É tude spectroscopique des états excitoniques liés dans les nanotubes de carbones monoparois
Dans ce manuscrit nous décrivons des études originales sur les propriétés photophysiques des nanotubes de carbone monoparois réalisées à l'échelle de la molécule unique. Nous nous sommes concentrés sur deux problématiques : l'effet du remplissage des nanotubes par de l'eau sur leurs propriétés de photoluminescence (PL) et la création de complexes multi-excitoniques en régime de forte excitation laser. Dans ce but nous avons utilisé une combinaison de microscopie, de spectroscopie et de mesures de déclin de PL. Nos résultats montrent pour des nanotubes de différents diamètres un décalage vers le rouge des énergies d'émission pour les nanotubes remplis d'eau. De plus, des déclins de PL biexponentiels sont obtenus pour des nanotubes individuels (6,4) vides et remplis d'eau. Les temps de déclin caractéristiques de ces deux espèces de nanotubes sont distincts, avec une réduction de la composante courte pour les nanotubes remplis. Ces résultats sont expliqués par une augmentation de la constante diélectrique dans les nanotubes remplis d'eau. Notre résultat le plus conséquent a été l'observation de la génération de trions dans des nanotubes non dopés en utilisant des moyen tout optiques. L'émission du trion apparaît dans les spectres de PL comme une bande latérale décalée vers le rouge. Basé sur nos observations expérimentales, nous proposons le modèle de génération du trion suivant lequel dans un régime multiexcitonique les interactions d'annihilation exciton-exciton créent des porteurs de charge qui sont piégés dans les fluctuations de potentiel électrostatique induites par les inhomogénéités de l'environnement. L'absorption subséquente d'un photon amène à la formation d'un trion localisé sur les charges piégées.In this dissertation we report on original experimental investigations of the photophysical properties of individual single-walled carbon nanotubes (SWCNTs) at the single molecule level. We focused on two problems: the effect of water-filling nanotubes on their photoluminescence (PL) properties and generation of multi-excitonic complexes in regimes of strong laser excitation. To do so we used a combination of microscopy, PL spectroscopy and PL time decay measurements. Our results show, for different nanotube diameters, a red-shift of the PL emission energy for water-filled nanotubes. Furthermore, biexponential PL time decay behaviors are obtained for individual water-filled and empty (6,4) SWCNTs. The characteristic decay times for both species of nanotubes are distinct, with a reduction of the short component in water-filled SWCNTs. These results are explained by an increase of the dielectric constant for water-filled nanotubes. Our most consequential experimental result was the observation of all optical trion generation in undoped nanotubes. Trion emission appears in the PL spectrum as a red-shifted sideband. Based on our experimental observations, we propose a trion generation model according to which, at a multiexcitonic regime, exciton-exciton annihilation interactions create charge carriers that are transiently trapped at electrostatic potential fluctuations induced by the inhomogeneities of the environment. Subsequent photon absorption leads to trion formation localized at the trapped carriers
Microscopy of single nano-objects : study of integrins diffusion in focal adhesions in live cells
L’effet photothermique permet de détecter efficacement des nanoparticules d’or avec un microscope en champ lointain grâce à leur forte absorption de la lumière. L’absence de problème photophysique fait des nanoparticules d’or une alternative au marquage de biomolécules par des sondes fluorescentes. La méthode PhACS (Photothermal Absorption Correlation Spectroscopy) utilise les fluctuations de signal photothermique dues au passage de nanoparticules dans le volume de détection pour étudier leur diffusion. Cette méthode permet également la mesure précise de diamètres hydrodynamiques de nanoparticules fonctionnalisées. La méthode SNaPT (Single Nano-Particle Tracking) réalise le suivi bidimensionnel de nanoparticules individuelles grâce à une localisation effectuée par triangulation. Nous avons appliqué cette méthode pour étudier la diffusion des intégrines alphaV-beta3 marquées par des nanoparticules d’or de 5 nm dans les adhérences focales, points d’ancrage entre le cytosquelette de la cellule