    Timoria Novalinda, 2022. Effect of Plant Growth Regulators on Micropropagation of Local Red Ginger Cultivars (Zingiber officinale var. Rubrum) Through In Vitro Culture, supervised by Luthfi Aziz Mahmud Siregar and Emmy Harso Kardhinata. This research aims to explore the combination of BAP and NAA with a specific concentration on MS medium toward micropropagation of local red ginger cultivars (Zingiber officinale var. Rubrum) through in vitro culture. This research was conducted at the Tissue Culture Laboratory of the ALIFA Agricultural Research Center (ALIFA-ARC) Medan from September 2021-February 2022. This research used a completely randomized design (CRD) with two treatment factors: plant growth regulators (without plant growth regulators, 2 mg/l BAP + 0.5 mg/l NAA, 3 mg/l BAP + 1 mg/l NAA, 3 mg/l BAP + 0.5 mg/l NAA and 5 mg/l BAP + 0.5 mg/l NAA) and local cultivars (local cultivar 1 and local cultivar 2). The results showed that at the stage of shoot induction, the treatment plant growth regulators had a significant effect on the number of shoots, color of shoots and length of shoots, but not a significant effect on the number of roots and length of roots. The treatment of local cultivars had no significant effect on each observation parameter. The interaction of the two treatments significantly affected the color of shoots. The combination of 3 mg/l BAP + 1 mg/l NAA responded to the number of shoots and length of shoots.Timoria Novalinda, 2022. Penelitian ini menggunakan Rancangan Acak Lengkap (RAL) dengan dua faktor perlakuan: zat pengatur tumbuh (tanpa zat pengatur tumbuh, 2 mg/l BAP + 0.5 mg/l NAA, 3 mg/l BAP + 1 mg/l NAA, 3 mg/l BAP + 0.5 mg/l NAA dan 5 mg/l BAP + 0.5 mg/l NAA) dan kultivar lokal (kultivar lokal 1 dan kultivar lokal 2). Hasil penelitian menunjukkan bahwa pada tahap induksi tunas, perlakuan zat pengatur tumbuh berpengaruh nyata terhadap jumlah tunas, warna tunas dan panjang tunas, akan tetapi berpengaruh tidak nyata terhadap jumlah akar dan panjang akar. Perlakuan kultivar lokal berpengaruh tidak nyata pada setiap parameter pengamatan. Interaksi kedua perlakuan berpengaruh nyata terhadap warna tunas. Revandy I.M Damanik M.Si., M.Sc., Ph.D. The puspose of this research is to identify the morphological characters of the crew banana plant (Musa paradisiaca cv. Awak) in Percut Sei Tuan District. This research was conducted in Percut Sei Tuan Subdistrict in the villages of Bandar Setia, Bandar Khalippa, Bandar Klippa, Saentis, and Sampali from July to September 2020 using a descriptive survey method using descriptors for banana (International Plant Genetic Resources Institute) guides with retrieval techniques sample by purposive sampling 15 samples of the awak bananas were obtained from 5 villages and 3 different samples were taken for each village.     KHOSMALINDA ANGGRAENI.S, 2020, Identifikasi dan Karakter Morfologis Tanaman Kopi (Coffea Sp.) di Kabupaten Dairi. Dibimbing oleh Prof.Dr.Ir. Rosmayati M.S dan Luthfi Aziz Mahmud Siregar,SP, M.Sc.Ph.D. Penelitian ini bertujuan untuk mengidentifikasi karakterisasi tanaman kopi (Coffea sp.) secara morfologis dan hubungan kekerabatan beberapa genotipe di Kabupaten Dairi Sumatera Utara. Penelitian ini dilakukan di tiga Kecamatan yaitu Kecamatan Sidikalang, Kecamatan Parbuluan dan Kecamatan Sitinjo pada bulan Maret-Agustus 2020 dengan metode survei menggunakan panduan deskriptor kopi IPGRI (International Plant Genetic Resources). Teknik pengambilan sampel secara acak. Parameter yang diamati adalah penampilan menyeluruh, tinggi tanaman, perwatakan tanaman, perkembangan vegetatif, bentuk stipule, bentuk daun, bentuk ujung daun, panjang daun, lebar daun, panjang petiol daun, warna daun muda, warna petiol daun, warna daun pucuk muda, warna daun dewasa, pola venation, posisi pembungaan, pembungaan batang tua, jumlah bunga axil-1, jumlah bunga fascicle-1, warna buah, bentuk buah, ribs buah, keteguhan calyx, tekstur endocarp, panjang buah, lebar buah, ketebalan buah, ketebalan pulp, panjang biji, lebar biji, ketebalan biji, warna biji, bentuk biji, berat biji kering. Hasil penelitian menunjukkan bahwa di Kabupaten Dairi Sumatera Utara terdapat 27 genotipe kopi yang diidentifikasi. Berdasarkan dendogram yang terbentuk hubungan kekerabatan pada skala jarak kekerabatan 16, 18, dan 25 dengan hubungan kekerabatan terdekat pada genotipe D20 dan D21 dengan koefisien ketidaksamaan sebesar 1,122% dan hubungan kekerabatan terjauh pada genotipe D1 dan D19 dengan koefisien ketidaksamaan sebesar 8,618%, dengan kadar kafein tertinggi di D19 yaitu 13,95% dan kadar kafein terendah D9 yaitu 7,775%.KHOSMALINDA ANGGRAENI.S, Identification and Morphological Characteristics of Coffee Plants ( Coffea sp.) in Dairi Regency.     