Going Beyond Counting First Authors in Author Co-citation Analysis
The present study examines one of the fundamental aspects of author co-citation analysis (ACA) - the way co-citation
counts are defined. Co-citation counting provides the data on which all subsequent statistical analyses and mappings
are based, and we compare ACA results based on two different types of co-citation counting - the traditional type that
only counts the first one among a cited work's authors on the one hand and a non-traditional type that takes into
account the first 5 authors of a cited work on the other hand. Results indicate that the picture produced through this non-traditional author co-citation counting contains more coherent author groups and is therefore considerably clearer. However, this picture represents fewer specialties in the research field being studied than that produced through the traditional first-author co-citation counting when the same number of top-ranked authors is selected and analyzed. Variations on the Author
“Variations on the Author” discusses two of Eduardo Coutinho’s recent films (Um Dia na Vida, from 2010, and Últimas Conversas, posthumously released in 2015) and their contribution to the general question of documentary authorship. The director’s filmography is characterized by a consistent yet self-effacing form of authorial self-inscription: Coutinho often features as an interviewer that rather than express opinions propels discourses; an interviewer that is good at listening. This mode of self-inscription characterizes him as an author who is not expressive but who is nonetheless markedly present on the screen. In Um Dia na Vida, however, Coutinho is completely absent form the image, while Últimas Conversas, on the contrary, includes a confessional prologue that moves the director from the margins to the center of his films. Appropriate Similarity Measures for Author Cocitation Analysis
We provide a number of new insights into the methodological discussion about author cocitation analysis. We first argue that the use of the Pearson correlation for measuring the similarity between authors’ cocitation profiles is not very satisfactory. We then discuss what kind of similarity measures may be used as an alternative to the Pearson correlation. We consider three similarity measures in particular. One is the well-known cosine. The other two similarity measures have not been used before in the bibliometric literature. Dispelling the Myths Behind First-author Citation Counts
We conducted a full-scale evaluative citation analysis study of scholars in the XML research field to explore just how different from each other author rankings resulting from different citation counting methods actually are, and to demonstrate the capability of emerging data and tools on the Web in supporting more realistic citation counting methods. Our results contest some common arguments for the continued
use of first-author citation counts in the evaluation of scholars, such as high correlations between author rankings by first-author citation counts and other citation
counting methods, and high costs of using more realistic citation counting methods that are not well-supported by the ISI databases. Molecular characterization of human spermatogonia
Spermatogonien sind die frühesten Vorläuferzellen der Spermatiden im adulten männlichen Organismus. Sie umfassen eine adulte Population mit Stammzell-Eigenschaften. Diese bilden durch ständige Selbsterneuerung und Differenzierung den Ausgangspunkt für eine lebenslange Spermatogenese im erwachsenen männlichen Organismus.
Im Rahmen der vorliegenden Doktorarbeit wurden durch die Auswertung von Mikroarray-Daten und deren Validierung auf mRNA- und Proteinebene spermatogonienspezifische Gene identifiziert. Zusätzlich wurden mittels Immunhistochemie weitere Spermatogonien-Marker untersucht. Mit Immun-Mehrfachfärbungen wurden anschließend verschiedene Marker-Kombinationen getestet, um Erkenntnisse über den molekularen Phänotyp verschiedener Subpopulationen und über die bislang noch nicht identifizierten spermatogonialen Stammzellen zu gewinnen. Hierbei zeigte sich eine unerwartete Heterogenität der Spermatogonien in Bezug auf die exprimierten Marker und keine einfache Korrelation des molekularen Phänotyps mit der bereits in den 1960- und 1970er Jahren festgelegten morphologischen Zelltypisierung. Neben einer Vielzahl an koexprimierten bzw. überlappenden Markern wurden einige sich hinsichtlich ihrer Expression ausschließende Marker detektiert.