et la matrice extracellulaire. Nous observons que ces intégrines ont tendance à former des agrégats qui alternent entre un mouvement diffusif et un mouvement confiné. Ce résultat appelle maintenant à un nouveau modèle où nous aurions une redistribution continue des intégrines au sein des adhérences focales.Gold nanoparticles may be detected with optical far-field microscopy by use of the photothermal effect due to their strong light absorbance. With no photophysic issues, gold nanoparticles are an alternative to fluorescent probes for use in biological systems. The PhACS method (Photothermal Absorption Correlation Spectroscopy) is used to study diffusion by measuring the autocorrelation of photothermal signal fluctuations due to nanoparticles passing through the detection volume. This method is sensitive enough to mesure the precise hydrodynamic diameter of functionalised nanoparticles. The SnaPT method (Single Nano-Particle Tracking) can track 2-dimensional motion of individual nanoparticles by pinpointing the localization with a triangulation method. The SNaPT method was used to study motion of alphaV-beta3 integrins that were bound to a 5 nm gold nanoparticle inside focal adhesion, where the cell cytoskeleton is linked to the extracullular matrix. The integrin was found to organize into clusters oscillating between the bound and diffuse states. These observations require new working models where integrins would be constantly redistributed

This thesis examines the interaction of light with single dibenzanthanthrene (DBATT) dye molecules. DBATT serves as a model for a quantum-mechanical two-level system. By means of strong focusing of the incident light and cooling of the dye molecules to temperatures below 2 K, a particularly efficient light-matter interaction can be realized. This enables observation of the nonlinearity inherent to a two-level system, e.g., in the form of saturation of the fluorescence signal with light beams containing only a few photons per lifetime of the excited molecular state.
Various nonlinear phenomena arise when two light beams of different frequency are sent to a single molecule. These processes can be exploited to coherently manipulate the transmission of a beam focused onto a single molecule via a second light beam. The occurrent effects, i.e., the AC-Stark shift, stimulated Rayleigh scattering, and three-photon amplification, are detected in the transmitted signal. In addition, four-wave mixing and the dependence of the excited state population on the phase difference of the two incident beams are demonstrated by the use of measurements with subnanosecond time resolution. These results show the possible application of organic dye molecules in the field of quantum information processing where nonlinearities on the single photon and single emitter level are highly sought-after.
In this work, the experimental and theoretical principles of single molecule spectroscopy are discussed. Particular attention is placed on the investigation of the coherent light-matter interaction using transmission measurements. A significant change in this transmitted signal, via the scattering of a single molecule, requires a strong light-matter coupling. To quantify the efficiency of this interaction, the maximum possible coupling of a focused beam and a single emitter is discussed. The results presented herein show that the achieved coupling is typically 5% of the theoretical maximum. Thus, the interaction of a molecule with two light fields with different frequencies is investigated. The nonlinear effects that arise are described qualitatively within the dressed-atom model and quantitatively with a Fourier ansatz. The experimental techniques are explained in detail and the measurement results are presented and discussed.Die vorliegende Dissertation behandelt die Wechselwirkung von Licht mit einzelnen Dibenzanthanthren (DBATT) Farbstoffmolekülen. DBATT dient hierbei als Modell für ein quantenmechanisches Zweiniveau-System. Durch starke Fokussierung des einfallenden Lichts und Kühlung der Farbstoffmoleküle auf eine Temperatur unter 2 K kann eine besonders effiziente Licht-Materie-Wechselwirkung realisiert werden. Diese ermöglicht es bereits mit einem Lichtstrahl, der nur wenige Photonen pro Lebenszeit des angeregten Molekülzustandes enthält, die inhärente Nichtlinearität eines Zweiniveau-Systems, zum Beispiel in Form von Sättigung des Fluoreszenzsignals, zu beobachten.