Muhammad Amri Yusra Simatupang, 2021. Reciprocal Cross Test of Glutinuous Corn (Zea mays L. var. URI) and Sweet Corn (Zea mays L. var. Bonanza Now), supervised by Luthfi Aziz Mahmud Siregar, SP, M,Sc, Ph.D and Dr. Diana Sofia Hanafiah SP, MP. The purpose of this study was to determine the maternal effect on the inheritance of maize traits through a reciprocal cross test between glutinuous corn (Zea mays L. var. URI) and sweet corn (Zea mays L. var. bonanza Now). This research was conducted on Jalan Sei Brantas, Medan with an altitude of ± 32 meters. This research was conducted from October 2020 to January 2021. The experimental design used was Reciprocal Cross, namely planting and observing each individual plant of the F1 generation. Crosses were carried out between the female elders of sweet corn and the male elders of sweet corn (F1), the male elders of sweet corn and the female elders of glutionuous corn (F1R) in 8 plots where each plot was planted with 4 plants. The total number of plants is 128 plants, with a sample of 1 plant per plot in the total sample is 32 samples. Based on the data obtained from the experiment, a t-test was performed at the 5% level and chi square test.The results showed that the percentage of crossing between sweet corn and glutinuous corn was 94%. Qualitative factors that affect the cross, namely the shape of the seeds from the crosses are round, while the color of the seeds from the dominant cross is yellow. The quantitative factor affected by the cross was significantly difference in the results on the the length of the seedless cobs, the diameter of the seedless cobs and weight of 100 seeds However, there was no significant difference in the parameters of the weight of the dry cobs.Muhammad Amri Yusra Simatupang, 2021 dengan judul penelitian “Uji Persilangan Resiprokal Jagung Pulut (Zea mays L. var. Sehingga total keselurahan berjumlah 128 tanaman, dengan sampel berjumlah 1 tanaman per plot sehingga total keseluruhan sampel berjumlah 32 sampel. Berdasarkan data yang diperoleh dari percobaan, dilakukan uji t pada taraf 5 %. dan uji chi square. Hasil penelitian menunjukkan bahwa persentase keberhasilan persilangan jagung manis dan jagung pulut adalah 94 %. Faktor kualitatif yang mempengaruhi persilangan yaitu bentuk biji hasil persilangan berbentuk membulat, sedangkan warna biji hasil persilangan dominan berwarna kuning. Faktor kuantitatif yang mempengaruhi persilangan yaitu terdapat perbedaan yang nyata hasil persilangan pada parameter panjang tongkol, diameter tongkol dan berat 100 biji dan. TOLERAN TERHADAP KONDISI HIPOKSIA SECARA IN VITRO, dibimbing oleh Revandy I. M. Damanik dan Luthfi Aziz M.Siregar. Penelitian ini bertujuan untuk mengetahui respon pertumbuhan dan identifikasi kalus embriogenik pada beberapa varietas kedelai (Glycine max (L.) Merill) toleran terhadap kondisi hipoksia secara in vitro. Penelitian ini dilaksanakan di Laboratorium Kultur Jaringan dan Laboratorium Teknologi Pangan Fakultas Pertanian Universitas Sumatera Utara, Medan dari bulan Maret sampai September 2016, Rancangan yang digunakan adalah rancangan acak lengkap (RAL) faktorial dengan 2 faktor perlakuan. Faktor pertama adalah varietas kedelai yang terdiri dari 3 taraf yaitu Baluran, Gepak kuning, Wilis. Faktor kedua adalah media dengan zat pengatur tumbuh yang terdiri dari 3 taraf yaitu 2,4-D, NAA, IAA 10 mg/l. Parameter yang diamati adalah persentase eksplan membentuk kalus embriogenik, warna kalus, struktur kalus, tipe perkembangan kalus, analisis histologi kalus, bobot segar kalus total, pengukuran kandungan klorofil total, pengukuran konsentrasi protein, pengukuran nilai aktifitas enzim Super Oksida Dismutase (SOD) dan pengukuran nilai aktivitas enzim peroksidase (POD). Hasil penelitian menunjukkan bahwa interaksi varietas Gepak kuning dan Wilis dengan pemberian ZPT 2,4-D 10 mg/l berpengaruh terhadap pertumbuhan kalus embriogenik, bobot segar kalus total, kandungan klorofil total, konsentrasi protein, nilai aktivitas enzim Super Oksida Dismutase (SOD) dan nilai aktivitas enzim Peroksidase (POD), tetapi perlakuan varietas Baluran terhadap ketiga jenis ZPT dan perlakuan ZPT NAA dan IAA 10 mg/l terhadap ketiga jenis varietas tidak berpengaruh terhadap semua parameter yang diamatiSopa Putri Tanjung, 2016: Potential formation in some variety embryogenic callus soybean (Glycine max (L.) The first factor is soybean varieties which consists of three levels ie baluran, Gepak yellow, Wilis. The second factor is the media with growth regulators which consists of three levels ie 2,4-D, NAA, IAA 10 mg/l. The parameters measured were the percentage of explants forming embryogenic callus, color callus, structure callus, types of development callus, histological analysis of callus fresh weight callus total, the measurement of chlorophyll content in total, measurement of protein concentration, measuring the value of the activity of enzyme Super oxide dismutase (SOD) and measurement value of the activity of peroxidase (POD).     