Multifunctional hydrogels in stereolithography for tissue reconstruction
Hydrogels multifonctionnels en stéréolithographie pour la reconstruction tissulaireL’ingénierie tissulaire a connu un nouvel essor avec la fabrication additive en permettant la construction de structures sophistiquées et un contrôle de la porosité de matériaux inatteignables avec les procédés conventionnels. L’élaboration de matériaux hydrogels biofonctionnels par ces procédés de fabrication additive permet d’améliorer considérablement le comportement cellulaire. Cependant, le développement d‘hydrogels en fabrication additive reste un grand défi. L’objectif de ces travaux de thèse a ainsi été de proposer de nouveaux hydrogels en stéréolithographie pour la conception de dispositifs médicaux. Afin d’élargir la gamme des hydrogels, plusieurs polymères ont été développés et adaptés au procédé de stéréolithographie avec (i) des hydrogels hybrides à base de gélatine et de poly(triméthylène carbonate), (ii) des hydrogels de synthèse de type polyoxazoline et (iii) des hydrogels thermostimulables pour l’impression 4D. La maitrise de la polymérisation par ouverture de cycle des carbonates et des oxazolines et la fonctionnalisation de polymères naturels et de synthèse a permis de moduler les propriétés mécaniques des hydrogels allant du kPa (tissus mous) au MPa (cartilage). Plusieurs architectures complexes ont été conçues en stéréolithographie sur la base de ces différents (co)polymères. Ces objets 3D ont montré d’excellentes viabilités cellulaires dans le temps. L’ensemble de l’étude a montré des résultats très prometteurs pour l’utilisation de ces polymères en ingénierie tissulaire, avec une perspective particulièrement prometteuse pour une reconstruction de tissu complexe et anisotropique tel que le disque intervertébral.Stereolithography multifunctional hydrogels for tissue reconstructionAdditive manufacturing has given new horizons to tissue engineering, allowing the construction of sophisticated structures and controlled material porosity that was unattainable via conventional processes. The development of biofunctional hydrogels using these additive manufacturing processes has allowed for significant improvements in cellular behaviour. However, the elaboration of hydrogels by additive manufacturing remains truly challenging. Thus, the objective of this thesis was to propose new hydrogels by stereolithography for the design of medical devices. As to expand the range of hydrogels, several polymers were developed and adapted to stereolithography such as (i) hybrid hydrogels based on gelatin and poly(trimethylene carbonate), (ii) synthetic hydrogels of polyoxazoline and (iii) thermoresponsive hydrogels for 4D printing. The controlled ring-opening polymerization of carbonates and oxazolines combined with the functionalization of both natural and synthetic polymers, allowed for the modulation of these hydrogels’ mechanical properties from kPa (soft tissues) to MPa (cartilage) scales. Based on these different (co)polymers, several complex architectures were constructed by stereolithography. These 3D objects demonstrated excellent cellular viability. This study shows highly promising results for the use of such polymers in tissue engineering, with a particularly promising perspective for the reconstruction of complex and anisotropic tissue such as the intervertebral disc. These results are already being used in industrial projects in the biomedical field and pave the way for new lines of research
Going Beyond Counting First Authors in Author Co-citation Analysis
The present study examines one of the fundamental aspects of author co-citation analysis (ACA) - the way co-citation
counts are defined. Co-citation counting provides the data on which all subsequent statistical analyses and mappings
are based, and we compare ACA results based on two different types of co-citation counting - the traditional type that
only counts the first one among a cited work's authors on the one hand and a non-traditional type that takes into
account the first 5 authors of a cited work on the other hand. Results indicate that the picture produced through this non-traditional author co-citation counting contains more coherent author groups and is therefore considerably clearer. However, this picture represents fewer specialties in the research field being studied than that produced through the traditional first-author co-citation counting when the same number of top-ranked authors is selected and analyzed. Variations on the Author
