Protocol for Investigating the Interactions Between Intrinsically Disordered Proteins and Membranes by Neutron Reflectometry
Several intrinsically disordered proteins (IDPs) exhibit high affinity for lipid membranes. Among the different biophysical methods to probe protein-lipid interaction, neutron reflectometry (NR) can provide direct and structural detailed information on the location of the IDP with respect to the membrane. Supported lipid bilayers are commonly used as cell membrane models in such experiments. NR measurements can be collected on the supported lipid bilayer before and after the interaction with the IDP to characterize whether the protein molecules are mainly located on the membrane surface (interaction with the lipid headgroups), are penetrating into the hydrophobic region of the membrane (interaction with the lipid acyl chains), or are not interacting at all with the membrane. The lipid composition of the supported lipid bilayer can easily be tuned; hence the NR experiments can be designed to investigate selective IDP-lipid interactions.This chapter will describe the fundamental steps for performing an NR experiment and the subsequent data analysis aimed at characterizing IDP-lipid bilayer interactions. The specific case of an intrinsically disordered region (IDR) from the membrane protein Na+/H+ exchanger isoform 1 (NHE1) will be used as an example, but the same protocol can be easily adapted to other IDPs
Thermodynamics properties of microemulsions involved in ion separation methods: a mesoscopic modelling approach
International audienceRecycling of metals, such as rare earths, lanthanides or actinides, into valuable material relies on ion specific separation, basis of the hydrometallurgy. Most of efficient methods known for separating ions are based on equilibria between complex fluids, i.e. water in oil (w/o) microemulsions. In the diluted regime, w/o reverse micelles, that selectively up-take some ions, are generally considered as spheres containing extractant molecules, water molecules, and complexed ions. However, in industrially relevant cases of liquid – liquid extraction, it is known that extractant aggregates cannot be described as simple metal-ion complexes spherical on average, since the high conductivity observed in the oil phase proves a bicontinuity degree of the system.Examples of nonionic w/o microemulsions have been simulated using the wavelet method developed by Arleth and Marčelja [1]. The thermodynamics of the interface is based on the Helfrich formalism [2]. In our model, the Helfrich free energy is minimized as a function of the bending and Gaussian elastic constants, which refer to the rigidity of the surfactant film in term of energies, and the spontaneous curvature of the surfactant film, which corresponds to its preferred curvature [3].Influence of the bending and Gaussian elastic constants is observed on frustrated and unfrustrated microemulsion structures, their scattering properties, and predicted ternary phase diagrams.However, it is well known that efficient surfactants used for such processes are mainly ionic. Therefore, it is crucial to develop a more realistic model that takes into account (i) the ionic character of the surfactant, and (ii) the presence of cations in solutions. Factores que intervienen en ausentismo y rotación del personal que labora en Call Centers de Barranquilla
The general objective of this research is to analyze the factors involved in the absenteeism and rotation of the staff working in Call Centers of Barranquilla. To achieve the development of this research, the use of a quantitative methodology of correlational type was carried out, in which through the selection of a sample of 65 subjects working in a call center in the city of Barranquilla and the application from a questionnaire on the various dimensions of rotation and absenteeism, we sought to resolve the general objective of this study. The results obtained during the investigation allowed us to observe the relationship between factors such as the position within the company, the age of the worker and working time with the different dimensions of absenteeism and rotation within the call centers of the city of Barranquilla. As one of the most representative elements of the final conclusions of this work, the importance of generating a high level of engagement and an organizational culture rooted in order to reduce the rates of rotation and absenteeism within this organization that suffer from this problem of worrisome way.El objetivo general de la presente investigación es el de analizar los factores que intervienen en el ausentismo y rotación del personal que labora en Call Centers de Barranquilla. Para lograr el desarrollo de esta investigación, se llevó a cabo el uso de una metodología cuantitativa de tipo correlacional, en la cual a través de la selección de una muestra de 65 sujetos que laboran en un call center de la ciudad de Barranquilla y la aplicación de un cuestionario sobre las diversas dimensiones de la rotación y el ausentismo se buscó dar resolución al objetivo general de este estudio. Los resultados arrojados durante la investigación permitieron observar la relación existente entre factores como el cargo dentro de la empresa, edad del trabajador y tiempo laborando con las diferentes dimensiones del ausentismo y la rotación dentro de los call center de la ciudad de Barranquilla. Como unos de los elementos más representativo de las conclusiones finales de este trabajo, se destaca la importancia en generar un alto nivel de engagement y una cultura organizacional arraigaida en aras de reducir los índices de rotación y ausentismo dentro de esta organización que sufren esta problemática de manera preocupante.Paredes Niz, Arleth Patrici
