REACTION-MECHANISM OF THE RECONSTITUTED TRICARBOXYLATE CARRIER FROM RAT-LIVER MITOCHONDRIA

BIOCHIMICA ET BIOPHYSICA ACTA. 1993;1142(1-2):139-145.Transport of citrate and malate by the tricarboxylate carrier from rat liver mitochondria has been studied in a reconstituted system. Homologous citrate/citrate antiport and heterologous (electroneutral) citrate/malate antiport was kinetically analyzed. The maximal rates of the two exchange modes did not vary significantly within pH 7.0 to 7.8 which is the optimum pH-range for transport activity. On the other hand, the apparent transport affinity varied considerably within this range. Calculations on the basis of the different pK values for citrate and malate indicate that only H-citrate 2 -and malate 2 -are accepted as transport species by the tricarboxylate carrier. A complete set of half-saturation constants was established for citrate and malate on both the external and the internal side of the membrane. Both the K(m) and V(max) for citrate and malate were independent of the nature of the countersubstrate at the other side of the membrane. Bisubstrate initial velocity analyses of the exchange reaction resulted in a kinetic pattern which is consistent with a sequential antiport mechanism. This type of mechanism implies formation of a ternary complex of the carrier with two substrate molecules before the transport reaction occurs. Thus the tricarboxylate carrier falls into the functional family of mitochondrial carrier proteins showing sequential transport mechanisms
Data visualisation in R
Cílem práce je představit možnosti grafického zobrazení dat a ukázat implementaci těchto zobrazení v R. Grafická zobrazení jsou rozdělena dle počtu a charakteru proměnných. Jednotlivá zobrazení jsou popsána a je ukázán postup jejich tvorby. Zobrazení jsou porovnávána mezi sebou a je diskutován jejich přínos stejně tak jako výhody a nevýhody oproti ostatním. Je předvedena jejich implementace v R. Dále je rozvedena tvorba grafů v R obecně, včetně druhotného přizpůsobování a kombinování těchto grafů. Jsou prezentovány autorem vytvořené funkce umožňující tvorbu některých z méně tradičních grafů.The aim of this thesis is to present ways of visualising data using R. Based on the number and types of variables suitable visualisation methods are presented. These methods are described and their creation is explained. They are further discussed and compared. Implementation of these methods in R is shown. Finally, the ways of customizing and combining graphs in R are presented, including some custom author-created functions

A new genus of Lindsaeoid ferns
In revising the New World representatives of the genus Lindsaea, the author came across a fern specimen from Borneo preserved in the Rijksherbarium, Leiden, that did not seem to fit into any described genus. It had been described as Schizoloma stortii v. A. v. R., but in the author’s opinion the genus Schizoloma Gaud., typified by Lindsaea ensifolia Sw., cannot be maintained (Kramer, 1957). Almost all species that in modern works have been left in it (e.g. HOLTTUM, 1954; ALSTON, 1956, as Schizolegnia) should be put back in Lindsaea. This, however, cannot be done with Sch. stortii, as this species possesses a peculiar combination of characters unlike anything found in Lindsaea. Its pinnae have a median main vein as in L. ensifolia, but are freeveined, and they have a large basal acroscopic auricle as in Isoloma, but are non-articulate. There is a remote resemblance to Lindsaea walkerae Hook., which may be its closest relative, as stipulated by van ALDERWEREKT van ROSENBURGH in his original description; the natural place of that species too would perhaps be in a separate genus. Sch. stortii differs from it, a.o., by its pale axes and alternate auriculate pinnae. Because of its apparently rather isolated position, it seems indicated to create a separate genus for this fern.

Going Beyond Counting First Authors in Author Co-citation Analysis
The present study examines one of the fundamental aspects of author co-citation analysis (ACA) - the way co-citation
counts are defined. Co-citation counting provides the data on which all subsequent statistical analyses and mappings
are based, and we compare ACA results based on two different types of co-citation counting - the traditional type that
only counts the first one among a cited work's authors on the one hand and a non-traditional type that takes into
account the first 5 authors of a cited work on the other hand. Results indicate that the picture produced through this non-traditional author co-citation counting contains more coherent author groups and is therefore considerably clearer. However, this picture represents fewer specialties in the research field being studied than that produced through the traditional first-author co-citation counting when the same number of top-ranked authors is selected and analyzed. Reasons for these effects are discussed