Verwendet man zwei Lichtstrahlen mit unterschiedlichen Frequenzen, so ergeben sich verschiedene nichtlineare Phänomene. Diese Prozesse können dazu genutzt werden, die Transmission eines Strahls, der auf ein einzelnes Molekül fokussiert ist, mithilfe eines zweiten Lichtstrahls kohärent zu beeinflussen. Die hierbei auftretenden Effekte, die AC-Starkverschiebung, stimulierte Rayleigh-Streuung und drei-Photonenverstärkung werden im Transmissionssignal nachgewiesen. Durch Messungen mit subnanosekunden Zeitauflösung wird außerdem Vier-Wellenmischung und die Abhängigkeit der Population im angeregten Zustand von der relativen Phase der beiden einfallenden Lichtstrahlen demonstriert. Diese Ergebnisse zeigen die Einsatzmöglichkeit organischer Farbstoffmoleküle im Bereich der Quanteninformationsverarbeitung, wo Nichtlinearitäten auf dem Niveau einzelner Photonen und einzelner Emitter benötigt werden.
In dieser Arbeit werden die experimentellen und theoretischen Grundlagen der Einzelmolekülspektroskopie besprochen. Ein besonderes Augenmerk liegt hierbei auf der Untersuchung der kohärenten Licht-Materie-Wechselwirkung mit Hilfe von Transmissionsmessungen. Eine signifikante Änderung dieses Signals durch die Streuung eines einzelnen Moleküls erfordert eine starke Licht-Materie-Wechselwirkung. Um die Effizienz dieser Interaktion zu quantifizieren, wird die maximal erreichbare Kopplung eines fokussierten Lichtstrahls an einen einzelnen Emitter erörtert. Dabei zeigt sich, dass die Kopplung im verwendeten System typischerweise bei 5% des theoretischen Maximums liegt. Anschließend wird die Wechselwirkung eines Moleküls mit zwei Lichtfeldern unterschiedlicher Frequenz untersucht. Die sich dabei ergebenden nichtlineare Effekte werden im Rahmen des dressed atom-Modells qualitativ und mittels eines Fourieransatzes quantitativ beschrieben. Die experimentellen Techniken werden ausführlich dargelegt und die Ergebnisse der Messungen präsentiert und diskutiert
Nanoscopie RESOLFT appliquée à l'étude de l'adhésion cellulaire
Les cellules peuvent ajuster leur adhésion au substrat et leur cytosquelette en fonction des variations de nature biochimique et physique de leur environnement. En retour, les cellules peuvent également contrôler leur microenvironnement en adhérant et en générant des forces sur les cellules voisines et la matrice extracellulaire. Les adhérences focales (AF) dépendantes des intégrines sont ainsi les zones de convergences de ces interactions, intégrant les signaux biochimiques et biomécaniques entre les composants intracellulaires et extracellulaires. Diverses fonctions cellulaires liées aux AF sont associées à la formation et à la cohésion des tissus. Les cellules cancéreuses qui se propagent dans le système circulatoire utilisent des mécanismes associés à l'adhésion cellulaire pour établir de nouvelles tumeurs dans le corps.Les techniques nanoscopiques ont révolutionné l'étude des structures biologiques en permettant à la microscopie optique d’imager sous la limite de diffraction. Le RESOLFT (Reversible Saturable OpticaL Fluorescent Transitions) permet de dépasser cette limite de diffraction en réduisant le volume d'émission par un procédé réversible On-Off. Après avoir étudié la photophysique de la protéine fluorescente rsEGFP2 qui a la propriété d’être réversiblement commutable, pour déterminer son potentiel d'utilisation pour l'imagerie RESOLFT, nous avons utilisé les paramètres photophysiques obtenus dans la construction d’un nanoscope RESOLFT. L’utilisation de cette approche a permis d’acquisition d'image à une résolution 4 fois supérieure à la limite de diffraction (~50 nm). Le RESOLFT présente aussi l’avantage de ne nécessiter qu’une faible intensité laser, ce qui réduit l'effet de photo-blanchiment et permet des acquisitions longues sur cellules vivantes.Nous avons ensuite utilisé le RESOLFT pour étudier l'initiation, la stabilisation et le désassemblage des AF. Nous avons pu étudier la réorganisation dynamique de diverses protéines impliquées dans la formation d’AF sur des fibroblastes embryonnaires murin et des tissus ovariens de drosophila melanogaster. Nous avons observé au sein des AF des amas de β3-integrin-rsEGFP2 dont la taille est inferieurs à la limite de diffraction, prouvant ainsi la capacité du RESOLFT à étudier la réorganisation