identified significant potential savings. Finally, the author also used a steam turbine as an example to present a multi-regression method on constructing simulation models by using basic statistics and optimization algorithms. This report presents a survey of the author??s working experience at the Energy Systems Laboratory (ESL) at Texas A&M University during the period of January 2002 through March 2004. The purpose of the above work was to allow the author to become familiar with the practice of engineering. The result is that the author knows how to complete a project from start to finish and understands how both technical and nontechnical aspects of a project need to be considered in order to ensure a quality deliverable and bring a project to successful completion. This report concludes that the objectives of the internship were successfully accomplished and that the requirements for the degree of Degree of Engineering have been satisfied

    Classical transportation models aim to minimize the total costs of homogeneous goods transport from supply points to demand points, taking into account unit transportation costs. They constitute a special case of network models and employ a technique based on linear programming. Suggested in 2015 and one of the early distribution methods, Tuncay Can’s Approximation Method (TCAM) is based on the geometric averages of unit transportation costs, although it is stated in the theorem that other means than geometric can be used. The aim of this study is to compare the total costs of a transportation model by solving a problem using geometric, arithmetic, square, and harmonic means based on TCAM. The coefficients of the transportation model were obtained randomly by simulation, and the method was repeated on the problem according to the different means and the appropriate means determined.Klasik ulaştırma modelleri birim taşıma maliyetlerini göz önüne alarak homojen malların arz noktalarından talep noktalarına taşınma maliyeti toplamını minimize etmeyi amaçlamaktadır. Ulaştırma problemi, ağ modellerinin özel bir halidir ve doğrusal programlama temelli bir tekniktir. Başlangıç dağıtım yöntemlerinden Tuncay Can yaklaşım metodu 2015 yılında geliştirilmiş bir metottur. Yöntem, birim taşıma maliyetlerinin geometrik ortalamalarının alınması esasına dayanmakla birlikte teoremde yöntem uygulanırken geometrik ortalamalar yerine farklı ortalamaların da kullanılabileceği belirtilmiştir. Bu çalışmanın amacı, Tuncay Can Yaklaşım Metodunu (TCYM) temel alarak, yöntemin belirttiği şekilde birim maliyetlerin geometrik ortalamalarının alınması ve ayrıca aritmetik, kareli ve harmonik ortalama kullanılarak da yöntemin uygulanması ile elde edilen toplam maliyetleri minimize eden başlangıç dağıtım planı incelenerek hangi ortalamada optimal sonuç verdiğini ortaya koymaktır. Bu amaca yönelik olarak kurulan ulaştırma modelinin katsayıları simülasyon yardımıyla rassal olarak değiştirilmiş ve yöntem farklı ortalamalara göre problem üzerinde tekrarlanarak, optimal toplam maliyet değerleri karşılaştırılmış ve uygun ortalama tespit edilmiştir
