    Variation of in vivo efficiency of the bacteriophage T7 terminator depending on terminator-upstream sequences

    The 40-kb linear genome of bacteriophage T7 contains only one terminator T phi, for the phage T7 RNA polymerase that is located between genes 10 and 11, Its in vitro termination efficiency has been previously found to vary upon any sequence within a transcription unit [Lee, J. T., Kim, H., Moon, K;., Kim, S., and Kang, C. (1991) Mel. Cells 1, 203-209]. In order to see if this variation takes place also in vivo, termination efficiencies within E. coli cells of the same terminator in the three different transcription units were measured in this study. The three recombinant plasmids, pNNTcat, pSNTcat, and pSSTcat, are different only in promoter-proximal, terminator distal sequence, and/or terminator-proximal upstream sequence. They have shown substantially different in vitro termination efficiencies. In vivo termination efficiencies were measured by two different methods. One method was to compare the expression product levels of a terminator-downstream caf gene (CAT activity) in the presence and absence of the terminator. The other was to compare the RNA levels of terminator-upstream and -downstream DNA. The in vivo termination efficiencies were different in three different plasmids when measured by RNA levels: levels: 97% in pNNTcat, 79% in pSNTcat, and 52% in pSSTcat (the latter tao plasmids showed significantly lower values when measured by protein levels). These in vivo efficiencies were substantially higher than the previously measured in vitro values (77%, 35%, and 17%, respectively). Nevertheless, the order of termination efficiency among the three plasmids was the same within E. call cells as in vitro: pNNTcat > pSNTcat > pSSTcat. Thus, termination efficiency of phage T7 RNA polymerase at the T7 terminator T phi varies upon both terminator-proximal and -distal upstream sequences not only in vitro, but also within E. coli cells

The strength of using case-wise bootstrapping method, according to Davidson and Hinkley (1999), is the robustness to nonnormality and heteroskedasticity. The main assumption of this research is that there exists a structural break in the macroeconomic data of Korean economy, and as mentioned by Hatemi-J (2011), it needs to be considered in order to find a full Fisher effect. The Korean economy was critically affected by the East Asian financial crisis from 1997 to 1998, and this gives a good reason to assume a structural break in the Korean market data. Although past Korean literatures have suggested its possible existence, it was not tested in detail. This research examines the possibility of such break, and confirms that there is a break in the Korean macroeconomic data. The empirical results from this research show that the bootstrapping method gives accurate estimates of real interest rates before and after the structural break point. However, the data showed that structural breaks do not affect the relationship between the nominal interest rate and inflation and the regression becomes distorted when the data directly related to the structural break period are not excluded in the analysis. The main question is whether the Fisher effect can be noticed before and after the structural break point, and it is shown in this study that the Fisher effect cannot be found both before and after the structural break point. However, the Fisher effect is discovered when no data are excluded, and the assumption of a structural break is not necessary

Kang & Yang (1994) described Choroterpides nigella based on larvae and eggs. They did not compare Ch. nigella with Ch. minor. As characters distinguishing Ch. nigella from Ch. major, only two ones are named: (1) transversal curved row of long setae on labrum and (2) tooth-like maxillary tusk with three comb-like setae at its base. Actually, both characters are present in all Dilatognathus, including Ch.