Als gute Kandidaten für die Subpopulation der adulten Stammzellen innerhalb der gesamten Spermatogonienpopulation wurden Zellen mit dem molekularen Phänotyp UTF1+/FGFR3+/DMRT1-/KIT- ermittelt. Die Ergebnisse dieser Arbeit liefern Argumente für UTF1 und FGFR3 als humane spermatogoniale Stammzellmarker: FGFR3 wurde niemals in proliferierenden Spermatogonien gefunden. Auch kam dieser Marker nicht zusammen mit DMRT1 vor. DMRT1 wurde als weitgehend mit dem Differenzierungsmarker KIT koexprimiert gefunden und könnte in der humanen Spermatogoniogenese funktionell eine ähnliche Rolle wie KIT spielen. UTF1 wurde kaum in differenzierenden (KIT+) Spermatogonien detektiert und war nur in seltenen Fällen als mit DMRT1 koexprimiert zu finden. Dagegen wurde FGFR3 in einer Subpopulation der UTF1+ Spermatogonien beobachtet. Untersuchungen an fetalem Testisgewebe zeigten außerdem, dass FGFR3 im humanen Hoden bereits von fetalen Keimzellen exprimiert wird.
Spermatogoniale Stammzellen sind näher an der Lamina propria gelegen bzw. sitzen dieser direkt auf. Hierbei handelt es sich um eine Subpopulation der spermatogonialen Zelltypen A dark und A pale. Hingegen stellen die B-Spermatogonien eine differenzierte Form der Spermatogonien dar. Diese nehmen innerhalb der Tubuli seminiferi eine von der Basallamina weiter entfernte Position ein. Die in dieser Arbeit identifizierten molekularen Marker FGFR3 und UTF1 wurden größtenteils in Zellen detektiert, die sich unmittelbar an der Lamina propria befinden, also in „früheren“, „undifferenzierteren“ Zellen. Dagegen wurden DMRT1 und KIT niemals in A dark-Spermatogonien detektiert, aber oftmals in Zellen, die weiter von der Basallamina entfernt lokalisiert waren.
Die humanen A dark-Spermatogonien enthalten innerhalb des nukleären Chromatins eine vakuolenähnliche Struktur (Rarefaktionszone), von der man weiß, dass sie sich nicht mit DNA-Farbstoffen, jedoch schwach mit Eosin anfärben lässt. Im Rahmen dieser Doktorarbeit konnte der A dark-„Vakuole“ erstmalig eine Substanz, das Protein EXOSC10, zugeordnet werden. Dieses Protein stellt eine bekannte eukaryotische Exosomkomponente dar. Für die A dark-Spermatogonien könnte dies bedeuten, dass sich das nukleäre RNA-Exosom innerhalb der „vakuolen“-ähnlichen Struktur dieses Zelltyps befindet. Andererseits könnte die A dark-„Vakuole“ auch ein Speicherort für das Protein EXOSC10 sein. Gleichzeitig deutete sich ein funktioneller Zusammenhang des EXOSC10-Proteins mit der Umwandlung von ruhenden in proliferierende Spermatogonien an.
Aufgrund der klonalen Vermehrung sollten „undifferenziertere“ Spermatogonien in kleinen Klonen und „differenzierte“ auch in größeren Klonen vorkommen. Die Tatsache, dass FGFR3 in der whole mount-Analyse in kleineren Zellklonen detektiert wurde, unterstützt die oben aufgeführte Argumentation hinsichtlich einer potentiellen Stammzellfunktion dieses Proteins. Hingegen wurde der Differenzierungsmarker KIT auch in größeren Zellklonen, also „späteren“ Entwicklungsstadien von Spermatogonien, beobachtet.
Der im Rahmen dieser Studie detektierte spermatogoniale Oberflächenmarker FGFR3 diente deshalb abschließend zur Isolierung der FGFR3-positiven spermatogonialen Subpopulation. Hiermit wurde die Grundlage für die Kultivierung dieser Zellpopulation gelegt.Spermatogonia as the earliest precursors of spermatids in the male adult contain a cell population with stem cell characteristics. Due to the ability for self renewal and differentiation, this cell population forms the basis for life-long spermatogenesis.
In this thesis, data from microarray analysis, real-time RT-PCR and immunohistochemistry were employed to detect spermatogonia-specific genes. Additionally, further markers for spermatogonia were examined by immunohistochemistry. Double and triple immunofluorescence staining with different marker combinations revealed an unexpected heterogeneity of the spermatogonial cells in terms of marker expression. No simple correlation with earlier morphological charactarizations of the cells could be confirmed. Besides a multitude of coexpressed respectively overlapping markers, some markers were detected which behaved mutually exclusive.
