Male mice lacking three germ cell expressed genes are fertile
In recent years, much knowledge about the functions of defined genes in spermatogenesis has been gained by making use of mouse transgenic and gene knockout models. Single null mutations in mouse genes encoding four male germ cell proteins, transition protein 2 (Tnp-2), proacrosin (Acr), histone H1.1 (H1.1), and histone H1t (H1t), have been generated and analyzed. Tnp-2 is believed to participate in the removal of the nuclear histones and initial condensation of the spermatid nucleus. Proacrosin is an acrosomal protease synthesized as a proenzyme and activated into acrosin during the acrosome reaction. The linker histone subtype H1.1 belongs to the group of main-type histones and is synthesized in somatic tissues and germ cells during the S-phase of the cell cycle. The histone gene H1t is expressed exclusively in spermatocytes and may have a function in establishing an open chromatin structure for the replacement of histones by transition proteins and protamines. Male mutant mice lacking any of these proteins show no apparent defects in spermatogenesis or fertility. To examine the synergistic effects of these proteins in spermatogenesis and during fertilization, two lines of triple null mice (Tnp-2(-/-)/Acr(-/-)/H1.1(-/-) and Tnp-2(-/-)/Acr(-1-)/H1t(-/-)) were established. Both lines are fertile and show normal sperm parameters, which clearly demonstrate the functional redundancy of these proteins in male mouse fertility. However, sperm only deficient for Acr (Acr(-/-)) are able to compete significantly with sperm from triple knockout mice Tnp-2(-/-)/Acr(-/-)/H1.1(-/-) (70.7% vs. 29.3%) but not with sperm from triple knockout mice Tnp-2(-/-)/Acr(-/-)/H1t(-/-) (53.6% vs. 46.4%). These results are consistent with a model that suggests that some sperm proteins play a role during sperm competition

Verhaltenstherapie. 2003;13(4):244-252.The Impact of Psychological Psychotherapy on Anxiety Provoking Dizziness in Panic Disorder with Agoraphobia Introduction: Dizziness is frequently reported as a typical somatic complaint in panic disorder with agoraphobia. However, it is unclear, how often dizziness is experienced as anxiety provoking, and to what extent fear of dizziness affects treatment success. The present study examined the frequency and the influence of behavior therapy on anxiety provoking dizziness in patients with panic disorder with agoraphobia. Patients and Methods: 398 agoraphobics with panic disorder participated in the study. To analyze the impact of psychotherapy on dizziness, patients were classified into four groups depending on anxiety provoking dizziness (strong/weak) before or after treatment (pre/post). Results: Fear of dizziness was the most frequent somatic complaint in these patients at the beginning of the treatment. According to different self-rating scales, all four groups improved considerably with treatment. However, the group that reported dizziness as very anxiety provoking before treatment but no longer anxiety provoking after treatment yielded the strongest overall therapy effect. The other three groups did not differ in their overall benefit from treatment. Similar effects were found if different somatic sensations such as heart palpitations or hard breathing were selected. Discussion: The frequency of fear of dizziness emphasizes its relevance in this patient sample. It is therefore important that practitioners be informed about possible differential diagnoses that may include this fear. However, fear of dizziness did not take a special role in treatment outcome because treatment efficacy depended on losing the fear of disorder-typical physical symptoms in general, not specifically the fear of dizziness

The Impact of Halogenated Phenylalanine Derivatives on NFGAIL Amyloid Formation
The hexapeptide hIAPP22–27 (NFGAIL) is known as a crucial amyloid core sequence of the human islet amyloid polypeptide (hIAPP) whose aggregates can be used to better understand the wild-type hIAPP′s toxicity to β-cell death. In amyloid research, the role of hydrophobic and aromatic-aromatic interactions as potential driving forces during the aggregation process is controversially discussed not only in case of NFGAIL, but also for amyloidogenic peptides in general. We have used halogenation of the aromatic residue as a strategy to modulate hydrophobic and aromatic-aromatic interactions and prepared a library of NFGAIL variants containing fluorinated and iodinated phenylalanine analogues. We used thioflavin T staining, transmission electron microscopy (TEM) and small-angle X-ray scattering (SAXS) to study the impact of side-chain halogenation on NFGAIL amyloid formation kinetics. Our data revealed a synergy between aggregation behavior and hydrophobicity of the phenylalanine residue. This study introduces systematic fluorination as a toolbox to further investigate the nature of the amyloid self-assembly process

