Alkalispirillum mobile gen. nov., spec. nov., an alkaliphilic non-phototrophic member of the Ectothiorhodospiraceae
No full textFrom cultures of the anoxygenic phototroph Halorhodospira halophila SL-1, an aerobic, gram-negative spirillum was isolated. This moderately halophilic, alkaliphilic bacterium was motile by means of a single polar flagellum. It is described here as Alkalispirillum mobile gen. nov., spec. nov. Phylogenetic analysis of the Alkalispirillum mobile 16S rRNA gene led to its classification in the %-subclass of the Proteobacteria, as it appears closely related to phototrophic purple sulfur bacteria of the genera Ectothiorhodospira and Halorhodospira. Surprisingly, A. mobile is an obligate aerobe. The organism grows optimally with a number of carboxylic acids (such as sodium acetate) as carbon source, at 2% (i.e. approximately 0.34 M) sodium chloride, at pH 9-10, and at temperatures ranging from 35 to 38 °C. The dominant cellular fatty acids of Alkalispirillum mobile are C12:0, C16:0, C18:1cis11, and C18:0; its G+C content is 66.2-0.5 mol%

De microbemens: Het belang van het onzichtbare leven
No full textDe mens is een wandelend ecosysteem. Ons leven lang verschaffen we de juiste omstandigheden aan een gigantische gemeenschap van micro-organismen in ons lichaam. Toch zien we microben voornamelijk als veroorzakers van ziekte en bederf en proberen we ze juist uit alle macht te verdelgen met antibiotica en ontsmettingsmiddelen. Maar we kunnen er niet omheen. Vanaf de kolonisatie van ons lichaam bij onze geboorte tot de ontbinding na onze dood: bacteriën spelen een onmisbare rol in ons leven. Deze cruciale rol wordt in De microbemens op onnavolgbare wijze onder de loep genomen. Microben zijn net mensen: ze zoeken naar eten, hebben seks en wisselen van lifestyle. Wij treden op als hun gastheer. We lezen waar ze in ons lichaam zitten, hoe we ze voeden en wat zij ons bieden. We blijken niet meer dan een slaaf te zijn van onze microben. We moeten onze eetlust de baas zien te worden en ons bevrijden van onze onzichtbare ketenen. Als een ware microbenmeester moeten we in deze moderne tijd niet te eenzijdig en ongericht inzetten op het verdelgen van de microben – met opkomende immuunziekten en antibioticum resistentie als gevolg. Microbenmissionaris Remco Kort brengt gezonde bacteriën naar het Afrikaanse continent, waar gezonde drinkyoghurt wordt gemaakt. We reizen met hem mee. Eindelijk, na meer dan drie eeuwen duisternis, krijgen microben de erkenning die ze verdienen. Vanaf nu staan ze vol in de schijnwerpers. We zullen ze nooit meer over het hoofd te zien

Design Considerations for Building a Scalable Digital Version of a Multi-player Educational Board Game for a MOOC in Logistics and Transportation
No full textWith more flexible and large-scale learning environments, new design requirements for games emerge. Massive Open Online Courses (MOOCs) are one of the most important innovations in the learning field. Still, it is a challenge to motivate learners and to keep them motivated in such huge learning environments. To address this challenge, we redesigned a board game targeting at an integrated view on disruption and communication management in an intermodal transportation situation. From the redesign, we have learned that an online game works better with fewer roles, requires immediate feedback, and an engaging way of challenge to keep players motivated. Using space syntax to assess accessibility of multimodal urban hubs and seamless mobility within the hubs Case study Delft-Campus train station
No full textMultimodal hubs connect different modes, allowing people to switch from one mode to another and seamlessly access/egress to/from their origin/destinations. However, there are limited methods to estimate the impact of future measures on the accessibility of multimodal hubs. These future measures could be wide reaching, among others, introduction of different schemes in the planning of the hub or introduction of new modes at multimodal hubs. This study provides interdisciplinary cross-cutting method including transport and urbanism literatures, by using Space Syntax method. This method allows the transport planners to analyse and compare the current situation and the proposed development plan. In this research, we show how changes made in urban spatial configuration can impact the accessibility of intermodal hubs. To learn about the impacts of the spatial configuration of a city on station design, we performed a case study at Delft Campus train station in the Netherlands, which is under construction to become a multimodal hub. Firstly, we analyse the current situation and then the proposed development plan in terms of, integration, choice, accessibility and intelligibility, using segment analyses. Furthermore, the immediate surrounding areas of the station have been analysed by Visual Graphic Analysis to find visual accessibility within the hub. Although the municipality proposed plans claim that the station’s accessibility would be enhanced, our method indicate otherwise and the proposed changes are not highly effective in increasing accessibility. Finally, some design recommendations for multimodal hubs are proposed.