UTF1+/FGFR3+/DMRT1-/KIT- cells were identified as good candidates for the subpopulation of adult stem cells within the whole spermatogonial cell population. Arguments that UTF1 and FGFR3 constitute human spermatogonial stem cell markers are: FGFR3 was never present in proliferating spermatogonia and never coexpressed with DMRT1. To a large extent, DMRT1 was detected together with KIT, a marker for differentiation of spermatogonial cells, suggesting a similar function. UTF1 was barely expressed in differentiating (KIT+) or DMRT1+ spermatogonia. FGFR3 expression was encountered in a subpopulation of UTF1+ spermatogonia. Examination of fetal testis tissue displayed FGFR3 expression in human testis already during fetal stage.
Spermatogonial stem cells that comprise a subpopulation of the spermatogonial types A dark and A pale are located in direct contact to the basement membrane of the testicular seminiferous tubules. Type B spermatogonia, a further, differentiating spermatogonial subtype exhibit a more central location within the seminiferous tubule. The identified stem cell markers FGFR3 and UTF1 were predominantly detected in cells residing directly at the basement membrane, presumably “early”, “undifferentiated” cells. Contrasting this, DMRT1 and KIT were never observed in A dark spermatogonia, but largely in cells distant to the basement membrane.
Human A dark spermatogonia contain a vacuole-like structure inside their nuclear chromatin (region of rarefaction) which is faintly stainable with eosin. In this thesis, the eukaryotic exosome protein EXOSC10 could be identified within this structure. For A dark spermatogonia, this result could implicate the presence of the nuclear RNA-exosome in the vacuole. Alternatively, the A dark vacuole could provide a storage compartment for the EXOSC10 protein. These results point to a putative connection of the protein with the transformation of quiescent to proliferating cells.
Based on clonal expansion theory, “undifferentiated” spermatogonia should appear in small cell clones in contrast to “differentiated” spermatogonia featuring larger cell clones. Detecting FGFR3 in smaller cell clones by whole mount approaches supports a potential stem cell function of the protein. On the other hand, the differentiation marker KIT was also observed in “later” stages of spermatogonial development.
To isolate a FGFR3+ subpopulation, magnetic-activated cell sorting using FGFR3 as a surface marker was performed. This is the prerequisite for cultivation of this cell population

We have done our best to complete the author checklist relating to the use of animals in the hut study. Note that the objective for the hut study was to evaluate the IRS treatment applications for residual efficacy against Anopheles mosquitoes, including the local An. coluzzii mosquito population. Cows were only used to attract mosquitoes into the huts and no tests were carried out directly on the cows. The author checklist is intended for use with studies where experiments are carried out on animals, which is why we have had such difficulty in completing this for the hut study, as many of the questions do not relate to how the cows were used
Protein-Protein-Wechselwirkungen im männlichen Genitaltrakt der Säuger (Mammalia) am Beispiel der Profiline und eines G-Protein-gekoppelten Rezeptors
Die Dynamik des Aktin-Zytoskletts bildet die Grundlage für die Regulation zweier zentraler Vorgänge des Fortpflanzungsgeschehens, nämlich der Spermatogenese und des transepithelialen Flüssigkeitstransportes in den ableitenden Samenwegen.
Ein Aspekt der Arbeit war die Rolle des HE6-Rezeptors (GPR64) beim Flüssigkeitstransport im proximalen Nebenhoden. Aufgrund seiner Lokalisation in unmittelbarer Nähe zum apikalen Aktin-Zytoskelett der Epithelzellen könnte HE6 am Anfang eines Signaltransduktionsweges stehen, der ein Signal von der adluminalen Seite des Epithels auf das apikale Aktin-Zytosklett und die tight junctions weiterleitet und somit den transzellulären und parazellulären Flüssigkeitstransport reguliert. In dieser Arbeit konnte mit Claudin-10, das durch die gezielte Mutation des HE6-Rezeptors in seiner Expression beeinflusst wird, eine Verbindung des HE6-Rezeptors zu den tight junctions und dem apikalen Aktin-Zytoskelett hergestellt werden.. Eine zunächst vermutete direkte Interaktion des HE6-Rezeptors mit den Profilinen, vor allem dem Profilin IV, konnte in dieser Arbeit dagegen nicht bestätigt werden, so dass die Regulation der Aktin-Dynamik mittels Profilinen, wie in Kirchhoff et al. (2006) angenommen, unwahrscheinlich erscheint. Jedoch wurde mit CRIPT auf Proteinebene ein anderes Zytoskelett-Protein als Interaktionspartner des HE6-Rezeptors ermittelt, das eine Schnittstelle zwischen HE6 und Zytoskelett herstellen und in die intrazelluläre Signaltransduktion involviert sein könnte.