“Variations on the Author” discusses two of Eduardo Coutinho’s recent films (Um Dia na Vida, from 2010, and Últimas Conversas, posthumously released in 2015) and their contribution to the general question of documentary authorship. The director’s filmography is characterized by a consistent yet self-effacing form of authorial self-inscription: Coutinho often features as an interviewer that rather than express opinions propels discourses; an interviewer that is good at listening. This mode of self-inscription characterizes him as an author who is not expressive but who is nonetheless markedly present on the screen. In Um Dia na Vida, however, Coutinho is completely absent form the image, while Últimas Conversas, on the contrary, includes a confessional prologue that moves the director from the margins to the center of his films. This article examines the ways in which these works stand out in the filmography of a director who offers new insights into the notion of cinematic authorship
Design of innovative nanoformulations based on polyoxazoline for topical delivery of antioxidants
La peau, principale barrière protectrice avec l’extérieur, est de plus en plus agressée par des sources environnementales (UV, pollution, tabac…). Ces agressions génèrent une surproduction de radicaux libres conduisant à terme au vieillissement prématuré de la peau et parfois à l’apparition de cancers cutanés. Afin de neutraliser ces radicaux présents en excès dans l’épiderme, nous avons élaboré des nanoformulations à base de polyoxazolines pour la délivrance topique d’antioxydants tels que la quercétine. Ces polyoxazolines amphiphiles sont une alternative intéressante au poly(éthylène glycol) dont l’utilisation accrue induit des problèmes de santé chez l’homme. Plusieurs architectures macromoléculaires de polyoxazolines ont été synthétisées et deux nanoformulations innovantes : micelles mixtes et nanocapsules lipidiques ont été mises au point. Ces formulations présentent une grande stabilité, un bon taux de chargement et maintiennent l’activité antioxydante de la quercétine encapsulée. La pénétration cutanée des polyoxazolines seuls et formulés a été étudiée sur plusieurs modèles de peau (GUV, modèle non biologique, oreille de souris, explant de peau humaine). Il a été montré que les nanoformulations sont capables de modifier les propriétés physico-chimiques de surface du modèle de peau non biologique, favorisant leur étalement et que les polymères peuvent pénétrer dans les couches lipidiques de modèles de membranes simples à plus complexes et in vivo sur oreille de souris. En conclusion, les polyoxazolines peuvent substituer le poly(éthylène glycol) dans les formulations et permettre la délivrance topique d’antioxydants dans l’épiderme de souris, ouvrant ainsi la voie vers des applications cutanées pour traiter des pathologies comme le psoriasis ou les mélanomes.The human skin, considered as the main barrier from the outside, is more and more assaulted by environmental sources (UV, pollutants, tobacco…). These aggressions over generate free radicals within the skin then leading to skin aging and potentially skin cancer. Therefore, nanoformulations based on polyoxazolines for topical delivery of antioxidants were investigated in order to quench the excess of free radicals located in the epidermis. As a side objective, this research project intends to demonstrate the potential of amphiphilic polyoxazolines in formulation as an alternative to poly(ethylene glycol), which overuse has led to clinical awareness. Various macromolecular architectures of polyoxazolines were synthesized and two main nanoformulations were elaborated: mixed micelles and lipid nanocapsules. These last showed a high stability overtime, a good drug loading and maintained the antioxidant activity of the encapsulated quercetin. The skin penetration study of polyoxazolines and polyoxazolines based formulations was conducted on various skin models (GUV, nonbiological skin model, mice ears, human skin explants). Nanoformulations were able to modify physicochemical skin surface properties to enhance their spreading and polyoxazolines penetrated from simple to complex lipids membranes and in vivo in mice skin. As a conclusion, polyoxazolines were proved to be an excellent alternative to poly(ethylene glycol) overuse for designing formulations and show promise for delivering topically antioxidants into epidermis creating new possibilities for skin treatment such as psoriasis or melanomas
Polymeric complexing materials based on phosphorus and poly(2-oxazoline) for the sorption of metal ions
