    Antinociceptive activity of cyclopeptide alkaloids isolated from Ziziphus oxyphylla Edgew (Rhamnaceae)

    The current study was designed to evaluate the antinociceptive profile of five cyclopeptide alkaloids isolated from Ziziphus oxyphylla, including Oxyphylline-B 1, Oxyphylline C 2 Oxyphylline-D 3, Nummularin-C 4, and Nummularin-R 5. The effect was studied in acetic acid induced writhing and formalin induced flinching behavior tests, at 2.5 and 5 mg/kg i.p. In the post-acetic acid induced writhing test, the compounds significantly ameliorated abdominal constrictions in a dose dependent manner, with compounds 1 and 5 showing 80.98% and 77.87% protection, respectively. When challenged in the formalin induced test, pretreatment of compounds significantly attenuated painful sensation in both phases. Moreover, compounds 1 and 5 were more effective with 45.32% and 75.32% for 1 and 36.77% and 71.10% protection for 5, in the 1st and 2nd phases respectively. The peripheral analgesia was strongly augmented by the central effects of these compounds. The current finding strongly supports the ethnomedicinal use of this valuable medicinal plant in various painful conditions

    Fabrication, Characterization, and In Vivo Evaluation of Famotidine Loaded Solid Lipid Nanoparticles for Boosting Oral Bioavailability

    Famotidine as H-2 receptor has antagonistic effects on gastric secretion. Unfortunately, its hydrophobic nature contributes to its variable and poor oral bioavailability. In the current study efforts are being made to fabricate famotidine loaded solid lipid nanoparticles with narrow size distribution. Prepared nanoformulations were pharmaceutically evaluated to confirm the desired boosted oral bioavailability. Famotidine loaded nanoformulation (FFSe-4) showed particle size 111.9 +/- 1.3 nm, polydispersity index 0.464 +/- 0.03, zeta potential -33.46 +/- 2mV, entrapment efficiency 84 +/- 2.7%, and drug loading capacity 2.709 +/- 0.13%. Drugexcipients compatibility was confirmed by Fourier transformed infrared spectroscopy. Scanning electron microscopy confirmed spherical shaped, nanosized particles. Differential scanning calorimetry and powder X-ray diffractometry confirmed the change in crystalline nature. Prepared nanoformulation was more stable at refrigerated temperature. In vitro study showed that drug release time is proportional to drug pay load and followed zero order kinetics. Release exponent (n > 0.5) confirmed non-Fickian-diffusion mechanism for drug release. In vivo pharmacokinetic studies showed 2.06-fold increase in oral bioavailability of famotidine dispersed in solid lipid nanoparticles compared to commercial product. These results authenticate solid lipid nanoparticles as drug delivery system and propose prolonged release with improved oral bioavailability for famotidine

    Analyzing the impact of environmental collaboration among supply chain stakeholders on a firm’s sustainable performance

    In the era of industrialization, environmentalists are more concerned with the environment and so are continuously interested in investigating organizational factors that can facilitate the transition towards sustainability. This research systematically investigates the impact of supply chain partner’s collaborative approach towards green practices on a firm's sustainability performance. Stakeholder and coordination theories are used to underpin the study. A structural equation modeling technique is adopted to analyze data collected from 126 green supply chain professionals working at various manufacturing firms operating in Pakistan using a survey questionnaire. The results indicate significant and positive impacts of institution pressure and customer monitoring on the adoption of green supply chain management (GSCM) practices by organizations. This study also explains that organizational GSCM practices, external GSCM practices and performance measures have positive and significant relationships. These findings reveal that it is important for managers to address external GSCM pressures by adopting green practices and being a focal firm should undertake GSCM initiatives in collaboration with their suppliers and customers to achieve a holistic impact which ultimately leads to betterment in overall sustainability performance
