Validation of a Polymerase Chain Reaction technique for Kidd blood group genotyping
The Kidd blood group antigens, Jkª and Jkᵇ , are two of the
main surface markers which are found on the membrane of red blood
cells. The determination of whether a donor or a recipient has the
Jkª and/or the Jkᵇ antigens is crucially important to have a successful
transfusion without the development of adverse incompatibility-related
reactions. In Malta, routine serological-based tests are applied with
the purpose of differentiating between homozygous and heterozygous
states for the Jk antigens respectively. Although these tests are
highly specific and sensitive, there are particular clinical scenarios
where haemagglutination assays are not suitable for determining the
individual’s Kidd blood group status. Additionally, the alternative
genotyping procedure has never been applied in Malta within the
context of blood grouping. The current study was therefore carried out
to determine whether a molecular-based technique such as Polymerase
Chain Reaction – Restriction Fragment Length Polymorphism analysis
(PCR-RFLP) is a suitable alternative procedure for distinguishing
amongst the three different Kidd phenotypes. After extracting
deoxyribonucleic acid (DNA) from 50 blood samples obtained from
serologically-tested healthy blood donors who expressed at least one of
the Kidd antigens, PCR-RFLP analyses were carried out. The results
of the latter were then compared with those previously obtained with
haemagglutination and a complete match was observed between the
two. Therefore, this PCR-RFLP method was confirmed as a suitable
alternative laboratory technique that can be used to determine
efficiently the Kidd blood group of both donors and recipients, in an
accurate manner without subjectivity as encountered in the case of
haemagglutination. This research further facilitates the introduction of
molecular-based techniques in molecular blood transfusion.peer-reviewe

A fast heuristic approach for train timetabling in a railway node
We consider a conflict-free scheduling problem which arises in railway networks, where ideal timetables have been provided for a set of trains, but where these timetables may be conflicting. We use a space-time graph approach from the railway scheduling literature in order to develop a fast heuristic which resolves conflicts by adjusting the ideal timetables while attempting to minimize the deviation from the ideal timetable. Our approach is tested on realistic data obtained from the railway node of Milan. © 2013 Elsevier B.V

A review of novel technologies and techniques associated with identification of bloodstream infection etiologies and rapid antimicrobial genotypic and quantitative phenotypic determination
Introduction: The antimicrobial aspect of management of patients with blood stream infections (BSI) and sepsis is time critical. In an era of increasing antimicrobial resistance, rapid detection and identification of bacteria with antimicrobial susceptibility is crucial to direct therapy early in the course of illness. Molecular techniques offer a potential solution to this. Areas covered: In the present review the authors have discussed a number of novel solutions utilizing a variety of molecular techniques for pathogen detection, identification and antimicrobial susceptibility. The review is not designed to be an exhaustive literature review covering all diagnostic solutions ever developed, instead the authors have focused on what they have had experience using, evaluating or currently view as new and exciting with potential to revolutionize BSI diagnosis. The authors searched PubMed (Medline) and Google Scholar with terms: BSI, Bacteraemia, Candidaemia, Diagnostics, AST, Rapid, AMR, Novel and Blood Culture. The authors attended recent clinical microbiology technology congresses. Expert commentary: There are multiple exciting novel technologies at differing stages of development with potential to revolutionize diagnosis of BSI. More work is needed as well as a standardized assessment of different platforms in order to better understand the clinical and financial impacts these will have in clinical microbiology laboratories.</p