M. BINTANG ANGGARA : Analysis of Morphological and Agronomic Characteristics of Cassava (Manihot esculenta Crantz) in Serdang Bedagai Regency, North Sumatra. Supervised by Dr. Ir. Emmy Harso Kardhinata, M.Sc and Luthfi Aziz Mahmud Siregar, SP., M.Sc., Ph.D. This study aims to analyze the morphological and agronomic characters of cassava plants in Serdang Bedagai Regency, North Sumatra. The research was carried out on the land of Silau Rakyat Village, Sei Rampah District, Serdang Bedagai Regency, North Sumatra, Indonesia (± 11 masl) from July 2020 to February 2021. The description of the morphological character analysis was carried out based on Fukuda et al. (2004) and analysis of agronomic characters using a randomized block design of one factor, namely the cassava genotype obtained in Sei Rampah District. From the results of the study obtained 10 genotypes of cassava with various local names depending on the area where the plant is found, namely G1 (Malaysia), G2 (Klanting Merah), G3 (Genderuwo Hitam), G4 (Bread), G5 (Keling), G6 ( Sawit Gading), G7 (Genderuwo Merah), G8 (Lampung), G9 (Simerah) and G10 (Klanting Putih). Each genotype shows a distinctive character that can distinguish it from other genotypes, including the character color of shoot leaves, color of old leaves, young stem color and old stem color. Analysis of agronomic characters showed no significant difference among genotypes in the number of cassava/tree. The highest cassava length was in G7 and G3, and the lowest was in G4. The average diameter of cassava was almost the same for all genotypes , as well as the average weight of cassava per tree except for genotypes G5, G4 and G1 which had lower cassava weight per tree compared to other genotypes.M. Penelitian dilaksanakan di lahan Desa Silau Rakyat Kecamatan Sei Rampah Kabupaten Serdang Bedagai, Sumatera Utara, Indonesia (± 11 mdpl) dari bulan Juli 2020 sampai Febuari 2021. Deskripsi analisis karakter morfologis dilakukan berdasarkan Fukuda et al. (2004) dan analisis karakter agronomis menggunakan Rancangan Acak Kelompok satu faktor yaitu genotip ubikayu yang diperoleh di Kecamatan Sei Rampah. Dari hasil penelitian diperoleh 10 genotip ubikayu dengan nama lolal yang bervariasi bergantung pada daerah dimana tanaman tersebut dijumpai, yaitu G1 (Malaysia), G2 (Klanting Merah), G3 (Genderuwo Hitam), G4 (Roti), G5 (Keling), G6 (Sawit Gading), G7 (Genderuwo Merah), G8 (Lampung), G9 (Simerah) dan G10 (Klanting Putih). Masing-masing genotip menunjukkan karakter yang khas yang dapat membedakan dengan genotip lainnya, antara lain pada karakter warna daun pucuk, warna daun tua, warna batang muda dan warna batang tua. Analisis karakter agronomi menunjukkan tidak ada perbedaan yang signifikan antar genotip pada karakter jumlah ubikayu per pohon. Panjang ubikayu tertinggi terdapat pada G7 dan G3, dan terendah pada G4. Diameter rata-rata ubikayu hampir sama untuk semua genotip, demikian juga berat rata-rata ubikayu per pohon kecuali genotip G5, G4 dan G1 yang memiliki berat ubikayu per pohon yang rendah dibandingkan genotip lainnya.139 HalamanSkripsi Sarjan

    LYLI HERAWATI SIREGAR : The Micropropagation of Boesenbergia flava bud by BAP and NAA for in vitro, guided by oleh Luthfi Aziz Mahmud Siregar dan Lollie Agustina P. Putri. The research aimed to know the influence of BAP and NAA concentration on micropropagation of Boesenbergia flava bud for in vitro. The research was carried out in the Tissue Culture Laboratory, Agriculture’s facuity of Nort Sumatera University from March to May 2012. This research used Completely Randomezed Design with two treatment factors. First factor was NAA concentration consist of four level; 0 mg/l; 1 mg/l; 2 mg/l; 3 mg/l. The second factor was BAP concentration consist of four level are 0 mg/l; 1,5 mg/l; 3 mg/l; 4,5 mg/l. The results of research showed that NAA concentration give significantly affected on long of roots and plant height and BAP concentration give significantly affected on long of roots and plant height but that was not significantly affected on roots number and leaves number.