Zadáním bakalářské práce byl návrh rodinného domu v obci Romanov. Parcela se rozléhá na svažitém terénu, v jižním části obce nad údolím Debř. Okolí tvoří převážně smíšené lesy. Území spadá do chráněné krajinné oblasti, proto je nutné mít na paměti regulativy pro výstavbu a přestavbu na území CHKO Kokořínsko - Máchův kraj. Koncept se zakládá na minimalizaci zásahu do stávající krajiny. To vedlo k umístění stavby na půdorysu původního rekreačního objektu, založení stavby nad terénem a zakomponování stávající vzrostlé zeleně do návrhu zahrady. Dům je navržen jako dřevostavba, kombinace systému two by six a hrázděné stavby, s tradičním sedlovým zastřešením. Z jižní a východní strany obepíná dům skládaná dřevěná konstrukce terasy, jejíž systémové řešení je použito k výstavbě zahradních objektů joga ateliéru a sauny.The main goal of this bachelor thesis is to design a family house in the village of Romanov. House is situated in slopping terrain, in southen part of the villagev, above a valley Debř. Surroundings contains mostly mixed forest. It is a part of the protected landscape area therefore it is necessarry to respect regulations for construction and reconstruction in the protected landscape area of Kokořínsko - Máchův kraj. The concept is based on minimazing the intervation into the existing landscape. Therefore the house is designed on the spot of an existing recreational object, founded above the surrouding ground, and considers placement of existing trees. The house is designed as a wooden construction, a combination of two by six system and half-timbered building, with tradition saddle roof. From south and east surrounds the house a stacked wood construction of a terrace, whose construction system is used also to build a yoga studio and a sauna in the garden
Die Beziehung der Europäischen Union zu den AKP-Staaten und ihre Auswirkungen auf die Entwicklung von Menschenrechten, Rechtsstaatlichkeit und Good Governance
Die folgende Master-Thesis „Die Beziehung der Europäischen Union zu den AKP-Staaten und ihre Auswirkungen auf die Entwicklung von Menschenrechten, Rechtsstaatlichkeit und Good Governance“ versucht eine Darstellung der Beziehungen zwischen den Europäischen Staaten und den AKP- Staaten ab 1957 zu bieten, und geht der Frage nach, inwiefern das Prinzip der „Good Governance“ in den zwischenstaatlichen Verträgen eingearbeitet wurde und ob es tatsächliche Auswirkungen zeigt.
Der Autor versucht hierzu zunächst die vertraglichen Entwicklungen über die Abkommen von Yaoundé, Lomé, Cotonou, den European Consensus, den Vertrag von Lissabon und etwaige zukünftige Entwicklungen aufzuzeigen. Hierbei wird auf die Spannungsfelder zwischen der Europäischen Union/EWG und ihren ehemaligen Kolonien eingegangen, da jeder Vertrag, der innerstaatliche Auswirkungen zeigen soll, auch gleichzeitig in die Souveränität der AKP-Staaten eingreift.
Im zweiten Teil wird dem Ursprung des Begriffs der „Good Governance“ nachgegangen, der ursprünglich aus einer Studie der Weltbank hervorging und sich schließlich zu einem der wichtigsten Prinzipien der Europäischen Union, sowohl im Inneren, als auch nach außen hin entwickelt hat. Infolgedessen wurde auch versucht dieses Prinzip in den AKP-Staaten zu verwirklichen, wobei es insbesondere in den Ländern der Sub-Sahara zu Schwierigkeiten gekommen ist.
Wie erfolgreich die Entwicklungshilfepolitik der Europäischen Union zukünftig sein wird, hängt stark davon ab, wie sie mit diesen Schwierigkeiten umgeht, und ob sie eine effektive Strategie besitzt sich gegen andere Staaten, die ihren Einfluss auf die AKP-Region ausweiten wollen, vorzugehen. Es ist nämlich klar ersichtlich, dass diese Regionen zunehmend an geopolitischem Einfluss gewinnen und deshalb immer mehr umworben werden.This master-thesis „Die Beziehungen der Europäischen Union zu den AKP-Staaten und ihre Auswirkungen auf die Entwicklung von Menschenrechten, Rechtsstaatlichkeit und Good Governance“ tries to show the connections between the European Union and its former organisation forms with the states of the african, caribbean and pacific region. The author emphasizes hereby on the development of these trade agreements and development policies since 1957. Furthermore he tries to outline the importance of the principle of Good Governance regarding this relationships and its implementation into the international contracts.
Since the decolonization of the ACP-countries a lot of agreements were signed between the European Union and its former colonies, like the treaty of Yaoundé, Lomé, Cotonou, the European Consensus on Development and the treaty of Lisboa. During all of this time there have been tensions between the countries and the Union regarding the sovereignty of the ACP-nations and the attempts to influence them via political principles and a western idea of human rights, rule of law and good governance.
In the second part this thesis tries to present the origin of the concept of good governance and its implementation in the ACP-states, especially in the sub-Sahara, where the level of statehood and governance was traditionally very low.
It is pinned out that for the future of the European Development Policy it is very important to search for an effective strategy dealing with the ACP-countries, because development and aid policy has reached a global geo-political importance and today is influenced by many powerful countries
Shape Evolution in Neutron-Rich Krypton Isotopes beyond N=60: First Spectroscopy of Kr 98,100
F. Flavigny et al. -- 6 pags., 5 figs.We report on the first γ-ray spectroscopy of low-lying states in neutron-rich Kr98,100 isotopes obtained from Rb99,101(p,2p) reactions at ∼220 MeV/nucleon. A reduction of the 21+ state energies beyond N=60 demonstrates a significant increase of deformation, shifted in neutron number compared to the sharper transition observed in strontium and zirconium isotopes. State-of-the-art beyond-mean-field calculations using the Gogny D1S interaction predict level energies in good agreement with experimental results. The identification of a low-lying (02+, 22+) state in Kr98 provides the first experimental evidence of a competing configuration at low energy in neutron-rich krypton isotopes consistent with the oblate-prolate shape coexistence picture suggested by theory.We express our gratitude to the RIKEN Nishina Center