à l'échelle nanométrique des protéines dans les AF. Nous avons aussi observé une organisation rapide des amas de β3-integrin-rsEGFP2 à l'intérieur des AF, la durée de vie des amas n’étant que de quelques dizaines de secondes. Ces données sous-tendent un modèle de remodelage constant des amas des protéines des AF. Il était également évident que seule une faible densité d’amas d'intégrines est nécessaire pour maintenir les AF. De plus, nous avons observé un flux rétrograde d’amas de talin-rsEGFP2 dans les AF. La vitesse mesurée de ce flux s'est avérée similaire à celle obtenue par suivi de molécules uniques. Nous avons également étudié la dynamique des amas de zyxine dans les AF et observé des fluctuations du taux de zyxine au sein des AF. Nos expériences indiquent un lien entre ces fluctuations et les forces mécaniques existantes dans les AF.Enfin, pour augmenter la vitesse d’acquisition, nous avons parallélisé notre configuration RESOLFT à l'aide d’un maillage optique. Les maillages optiques sont des configurations périodiques de lumière formées par interférence. Comparé à notre première implémentation du RESOLFT, qui permet de sonder qu’un seul point à la fois, les maillages optiques peuvent être utilisés pour sonder simultanément plusieurs points. Nous avons également mis en place un deuxième maillage optique permettant la commutation du fluorophore, ce qui nous a permis de réduire les effets de photo-blanchiment lors de l'imagerie. Cette configuration en double maillage nous a permis d'obtenir une série d’images (>40 images) de nanostructures avec des résolutions proches de 55 nm sur des cellules vivantes à une fréquence de 0,3 kHz sur un champ de vision 15x15μm.Cells can adjust their adhesive and cytoskeletal organizations depending on the changes in the biochemical and physical nature of their surroundings. In return, cells can also control their microenvironment by adhering and generating forces on neighboring cells and extracellular matrices. Integrin-dependent adhesion sites (AS) are the converging zones of these interactions, integrating biochemical and biomechanical signals between intracellular and extracellular components. Various cellular functions linked to AS are associated with the formation and cohesion of tissues. Cancer cells that spread through the circulatory system use mechanisms associated with cell adhesion to establish new tumors in the body.Nanoscopic techniques have revolutionized the study of biological structures by bringing sub-diffraction imaging into the realm of light microscopy. RESOLFT (Reversible Saturable Optical Fluorescent Transitions) break the diffraction barrier by suppressing the volume of emission through a reversible on-off process. We studied the photophysics of a reversible switchable protein called rsEGFP2 expressed in living cells. The parameters for photoswitching obtained from these studies were used to implement a RESOLFT nanoscope. Using this system, we have demonstrated an imaging resolution close to 50 nm, a 4-fold improvement over the diffraction limit. RESOLFT nanoscopy requires less intensity to obtain a super-resolved image, thereby reducing the effect of photobleaching and enabling long term live cell imaging.We then used the RESOLFT nanoscope to study the initiation, stabilization, and disassembly of AS. Using this technique, we were able to study the dynamic reorganization of various proteins involved in the formation of AS on mouse embryonic fibroblasts and ovarian tissues of drosophila melanogaster. We observed clusters of β3-integrin-rsEGFP2 within the AS that are smaller than the diffraction limit, demonstrating RESOLFT's ability to study the nanoscale organization of proteins in the AS. We also observed a rapid reorganization of the β3-integrin-rsEGFP2 clusters within the AS, with the cluster lifetime only being a few tens of seconds. These data suggest a model of constant remodeling of AS protein clusters. In the case of Talin rsEGFP2, we observed a rearward flow of clusters towards the interior of the cell. The speed of this flow was measured and was found to be close to the speed measured by other techniques like single-molecule tracking. We also studied the dynamics of zyxin nanoclusters in AS. We recorded nanoscale fluctuations of zyxin levels in AS, and our experiments