Ce travail de thèse a pour objectif le développement de matériaux polymères complexants pour la sorption et/ou la récupération d’ions de métaux lourds présents dans les effluents industriels. Trois types de matériaux à base de phosphore et de poly(2-oxazoline) ont été synthétisés et étudiés avec en premier lieu des polymères hydrosolubles complexants à base de poly(2-éthyl-2-oxazoline) fonctionnalisés par des groupements complexants phosphorés. La capacité de sorption de ces polymères vis-à-vis d’ions métalliques tels que le nickel et l’aluminium a été mise en évidence.Dans un second temps, des nanogels polymères complexants et photo-stimulables ont été préparés à partir d’un terpolymère hydrosoluble auto-assemblable à base de poly(2-méthyl-2-oxazoline), de groupements acide phosphonique et de coumarine photo-dimérisable. L’auto-assemblage dans l’eau de ces polymères a conduit à l’obtention de nanoparticules hydrosolubles capables après irradiation UV de former des nanogels à cœur photoréticulé. L’effet de la photo-stimulation et du pH a été évalué sur la taille des nanogels, leur stabilité dans le temps et leur capacité de sorption vis-à-vis du néodyme et du nickel ont été évaluées, mettant ainsi en exergue les avantages des nanogels par rapport aux nanoparticules.Enfin, une méthode de préparation simple, rapide et originale d’hydrogels complexants a été mise au point par photo-polymérisation d’un monomère acrylique porteur d’un groupement phosphoné (APC1) en solution aqueuse en présence de poly(2-éthyl-2-oxazoline)-co-poly(éthylèneimine) (POx-PEI). L’originalité de cette méthodologie est de permettre l’élaboration d’hydrogels homogènes à partir de polymères immiscibles. La tenue mécanique de ces matériaux résulte de l’enchevêtrement physique des chaînes polymères P(APC1) et POx-PEI ainsi que les nombreuses interactions coopératives, en particulier les liaisons hydrogène, entre les groupements ester, amine, amide, phosphores présents sur les polymères. Les études de gonflement, mécanique et de sorption des hydrogels dans des solutions contenant des ions métalliques de zinc, cuivre, calcium et nickel ont mis en évidence à la fois la capacité de complexation des hydrogels et le renfort des propriétés mécaniques des hydrogels après sorption des ions métalliques.This PhD work investigates complexing polymeric materials for the sorption and/or the recovery of heavy metal ions coming from industrial effluents. Three types of materials based on phosphorus and poly(2-oxazoline) were studied with firstly, water-soluble complexing polymers based on poly(2-ethyl-2-oxazoline) functionalized by phosphorus-based complexing groups. The sorption capacity of these polymers against metal ions such as nickel and aluminum was demonstrated.Secondly, photo-active and complexing polymeric nanogels were prepared from a water-soluble terpolymer based on poly(2-methyl-2-oxazoline), phosphonic acid moieties and photo-dimerizable coumarin groups. The self-assembly of these polymers in water allows the obtaining of water-soluble nanoparticles able to produce nanogels with a photo-crosslinked core under UV irradiation. The influence of UV irradiation and pH on the size of the nanogels was evaluated, their stability over time and their sorption capacity against neodymium and nickel were measured, demonstrating the advantages of nanogels towards nanoparticles.Finally, a simple, fast and original method of elaboration of complexing hydrogels was successfully reached by photopolymerization of an acrylic monomer bearing a phosphonated group (APC1) in aqueous solution in the presence of poly(2-ethyl-2-oxazoline)-co-poly(ethyleneimine) (POx-PEI). The originality of this methodology is the elaboration of homogeneous hydrogels from incompatible polymers. The mechanical strength of these materials results from physical entanglement of P(APC1) and POx-PEI chains and from the numerous cooperative interactions, in particular hydrogen bonds between ester, amine, amide, phosphorus groups of the polymers. The study of swelling, mechanics and sorption of hydrogels in solutions containing metal ions such as zinc, copper, calcium, and nickel showed the complexation ability of the hydrogels and the enhancement of the mechanical properties of the material after sorption of metallic ions
Appropriate Similarity Measures for Author Cocitation Analysis
We provide a number of new insights into the methodological discussion about author cocitation analysis. We first argue that the use of the Pearson correlation for measuring the similarity between authors’ cocitation profiles is not very satisfactory. We then discuss what kind of similarity measures may be used as an alternative to the Pearson correlation. We consider three similarity measures in particular. One is the well-known cosine. The other two similarity measures have not been used before in the bibliometric literature. High energy density batteries using a gelled electrolyte, providing a long life and enhanced security
