Typed correspondence from Hideo Hashimoto, Pastor of the Japanese Methodist Church, to his congregation and friends. He expresses his feelings towards the current state of the country the day before the mass removal order. The Bishop James Chamberlain Baker Collection includes letters, documents, and articles about Japanese Americans during World War II. Subjects in the collection include Japanese Americans mass removal, Pearl Harbor and the aftermath, religion, and support from the non-Japanese American community. The collection was digitized and made accessible online by CSUDH Gerth Archives and Special Collections

Improved quantification of DNA methylation using methylation-sensitive restriction enzymes and real-time PCR

Heterogeneity of cells with respect to the DNA methylation status at a specific CpG site is a problem when assessing methylation status. We have developed a simple two-step method for the quantification of the percent of cells that display methylation at a specific CpG site in the promoter of a specific gene. The first step is overnight digestion of genomic DNA (optimal conc. 20 ng/5 microl) with a relevant methylation-sensitive restriction enzyme (optimal 2 units). This is followed by real time PCR, using the SYBR Green method, with primers that bracket the site cleaved by the enzyme. By including fully methylated and fully non-methylated DNA in each PCR plate, the errors caused by non-specific digestion or incomplete digestion can be measured and used to adjust the raw results and thus increase specificity. The method can detect differences in methylation status if these are more than 10%. No specialized equipment is required beyond the real-time PCR system and the method can be adapted for any of the 53 commercially available methylation-sensitive restriction enzymes

DNA demethylation at specific CpG sites in the IL1B promoter in response to inflammatory cytokines in human articular chondrocytes

Objective: to determine whether changes in the DNA methylation status in the promoter region of the gene encoding interleukin-1? (IL-1?) account for expression of IL1B mRNA after long-term treatment of human articular chondrocytes with inflammatory cytokines. Methods: IL-1? or TNF? combined with oncostatin M (OSM), or 5-aza-deoxycytidine (5-aza- dC) were added twice weekly for 4-5 weeks to primary cultures of normal human articular chondrocytes, obtained from patients with a femoral neck fracture. Expression of MMP13, IL1B??TNFA and DNMT1 were determined by SybrGreen-based qRT-PCR on genomic DNA and total RNA extracted from the same sample before and after culture. Bisulfite modification was used to identify which CpG sites in the IL1B promoter showed differential methylation between expressing and non-expressing cells. The percentages of cells that were methylated at that critical CpG site (-299bp) were quantified by a method that depended on methylation-sensitive restriction enzymes and real-time PCR. Secretion of IL-1? was assessed by enzyme-linked immunoabsorbent assay (ELISA) of the culture media. Results: healthy chondrocytes did not express IL1B mRNA, but the levels were increased 5-fold by 5-aza-dC and 100- to 1000-fold by or TNF?/OSM. The % CpG methylation was decreased by 5-aza-dC, but reduced considerably more by IL-1? and almost abolished by TNF?/OSM. The mRNA was translated into protein in cytokine-treated chondrocytes. Conclusion: these novel findings indicate that inflammatory cytokines can change DNA methylation status at key CpG sites, resulting in long-term induction of IL1B in human articular chondrocytes.<br/

Letter from Frances K. Hashimoto, 1982 General Chairperson, Nisei Week Japanese Festival, to Jim Matsuoka, regarding Nisei Week. The Jim Matsuoka Nikkei for Civil Rights and Redress Collection includes brochures, meeting notes and agendas, publications, booklets, and other material related to the Nikkei for Civil Rights and Redress (NCRR), formally known as the National Coalition for Redress/Reparations. The National Coalition for Redress/Reparations was officially formed on July 12, 1980, and included members of the Los Angeles Community Coalition for Redress/Reparations (LACCRR), Japanese Community Progressive Alliance (JCPA), Tule Lake Committee, Nihonmachi Outreach Committee, the Asian/Pacific Student Union, and other members of the community. The material was collected by Jim Matsuoka, a founding member of the organization. Matsuoka also served on the board and was the treasurer. In addition to the NCRR material, the collection also contains event flyers and Day of Remembrance material. For issues of the Nikkei for Civil Rights and Redress newsletter "Banner" published after 2007, visit the NCRR website at https://ncrr-la.org/

Letter written from Fresno Assembly Center to friends in the "outside" world describing the harsh effects of incarceration including quotes from the letters of incarcerees in other camps as well as from cited publications. Lists ways and actions to help the incarcerees. Also includes descriptions of recreation, education, and church activities. The Japanese American Relocation Collection is composed of ephemera related to the relocation program during World War II. Items include the official government report of Manzanar Relocation Center, a photo album, post-war activism materials related to preserving and remembering the camps, various clippings, and documents. The strength of this collection is found in its many perspectives on the controversial relocation program and how it has been presented since World War II

Introduction [to The Japanese Labor Market in a Comparative Perspective with the United States]

    No full text
This study offers a comparative analysis of a number of Japanese labor market features in relation to the U.S. The author examines employer-employee attachment, workforce adjustment, and industrial relations including unique Japanese institutions such as joint consultation and consensus-based decision making. Hashimoto argues that cultural-traditional influences, which shape the transaction-cost environment, interacted with technological progress in shaping the various uniquely-Japanese labor market features.https://research.upjohn.org/up_press/1101/thumbnail.jp