Analysis of mechanisms, molecules and secondary metabolites related to the probiotic potential of leuconostoc mesenteroides, supporting its implementation and evaluation in new studies
Leuconostoc mesenteroides es una bacteria ácido-láctica de gran interés en las últimas décadas por sus diversas características tecnológicas y su potencial probiótico. Este estudio se propuso analizar los mecanismos, moléculas y metabolitos secundarios responsables de sus efectos beneficiosos. La revisión sistemática de la literatura científica reveló una creciente tendencia en el estudio de L. mesenteroides desde el año 2016, principalmente en áreas como agricultura, inmunología, microbiología y biomedicina. Se identificaron diversas actividades probióticas de L. mesenteroides, siendo la inhibición de microorganismos patógenos la más prevalente, atribuida a la producción de sustancias antibacterianas, la exclusión competitiva por sitios de adhesión y la capacidad de co-agregación. L. mesenteroides también demostró una notable actividad inmunomoduladora, evidenciada por la inducción de citoquinas, mejora de la función de barrera intestinal y modulación de la expresión de genes relacionados con la respuesta inmune en los diferentes modelos probados, ya fueran líneas celulares o seres vivos. Además de la inhibición de patógenos y la inmunomodulación, L. mesenteroides presentó otras actividades beneficiosas, aunque menos estudiadas, como la reducción del colesterol, la actividad antioxidante, detoxificante, antidiabética, antihiperucémica, antiviral, neuroprotectora y protectora del ADN. A pesar de la evidencia acumulada, persisten vacíos en el entendimiento de los mecanismos moleculares subyacentes a estas actividades.
En cuanto a los genes y secuencias relacionados con los mecanismos y moléculas producidas por L. mesenteroides, se encontró información limitada en los estudios analizados. Sin embargo, se logró proporcionar las secuencias de las cepas y algunas moléculas producidas por L. mesenteroides almacenadas en el Gen Bank de NCBI, junto con algunas características de interés. En relación con las características estructurales y bioquímicas de las moléculas producidas por L. mesenteroides, se pudieron identificar diversidad de características estructurales, sin embargo, los estudios analizados se centraron en la actividad de las moléculas, más que en su estructura. A pesar de la información disponible, aún existen vacíos en la comprensión de los mecanismos moleculares específicos y las sustancias responsables de las actividades de L. mesenteroides. Se requieren más estudios para dilucidar completamente las interacciones entre L. mesenteroides y sus huéspedes, así como para identificar y caracterizar completamente las moléculas bioactivas involucradas en sus efectos beneficiosos junto con los genes y secuencias relacionados a su expresión. Esta revisión bibliográfica proporciona una base sólida para futuras investigaciones que busquen explorar el potencial de L. mesenteroides como probiótico y desarrollar nuevas aplicaciones biotecnológicas en diversas industrias.Universidad Libre - Facultad Ciencias de la Salud, Exactas y Naturales - MicrobiologíaLeuconostoc mesenteroides is a lactic acid bacterium of great interest in recent decades due to its various technological characteristics and its probiotic potential. This study set out to analyze the mechanisms, molecules, and secondary metabolites responsible for their beneficial effects. The systematic review of the scientific literature revealed a growing trend in the study of L. mesenteroides since 2016, mainly in areas such as agriculture, immunology, microbiology and biomedicine. Various probiotic activities of L. mesenteroides were identified, with inhibition of pathogenic microorganisms being the most prevalent, attributed to the production of antibacterial substances, competitive exclusion by adhesion sites and co-aggregation capacity. L. mesenteroides also demonstrated remarkable immunomodulatory activity, evidenced by cytokine induction, improved intestinal barrier function, and modulation of the expression of genes related to the immune response in the different models tested, whether cell lines or living beings. In addition to pathogen inhibition and immunomodulation, L. mesenteroides presented other beneficial activities, although less studied, such as cholesterol reduction, antioxidant, detoxifying, antidiabetic, antihyperukemia, antiviral, neuroprotective, and DNA-protective activity. Despite the accumulated evidence, gaps persist in understanding the molecular mechanisms underlying these activities.