indicate a link between this fluctuation and mechanical forces existing inside AS.To enable faster imaging, we parallelized our RESOLFT setup using optical lattices. Optical lattices are periodic patterns of light formed through interference. Compared to our first implementation of RESOLFT, which probes only a single point at a time, optical lattices can be used to probe multiple points simultaneously. We also implemented a second lattice for fluorophore on switching, which helped us to reduce the effects of photobleaching during imaging. The dual lattice setup allowed us long term imaging (>40 frames) of nanostructures with resolution close to 55 nm inside living cells, at a frame rate of 0.3kHz over a 15x15μm field of view
Fundamental optical properties of single semiconductor nanocrystals at cryogenic temperatures
Les nanocristaux de semi-conducteurs présentent des propriétés optiques et électroniques remarquables en raison du confinement quantique de leurs porteurs de charge, ce qui les rend avantageux pour diverses applications en optoélectronique, dans les dispositifs émetteurs de lumière et dans les technologies basées sur le spin. La compréhension de la physique de l’exciton de bord de bande, dont la recombinaison est à l’origine de leur photoluminescence, est cruciale pour le développement de ces applications. Cette thèse porte sur l’étude expérimentale des propriétés optiques des nanocristaux de phosphure d’indium et de pérovskites d’halogénure de plomb. En utilisant une méthode de spectroscopie de magnéto-photoluminescence sur des nanocristaux uniques à basse température, nous révélons des empreintes spectrales très sensibles à la morphologie des nanocristaux et élucidons la structure fine de l’exciton de bord de bande et les énergies de liaison des complexes de charge. Dans les nanocristauxd’InP/ZnS/ZnSe, l’évolution des spectres et des déclins de luminescence sous champ magnétique montrent l’existence d’un niveau d’exciton noir situé à moins d’un millielectronvolt en dessous du triplet brillant de l’exciton, résultats étayés par un modèle tenant compte de l’anisotropie de forme du coeur d’InP. Dans les pérovskites d’halogénure de plomb, nous démontrons que l’état fondamental de l’exciton est noir et se situe plusieurs millielectronvolts en dessous des sous-niveaux d’exciton brillants les plus bas, résolvant ainsi le débat sur l’ordre des niveaux brillants et noirs de l’exciton dans ces matériaux. En combinant nos résultats avec des mesures spectroscopiques sur divers composés de nanocristaux de pérovskite, nous établissons des lois d’échelle universelles qui relient l’éclatement de la structure fine de l’exciton et les énergies de liaison du trion et du biexciton à l’énergie de l’exciton de bord de bande dans les nanostructures de pérovskite d’halogénure de plomb, quelle que soit leur composition chimique. Enfin, des analyses préliminaires de spectroscopie sur des nano-bâtonnets de pérovskite avec un grand rapport d’aspect suggèrent leur potentiel en tant qu’émetteurs de lumière quantique grâce à leur émission composée d’une raie unique.Semiconductor nanocrystals exhibit outstanding optical and electronic properties due to the quantum confinement of their charge carriers, making them valuable for various applications in optoelectronics, light-emitting devices, and spin-based technologies. Understanding the physics of the band-edge exciton, whose recombination is at the origin of their photoluminescence, is crucial for developing these applications. This thesis focuses on the experimental study of the optical properties of indium phosphide and lead halide perovskites nanocrystals. Using magneto-photoluminescence spectroscopy onsingle nanocrystals at low temperatures, we reveal spectral fingerprints highly sensitive to nanocrystal morphologies and elucidate the entire band-edge exciton fine structure and charge-complex binding energies. In InP/ZnS/ZnSe nanocrystals, the evolution of photoluminescence spectra and decays under magnetic fields show evidence for a ground dark exciton level lying less than a millielectronvolt below the bright exciton triplet, findings supported by a model accounting for the shape anisotropy of the InPcore. In lead halide perovskites, we demonstrate that the ground exciton state is dark and lies several