Ein weiterer Aspekt der Arbeit waren die Testis-exprimierten Profiline III und IV und ihre Rolle bei der Ausbildung der spezifischen Zellgestalt der Spermien. Hauptexpressionsort beider Profiline sind die männlichen Keimzellen. Für die mRNAs konnten distinkte Expressionsprofile gezeigt werden, die mit dem Verlauf der Spermatogenese korreliert sind. Während das Profilin III-Gen ein post-meiotisches Expressionsmuster aufweist und erst in den haploiden Spermatiden zu finden ist, sind Profilin IV-Transkripte bereits prä-/meiotisch in Spermatozyten nachzuweisen. Auf Proteinebene sind beide Profiline mit der Phase der Spermiogenese korreliert und erst post-meiotisch in Spermatiden vorzufinden. Jedoch sind beide in unterschiedlichen Keimzellstrukturen lokalisiert. Profilin III ist im kompletten Akrosom runder, elongierender und reifer Spermatiden vorzufinden, wohingegen Profilin IV in runden Spermatiden zunächst im akrosomalen Vesikel und in reifen Spermatozoen nur im apikalen Akrosomenbereich lokalisiert ist. Zusätzlich konnte es in einer post-akrosomalen Struktur zwischen Akrosom und Manschette detektiert werden. Diese zeitlich und räumlich unterschiedlichen Expressionsprofile sprechen für distinkte Funktionen der Profiline während der Spermiogenese. Diese Vermutung wurde durch die funktionellen Studien und die Ergebnisse der Yeast Two-Hybrid-Library-Screenings unterstrichen. Hier zeigte sich Profilin III als ein „typisches“ Profilin mit den charakteristischen Aktin- und Poly-Prolin-Bindungseigenschaften, Profilin IV hingegen nicht. Die Interaktion des Profilin III mit Aktin und dem FH-Protein Diaph3 lassen vermuten, dass es sich bei Profilin III um ein regulatorisches Protein des Aktin-Zytoskeletts der Keimzellen handelt. Ebenso wie die Profiline I und II könnte es die Aktin-Dynamik beeinflussen, in dem es in vivo durch Bildung von Profilin-Aktin-Komplexen oder die Bindung an F-Aktin-gebundenes Diaph3, die Aktin-Polymerisationsrate steigert. Bei Profilin III könnte es sich um das „typische“ Aktin-bindende Profilin der männlichen Keimzellen handeln, das durch direkte und indirekte Regulation des Aktin-Zytoskelettes an der morphologischen Differenzierung der Spermatiden, die für die Fertilität des Spermium von grundlegender Bedeutung ist beteiligt ist.
Profilin IV scheint im Unterschied zu Profilin III kein „typisches“ Profilin mit den charakteristischen Aktin- und Poly-Prolin-Bindungseigenschaften zu sein. Schon der phylogenetische Vergleich und die Sequenz-Analyse ließen vermuten, dass Profilin IV aufgrund seiner geringen Homologie zu den anderen Profilinen und kaum konservierten funktionellen Aminosäuren keine charakteristischen Profilin-Interaktionen eingehen kann. Dementsprechend konnte weder in den funktionellen Studien noch im Y2H-Library Screen eine Interaktion mit Aktin oder anderen charakteristischen Profilin-Liganden, wie z.B. Poly-Prolin-Proteinen ermittelt werden. Möglicherweise handelt es sich um eine Profilin-Isoform, die ihre hauptsächliche Funktion in der Signaltransduktion und weniger in der direkten Zytoskelett-Regulation hat