Actuellement, les batteries Li-ion sont massivement utilisées dans les dispositifs mobiles de notre quotidien, en raison de leur performance élevée et leur flexibilité accrue. Du fait d’une croissance intensive de ces systèmes de stockage, la recherche scientifique s’est intéressée à l’amélioration de leur densité d’énergie, durée de vie et au renforcement de leur sécurité. L’objectif double de cette thèse, en collaboration avec l’ADEME et Total, est d’élaborer de nouveaux matériaux d’électrodes négatives de forte capacité et de développer des électrolytes polymères innovants pour le fonctionnement de cette technologie dans des conditions plus sécurisées.Les matériaux d’électrodes négatives de composition MSnSb, avec M un métal de transition, ont particulièrement retenu notre attention en raison des performances électrochimiques remarquables et inattendue de TiSnSb en température. Dans l’optique de comprendre l’influence du métal de transition, l’étude a porté sur la compréhension des comportements électrochimiques des matériaux d’électrodes NbSnSb et ZnSnSb2. De ce fait, les mécanismes réactionnels de lithiation / délithiation des matériaux ont été étudiés en profondeur, grâce à des mesures operando (DRX, Spectroscopie Mössbauer et XAS). L’évolution de l’interface électrode/électrolyte, pour différents électrolytes et à différentes températures de cyclage, a été réalisée par différentes analyses de surface (XPS, Auger…). Ainsi la nature et l’évolution de la couche de passivation (SEI), ont pu être déterminées. Cette dernière étant à l’origine du vieillissement prématuré des batteries Li-ion, nous nous sommes orientés vers l’élaboration d’électrolyte polymère, qui est un défi majeur pour améliorer la cyclabilité et la sécurité de ces dispositifs. Un intérêt singulier, s’est porté sur l’intégration des polycarbonates cycliques, et plus spécifiquement sur le glycérol carbonate méthacrylate, comme brique élémentaire des matériaux polymères. Des électrolytes polymères gélifiés, réticulés et tout solides, pour batteries Li-ion et Na-ion, ont été réalisés afin d’en évaluer leur performance électrochimiqueCurrently, Li-ion batteries are widely used to power mobile devices, owing to their high performance and huge flexibility. Societal issues lead the scientific research to improve the energy density, lifetime and enhance the safety of this storage system. In this way, twice thesis objectives are develop: high capacity negative electrode materials for the high energy density aspect, and innovative polymer electrolytes for more secure conditions, in collaboration with ADEME and Total.MSnSb negative electrode materials, with M a transition metal, have particularly caught our attention because of the remarkable electrochemical performance and unexpected TiSnSb cycling in temperature. In order to understand the transition metal influence, the study focused on the determination of the electrochemical behavior of NbSnSb and ZnSnSb2 electrode materials. The materials lithiation / delithiation reaction mechanisms have been depth highlighted by operando measurements (DRX, Spectroscopie Mössbauer and XAS). The electrode / electrolyte interface evolution, for different electrolytes and temperatures cycling, was investigated by several surface analyzes (XPS, Auger ...) to determine the solid electrolyte interphase (SEI) composition. Because the Li-ion batteries aging is caused by the formation of this passivation layer, we started to develop polymer electrolyte, which is a major challenge to improve the cyclability and safety of these devices. Cyclic polycarbonates are notably, and more specifically polymeric materials based on glycerol carbonate methacrylate. Dispelling the Myths Behind First-author Citation Counts
We conducted a full-scale evaluative citation analysis study of scholars in the XML research field to explore just how different from each other author rankings resulting from different citation counting methods actually are, and to demonstrate the capability of emerging data and tools on the Web in supporting more realistic citation counting methods. Our results contest some common arguments for the continued
use of first-author citation counts in the evaluation of scholars, such as high correlations between author rankings by first-author citation counts and other citation
counting methods, and high costs of using more realistic citation counting methods that are not well-supported by the ISI databases. It is argued that increasingly available digital full text research papers make it possible for citation analysis studies to go beyond what the ISI databases have directly supported and to employ more