millielectronvolts below the lowest bright exciton sublevels, settling the debate on the bright-dark exciton level ordering in these materials. Combining our results with spectroscopic measurements on various perovskite nanocrystal compounds, we establish universal scaling laws relating exciton fine structure splitting, trion and biexciton binding energies to the band-edge exciton energy in lead-halide perovskitenanostructures, regardless of their chemical composition. Lastly, preliminary spectroscopy analyses on perovskite nanorods with a high aspect ratio suggest their potential as candidates for quantum light emitters due to their characteristic single emission line
Going Beyond Counting First Authors in Author Co-citation Analysis
The present study examines one of the fundamental aspects of author co-citation analysis (ACA) - the way co-citation
counts are defined. Co-citation counting provides the data on which all subsequent statistical analyses and mappings
are based, and we compare ACA results based on two different types of co-citation counting - the traditional type that
only counts the first one among a cited work's authors on the one hand and a non-traditional type that takes into
account the first 5 authors of a cited work on the other hand. Results indicate that the picture produced through this non-traditional author co-citation counting contains more coherent author groups and is therefore considerably clearer. However, this picture represents fewer specialties in the research field being studied than that produced through the traditional first-author co-citation counting when the same number of top-ranked authors is selected and analyzed. Reasons for these effects are discussed
Variations on the Author
“Variations on the Author” discusses two of Eduardo Coutinho’s recent films (Um Dia na Vida, from 2010, and Últimas Conversas, posthumously released in 2015) and their contribution to the general question of documentary authorship. The director’s filmography is characterized by a consistent yet self-effacing form of authorial self-inscription: Coutinho often features as an interviewer that rather than express opinions propels discourses; an interviewer that is good at listening. This mode of self-inscription characterizes him as an author who is not expressive but who is nonetheless markedly present on the screen. In Um Dia na Vida, however, Coutinho is completely absent form the image, while Últimas Conversas, on the contrary, includes a confessional prologue that moves the director from the margins to the center of his films. This article examines the ways in which these works stand out in the filmography of a director who offers new insights into the notion of cinematic authorship
Appropriate Similarity Measures for Author Cocitation Analysis
We provide a number of new insights into the methodological discussion about author cocitation analysis. We first argue that the use of the Pearson correlation for measuring the similarity between authors’ cocitation profiles is not very satisfactory. We then discuss what kind of similarity measures may be used as an alternative to the Pearson correlation. We consider three similarity measures in particular. One is the well-known cosine. The other two similarity measures have not been used before in the bibliometric literature. Dispelling the Myths Behind First-author Citation Counts
We conducted a full-scale evaluative citation analysis study of scholars in the XML research field to explore just how different from each other author rankings resulting from different citation counting methods actually are, and to demonstrate the capability of emerging data and tools on the Web in supporting more realistic citation counting methods. Our results contest some common arguments for the continued
use of first-author citation counts in the evaluation of scholars, such as high correlations between author rankings by first-author citation counts and other citation
counting methods, and high costs of using more realistic citation counting methods that are not well-supported by the ISI databases. It is argued that increasingly available digital full text research papers make it possible for citation analysis studies to go beyond what the ISI databases have directly supported and to employ more