Regarding the genes and sequences related to the mechanisms and molecules produced by L. mesenteroides, limited information was found in the studies analyzed. However, it was possible to provide the sequences of the strains and some molecules produced by L. mesenteroides stored in the NCBI Gene Bank, along with some characteristics of interest. In relation to the structural and biochemical characteristics of the molecules produced by L. mesenteroides, a diversity of structural characteristics could be identified, however, the studies analyzed focused on the activity of the molecules, rather than on their structure. Despite the available information, there are still gaps in the understanding of the specific molecular mechanisms and substances responsible for the activities of L. mesenteroides. Further studies are required to fully elucidate the interactions between L. mesenteroides and its hosts, as well as to fully identify and characterize the bioactive molecules involved in its beneficial effects along with the genes and sequences related to their expression. This literature review provides a solid foundation for future research seeking to explore the potential of L. mesenteroides as a probiotic and develop new biotechnological applications in various industries
Structure Parameters of Synaptic Vesicles Quantified by Small-Angle X-Ray Scattering
Synaptic vesicles (SVs) are small, membrane-bound organelles that are found in the synaptic terminal of neurons, and which are crucial in neurotransmission. After a rise in internal [Ca(2+)] during neuronal stimulation, SVs fuse with the plasma membrane releasing their neurotransmitter content, which then signals neighboring neurons. SVs are subsequently recycled and refilled with neurotransmitter for further rounds of release. Recently, tremendous progress has been made in elucidating the molecular composition of SVs, as well as putative protein-protein interactions. However, what is lacking is an empirical description of SV structure at the supramolecular level-which is necessary to enable us to fully understand the processes of membrane fusion, retrieval, and recycling. Using small-angle x-ray scattering, we have directly investigated the size and structure of purified SVs. From this information, we deduced detailed size and density parameters for the protein layers responsible for SV function, as well as information about the lipid bilayer. To achieve a convincing model fit, a laterally anisotropic structure for the protein shell is needed, as a rotationally symmetric density profile does not explain the data. Not only does our model confirm many of the preexisting ideas concerning SV structure, but also for the first time, to our knowledge, it indicates structural refinements, such as the presence of protein microdomains

Este trabajo se hace con el propósito de crear un manual de procedimientos contables que facilitara la manera de cómo se registran las transacciones y la responsabilidad que conlleva el proceso de compra y despacho del Combustible.
Todo esto mediante la observación de los procedimientos y la forma en cómo se registran dichas transacciones actualmente en la empresa y lo que indica la norma para la organización de un proceso contable.
Este trabajo contribuirá a aportar las herramientas necesarias para que los colaboradores de la empresa puedan realizar sus funciones de manera eficiente y eficaz, ganando tiempo y a su vez generando confianza en la información que se emita del sistema contable.This work is done with the purpose of creating a manual of accounting procedures that will facilitate the way in which transactions are recorded and the responsibility involved in the process of purchase and dispatch of fuel.
All this by means of the observation in first person of the procedures and the form in how these transactions are registered at the present time in the company, it presents some faults and emptiness’s that cause that the information verified in the systems presents errors and the balances of the accounts are not the correct ones.

