Lifespan of neurons is uncoupled from organismal lifespan.
Neurons in mammals do not undergo replicative aging, and, in absence of pathologic conditions, their lifespan is limited only by the maximum lifespan of the organism. Whether neuronal lifespan is determined by the strain-specific lifetime or can be extended beyond this limit is unknown. Here, we transplanted embryonic mouse cerebellar precursors into the developing brain of the longer-living Wistar rats. The donor cells integrated into the rat cerebellum developing into mature neurons while retaining mouse-specific morphometric traits. In their new environment, the grafted mouse neurons did not die at or before the maximum lifespan of their strain of origin but survived as long as 36 mo, doubling the average lifespan of the donor mice. Thus, the lifespan of neurons is not limited by the maximum lifespan of the donor organism, but continues when transplanted in a longer-living host
Calpurnius Agricola in Ti.
Circulating cathepsin K and cystatin C in patients with cancer related bone disease: Clinical and therapeutic implications
The clinical significance of serum cathepsin K and cystatin C was assessed in patients with breast cancer (BCa) or prostate cancer (PCa) with confined disease (M0) or bone metastasis (BM). Cathepsin K and cystatin C circulating levels were determined by ELISAs in 63 cancer patients, in 35 patients with nonmalignant diseases and in 42 healthy blood donors (control group). In BCa patients, cathepsin K serum levels were sig- nificantly lower than in sex matched control group (HS; p 1⁄4 0.0008) or in patients with primary osteoporosis (OP; p 1⁄4 0.0009). On the contrary, cystatin C levels were significantly higher in BCa patients than in HS ( p 1⁄4 0.0001) or OP ( p 1⁄4 0.017). In PCa patients, cathepsin K concen- trations did not significantly differ from those measured in sex matched HS or in patients with benign prostatic hyperplasia (BPH). Conversely, cystatin C was more elevated in cancer patients than in controls ( p 1⁄4 0.0001) or BPH patients ( p 1⁄4 0.0078). Furthermore, in PCa patients, a pos- itive correlation was observed between cystatin C and cathepsin K (rS 1⁄4 0.34; p 1⁄4 0.047). No further relationship was highlighted between these molecules and the clinicobiological parameters of BCa or PCa progression including the number of bone lesions. Moreover, ROC curve analysis showed a poor diagnostic performance of cathepsin K and cystatin C in the detection of BM patients. Interestingly, the administration of zole- dronic acid (ZA), a bisphosphonate derivative endowed with a potent antiosteoclastic activity, induced in BM patients a marked increase of cathepsin K and cystatin C serum levels compared to baseline values. However, this phenomenon was statistically significant only in the PCa group. In conclusion Cystatin C and cathepsin K may be regarded as possible markers to monitor the therapeutic response to bisphosphonate treatments. Nevertheless, their clinical value as specific gauges of skeletal metastasis remains questionable
Of Leto: a staged concert reading
abstract: Of Leto: a staged concert reading is a new work development created by Alexander Tom and Daniel Oberhaus focusing on collegiate collaboration, production process, and creative intuition. An original story was adapted by Daniel Oberhaus into a working libretto. Alexander Tom created a two-act musical-drama and utilized the colleges on the Arizona State University \u2014 Tempe campus: Barrett, the Honors College, W.P. Carey School of Business, the College of Liberal Arts and Sciences, and the Herberger Institute for Design and the Arts: School of Music and School of Theatre, Film and Dance. This cross-discipline staged concert reading was comprised of a libretto by Daniel Oberhaus, music, additional lyrics and orchestrations by Alexander Tom, and orchestrations by Drew Nichols. The performance included a thirteen-piece orchestra and fourteen vocalists in undergraduate and graduate programs. This paper includes research on Benjamin Britten and Myfanwy Piper's Death in Venice and Stephen Sondheim and Hugh Wheeler's Sweeney Todd, the Demon Barber of Fleet Street. Its purpose is to impart a comparative analysis on the process of collaboration in opera, musical theatre, and the newly determined "musical-drama" \u2014 the genre in which Of Leto resides. Use of historical research will expound on the evolution of musical theatre along with each team's collaborative processes in relation to the music (lyrics and melody respectively), the libretto, and the production. The research permits conclusions regarding the possible practices to utilize in creating new student works like Of Leto
The genesis of cerebellar GABAergic neurons: fate potential and specification mechanisms
The variety of neuronal phenotypes that populate the cerebellum derives from progenitors that proliferate in two germinal neuroepithelia: the ventricular zone generates GABAergic neurons, whereas the rhombic lip is the origin of glutamatergic types. Progenitors of the ventricular zone produce GABAergic projection neurons (Purkinje cells and nucleo-olivary neurons) at the onset of cerebellar neurogenesis. Later on, however, these progenitors migrate into the prospective white matter, where they continue to divide up to postnatal development and generate different categories of inhibitory interneurons, according to precise spatio-temporal schedules. Projection neurons derive from discrete progenitor pools located in distinct microdomains of the ventricular zone, whereas interneurons originate from a single population of precursors, distinguished by the expression of the transcription factor Pax-2. Heterotopic/heterochronic transplantation experiments indicate that interneuron progenitors maintain full developmental potentialities up to the end of cerebellar development and acquire mature phenotypes under the influence of environmental cues present in the prospective white matter. Furthermore, the final fate choice occurs in postmitotic cells, rather than dividing progenitors. Extracerebellar cells grafted to the postnatal cerebellum are not responsive to local neurogenic cues and fail to adopt clear cerebellar identities. On the other hand, cerebellar cells grafted to extracerebellar regions retain typical phenotypes of cerebellar GABAergic interneurons, but acquire specific traits under the influence of local cues. These findings indicate that interneuron progenitors are multipotent and sensitive to spatio-temporally patterned environmental signals that regulate the genesis of different categories of interneurons, in precise quantities and at defined times and places
The Role of Vitamin K in Bone Remodeling and Osteoporosis
Vitamin K is an essential fat soluble vitamin involved in the regulation of normal coagulation. However, growing
evidence highlights that this molecule appears to be also implicated in the regulation of other important
biological functions such as bone mineralization, calcium homeostasis, apoptosis, cell growth and signal
transduction. In particular, many studies have focused their attention on the protective effects of vitamin K on
bone tissue in the outlook of its use in the prevention and treatment of osteoporosis in post-menopausal women.
The objective of the present paper is to review data of the literature regarding the metabolic effects of Vitamin K
in bone tissue and its clinical role in the prevention and treatment of osteoporosis
Cerebellar granule cells transplanted in vivo can follow physiological and unusual migratory routes to integrate into the recipient cortex.
CNS repair by cell transplantation requires new neurons to integrate into complex recipient networks. We assessed how the migratory route of transplanted granule neurons and the developmental stage of the host rat cerebellum influence engraftment. In both embryonic and postnatal hosts, granule cells can enter the cerebellar cortex and achieve correct placement along their natural migratory pathway. Donor neurons can also reach the internal granular layer from the white matter and integrate following an unusual developmental pattern. Although the frequency of correct positioning declines in parallel with cortical development, in mature recipients correct homing is more frequent through the unusual path. Following depletion of granule cell precursors in the host, more granule neurons engraft, but their ability for achieving correct placement is unchanged. Therefore, while the cerebellar environment remains receptive for granule cells even after the end of development, their full integration is partially hindered by the mature cortical architecture