Enzymes associated with the probiotic activity of microorganisms in cattle
La Agenda 2030 de las Naciones Unidas y sus Objetivos de Desarrollo Sostenible
(ODS), han priorizado la seguridad alimentaria como un objetivo fundamental para
erradicar el hambre, mejorar la nutrición y promover la agricultura sostenible en todo el
mundo (ODS2: Hambre cero). El uso excesivo del agua y la tierra, sumado a prácticas
agrícolas no sostenibles, han ocasionado daños en los ecosistemas que impiden
mantener una producción limpia y sostenible de alimentos. Sin embargo, la demanda de
alimentos en general y el consumo de carnes en particular es una línea que se mantiene
constante, exigiendo prácticas como el uso de antibióticos y otros aditivos alimentarios,
para promover el crecimiento en animales de sacrificio. Estas prácticas, aunque son
permitidas, pueden generar efectos adversos asociados al uso inadecuado de antibiótico,
razón por la cual cada vez son más las entidades reguladoras que han restringido el uso
de antibióticos en animales para consumo, fomentando alternativas viables como la
utilización de probióticos. Los probióticos son microorganismos que actúan mediante
varios mecanismos: inhiben la adhesión de patógenos, producen sustancias
antimicrobianas, excluyen competitivamente a microorganismos nocivos, y mejoran la
función de barrera y el sistema inmunológico. Bacterias como Lactobacillus rhamnosus
y Lactobacillus plantarum han mostrado efectividad en la prevención de la adhesión de
Escherichia coli a la superficie intestinal. En el caso de hongos, algunas levaduras como
Saccharomyces boulardii produce enzimas y metabolitos beneficiosos, que ayudan a
regular la flora intestinal e inhibir el crecimiento de patógenos. En este contexto, se
presenta a continuación una revisión que describe las propiedades bioquímicas y
estructurales de algunas enzimas implicadas en la actividad probiótica de ciertos
microorganismos sobre el ganado bovino. Para ello, se realizó una búsqueda de
información utilizando bases de datos tales como PubMed, Scopus y Google
Académico, siguiendo la metodología PRISMA. Se identificaron moléculas de interés
especificando detalles de los procesos bioquímicos en los cuales intervienen.
Adicionalmente se analizaron secuencias determinando motivos y dominios
conservados que estuvieran relacionados con las propiedades de los microorganismos.
Los resultados obtenidos con esta revisión constituyen una contribución al estado de
arte de enzimas asociadas a la actividad probiótica de microorganismos en el ganado

prioritized food security as a fundamental objective to eradicate hunger, improve
nutrition, and promote sustainable agriculture worldwide (SDG 2: Zero Hunger).
Excessive use of water and land, combined with unsustainable agricultural practices,
has caused damage to ecosystems, hindering the ability to sustain clean and sustainable
food production. However, the demand for food in general, and meat consumption in
particular, remains constant, necessitating practices such as the use of antibiotics and
other feed additives to promote growth in livestock. While these practices are permitted,
they can lead to adverse effects associated with the misuse of antibiotics. As a result,
regulatory agencies are increasingly restricting antibiotic use in animals intended for
consumption, encouraging viable alternatives such as probiotics. Probiotics are
microorganisms that act through various mechanisms: they inhibit pathogen adhesion,
produce antimicrobial substances, competitively exclude harmful microorganisms, and
enhance barrier function and the immune system. Bacteria such as Lactobacillus
rhamnosus and Lactobacillus plantarum have demonstrated effectiveness in preventing
the adhesion of Escherichia coli to intestinal surfaces. Regarding fungi, certain yeasts
like Saccharomyces boulardii produce enzymes and beneficial metabolites that help
regulate gut flora and inhibit pathogen growth.
In this context, this review presents an analysis of the biochemical and structural
properties of specific enzymes involved in the probiotic activity of certain
microorganisms in cattle. The study involved a systematic information search using
databases such as PubMed, Scopus, and Google Scholar, following the PRISMA
methodology. Molecules of interest were identified, detailing the biochemical processes
in which they participate. Additionally, sequence analyses were conducted to determine
conserved motifs and domains associated with the microorganisms' properties. The
findings from this review contribute to the state of the art regarding enzymes associated
